1.Apoptosis of bladder cancer cells induced by oligonucleotide telomerase inhibitor

Objective To study the potential effects of telomerase inhibitor on the apoptosis of bladder cancer cells. Methods A phosphorothioate oligonucleotide (PS ODN) with sequence 5′ d(TTAGGG) 3′, as the telomerase inhibitor, was incubated with a bladder cancer cell line EJ. Such treated cells were studied with many approaches for the telomerase activity, the growth status and the morphological changes. Results The PS ODN inhibited the telomerase activity in the cells, arrested the cell growth, and induced the apoptosis of the cells. Conclusions The apoptosis of bladder cancer cells could be induced by PS ODN with 5′ d(TTAGGG) 3′ sequence as a telomerase inhibitor.
2.Telomerase activity in the tissue adjacent to bladder cancer and its significance

Objective To study the telomerase activity in the tissue adjacent to bladder cancer and to investigate its clinical significance. Methods Telomerase activity was detected with telomeric repeat amplification PCR (TRAP) assay. The telomerase activity in the tissue adjacent to bladder cancer was evaluated. Results Telomerase activity was positive in the tissue samples adjacent to bladder cancer in 10 of the 24 cases(42%). Telomerase activity in the adjacent tissue has been related with the tumour grades and stages. The tumour recurrence was also related with the telomerase activity in the adjacent tissue. Conclusions The detection of telomerase activity in the tissue adjacent to bladder cancer could be a prognostic marker for bladder tumor recurrence.
3.Study on the related factors for mineral metabolism in elderly patients with maintenance hemodialysis

Objective To investigate the present state of mineral metabolism and its related factors in elderly patients with maintenance hemodialysis (MHD),in order to improve the quality of life and survival rate in elderly patients with MHD in Xinxiang.Methods Clinical data of 466 patients with treatment of MHD for over 3 months in 4 hospitals in Xinxiang district were enrolled from January 2012 to August 2013 in this study,and the patients were divided into the elderly group (n=159,aged ≥ 60 years) and the non-elderly group (n =307,aged < 60 years).The evaluation indexes for mineral metabolism were determinated,such as serum levels of calcium,phosphorus,intact parathyroid hormone (iPTH) and alkaline phosphatase (ALP).The present state of mineral metabolism and its relationships with anemia,dialysis ages,adequacy of dialysis,malnutrition and inflammatory factors were analyzed in elderly patients with MHD.Results Among 466 patients,the serum levels of calcium,phosphorus and iPTH in MHD patients were (1.95 ± 0.34) mmol/L,(2.54±1.38) mmol/L and (409.5±345.6) ng/L respectively,and their control rates were 34.3%,20.4% and 25.5% respectively.And in patients with hemodialysis three times a week,the control rates were 50.8%,31.4% and 32.2% respectively.The serum levels of phosphorus,iPTH,ALP,albumin and transferrin saturation were lower in elderly group than in non-elderly group (all P<0.01),and the serum levels of pre-albumin,hemoglobin and feritin were lower in elderly group than in non-elderly group (all P<0.05).But the serum levels of calcium and C-reactive protein (CRP)were higher in elderly group than in non-elderly group (all P<0.05).Conclusions The elderly patients with MHD exhibit unique clinical manifestations of abnormal mineral metabolism,and low bone transformation osteopathy is the main manifestation.The micro-inflammation,malnutrition and anemia are more severe in elderly patients than in non elderly patients.
4.EFFECT OF ATIFICIAL FIBER COMPLEXES ON GROWTH AND ERYTHROCYTE MEMBRANE FLUIDITY IN RATS

Objective: To examine the effect of artificial fiber complexes (AFC) on growth and RBC membrane fluidity in comparison with three single fibers. Methods: 48 healthy, female Wistar rats (50-80 g) were randomly divided into 6 groups based on body weight. All five kinds of DF (cellulose, pectin, sodium alginate, cellulose pectin complex, cellulose sodium alginate complex) were given at a level of 10% in a lipid rich diet. The control group was fed lipid rich diet alone without DF. After 8 weeks, the effect on growth and development, lipid peroxidation, RBC membrane fluidity were studied. Results: 1.All DF groups did not affect the growth and development of the rats. 2.The fecal weight and its fat content in all DF groups were significantly higher than that in control group, especially for AFC group.3. All kinds of DF could increase serum SOD and GSH Px activities and reduce MDA content (except cellulose and sodium alginate group). RBC membrane fluidity was significantly increased in DF groups, and AFC groups were the most effective. Conclusion: All these kinds of DF have the effect of increasing RBC membrane fluidity, and AFC is the best.
5.Clinical manifestation and outcome of severe ulcerative colitis

Objective To evaluate the clinical outcome of severe ulcerative colitis(UC)and to find the factors related to treatment and outcome.Methods Forty one hospitalized patients with UC during 1988-2004 were retrospectively reviewed.Data were recorded including the onset,symptoms,signs, laboratory,endoscopic,radiologic and pathologic findings,as well as the processes of clinical treatment. The patients who undergone surgery were also analysed.Results Forty one of 144(28.5%)hospital ized patients were suffered from severe UC,and among them 92.7%(38/41)had pancolitis.The patients who had first onset,chronic persistent,chronic recurrent type were account for 36.9%(15/41),36.9%(15/41) and 26.8%(11/41),respectively.The steroids treatment played the main role in the inducing remission of severe UC(61.0%).Thirty one cases(75.6%)could be relieved by drug therapy.Seven cases(17.1%) were progressed to have operation.The age of early onset,pancolitis,low hemoglobin and serum albumin levels and need of intravenous steroids treatment were associated with the need of surgery.Conclusions Most of the severe UC patients respond well to the medical therapy,but for some non-responding or steroids depending individuals,after a reasonable duration of treatment,surgery should be considered.
6.Establishement of HPLC fingerprints of flavonoids in Ziziphi spinosae Semen and determination of two constituents

