1.Study of differentially expressed cDNA on hepatocellular carcinoma

Objective:To search for gene related to hepatocellular carcinoma ( HCC), and study its, potential significance in liver oncogenesis. Methods:The difference in mRNA expression among HCC, nontumorous liver tissue, fetal liver tissue and normal liver tissue was investigated by differential display technique. Ten hepatic tissues of HCC versus surrounding noncancerous liver tissues were examined with MRG98.2 as a probe in Dot blot analysis. Expression of vascular endothelial growth factor (VEGF)mRNA in above mentioned samples was examined with reverse transcription- polymerase chain reaction(RT~ PCR). Results:Fifty five differentially expressed cDNA fragments were isolated from HCC samples, 24 of them were present in HCC and fetal liver tissues simultaneously. Dot blot analysis showed that differential display fragment MRG98.2 expressed in 7 cases of HCC samples (7/10)but only slightly in 2 of the surrounding noncancerous hepatic tissues (2/10). The expression of VEGFmRNA was upregulation in 6/7 of HCC samples expressed MRG98. 2. Gmclusian :The activation of some genes expressed in fetal period may have an important role in the genesis of HCC. MRG98.2 may be a gene related to HCC. Its expression in HCC correlates with VEGFmRNA. MRG98.2 expression is helpful in predicting a tendency toward invasion and metastasis of HCC.
2.Construction and practice of psychological sitcom teaching model in Psychiatric Nursing

Objective To construct psychological sitcom teaching model and explore the effect of its application in Psychiatric Nursing teaching.Methods Through literature review,study of the theories,summary of clinical practice and educational experience,we constructed a theoretical framework of psychological sitcom teaching model for Psychiatric Nursing.This model is composed of four contents (teaching aim,teaching modules,teaching process,teaching basic program) and four sessions (initial calculation,acting scripts,classroom observation and sharing,writing experience and internalization of knowledge).A total of 192 nursing students of grade 2012 from the nursing department were selected as the research objects.At the end of the course,questionnaires were employed to evaluate the effects of the students.Results The questionnaires demonstrated that 99.0% studentsprefer this teaching model and more than 97.9% students considered it helpful to improve four aspects of comprehensive abilities including theoretical learning abilities,leadership and decision making skills,nurse-patient communication skills and psychological nursing abilities.Conclusions Psychological sitcom teaching model is helpful to improve learning effect of Psychiatric Nursing for nursing students and should be widely spread.
3.COMBINED ASSAY OF SERUM TUMOR ENZYME MARKERS IN THE DIAGNOSIS OF HEPATOCELLULAR CARCINOMA

8.Impact of Recent Changes in Domestic Policies and Regulations on the Development and Application of Ge-neric Drugs

Objective:To explore the impact of recent changes in domestic policies and regulations on the development and appli-cation of generic drugs. Methods:Through the collation of the recent regulatory documents, the policy changes for generic drugs were analyzed, and the corresponding development proposals were provided. Results and Conclusion:According to the possible impact of current policy changes on generic drug development, the relative solutions are proposed with the aim to provide certain reference for ge-neric pharmaceutical R & D institutions.
9.Effect of long intergenic noncoding RNA-p21 knockdown on radiosensitivity of hypoxic tumor cells

Objective To investigate the effect and mechanisms of lincRNA-p21 knockdown on radiosensitivity of hypoxic tumor cells.Methods The expression of lincRNA-p21 in SMMC7721 and U251 MG cells after hypoxia treatment were detected by QRT-PCR.Lentivirus vector carrying lincRNA-p21 siRNA or scrambled oligos were constructed.SMMC7721 and U251MG cells with stable integration of lincRNA-p21 siRNA or scrambled sequence were generated through lentiviral-mediated gene transfer.Cell cycle and apoptosis of hypoxic tumor cells were analyzed by flow cytometry.Radiosensitivity of hypoxic tumor cells were measured by cloning formation assay.The expressions of HIF-1α and GLUT1 protein were detected by Western blot.Results Under hypoxia condition (1% O2) for 24 and 48 h, lincRNA-p21 strongly increased (t =5.13, 7.49, 8.90, 10.11, P < 0.05) in SMMC7721 and U251MG cells in a timedependent manner, but this increase was suppressed in the SMMC7721 and U251MG cells with stable integration of lincRNA-p21 siRNA (t =144.81, 334.32, P < 0.05).When SMMC7721 and U251MG cells were cultured under hypoxia condition (1% O2) for 48 h, knockdown of lincRNA-p21 induced G2/M phase arrest (t =7.05, 10.18, P < 0.05) and apoptosis (t =42.27, 24.79, P < 0.05), reduced the expressions of HIF-1α and GLUT1 protein, and enhanced radiosensitivity with a radiosensitization ratio of 1.23 and 1.31, respectively.Conclusions Hypoxia treatment elevate the expression of lincRNA-p21.Knockdown of lincRNA-p21 enhance radiosensitivity of hypoxic tumor cells accompanied with G2/M phase arrest, apoptosis induction and the decrease of HIF-1α protein expression.
10.Effect of trauma scoring system and combined score on the trauma response of acute trauma ;patients

Objective To investigate the performance of Injury Severity Score (ISS), New Injury Severity Score (NISS), Revised Trauma Score (RTS), CRAMS (circulation, respiration, abdomen, motor, speech)score and combined score on the trauma response of trauma patients. Methods Data of acute trauma patients from March 2014 to February 2015 were chosen as the research object. The clinical information at admission was recorded, and the ISS, NISS, RTS, CRAMS and acute physiology and chronic health evaluation Ⅱ(APACHE Ⅱ) were calculated. The optimal cut-off values were looked for the comparability between the four scores and APACHE Ⅱ score were figured out by ROC curve. The joint diagnosis combined physiological score with anatomical score in overlap mode was used for comparing sensitivity and specificity. Results There was a total of 1 020 patients included in the study. APACHEⅡscore ≥20 was found 711 cases, and APACHEⅡ<20 was 309 patients, and there were significant statistic differences in ISS score (U=11.347, P<0.05),NISS score (U=11.969, P<0.05),CRAMS score (U=8.194, P < 0.05) and RTS score (U=8.357, P < 0.05) between two groups. It was showed by ROC curve analysis that the area under the ROC curve (AUC) of ISS, NISS, CRAMS and RTS was 0.907, 0.941, 0.768 and 0.803 (all P<0.05). Compared with the trauma score, combined scores could increase the sensitivity of the prompt assessment of trauma severity in trauma patients, but the combined scores may also reduce the specificity. Conclusions Of these four scoring systems, NISS has the best correlation with APACHEⅡ. Compared with the trauma score, combined scores can increase the sensitivity of the prompt assessment of trauma severity in trauma patients, but the combined scores may also reduce the specificity.
