1.Characteristics and feasibility of vessel restenosis models produced by denuding arterial endothelium with domestic-made balloon catheter in rats

BACKGROUND:Researches on establishing restenosis models in rats emphasize the importancc of 2.0 Forgarty balloon catheter import,but little has been mentioned about domestic-made balloon catheter.OBJECTIVE:To explore the characteristics of vessel restenosis model by denuding arterial endothelium with domestic-made 2.0 Forgarty balloon catheter in rats.DESIGN:A randomized controlled animal experiment.SETTING:Laboratory of Pathophysiology,Hunan University of Traditional Chinese Medicine.MATERIALs:This study was performed at the Laboratory of Pathophysiology,Hunan University of Traditional Chinese Medicine between September 2006 and January 2007.Healthy male SD rats,weighing 300-350 g,were selected.The protocol was performed in accordance with ethical guidelines for the use and care of animals.Domestic-made 2.0 Forgarty balloon catheters were made of balloons and catheters.Proliferating cell nuclear antigen(PCNA),collagen I,and SM α-actin immunohistochemical staining kits were provided by Wuhan Boster Bioengineering Co.,Ltd,China.METHODS:The rats were randomly divided into 4 groups:sham-operated group(n=5),7th day model group(n=4),14th day model group(n=5),and 21st day model group(n=4).The vessel restenosis models were established in the latter 3 groups by denuding arterial endothelium with domestic-made balloon catheter in rats.In the sham-operated group,rats were without insertion of balloon catheter.Injured thoracic aortas were taken out for HE staining to observe histological changes of vascular intima.At the meantime,expressions of SM α-actin,PCNA and collagen I were determined by immunohistochemical method in each group.MAIN OUTCOME MEASURES:Histological changes of intima at the thoracic aorta as well as expressions of SM α-actin,PCNA and collagen I.RESULTS:Eighteen rats were included in the final analysis.The aortal vascular walls remained integrity without narrowness of the lumen areas and proliferation of the endothelium,vascular smooth muscle cells(VSMCs)ranged regularly in the mesolamella in the sham-operated group.The endothelium had little proliferation in the 7th day model group.The intima thickened and the area of lumen became narrow in the 14th day model group.The intimal hyperplasia was diffusive and progressive which mainly included VSMCs,the area of lumen narrowed markedly and cells ranged disorderly in mesolamella in the 21st day model group.In the 14th day model group,PCNA level was higher compared to the remaining 3 groups(P<0.01).SM α-actin level were significantly higher in the 14th and 21st day model groups than in the sham-operated group and 7th day model group(both P<0.01).There were no significant differences in collagen I among the groups(P>0.05).CONCLUSION:Vascular intima hyperplasia appeared markedly 14-21 days after domestic-made balloon catheter-induced rat aorta injury.The results suggested that intima hyperplasia is mainly induced by proliferation of VSMCs.
2.Investigaion about the mRNA expression of ASB2 and Jak3 in patients with acute lymphoblastic leuke-mia

Objective To explore the expression of ankyrin-repeat SOCS box containing protein 2 (ASB2)and Janus kinase 3 (Jak3)mRNA in bone marrow plasma mononuclear cells of patients with acute lymphoblastic leukemia (ALL)and discuss the correlation of them.Methods Bone marrow plasma was collected from 48 patients of newly diagnosed ALL (37 cases B-ALL and 11 cases T-ALL)and 34 non leukemia patients (control group),respectively.Expression levels of ASB2 and Jak3 mRNA were detected by real-time quantitative PCR.Results The expression of ASB2 mRNA in B-ALL and T-ALL were significantly different compared to control group (t =14.2,P <0.01;t =15.2,P <0.01),which were 68.5,32.7 folded more than control group,respectively.Moreover,the expression level of Jak3 mRNA in B-ALL group and T-ALL group were 2 336.1 and 7 131.5 folded more than control group (t =37.3,P <0.01;t =37.6,P <0.01). At last,the expression of ASB2 and Jak3 gene was positive correlation (r =0.523,P <0.001).Conclusion ASB2 and Jak3 are expressed abnormally in ALL patients and the correlation of them is positive.ASB2 and Jak3 may be related to the proliferation and differentiation in leukemia cells.
3.Postoperative recovery of different surgical approaches for patients with cardia cancer

