1.Expression and purification of human S100A2-GST fusion protein in prokaryotic cells

Objective:To obtain human S100A2-GST fusion protein for further research on human S100A2(hS100A2)protein function and its interactions with other proteins and systems.Methods:hS100A2 gene from pHAHA-hS100A2 was subcloned into an expression vector pGST-moluc to construct pGST-moluc-hS100A2.The new plasmid was identified by digestion with XhoⅠand EcoRⅠ.The recombinant BL21 was induced with IPTG to express recombinant fusion protein hS100A2-GST,which was purified by Glutathion-Sepharose 4B ball beads,identified by SDS-PAGE and Western Blot,and quantified by Bradford method.Results:After the digestion,the recombinant plasmid was cutted into two fragments,300 bp and 5kb.After treated with IPTG,the recombinant BL21 strain expressed a protein,which was about 36 kD and was recognized specifically by an-ti-hS100A2.Its yield was 5 mg/L bacterial culture.After isolated by Glutathion-Sepharose 4B ball beads,its purity was 92%.Conclusion:hS100A2-GST expression plasmid pGST-moluc-hS100A2 was constructed successfully.hS100A2-GST fusion pro-tein could be expressed in Escherichia coli with higher yield and isolated with higher purity,which lays the foundation for the follow-up of the hS100A2 research.
2.Therapeutic effect of the regimen containing Moxifloxacin in intensive phase for pulmonary tuberculosis patients

Objective To explore the effect and safety of the regimen containing Moxifloxacin in intensive phase for pulmonary tuberculosis patients. Methods 88 cases with initial tuberculosis(each group of 44 cases) were studied in comparison in the same period. The treatment group contained isoniazid, rifampicin and moxifloxacin of antituberculosis regime and the control group contained isoniazid,rifampicin and pyrazineamide. Results After 2 months intensive phase anti-tubercular treatment ,the sputum negative conversion rates was 95.0% and control's was 94.4%(P > 0.05); the absorb rate of lesions: the treatment group 77.3 % and the control group 79.5%; the liver damage rate: the treatment group and control group was 68.2% ,34.1% respectively(P < 0.05). Conclusion Anti-tuberculosis intensive phase regimes containing moxifloxacin for tuberculosis treatment was safe and effective in initial treatment of pulmonary tuberculose.
3.Contents of Rhodioloside and Polysaccharide in Different Parts of Cultivated Rhodiola sachalinensis

OBJECTIVE:To compare the contents of Rhodioloside and polysaccharide extracted from different part in cultivated Rhodiola sachalinensis.METHODS:The Rhodioloside was extracted with 70% alcohol and determined by HPLC;The polysaccharide was extracted with water and determined by sulphuric acid-phenol method.RESULTS:The content of the Rhodioside ranged between 0.026 5% and 0.443 2%,and the content of polysaccharide between 0.965% and 4.612% in the medicinal herbs,with the main root showing the highest contents.CONCLUSION:The contents of Rhodioside and polysaccharide are obviously different in different part of cultivated Rhodiola sachalinensis.The results provided reasonable theoretical evidence for the exploitation of cultivated Rhodiola sachalinensis.

8.Research of Correlation between Deep Vein Thrombosis and Ankle Fractures

Deep vein thrombosis (DVT) is a serious complication of fractures. The incidence of ankle fracture which occurs most frequently intra-articular is 200/100 000, about 3.9%of all fractures. Domestic and foreign reports of ankle fractures and DVT were allocated and analyzed to review their correlation. Furthermore, different methods of rou-tined thromboprophylaxis was discussed. DVT incident after ankle fractures and the necessity of routined thromboprophy-laxis were reviewed. DVT incidence after ankle fractures was low and routine thromboprophylaxis was not necessary to all ankle fracture patients except the ones who had risk factors of thrombosis which could induce DVT.
9.The relationship between serum 25OHD levels and semen parameters

Objective To analyze the relationship between serum 25 hydroxy vitamin D (25OHD) levels and semen pa?rameters. Methods The serum levels of 25OHD were examined in 68 healthy men, who were divided into three groups. Twenty-two cases were included in 25OHD normal group (30μg/L≤25OHD<40μg/L), 20 cases were in 25OHD relatively insufficient group (20μg/L≤25OHD<30μg/L), and 26 cases were in 25OHD deficiency group (25OHD<20μg/L). The se?rum reproductive hormones including follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E 2), prolactin (PRL) and testosterone (T) were detected by chemiluminescence immune analysis. Semen samples were collected by mastur?bation after a period of abstinence for 3 and 7 days. The total sperm count, sperm concentration, percentage of progressive motile sperm and percentage of normal morphology sperm were measured by sperm quality analyzer. Results There were no significant differences in FSH, LH, E2, PRL and T levels between three groups, and which were all in normal ranges. The total sperm count, sperm concentration, percentage of progressive motile sperm and percentage of normal morphology sperm were significantly lower in 25OHD relatively insufficient group and 25OHD deficiency group than those in 25OHD normal group. The total sperm count and the percentage of progressive motile sperm were significantly lower in 25OHD deficiency group than those in 25OHD relatively insufficient group (P<0.05). There were no statistical differences in sperm concentra?tion and percentage of normal morphology sperm between 25OHD deficiency group and 25OHD relatively insufficient group. Conclusion The decrease of semen quality may be related to serum 25OHD at the relatively insufficient and deficiency lev?els. The effect of 25OHD on semen parameters may not be mediated by reproductive hormones.
