2.Follow-up study of 1035 children with recurrent wheezing

Objective To investigate the long-term outcome of children with recurrent wheeze and to determine the effectiveness of inhaled hormone therapy. Methods One thousand and thirty-five children with recurrent wheezing were followed up for more than 4 years and the data were retrospectively evaluated. Results Of 1035 cases, 751 (72.56%) patients outgrew their wheeze during the follow-up period, whereas the other 284 (27.44%) patients had recurrence wheeze during the last two years. The age of wheezing onset was<3 years in 542 (52.37%) cases, from 3 to 7 years in 386 (37.29%) cases, and from 7 to 12 years in 107 (10.34%) cases. There was significant difference in clinical control rate among groups with different wheezing ages onset (χ2=45.27, P<0.001). Children with wheezing age onset from 7 to 12 years had the lowest clinical control rate. Among 1035 wheeze children, 343 (79.95%) children in 429 cases who received inhaled hormone therapy for more than one year outgrew their wheeze. Whereas 408 (67.35%) in 606 cases who did not receive inhaled hormone therapy outgrew their wheeze. There was significant difference of clinical control rate between inhaled group and non-inhaled group (P<0.01). Con-clusions The age of wheezing onset is<7 years in 89.66%of children with recurrent wheeze. Most of them can be clinicalycon-trolled. The long term inhaled hoemone therapy for children with recurrent wheeze can reduce the risk of developing adulthood asthma.

4.A study on detection method of lamivudine related mutations in hepatitis B virus polymerase gene.

OBJECTIVETo establish a simple and accurate method for rapid detection of lamivudine related mutations in hepatitis B virus (HBV) polymerase gene.
METHODSHBV polymerase gene fragments of covering B and C active region were amplified by nested polymerase chain reaction (nPCR) or nested mismatched PCR. The PCR products were digested with Nde I or Nia III and subjected to electrophoresis on agarose gel, respectively. The patterns of restriction fragment length polymorphism (RFLP) were distinguished. Using this method, thirty patients with chronic hepatitis B and treated with lamivudine for at least one year were analysed for the lamivudine related mutations in polymerase gene. Sixteen cases without lamivudine therapy were used as controls. Some of the patients were also analysed by clone sequencing.
RESULTSThe nested mismatched PCR-RFLP method was simple, accurate and rapid. The whole experiments could be finished in eleven hours. The least titers of HBV DNA which could be detected was 10.3 copies/ml. The wild or mutant strains judged by RFLP were identified by clone sequencing. Mutation in the tyrosine methionine aspartic aspartic acid (YMDD) motif of HBV polymerase gene was found in eight patients and mutations of YMDD motif associated with L526M were found in another three patients. However, there were no such mutations in the control cases.
CONCLUSIONThe nested PCR-RFLP is considered as a simple and accurate method for rapid detection of lamivudine related mutations in HBV polymerase gene. It is suitable for larger number of sample detection.

5.Preparation of budesonide-poly (ethylene oxide) solid dispersions using supercritical carbon dioxide and in vitro evaluation.

An application of supercritical fluids technology for processing of budesonide-poly (ethylene oxide) solid dispersions was presented. The correlations of the operation parameters in the preparation process were studied. Solid dispersions of budesonide in poly (ethylene oxide) were prepared using a static method for supercritical carbon dioxide and characterized by powder X-ray diffractometry, differential scanning calorimetry, intrinsic dissolution, and in vitro dissolution. It was found that the optimum condition of solid dispersions formation was as follows: temperature, 40 degrees C ; pressure, 20 MPa; the ratio of budesonide and poly (ethylene oxide) , 1: 10. Drug existed in amorphous state in hydrophilic poly (ethylene oxide) carriers and intrinsic solubility and dissolution rates were significantly enhanced. The mechanism of the enhanced dissolution may be attributed to the amorphous character of the budesonide, improvement of the wettability of the hydrophobic budesonide, together with the formation of hydrogen bond of budesonide and hydrophilic poly (ethylene oxide). The supercritical fluids process can be used as an alternative method for preparation of solid dispersions.