AIM To establish the HPLC fingerprints of flavonoids in Ziziphi spinosae Semen and to determine the contents of two constituents.METHODS With spinosin as a reference peak,the HPLC fingerprints of ten batches of samples were established.The analysis of methanol extract of flavonoids in Ziziphi spinosae Semen was performed on a 30 ℃ thermostatic Agilent TC-C18column (4.6 mm × 250 mm,5 μm),with the mobile phase comprising of acetonitrile-0.1% acetic acid flowing at 1 mL/min in a gradient elution manner,and the detection wavelength was set at 280 nm.RESULTS There were ten common peaks in the HPLC fingerprints,eight of which (vicenin-Ⅱ,glucosylvitexin,isospinosin,spinosin,6'-pyridyloylspinosin,6'-p-hydroxybenzoylspinosin,6'-feruloylspinosin and 6'-p-coumaroylspinosin) were identified.Spinosin and 6'-feruloylspinosin showed good linear relationships within the ranges of 15.00-40.00 μg and 5.00-14.00 μg,whose average recoveries (RSDs)were 100.5% (1.6%) and 100.4% (1.6%),respectively.CONCLUSION This accurate,simple and reliable method can be used for the quality control of flavonoids in Ziziphi spinosae Semen.
7.Adhesion levels in uropathogenic Escherichia coli with various degree of drug resistance

Objective To investigate the adhesion levels in uropathogenic Escherichia coli with various degree of drug resistance.Methods One hundred strains of Escherichia coli isolated from urine specimen were collected from patients admitted to 4 Grade A tertiary hospitals in Tianjin during March 2012 to October 2015.Escherichia coli were divided into drug sensitive group and drug resistant group by drug sensitivity tests with 50 strains in each group.The expressions of fimH,fimA,fimB genes of type I fimbriae and papA,papB,papC,papGII genes of P fimbriae were detected by polymerase chain reaction(PCR)and real-time fluorescence quantitative RCR (RT-PCR),respectively.Adhesion ability of type I fimbriae and P fimbriae were tested by yeast cell adhesion test and erythrocyte agglutination test.Chi square test and t(Z) test were used to analyze the data.Results The positive rate of papGII in drug resistant group (42.0%) was significantly higher than that in the drug sensitive group (16.0%)(χ2 =8.208,P <0.05),while there were no significant differences in the positive rates of fimH,fimA,fimB,papA,papB and papC genes between two groups(all P >0.05).The expression levels of fimH,fimB and papC genes in the sensitive group were higher than those in the resistant group(Z =3.427,t =5.182 and 8.120,all P <0.05).The adhesion ability of strains carrying type I fimbriae in sensitive group was stronger than that of resistant group (χ2 =5.769,P <0.05).Conclusions The decline in adhesion ability of type I fimbriae in drug resistant E.coli strains is possibly associated with the adaptive cost of bacteria,the transcription and deficiency of other genes encoded by fim and pap gene cluster will also affect the adhesion function of type I pili and type P pili.
8.Analysis of live newborn birth condition change trend in Hongjiang people′s hospital from 1987 to 2011

Objective To learn the change trend of live newborn birth condition from 1987 to 2011 in Hongjiang people′s hospi-tal .Methods Cluster sampling was adopted and altogether 2 711 singleton live newborns in 1987 ,1995 ,2005 ,2011 year were retro-spectively analyzed .Results From 1987 to 2011 ,the birth weight changes of live newborn overall showed an increasing trend .The sex ratio of birth appeared rising after 1995 ,especially apparent in parous wowen and rural areas .Incidence of macrosomia showed an increasing trend ,the post-term infant delivery rate decreased after 1995 .Low birth weight infant and premature infant birth rate trend was not obvious .Conclusion 25 years in Hongjiang people`s hospital ,the overall trend of live newborn birth outcomes is good which prompts that the quality of perinatal care in the hospital is improved .In the future ,the perinatal health care should focuse on taking effective comprehensive measures to control the growth of the birth sex ratio and the incidence of macrosomia .

10.Three-dimensional Finite Element Analysis of Biomechanical Effect of Rigid Fixation and Elastic Fixation on Lumbar Interbody Fusion.

This study was aimed to compare the mechanical characteristics under different physiological load conditions with three-dimensional finite element model of rigid fixation and elastic fixation in the lumbar. We observed the stress distribution characteristics of a sample of healthy male volunteer modeling under vertical, flexion and extension torque situation. The outcomes showed that there existed 4-6 times pressure on the connecting rod of rigid fixation compared with the elastic fixations under different loads, and the stress peak and area of force on elastic fixation were much higher than that of the rigid fixations. The elastic fixation has more biomechanical advantages than rigid fixation in promoting interbody lumbar fusion after surgery.
