Objective To study the effects of tumor necrosis factor-?(TNF-?) on hypermetabolism of skeletal muscle protein in rats with chronic obstructive pulmonary disease(COPD) and explore the mechanisms. Methods Forty-five SD rats were randomly divided into normal control group,COPD group and COPD +TNF-? group(n=15).Rat COPD models were established by passive cigarette smoking in COPD group and COPD +TNF-? group. After dissecting and isolating the extensor digitorium longus(EDL) muscles, the EDL muscles were either cultured with media containing 10 ug/L recombinant rat TNF-? or without TNF-?.The subsequent changes in ubiquitin mRNA and protein levels were determined by real-time quantitative PCR and Western blot, respectively. Results The expressions of ubiquitin mRNA and protein of COPD group and COPD+TNF-? group were higher than those of normal contro1 group(P
6.Effects of Cyanide Poisoning on Myocardial Contractibility in Dogs

Journal of Third Military Medical University 1984;0(01):-
The effects of cyanide poisoning on the contractibility of the cardiac muscle were observed after the dogs were infused intravenously with a lethal dose of NaCN (2.5 mg/kg) within 2 minutes through a cannulated femoral vein. Heart rate (HR) , left ventricular pressure (LVP) , the largest changing rate of LVP (LVP?dp/dtmax), cardiac muscle contractibility (CMC) , the largest changing rate of CMC (CMC?dc/dtmax) , and ECG changes were recorded. It was found that there was an initial excitatory response of cardiac functions in the first minute of NaCN infusion, and then a drastic cardiac failure occurred, which resulted in the rapid death of the dogs within 10 minutes after NaCN administration. The changes of myocardial contractibility were slower than those of .other cardiac parameters, which implies that cyanide can exert an excitation on the myocardium initially. Our results indicate that the effects of cyanide on myocardial contractibility may be one of the important factors to induce serious arrhythmia, which in turn results fatal cardiac failure.

Objective To investigate the function of donor-derived dendritic cells (DCs) treated with NF-?B decoy in prolonging murine cardiac allograft survival time. Methods Donor bone marrow- derived DCs were treated with NF-?B decoy in vitro.Heterotopic abdominal heart transplantation was performed from BALB/c to C57BL/6 mice.Recipients were grouped according to different pretreat ments as follows:(1) Control group, infusion of PBS (0.2 ml) alone intravenously via the portal vein 7 days before heart transplantation;(2) CsA group, treated with sub-therapeutic CsA only for 7 days (10 mg?kg~(-1)?day~(-1)) through intraperitoneal injection after transplantation, and the same as control group before transplantation;(3) Control DCs group, infusion of only cultured 5th-day recipient DCs untreated with NF-?B ODN decoy;(4) Treated DCs group, infusion of recipient DCs pretreated with NF-~cB ODN decoy;(5) Combined treatment group, infusion of recipient DCs treated with NF-~cB ODN decoy before transplantation and intraperitoneal injection of sub-therapeutic CsA for 7 days (10 mg?kg~(-1)?day~(-1)) after transplantation;(6) Third party donor group,C3H/HeJ mice used as donor, and recipient (C57BL/6) was treated the same as combined treatment group.Every group had 8 mice and graft survival time was observed.Cytokines (IL-2,INF-?,IL-4 and IL-10) in recipient serums were analyzed by ELISA at 7th day after transplantation. Results The graft mean survival time (MST) in control group,CsA group,Control DCs group, treated DCs group, combined treatment group and third party donor group was 7 days,10.3 days,7.6 days,21.4 days,53.6 days and 9 days, respectively. There was significant difference in MST between treated DCs group and control group or control DCs group (P

Dioscin, a typical saponin, is widely present in the family of Dioscoreaceae, Liliaceae, Caryophyllaceae and Rosaceae, especially in Dioscoreaceae, including Discorea nipponica Makino, Dioscorea zingiberensis C. H. Wright and Dioscorea panthaica Prain et Burkill. Traditional Chinese medicine reported that dioscin plays a role in expectorant, relaxing the muscles and stimulating the blood circulation, aiding digestion and diuresis. With the development of science and technology in recent years, some new extraction and separation techniques and methods have been applied to the study of dioscin, and more and more pharmacological effects were found. Modern pharmacology studies have confirmed that dioscin had some activities on desensitization, anti-inflammatory, lipid-lowering, anti-tumor, hepatoprotection and anti-viral. After oral administration, dioscin is metabolized to diosgenin, which is the true active ingredient and is an important raw material to synthesize steroid hormone drugs. Therefore, the studies on dioscin are valueable and promising. In this review, we make a summary on the researches of dioscin including the extraction technology, separation and prepara- tion, chemical synthesis, drug metabolism, determination and pharmacological researches.

Objective To sum up experience with surgical excision of isolated local recurrence for renal cell carcinoma. Methods From March 2004 to November 2007, 7 patients (five cases un-derwent radical nephrectomy and two nephron-sparing surgery) with isolated local recurrence of renal cell carcinoma were treated at our department. All patients underwent extensive surgery for local re-currence. Results The mean patient age was 42 years (range 19 to 6). The mean time to local re-currence was 23.3 months (range 12 to 54). The Mean size of the recurrent tumor was 5.2 cm(range 2.5 to 10.5). Peritoneal exploration was performed in 7 patients and 5 had complete en bloc excision of the renal cell carcinoma mass. 2 patients gross disease was excised. The mean blood loss was 1050 (150-3000) ml. Surgical complications occurred in 2 patients, iliohypogastric nerve injure in one and ileus performation in another one. All patients recovered finally. Six patients were followed and one lost follow-up. Mean follow-up time was 13(8-27) months. One patient died of metastatic disease at 22 months after excision of the renal cell carcinoma mass. Conelusion En bloc excision of isolated locally recurrent renal cell carcinoma is possible, and complete surgical resection could lead to pro-longed disease-free survival.
