[Objective] To explore whether necroptosis is involved in the mechanism of lung injury induced by intestinal ischemia-reperfusion.[Method] Thirty-two healthy male Sprague-Dawley rats were randomly assigned into 4 groups (n--8):sham operation group (sham group),isehemia/ reperfusion group (I/R group),necroptosis inhibitor necrostatin-1 group (Nec-1 group) and solvent dimethyl sulfoxide (DMSO) group (DMSO group).Model of intestinal I/R injury was produced by clamping the superior mesenteric artery for 1.5 h followed by 6 h reperfusion in rats.Necrostatin-1 1.0 mg/kg was administered 30 min before occlusion in Nec1 group, while the equal volume of DMSO was given instead in DMSO group. The rats were sacrificed at 6 h of reperfusion and the lung tissues were removed for measurement of wet-dry ratio and microscopic examination and scored. The expression of receptor-interacting protein 1 (RIP1) and receptor-interacting protein 3 (RIP3) in lung tissues was detected using Western-blot and immunohistochemistry.[Result] Compared with sham group, lung morphology score and wet/dry ratio in I/R,DMSO group raised (P < 0.05).Lung morphology score and wet/dry ratio statistically declined in Nec-1 group compared with I/R and DMSO group (P < 0.05),while there was no statistical difference of wet/dry ratio between sham group and Nec-1 group (P > 0.05).As the result of westernblot and immunohistochemistry showed, the expression of RIP1 and RIP3 was up-regulated in I/R group and DMSO group (P <0.05),which was inhibited by Nec-1 in Nec-1 group (P < 0.05).[Conclusion] Necroptosis is involved in the mechanism of lung injury induced by intestinal ischemia-reperfusion,and Nec-1,the special inhibitor of RIP1,can reduce the injury.

Objective To investigate the difference in ocular surface microbiota between dry eye patients and healthy subjects, and discuss the role of microbiota in dry eye. Methods Twenty cases of dry eye patients and 90 cases of healthy subjects were collected in the PLA General Hospital and Zhongshan Ophthalmic Center. The samples of conjunctiva impression cytology were collected from all subjects, and then metagenomic shotgun sequencing was performed following the DNA extraction. The differences in alpha diversity and metabolic pathways of the ocular surface microbiota between dry eye patients and healthy subjects were evaluated. Results There was no significant difference in alpha diversity of the microbial community between dry eye patients and healthy subjects (P =0.13).However,an increase of 15 species and a decrease of 10 species were detected on the ocular surface of dry eye patients. The enriched antibiotic resistance genes in dry eye patients were more than healthy subjects. Conclusion Although the alpha diversity of the microbial community on ocular surface between dry eye patients and healthy subjects are not distinguishable,a significant difference could be found in relative abundance and metabolic pathways, suggest that these specific microbiome may be related to the pathogenesis and disease progression of dry eye.

Object To optimize the preparation procedure for Zhixuan Granula (ZXG). Methods The optimum extracting conditions of ZXG were selected by orthogonal test with the active components: 23-acetate alisol B, atractylenolide I, and dried extract as the index, it mice sedation of ZXG was clarified by pharmacodynamics. Results The optimum preparation procedure was as follows: Rhizoma Alismatis and Rhizoma Atractylodis Macrocephalae were extracted with alcohol first, adding 12-fold 70% alcohol by refluxing, extracting twice, 2 h once, then extracted with water, adding 14-fold water, extracting twice, 2 h once. The extract showed the obvious effect on sedation of mice. Conclusion The optimum preparation procedure is reliable, with higher extracting ratio of the active components.

Objective: The quality standards for Zhixuan Granule (Rhizoma Alismatis, Rhizoma Atractylodis Macrocephalae, etc.) were studied. Methods: The TLC methods for identification of Rhizoma Alismatis、 Rhizoma Atractylodis Macrocephalae were established. A simple HPLC was established for the determination of 23-acetate alisol B. The mobile phase was acetonitrile-water(70∶30). UV detecting wavelength was at 208nm. Results: Rhizoma Alismatis and Rhizoma Atractylodis Macrocephala could be detected. 23-acetate alisol B showed a linear relationship at the concentration range of 99~1388.8ng, r=0.9999. The average recovery was 103.05% and RSD was 2.41%(n=6). Conclusion: This method is suitable for the quality control of Zhixuan Granule.

Objective To evaluate the relationship between the clinicopathological factors, prognosis and the expression of p16 and c erbB 2 protein in primary breast cancer. Methods The expression of p16 and c erbB 2 by immunohistochemical method was observed in 50 patients with primary breast cancer and the detection of p16 by polymerase chain reaction(PCR) and the point mutation of p16 by PCR single strand conformational polymorphism(SSCP) were detected in 20 patients with breast cancer. Results Among the cancers, positive expression of p16 protein was found in 17(34.00%) cases, c erbB 2 protein positive expression in 24(48.00%) cases. No homozygous deletion in p16 gene was found. However, exon2 point mutation of p16 gene was found in 1 of 20 breast cancer. The results showed no relationship between p16 expression and clinicopathological factor or prognosis. Positive expressions of c erbB 2 protein were often found in breast cancer with lymph node metastasis(P=0.0237) with a poor 5 year survivalrate(P=0.0169). There was no consistent relationship between the expression of p16 and c erbB 2 protein. Neither p16 nor c erbB 2 protein expression could be as an independent prognostic factor. Conclusions The patients with breast cancer of positive expression of c erbB 2 protein has a high lymph node metastasis rate and a poor survival rate. The point mutation rate of p16 gene is lower in primary breast cancer, and it can be a molecular events in advanced primary breast cancer.

Sugarcane stem is an ideal organ for producing foreign pharmaceutical proteins and chemicals by genetic engineering.A perfect promoter driving foreign gene to express strongly and specifically in sugarcane stem is necessary for this purpose. In order to isolate a Sugarcane stem-specific promoter,a fragment of 1968bp nucleotide sequence(Ppst2a)upstream 5' of sugarcane pst2a gene, which was demonstrated to express specifically in sugarcane stem previously was isolated by using chromosomal walking.Bioinformatical analysis of this sequence shows that the sequence contains some typical elements of a promoter. To identify the stem-specific of this promoter,a construct was derived from pCAMBIA1301,which original CaMV 35S promoter was replaced by the 1968bp nucleotide sequence, and named as pCAMBIA1900.Transformations of pCAMBIA1900 and pCAMBIA1301 to leaves and stem pieces of sugarcane were carried out by using particle bombardment. The transient expression of gus showed that the gus expressed specifically in sugarcane with a little higher level compared with CaMV 35S.It is the first report that pst2a promoter is a potential stem-specific promoter which can further be used in transgenes into sugarcane.

A new phenethanol, (2'R)-4-(2', 3'-dihydroxy-3'-methyl-butanoxy)-phenethanol (1), along with other eleven known benzene derivatives (2-12) were isolated from the roots, stems and leaves of Clausena excavata (Rutaceae). Compounds 3 and 4 are new natural products, and compounds 5-8, 10-12 were isolated from C. excavata for the first time. Their structures were elucidated on the basis of MS, 1D and 2D NMR spectroscopic analyses including HSQC, COSY and HMBC experiments. 1 was tested for its cytotoxicities against A549, HeLa and BGC-823 cancer cell lines, and antimicrobial activities against Candida albicans and Staphylococcus aureus. The results showed that 1 did not exhibit cytotoxic and antimicrobial activities.
