1.Quality of root canal fillings using three gutta-percha obturation techniques.

OBJECTIVES: The goal of this study was to compare the density of gutta-percha root fillings obturated with the following techniques: cold lateral (CL) compaction, ultrasonic lateral (UL) compaction, and warm vertical (WV) compaction. MATERIALS AND METHODS: Thirty-three extracted mandibular first molars, with two separate mesial canals in each, were selected. After instrumentation, the canals were stratified into three groups based on canal length and curvature, and underwent obturation with one of the techniques. No sealer was used in order to avoid masking any voids. The teeth were imaged pre- and post-obturation using micro-computed tomography. The reconstructed three-dimensional images were analyzed volumetrically to determine the amount of gutta-percha present in every 2 mm segment of the canal. P values < 0.05 were considered to indicate statistical significance. RESULTS: The overall mean volume fraction of gutta-percha was 68.51 +/- 6.75% for CL, 86.56 +/- 5.00% for UL, and 88.91 +/- 5.16% for WV. Significant differences were found between CL and UL and between CL and WV (p < 0.05), but not between UL and WV (p = 0.526). The gutta-percha density of the roots treated with WV and UL increased towards the coronal aspect, but this trend was not noted in the CL group. CONCLUSIONS: WV compaction and UL compaction produced a significantly denser gutta-percha root filling than CL compaction. The density of gutta-percha was observed to increase towards the coronal aspect when the former two techniques were used.

3.Study on prevalence and correlation factors of bronchial asthma in Zaozhuang area, Shandong province.

OBJECTIVETo study the prevalence of asthma and its correlated factors in Zaozhuang area in 2003, to provide a basic consideration for prevention/treatment and control policy.
METHODS6 points were selected by stratified-clusterd-random sampling with a total of 16,725 persons expected, but only 10,610 subjects investigated.
RESULTSIn this survey, 128 asthma cases were identified with a overall prevalence of 1.21%. The prevalence for children was 2.02%, and for adult was 0.90% with the former significantly higher then the latter (chi(2) = 21.39, P < 0.01). Rates for male and female were 1.08%, 1.32% with a ratio of 1:1.22. For 77.97% of children with asthma. The initiative age of asthma was before 7 years old among children while among 36.23% of the adults, it was before 15 years of age. Correlation analysis showed that upper respiratory tract infection (OR = 17.81, 95% CI: 12.25-25.89), cold air exposure (OR = 3.43, 95% CI: 2.41-4.90), stimulation through cooking and by harmful gases (OR = 2.56, 95% CI: 1.80-3.63), allergic materials (OR = 2.74, 95% CI: 1.80-4.17) were main inducing factors. 65.63% of the asthma cases having had history of allergic disease while 25.78% having had family history with the OR of allergic history and family history as 21.69 vs. 73.96.
CONCLUSIONThe epidemic status of bronchial asthma was serious, with an assumption that asthma cases might have reached the number of 43 thousand in Zaozhuang area.

4.Clinical observation on abdominal electroacupuncture for treatment of poststroke constipation.

OBJECTIVETo compare therapeutic effects of abdominal electroacupuncture (EA) and western medicine on poststroke constipation.
METHODSEighty cases were randomly divided into an EA group and a medication group, 40 cases in each group. The EA group were treated with EA at Daheng (SP 15), Fujie (SP 14), Tianshu (ST 25), Shuidao (ST 28), etc., once a day, 30 min each session, and the medication group with oral administration of 10 mg Cisapride, thrice each day. Seven days constituted one course. After 2 courses, clinical therapeutic effects were evaluated by cumulative scores of symptoms.
RESULTSThe total effective rate of 92.5% in the EA group was significantly better than 72.5% in the medication group (P < 0.05). After treatment, the cumulative scores of clinical symptoms significantly decreased in the two groups (P < 0.05) and the improving degrees of symptoms in the EA group was significantly better than that in the medication group (P < 0.05).
CONCLUSIONAbdominal electroacupuncture has a definite therapeutic effect on poststroke constipation, accelerating gastrointestinal movement.

