ObjectiveTo investigate the effect of cardiopulmonary bypass (CPB) on the secretory function of islet cells in rabbits.MethodsTwenty adult New Zealand white rabbits of both sexes,weighing 2.5-3.0kg,were randomly divided into 2 groups ( n =10 each):sham operation group (group S) and CPB group.The rabbits were anesthetized with 3％ pentobarbital sodium 30 mg/kg.Blood samples were collected from the left femoral artery at 5 min after anesthesia (T1),immediately before CPB (T2 ),immediately after aortic clamping (T3 ),and at 5,35 and 75 min after aortic unclamping (T4-6) in the two groups for determination of levels of blood glucose,insulin and glucagons.Insulin resistance index was calculated.ResultsCompared with group S,the blood glucose concentration and levels of insulin and glucagons and insulin resistance index at T3-6 were significantly increased in group CPB ( P ＜ 0.05).ConclusionAlthough increase in blood glucose enhances the secretion of insulin in islet β cells,hyperglycemia cannot be compensated completely by the increased insulin during CPB in rabbits.The increase in blood glucose may be related to islet α cell resistance.

Objective To investigate the expression of miR-200c in endometrial carcinoma and its correlation with ZEB2 protein. Methods The expression levels of miR-200c and ZEB2 gene and protein in tissues were detected. The expression of miR-200c in endometrial carcinoma RL95-2 cell line was inhibited by using antisense oligonucleotides and its effect on cell invasiveness was tested. The expressions of ZEB2 gene and protein in cells were detected. Results The expression levels of miR-200c and ZEB2 mRNA and protein in endometrial tissues were significantly higher than those in adjacent tissues and control group (P < 0.05). The average penetrating number in antisense miR-200c transfected group was significantly less than negative control group and liposome group (P < 0.05). The expression levels of ZEB2 gene and protein in antisense miR-200c transfected group were lower than the negative control group and liposome group (P < 0.05). Conclusion The expression of mRNA-200c in endometrial carcinoma was high and it might be promoting tumor cell invasion and metastasis by regulating ZEB2.

Objective To study the effect and safety of controlled-release dinoprostone on the induced labor of late pregnancy . Methods 60 cases of late pregnancy puerpera were treated in our hospital from February 2014 to September 2015, and divided into the control group and the experimental group with reference to the stochastic indicator method, 30 cases in each group. The control group were given induced labor by oxytocin, and the experimental group were given induced labor by controlled-release dinoprostone, Changes of Bishop score, effects of promoting cervical mature, induced labor situation, and adverse reaction were compared between two groups. Results After medication, the cervical Bishop score of the group (8.35±0.93) points were significantly higher than (6.64±0.72) of the control group (P<0.05). The total effective rate 96.67％(29/30) of promoting cervical mature in the experimental group was significantly higher than 80.00％ (24/30) of the control group (P<0.05). Medicine to clinical time (14.89±1.57)h and total labor time (6.94±1.52) h of the experimental group were significantly better than (21.48±2.35) h and (9.30±1.31) h of the control group (P<0.05). The rate of delivery by vagina of the experimental group 83.33％ (25/30) was significantly higher than 60.00％ (18/30) of the control group (P<0.05). There was no different on adverse reaction rate between two groups. Conclusion There was good effects of Controlled-release dinoprostone on the induced labor of later pregnancy, with high rate of delivery by vagina and better safety.

Objective To evaluate the status of the influence factors of foundational course teachers' academic profession development in medical college.Methods The survey includes academic promotion and academic innovation.In aspect of academic innovation,a total of 68 foundational course teachers in medical college were selected.The subjects were tested by scale of exterior influencing factors on innovative behavior,the assessment scale of innovative behavior and the organizational innovation climate scale,the scientific spirit scale of the scientific and technological talents.The impact of demographic characteristics on academic promotion was also tested.Results In aspect of academic promotion,teachers with different department and education showed statistically significant difference in titles(P＜0.05).The age of teachers' promoted to professor is 40-50 years old,vice professor is 30-40 years old.Generally,it takes 6-10 years to promote to senior titles,and it takes less time for doctors.In aspect of academic innovation,scale was used in 5 points,the lowest 1 point,the highest 5 point.The mean score of the scale of exterior influencing factors on innovative behavior is mission design3.28 ±0.96,innovation investment 2.53±0.74,innovation environment 3.10±0.65.The mean score of the assessment scale of innovative behavior and the organizational innovation climate scale is organization target cognition 3.10 ± 0.74,colleagues relationship 3.44±0.82,team innovation motivation 3.34±0.83.The mean score of the scientific spirit scale of the scientific and technological talents in spiritual science dimension is 3.40±0.67 points.The projects with low scores are distributed in three dimensions of innovation investment,innovation environment and organization target cognition.Conclusions Teachers' academic promotion is related to the department and education.In aspect of academic innovation,teachers' self-evaluation of scientific spirit is higher,the evaluation of the mission design,colleagues relationship,team innovation motivation is also higher,the evaluation of the innovation environment,organization target cognition,innovation investment is lower.Material resources,research cooperation and so on especially should be improved.

As an important messenger molecule, NO exists in both peripheral and central nervous system (CNS). Recently, NO was proved to play a critical role in CNS, especially in synaptic plasticity. As a kind of retrograde messenger, it plays some roles in the induction and maintenance of LTP through activation of downstream pathway or molecules, such as NO-cGMP-PKG, ADPRT and C-FOS.

