1.Roles of reactive oxygen species in HepG_2 cell apoptosis induced by CDDP

Objective To investigate the effects of acivicin on HepG 2 cell apoptosis and the effects of acivcin with cis diaminedichloroplatinum (CDDP) on HepG 2 cell apoptosis and on production of intracellular ROS. Methods HepG 2 cells were treated with acivcin, CDDP, and acivicin combined with CDDP. The cytotoxicity was measured by MTT assay. Flow cytometry was used to detect the intracellular ROS, and the rate of apoptosis was determined by TUNEL assay. Results The concentration of acivicin to cause HepG 2 IC 50 was 1.4 mmol/L, and CDDP was 67 ?mol/L. Significant increases in intracellular ROS content were observed in HepG 2 cells 24 h after treatment with acivicin (1.4 mmol/L), CDDP (67 ?mol/L), or acivicin (1 4 mmol/L) plus CDDP (67 ?mol/L), especially with acivicin (1 4 mmol/L) plus CDDP (67 ?mol/L). HepG 2 cell apoptotic rates induced by acivicin (1 4 mmol/L) at 24, 48, and 72 h were (16 3?3 5), (27 9?4 3), (47 2?3 0), respectively, and a significant difference was observed compared to that of control group ( P
2.Changes and significance of P~(53) , C-myc, Bax, and Bcl-2 in surviving HepG_2 cells under continuous stress of cisplatin

Objective To observe the dynamic changes of P 53 , C myc, Bax, Bcl 2, and NF ?B in surviving HepG 2 cells under the continuous stress of cisplatin for the investigation of the potential role in the resistance of HepG 2 cells to cisplatin. Methods The apoptosis of HepG 2 cells and changes of the apoptosis associated factors including P 53 , C myc, Bax, Bcl 2, and NF ?B in the surviving HepG 2 cells were detected by immunohistochemical and RT PCR assays. Results No obvious change of P 53 and C myc was found in the surviving HepG 2 cells under cisplatin stress, but persistent increase of Bcl 2 and decrease of Bax and a continuous increase of Bcl 2/Bax ratio were found. Markedly positive expression of NF ?B mRNA was found at 7 d after the stress of cisplatin. Conclusion The mechanisms of the surviving HepG 2 cells under continuous stress of cisplatin may be associated with the increase in Bcl 2 and decrease in Bax, specially the continuous increase in Bcl 2/Bax ratio, while the positive NF ?B mRNA expression may be associated with the later events after HepG 2 cell survive.

5.Impact on cervical axial symptom of modified open-door laminoplasty with C_3 laminectomy

Objective To verify whether the modified open-door laminoplasty with C3 laminectomy is effectve in preventing postoperative axial symptom. Methods Since March 2001 to March 2005, 44 patients who underwent modified open-door laminoplasty with semispinalis cervicis insertion intact were included in this study(group A). 50 patients who underwent conventional open-door laminoplasty in the authors' department during the same period were served as control(group B). Preoperative and postoperative Japanese Orthopedic Association(JOA) scores, the degree of axial symptom, ranges of neck motion, cervical curvature index were recorded and compared. Results The average JOA recovery rate was 59.2%?11.3% for group A and 60.1%?19.5% for group B. There was no significant difference in JOA recovery rate between two groups. Objective To investigate the effects of sulfur dioxide(SO2) pollution in different concentrations on activity of SOD,levels of GSH and MDA in heart and lung tissue of exercised rats,in order to elucidate the toxicological mechanism of SO2 pollution.Methods Forty-eight SD rats were randomly divided into eight groups:a rest group(RG),an exercise group(EG),a low pollution exercise group(LEG),a low pollution rest group(LRG),a moderate pollution exercise group(MEG),a moderate pollution rest group(MRG),a high pollution exercise group(HEG) and a high pollution rest group(HRG).All groups,except RG and EG,were exposed to SO2 with different concentrations(5 mg/m3,10 mg/m3,15 mg/m3),meanwhile exercised rats run in a motor-driven wheel at a speed of 8 m/min,2 h/d for 10 days.Rats were sacrificed at 24 h after treatment and the anti-oxidative enzymes activity and lipid peroxidation(LPO) levels were measured.Results Compared with RG,SOD activity of lung tissue significantly decreased in HEG,MEG,LEG(P
7.Bibliometric Analysis of TCM Preserving and Health Care Based on CNKI