5.Screening of differential expression proteins in serum and detection of C-reactive protein in patients with chronic spontaneous urticaria

Objective:To investigate the relationship of C-reactive protein expression in serum with duration time of wheals in patients with chronic spontaneous urticaria(CSU).Methods:The differential proteins in serum in CSU patients with different duration time of wheals were identified using isobaric tag for relative absolute quantitation(iTRAQ)and the expression of CRP in serum was analyzed.The levels of serum CRP in CSU patients were detectedby immune nephelometry.Results:The level of CRP in serum was up-regulated in CSU patients(P<0.05).The expression of CRP in serum was significantly higher in CSU patients with longer duration of wheals than that in CSU patients with shorter duration of wheals(P< 0.05). Conclusions:The expression of serum CRP is related to duration time of wheals in CSU patients.
6.Change of early serum TNF-alpha and IL-6 levels in acute cerebral infarction and its significances.

OBJECTIVETo investigate the change of early serum TNF-alpha and IL-6 levels in acute cerebral infarction and its significances.
METHODSSerum TNF-alpha and IL-6 levels in 30 health subjects and 35 patients with acute cerebral infarction (ACI) within 6 hours of onset were measured by enzyme linked immunosorbent assay (ELISA); neurological deficits scores (NDS) in all cases were determined, and Spearman test was used for correlation.
RESULTSThe serum levels of TNF-alpha and IL-6 in ACI group were markedly higher than those in health subjects and there was a positive correlation of TNF-alpha and IL-6 levels with 6 h NDS (rs=0.89 and 0.93, P<0.001) and with NDS progression (rs=0.90 and 0.91, P<0.001). Early serum TNF-alpha and IL-6 levels in progressive cerebral infarction (PCI) group were evidently higher than those in stable cerebral infarction (SCI)[(49.56+/-12.12) pg/L compared with (24.30+/-7.4) ng/L and (39.76+/-7.88) ng/L compared with (20.78+/-6.28) ng/L, respectively, P<0.01)].
CONCLUSIONThe early serum levels of TNF-alpha and IL-6 in ACI markedly increase and are closely correlated with disease severity; which may be of value in PCI risk evaluation.

OBJECTIVETo investigate the availability of serum level of macrophage clony stimulating factor (M-CSF) as a marker for early diagnosis of Alzheimer's disease (AD).
METHODSThe serum levels of M-CSF in 70 patients with AD, 52 healthy controls, 22 patients with VAD (vascular dementia) were measured and the serum levels of IL-1 beta, IL-6, TNF-alpha in 32 patients with AD and 20 controls were measured as well.
RESULTSSerum levels of M-CSF were significantly elevated in patients with AD when compared with healthy controls (P < 0.01) and VAD controls (P < 0.05) respectively. At the early stage of mild dementia and middle dementia, serum levels of M-CSF were significantly elevated, but at the later stage of severe dementia, they returned to normal level. Serum levels of IL-1 beta were significantly elevated in AD patients compared with controls (P < 0.05), and serum levels of TNF-alpha and IL-6 were within the normal range in patients with AD.
CONCLUSIONSThe results suggest that serum M-CSF level may provide a convenient and sensitive means for the early diagnosis of Alzheimer's disease.

8.Expression and evaluation of porcine circovirus type 2 capsid protein mediated by recombinant adenoassociated virus 8

Background:
Porcine circovirus type 2 (PCV2) is an important infectious pathogen implicated in porcine circovirus-associated diseases (PCVAD), which has caused significant economic losses in the pig industry worldwide.
Objectives:
A suitable viral vector-mediated gene transfer platform for the expression of the capsid protein (Cap) is an attractive strategy.
Methods:
In the present study, a recombinant adeno-associated virus 8 (rAAV8) vector was constructed to encode Cap (Cap-rAAV) in vitro and in vitro after gene transfer.
Results:
The obtained results showed that Cap could be expressed in HEK293T cells and BABL/c mice. The results of lymphocytes proliferative, as well as immunoglobulin G (IgG) 2a and interferon-γ showed strong cellular immune responses induced by Cap-rAAV. The enzyme-linked immunosorbent assay titers obtained and the IgG1 and interleukin-4 levels showed that humoral immune responses were also induced by Cap-rAAV. Altogether, these results demonstrated that the rAAV8 vaccine Cap-rAAV can induce strong cellular and humoral immune responses, indicating a potential rAAV8 vaccine against PCV2.
Conclusions
The injection of rAAV8 encoding PCV2 Cap genes into muscle tissue can ensure long-term, continuous, and systemic expression.
9.Study on the Expression Level of Magnesiun Transporter Protein 1 in Peripheral Blood T Lymphocytes of Patients with Systemic Lupus Erythematosus and Its Clinical Value