Objective To study the postoperative recovery of different surgical approaches for patients with cardia cancer.Methods From January 2011 to March 2016,174 cases of patients with cardia cancer were selected and divided into observation group (76 cases) and control group (98 cases).The patients of control group received via thoracic approach surgery,while those of observation group were given abdominal approach surgery.The operationtime,intraoperative blood loss,dissected lymph node numbers,postoperative hospital stay,hospitalization expense,positive rate of surgical margin,radical operation rate,perioperative mortality,pulmonary complications,operative incision infection rate and anastomotic fistula rate of two groups were compared.Results There were no significant differences of intraoperative blood loss and operation time,positive rate of surgical margin,radical operation rate,perioperative mortality,postoperative pneumothorax rate,hydropneumothorax rate,pyothorax rate,operative incision infection rate and anastomotic fistula rate between two groups (P > 0.05).There were significant differences of the dissected lymph node numbers,postoperative hospital stay,hospitalization expense,postoperative hydrothorax rate,postoperative pneumonia rate,total pulmonary complications rate and total complications rate between two groups (P < 0.05).Conclusions Abdominal approach for patients with cardia cancer,compared to via thoracic approach surgery,presents advantages in terms of dissected lymph nodes,postoperative hospital stay,hospitalization expense,postoperative pneumonia rate,hydrothrax rate,total pulmonary complications rate and total postoperative complications rate.
4.Transformation and standardization of the inactivation test method for virus suspension in biosafety laboratory

Chemical disinfectants are generally used for virus inactivation and environment disinfection in biosafety laboratory, and the efficacy and evaluation of the disinfection are critical to ensure the laboratory biosafety.However, there is a current lack of applied standard to evaluate the virucidal efficacy of chemical disinfectants in our country.In this paper, a European Union standard“Method and Requirements of Virucidal Quantitative Suspension Test Method for Chemical Disinfectants Used in Human Medicine” was analyzed and a standard transformation scheme has been proposed.It is suggested that the model viruses should be increased from 3 to 6, including the surrogate viruses to substitute highly pathogenic viruses, and that the method to remove the residual chemical disinfectant and the calculation of 95%confidence interval should be incorporated into the standard.The suggestion will improve the scientific and operational standards related to disinfection and sterilization in biosafety laboratory in China.
5.Establishment the cut-off value of hTSH in dried venous blood spots of neonatal screening for CH in Guilin

Objective To investigate the distribution of human thyroid stimulation hormone(hTSH) in dried venous blood spots of neonatal screening for congenital hypothyroidism in Guilin ,and establish the hTSH cut‐off value for neonatal screening .Methods We selected 101 817 cases of neonatals with the local household registration from January 2011 to December 2013 ,and collected their venous blood to be made into dried venous blood spots on filter paper ,then the hTSH levels in dried blood spots were deter‐mined for congenital hypothyroidism ,According to screening results ,using the percentile method to calculate the hTSH P50 ,P99 of all the normal neonatals and neonatals that born in different quarters .Results Among 101 817 neonatals who were screened ,101 746 cases of normal neonatals ,71 cases of patients(including 47 cases of patients with CH ,24 cases of patients with high hTSH hy‐perlipidemia) ,the hTSH levels of normal neonatals showing a skewed distribution ,P50 ,P99 were 2 .45 μU/mL ,9 .86 μU/mL .the hTSH levels of the neonatal who were born in different quarters was changed with the change in the quarter ,P50 ,P99 were 2 .15μU/mL ,9 .20 μU/mL in The Spring ;P50 ,P99 were 1 .97 μU/mL ,8 .38 μU/mL in the summer ;P50 ,P99 were 2 .67 μU/mL ,10 .43μU/mL in the autumn ;P50 ,P99 were 2 .90μU/mL ,11 .15μU/mL in the winter .Through the analysis ,although the hTSH cut‐off of capillary blood spots that was used in the local center could be applied to dried venous blood spots ,but not ideal cut‐off value .Even‐tually ,the hTSH P99 9 .20μU /mL in the spring was identified as cut‐off value of dried venous blood spots for routine screening la‐boratory ,which had a sensitivity of 100 .00% ,a specificity of 36 .80% ,the diagnosis rate of 38 .96% ,and no missed phenomena .For the better service ,the hTSH value in the range of 9 .20-20 .00 μU/mL was defined as the boundary region ,and it was the danger region when the hTSH value was higher than 20 .00 μU/mL .Conclusion It is necessary to establish a reasonable hTSH cut‐off value to provide objective evidence for the use of dried venous blood spots of neonatal screening for congenital hypothyroidism ,ac‐cording to the boundary region and the danger region ,the recall chosen a different way ,the development of neonatal screening can be effectively promoted .
6.The lower urinary tract obstruction of urodynamic study on young and middle-aged patients with chronic prostatitis