Objective To understand the status of clinical study and development trend of TCM preserving and health care based on bibliometric analysis. Methods Literature related to TCM preserving and health care in CNKI was searched by computers. NoteExpress3.0.4.6732 was used to conduct metrology analysis on publication time, keywords, literature sources, high-frequency authors, study level, publication institutes and fund program, with a purpose to discuss the research condition of this field. Results Totally 2921 valid articles were achieved after screening, including 342 conference articles, 2624 journal articles, 115 academic dissertations. The publication amount increased each year; the cumulative amount of the literature showed a J-type growth trend, and did not translate into a logical pattern of growth curve; spiritual emotion, diet therapy, health intervention were high-frequency keywords; literature source distribution was in line with Brad law; the first eight core journals were Modern Health, Family Medicine, Health Magazine, Guangming Journal of Chinese Medicine, Health Science, China Journal of Basic Medicine in Traditional Chinese Medicine,Chinese Health Care, Chinese Medicine Modern Distance Education of China; publication institutes were Beijing University of Chinese Medicine, Guangzhou University of Chinese Medicine, China Academy of Chinese Medical Sciences, and Shanghai University of Traditional Chinese Medicine; fund programs were mainly national projects. Conclusion TCM preserving and health care show a good development trend, with large space for development, requires more researches.
8.Apoptosis of hepatic stellate cells induced by sulfasalazine

Objective To determine whether sulfasalazine stimulates hepatic stellate cell (HSC-T6) apoptosis and its possible mechanism. Methods CCK-8 assay, acridine orange/ethidium bromide(AO/EB) and Annexin Ⅴ FITC/PI were used to determine cell growth and cell apoptosis. The expression of NF-?B P65, phospho-IKK and phospho-I?B was detected by Western blotting. The nuclear translocation of HSC-T6 P65 was observed with laser confocal microscopy. Results Sulfasalazine displayed a strong growth inhibition and promoting apoptosis effect on HSC-T6 cells in a dose and time-dependent manner. Sulfasalazine, but not 5-aminosalicylic acid or sulfapyridine, inhibited the activation of NF-?B by down-regulating the expressions of P-IKK, P-I?B and the nuclear translocation of P65. Conclusion Sulfasalazine can inhibit NF-?B activity and promote apoptosis in HSC-T6 cells, where the Rel/NF-?B/I?B/IKK pathway plays an important role in HSC survival.
9.Phaeohyphomycosis Caused by Bipolaris Spicifera:First Case Report in China

Objective To isolate and identify pathogenic f ungus in a patient with intracranial infection.Methods Specimens were taken from the spinal fluid of the patient.Then,microsco py and fungal culture were done to identify the pathogen.The hi stopathologic features were reprod uced through animal pathogenicity s tudy in a mice model.Results According to the colony appearance i n culture medium,the morphological features in microscopy,such as conidia arrangement and size,germ tube forming site,this fungus was identified as Bipolaris spicifera.Hyphae and swollen hyphal cells resembling chlamydospores,septate pi gmented hyphae were observed in brain tissue specimen of mice experimental model,which were consiste nt with phaeohyphomycosis.Conclu-sion This is the first case of phaeohyphom ycosis caused by Bipolaris spicifera reported in China.
10.Relationship between HLA-DRB1 alleles and vascular anomalies in infants of Han nationality in Guangxi Zhuang Autonomous Region

Objective To investigate the relationship between HLA-DRB1 alleles and vascular anomalies in infants of Han nationality in Guangxi Zhuang Autonomous Region.Methods This study included 145 infants with vascular anomalies (99 cases of hemangioma (hemangioma group) and 46 cases of vascular malformation (vascular malformation group)) and 105 healthy infants (control group) of Han nationality residing in Guangxi Zhuang Autonomous Region.The genotypes of HLA-DRB1 alleles were determined by using PCR-sequence specific primers (PCR-SSP).Chi-square test was performed to analyze the difference in the frequency of HLA-DRB1 alleles between these groups by using the SPSS 16.0 software.Results There were significant differences in the frequency of HLA-DRB1*0901,*1401 and *16 alleles among the hemangioma group,vascular malformation group and control group (x2 =13.05,12.79,10.36,respectively,all P < 0.01).Paired comparison revealed significant differences in the frequency of HLA-DRB1*0901 allele between the hemangioma group and vascular malformation group (RR =4.84,P < 0.01) as well as between the hemangioma group and control group (RR =3.21,P < 0.01),and in the frequency of HLA-DRB1*16 allele between the hemangioma group and control group (RR =2.25,P < 0.01) as well as between the vascular malformation group and control group (RR =2.60,P < 0.01).The frequency of HLA-DR*1401 allele was significantly lower in the hemangioma group than in the control group (RR =0.30,P < 0.01).Conclusions HLA-DRB1*0901 and *16 may be the predisposing genes for hemangioma and vascular anomalies respectively,while HLA-DRB1*1401 appears to be protective against hemangioma,in infants of Han nationality in Guangxi Zhuang Autonomous Region.