Objective To investigate the expression of magnesiun transporter 1 protein(MagT1)in peripheral blood T lymphocytes of patients with systemic lupus erythematosus(SLE),and elucidate its clinical significance.Methods A total of 79 SLE patients admitted to the People's Hospital of Xishuangbanna Dai Autonomous Prefecture from January 2019 to June 2021 were selected as the study subjects,and SLE patients were divided into moderate to severe group(SLEDAI ≥ 10,n=32)and mild group(SLEDAI<10,n=47),according to SLE disease activity index(SLEDAI)score.Another 40 healthy patients who underwent physical examinations during the same period were selected as control group.The levels of clinical biochemical and serum immune-related indexes were recorded.The expressions of MagT1 mRNA and protein in peripheral blood T lymphocytes were detected by quantitative real-time PCR(qRT-PCR)and Western Blot methods.The correlation between the expression of MagT1 protein in peripheral blood T lymphocytes and biochemical and immunity-related indexes was analyzed by Pearson correlation analysis.Receiver working characteristic(ROC)curve was drawn to analyze the diagnostic value of MagT1 protein expression of peripheral blood T lymphocytes in diagnosing SLE and its severity.Results Compared with the control group,the expression levels of MagT1 mRNA[0.65(0.36,0.99),0.23(0.07,0.36)vs 1.20(0.83,1.37)]and MagT1 protein[0.35(0.22,0.42),0.22(0.15,0.27)vs 0.53(0.40,0.63)]of peripheral blood T lymphocytes in the mild group and the moderate to severe group were reduced,with significant differences(Z=5.247,7.078;5.128,7.257,all P<0.05).Compared with the mild group,the expression levels of MagT1 mRNA[0.65(0.36,0.99)vs 0.23(0.07,0.36)]and protein[0.35(0.22,0.42)vs 0.22(0.15,0.27)]of peripheral blood T lymphocytes in the moderate to severe group were reduced,with significant differences(Z=5.169,3.599,all P<0.05).Pearson correlation results showed that MagT1 protein expression in peripheral blood T lymphocytes was positively correlated with serum hemoglobin(HGB),complement 3(C3)and C4 levels(r=0.496,0.637,0.588,all P<0.05),but was negatively correlated with serum erythrocyte sedimentation rate(ESR),hypersensitive C-reactive protein(hs-CRP),IgG levels and SLEDAI(r=-0.598,-0.476,-0.646,-0.514,all P<0.05).The results of the ROC curve showed that the area under the curve in diagnostic SLE for the expression level of peripheral blood T lymphocyte MagT1 protein was 0.893(95%CI:0.838~0.948),with a sensitivity and a specificity of 90.0%and 67.1%,respectively,and the area under the curve in diagnostic SLE severity was 0.739(95%CI:0.631~0.848),with a sensitivity and a specificity of 57.4%and 84.4%,respectively.Conclusion The expressions of peripheral blood T lymphocyte and MagT1 were down regulated in SLE patients,and they had a negative trend with the severity of SLE.They may have a certain diagnostic value for the diagnosis and severity of SLE.
10.Impact of premature rupture of membranes on neonatal complications in preterm infants with gestational age <37 weeks.

OBJECTIVETo investigate the effect of premature rupture of the membrane (PROM) on neonatal complications in premature infants.
METHODSThe registration information of 7684 preterm infants with gestational age <37 weeks were collected from the cooperative units in the task group between January 1, 2014 to December 31, 2014. Specially trained personnel from each cooperative units filled in the unified form in a standardized format to record the gender, gestational age, birth weight, PROM, placental abruption, antenatal corticosteroid, Apgar score, amniotic fluid pollution, and complications of the infants. The data were analyzed comparatively between the cases with PROM and those without (control).
RESULTSThe preterm mortality rate was significantly lower but the incidences of ICH, NEC, ROP and BPD were significantly higher in PROM group than in the control group (P<0.05). The 95% confidence interval of the OR value was <1 for mortality, and was >1 for ICH, NEC, ROP and BPD. After adjustment for gestational age, birth weight, gender, mode of delivery, placental abruption, placenta previa, prenatal hormones, gestational diabetes mellitus (GDM), gestational period hypertension and 5-min Apgar score <7, the incidences of NEC, ROP and BPD were significantly different between the two groups (P<0.05) with 95% confidence interval of OR value >1, but the mortality rate and incidence of ICH were not significantly different between the two groups (P>0.05).
CONCLUSIONPROM is a risk factor for NEC, ROP and BPD in preterm infants, and adequate intervention of PROM can reduce the incidences of such complications as NEC, ROP and BPD in the infants.