Objective To study the urodynamic change of young and middle-aged patients with chronic prostatitis,and provide reference for clinical diagnosis and treatment.Methods Seventy young and middle-aged patients with chronic prostatitis were selected,the bladder capacity,bladder stability,bladder compliance,detrusor pressure and bladder pressure volume curve etc were measured.Results Unstable of detrusor contraction of 20 cases (28.6%,20/70),bladder sensitivity of 35 cases (50.0%,35/70),detrusor underactivity of 46 cases (65.7%,46/70).In 70 young and middle-aged patients with chronic prostatitis,the maximum bladder capacity was 100-497 (380 ± 80) ml,the detrusor pressure at maximum flow rate was 11.2-112.2 (38.4 ± 3.9) cm H2O (1 cm H2O =0.098 kPa),the drop of maximum flow rate was (10.53 ± 1.92) ml/s.Conclusions Through the urodynamic examination,the lower urinary tract obstruction syndrome can be found in young and middle-aged patients with chronic prostatitis,especially who have no significantly improvement after drug therapy.Now,the urodynamic examination for the lower urinary tract obstruction is a credible way to diagnosis,which plays an important role in diagnosis of chronic prostatitis and is useful for clinical practice.
7.The changes of pulmonary surfactant associated protein of sputum and oxygenation index in dysfunctional ventilatory weaning response patients

Objective To explore the expression of SP-A in surface active substance of sputum pulmonary of dysfunctional venti-latory weaning response patients and to study changes of expression of SP-A and oxygenation index ,arterial partial pressure of oxy-gen .Methods The mechanical ventilatory patients were divide into dysfunctional ventilatory weaning response group (group A) and easily weaning group(group B) ,then collected sputum from ventilators at different times :on the first day ,two and seven days after using ventilators .Used ELISA to test level of SP-A and oxygenation index ,arterial partial pressure of oxygen in sputum of group A and group B .Results For group A ,SP-A content oxygenation index and arterial partial pressure of oxygen were significantly less than those for group B(P<0 .05);after two days ,for group A ,SP-A content in sputum was significantly less than that on the first day ,and SP-A content in sputum was significantly less than that on the second day (P<0 .05);two days later ,oxygenation index and arterial partial pressure of oxygen were significantly lmore than those on the first day (P<0 .05) and there was no statistical difference in oxygenation index and arterial partial pressure of oxygen between the seventh day and the second day (P>0 .05) .Con-clusion Dysfunctional ventilatory weaning response and lung injury related to ventilator can be diagnosed early through the test of SP-A content in sputum ,which provides a brand-new way and method of treatment in dysfunctional ventilatory weaning response and lung injury related to ventilator .
8.Influenza vaccination in patients with systemic lupus erythematosus

Patients with systemic lupus erythematosus (SLE)are susceptible to influenza virus infection due to autoimmune disorder,use of glucocorticoids and the immunosuppressive agents.Influenza virus infection affects the disease stability,and the mortality of influenza virus infection in SLE patients is 5 times higher than that in general people infected by influenza virus.Vaccination is the most effective measure to prevent influenza virus infection.There is lack of studies on the influenza immunization in SLE patients in China,so this paper reviews prevention effect and safety of inoculation,the immune response to vaccine as well as the factors influencing immune response in influenza vaccination for SLE patients.
9.Inducing apoptosis effect by 188Re-k-ras-AGPNA on pancreatic cancer cells and biodistribution characteristics in nude mice bearing xenografts