6.The similarity between human embryonic stem cell-derived epithelial cells and ameloblast-lineage cells.

This study aimed to compare epithelial cells derived from human embryonic stem cells (hESCs) to human ameloblast-lineage cells (ALCs), as a way to determine their potential use as a cell source for ameloblast regeneration. Induced by various concentrations of bone morphogenetic protein 4 (BMP4), retinoic acid (RA) and lithium chloride (LiCl) for 7 days, hESCs adopted cobble-stone epithelial phenotype (hESC-derived epithelial cells (ES-ECs)) and expressed cytokeratin 14. Compared with ALCs and oral epithelial cells (OE), ES-ECs expressed amelogenesis-associated genes similar to ALCs. ES-ECs were compared with human fetal skin epithelium, human fetal oral buccal mucosal epithelial cells and human ALCs for their expression pattern of cytokeratins as well. ALCs had relatively high expression levels of cytokeratin 76, which was also found to be upregulated in ES-ECs. Based on the present study, with the similarity of gene expression with ALCs, ES-ECs are a promising potential cell source for regeneration, which are not available in erupted human teeth for regeneration of enamel.

7.Mineralized collagen based composite for bone tissue engineering.

OBJECTIVETo construct a mineralized collagen based composite by biomimetic synthesis for bone tissue engineering.
METHODSUsing the molecular collagen as the template, the calcium phosphate is deposited on it to produce a mineralized collagen based composite, then is combined with minute amount of poly lactic acid (PLA), the three-dimensional scaffold composite is prepared by liquid phase separation. Using osteoblast culture technique, the biocompatibility of this biomaterial in vitro is detected by x-ray diffraction, SEM, TEM, fluoroscopy and CLSM.
RESULTSBoth degree and the size of crystals in the composite are low, which are similar to that of nature bone. It possesses porous structure and the porosity of the composite is high. The typical fibrillar microstructure is self-assembled of the collagen and the nano-crystal hydroxyapatite (HA) in the composite, moreover, the x-ray diffraction graphic of HA crystal shows the [002]-oriented.
CONCLUSIONSThe biomimetic three-dimensional composite can serve as one of the optimal scaffold material for bone tissue engineering both on structure and on property.

9.The assessment of criminal responsibility of schizophrenic rape offenders.

OBJECTIVE:
To explore potential application of Diminished Criminal Responsibility Rating Scale (DCRRS) on rape offenders.
METHODS:
The DCRRS was used retrospectively to study 77 cases of rape offenders. They were divided into three groups according to the degree of diminished criminal responsibility.
RESULTS:
There were significant differences in rating scores among three groups (mild group 21.32+/-5.56, moderate group 25.92+/-5.19, and major group 29.54+/-4.62), and 16 of 18 items showed good correlation with the total scores of the scale (r=0.256 to 0.611), 7 factors were extracted by the factor analysis and their cumulative squared loadings was 75.784%.
CONCLUSION
The DCRRS is valid and reliable in a in assessment of criminal responsibility schizophrenic rape offenders.

10.Relative Expression of Indicators for Wound Age Estimation in Forensic Pathology.

OBJECTIVE:
In order to understand which kind of function genes play an important role for estimating wound age, the variation of difference genes' mRNA expression were compared after injury.
METHODS:
The mRNA expression levels of seven candidate genes (ICAM-1, NF-κB, MX2, MT1, MT2, sTnI, and Cox6c) were analyzed in contused rat skeletal muscle at different time points using real-time fluorescent quantitative PCR (RT-qPCR). The raw Ct values were normalized relative to that of RPL32 mRNA, and converted to standard Ct values. At each time point after injury, the standard deviations (SD) of the standard Ct values were calculated by SPSS.
RESULTS:
The expression trends of the seven genes were all found to be related to wound age, but there were lower variation coefficients and greater reliability of s TnI and Cox6c when compared with other genes.
CONCLUSION
The genes encoding structural proteins or proteins that perform basic functions can be suitable for wound age estimation.