HPLC fingerprint of Congrong Zonggan capsule was established in order to provide basis for quality evaluation. With acteoside as the reference, HPLC was adopted for fingerprint analysis on Congrong Zonggan capsule. The chromatographic conditions wereas follows. Waters C18 column (4.6 mm x 150 mm, 5 μm) was used, with methylalcohol-0.1% formic acid as the mobile phase for gradient elution at the flow rate of 1.0 mL x min(-1). The detection wavelength was 330 nm, and the column temperature was 30 °C. This method was highly accurate and reproducible. All of the 13 components in tested samples reached the baseline resolved peak, and 15 batches of finished products showed the similarity of above 0.95. The method was accurate and feasible and could be used as a simple and effective method to evaluate the quality of the traditional Chinese medicines.

Objective To investigate the effects of preconditioning with different concentrations of nerve growth factor (NGF)-β on ischemia-reperfusion (I/R) injury in isolated rat hearts.Methods Thirty-two male Sprague-Dawley rats weighing 200-300 g,aged 8-10 weeks,were anesthetized with intraperitoneal 10％ chloral hydrate 300 mg/kg and heparin 500 IU/kg.Their hearts were excised and perfused in a Langendorff apparatus with K-H solution oxygenated with 95％ O2-5％ CO2 at 37 ℃.After 10 min of stabilization,the isolated hearts were randomly divided into 4 groups (n=8 each): I/R group and different concentrations of NGF-β groups (groups N1-N3).In group I/R,the hearts were continuously perfused with K-H solution for 30 min,perfusion was suspended for 30 min followed by 120 min reperfusion.In groups N1-N3,the hearts were continuously perfused with K-H solution containing NGF-β0.1,0.2 and 0.4 ng/ml,respectively,for 20 min,followed by 10 min washout,and perfusion was suspended for 30 min followed by 120 min reperfusion.HR,Left ventricular end-diastolic pressure (LV-EDP),left ventricular developed pressure (LVDP) and + dp/dtmax were measured at the end of 10 min stabilization (baseline,T1),immediately before suspension of perfusion (T2),and at 5,30,60 and 120 min of reperfusion (T3-6).The activities of creatine kinase MB (CK-MB) and lactate dehydrogenase (LDH) in the coronary effluent were measured at T1 and T3-T6.Myocardial tissues were obtained at T6 for detection of apoptosis (by TUNEL) and for microscopic examination.Apoptotic index (AI) was calculated.Results Compared with the baseline value at T1,LVDP and + dp/dtmax were significantly decreased,and LVEDP and activities of LDH and CK-MB were significantly increased at T3-T6 in each group,HR was significantly decreased in groups I/R,N2 and N3,while HR was significantly increased in group N1 (P ＜ 0.05).Compared with group I/R,LVDP,+ dp/dtmax and HR were significantly increased,LVEDP,activities of LDH and CK-MB and AI were significantly decreased in group N1,LVDP,+ dp/dtmax and HR were significantly decreased,and activities of LDH and CK-MB were significantly increased in group N2,and LVDP,+ dp/dtmax and HR were significantly decreased,and LVEDP,activities of LDH and CK-MB and Al were significantly increased in group N3 (P ＜ 0.05).LVDP,+ dp/dtmax and HR were significantly lower,and LVEDP,activities of LDH and CK-MB and AI were significantly higher in groups N2 and N3 than in group N1,and in group N3 than in group N2 (P ＜ 0.05).The pathologic changes were reduced in group N1 compared with I/R,N2 and N3 groups.Conclusion Preconditioning with the optimum concentration of NGF-β can at-tenuate I/R injury in isolated rat hearts,however,the injury can be aggravated when the concentration is too large,and inhibition of apoptosis in myocardial cells is involved in the mechanism of myocardial protection.

BACKGROUND: NOV protein encoded by nephroblastoma overexpression gene(NOV) is IGF(insulin-like growth factor) -binding protein. What is its impact on human neural stem cell(hNSC) proliferation and differentiation?OBJECTIVE: To investigate the impacts of NOV protein on hNSCs proliferation and differentiation.DESIGN: A single factor analysis of variance experimental study using cells as subjectsSETTING: Department of histology and embryology, and department of neurobiology in a military medical university.MATERIALS: Study was conducted in the Department of Histology and Embryology of the Third Military Medical University of Chinese PLA. Subjects were hNSCs cultured from 10 to 14 weeks human embryo cerebral cortex.INTERVENTIONS: COS-7 cells were transfected by NOV gene recombined plasmid. COS-7 cell and COS-7 cell modified by NOV gene conditioned culture media(COS-CM and NOV-CM) were collected and reacted with the cultured HNSCs.MAIN OUTCOME MEASURES: hNSCs proliferation was detected by 3H-TdR scintillation analysis, and hNSCs differentiation was detected by immunocytochemistry and flow cytometer(FCM).RESULTS: Both COS-CM and NOV-CM could significant promote the intake of 3H-TdR by HNSCs, of which the 1/minute of NOV-CM group was significantly higher than that of COS-CM group(P ＜ 0.05), which indicated that NOV-CM contained component that could facilitate hNSCs proliferation, and moreover, there was certain dose-effect relationship in NOV-CM' s facilitation of cellular proliferation. The results of immunocytochemistry and FCM revealed that there were more NF-200 positive cells in NOV-CM group, while many glial fibrillary acidic protein positive cells could be seen in COS-CM group.CONCLUSION: NOV protein might have facilitative effects on hNSCs proliferation and differentiation into neurons.