Objective To investigate the depressing effect of antigene peptide nucleic acid (AGPNA)on the k-ras gene expression of human pancreatic cancer Patu8988 cells, the inducing apoptosis effect on Patu8988 cells, and the biodistribution characteristics in nude mice bearing xenografts using 188Re-k-ras-AGPNA.Methods The expression level of k-ras mRNA and the expression ratio of k-ras protein in Patu8988 cells transfected with AGPNA was measured by RT-PCR and flow cytometry ,respectively. The degree of cellular apoptosis 3 to 5 d after treating Patu8988 cells with 188Re-k-ras-AGPNA or 188ReO4- was determined by flow cytometry. For biodistribution study, 58 nude mice bearing Patu8988 cell xenografts were divided into two groups: intratumoral injection of 188Re-k-ras-AGPNA (Group A) and 188ReO4- (Group B). At different time points, the mice were sacrificed and organs of interest were excised, weighted and counted by a gamma counter. The organ uptake was calculated as a % ID/g and the absorbed doses of organs were calculated. One-way analysis of variance was used. Results After transfected with 1 nmol/ml AGPNA, the k-ras mRNA gray scale ratio and the expression ratio of k-ras protein were 1.00 ± 0.39 and (15.05 ± 5.07)%, respectively. They were significantly lower than those of the control group with 1.86 ± 0.07 and (24. 38 ± 5.40) % (F = 2. 545, 5. 327, P<0. 05). At 4 and 5 d after treatment in Group A, float cells' apoptosis ratios were (26.30 ± 7.45) % and (27.90 ± 10. 38) %, respectively. Tumors were the major distribution site in Group A with uptake of (37.47 ±21.31), (35.96 ±7.80) and (15.46 ±4.93) %lD/g at 1 h, 1 d and 7 d after intra-tumor injection, respectively. The absorbed dose of tumor was 15 569 mGy/MBq. Condusions Transfection with k-ras-AGPNA on Patu8988 cells may inhibit k-ras expression at mRNA and protein expression level, and 188Re-k-ras-AGPNA can induce apoptosis of Patu8988 cells.Tumor is the major distribution site in nude mice bearing human pancreatic cancer xenografts after intratumoral injection of 188Re-k-ras-AGPNA.
10.A comparative study of 11C-MET PET with MRI for target volume delineation in postoperative radiotherapy for brain high grade glioma

Objective To evaluate the value of L-(methyl-11C)-labeled methionine positron emissions tomography (MET PET) and MRI in target volume delineation for postoperative radiotherapy for brain high grade glioma (HGG).Methods Thirty-seven patients with supratentorial HGG were included.Both MRI and MET PET scan were performed in the same treatment position for all patients.The consistency to determine residual tumor between MRI and MET PET was analyzed.Imaging data of MET PET and MRI were coregistered using the BrainLAB image fusion software.The extension of the volume with high uptake (VMET) on MET PET were compared quantitately with the enhancing area on MRI T1W gadolinium enhancement (VGd) and the hyperintensity area on MRI T2W (VT2).Results Both MET PET and MRI were positive for 19 patients and negative for 7 patients.The consistency between these two scans was 70.3%.MET PET was integrated with MRI in 30 patients with positive MET uptake.VMET were partially or entirely outside VGd in 29 patients and VT2 in 17 patients, whereas VGd and VT2 were partially or entirely outside VMET in all patients.The maximal distance from the margin of VMET to VGd was ≥ 2.0 cm in 50%patients and the corresponding distance of VMET to VT2 was ≥ 1.0 cm in 33% patients.Conclusions The differences are existing between MET PET and MRI in determination and identification of the location and extension of residual tumor for patients with HGG.The integration of MET PET and MRI can accurately delineate radiation target volume.
