A simple and sensitive method was developed to screen ?-PL p ro duced strains from soils. 150mg/L K_2Cr_2O_7 was added to th e solid medium for actinomycetes enriching. A basic dye, methylene blue, incorp orated in the agar plate to detect alkali producers, because dye reacted with th e secreted basic polymers by electrostatic interaction with the secreted basic p dymers and formed special zoon. And then dragendorff regent was used to detect alkaloid producers. Four ?-PL producers were obtained by TLC analysis. Meanw hile, phylogenetic analysis of PL6-3 strain, including morphology, physiologica l and biochemical characters and chemotaxomy were performed and phylogenetic tre e was constructed based on the 16S rDNA sequences. And the results indicated th at PL6-3 strain is a member of Kitasatospora.

Objective To study the effect of low-dose FK778 in preventing chronic renal al- lograft rejection in rats. Methods The rat model of chronic renal allograft rejection was established by using micro-surgery technique. The recipients were divided into two groups. The recipients in the study group were treated with FK778 at a dose of 5mg?kg~(-1)?d~(-1)dissolved in carboxymethylcellulose by means of gavage and the controls were treated with carboxymethylcellulose.Urinary protein con- centrations were measured every 4 weeks for 24 weeks. On 24th week after operation, the rats were killed and the kidney grafts were taken out for histological and immunohistological examinations as well as quantitative real-time RT-PCR analysis. Results After 24 weeks of treatment, proteinuria, the severity of chronic rejection, glomeruIosclerosicytes and monocytes/macrophages in the study group were significantly milder than in control group. And the expression of TGF-?mRNA and PDGF-B mRNA was significantly reduced in the study group as compared with that in the control group. Conclusion Low-dose of FK778 might prevent the rats from chronic renal allograft rejection.

BACKGROUND: The permeability of blood-retinal barrier in rats can be increased due to the exposure under infrasound. There is rare research on ionic mechanism of such damage to barrier because of lacking the sources of retinal microvascular endothelial cells.OBJECTIVE: To investigate the impact of infrasound on calcium-activated potassium channel(BKca) of bovine retinal microvascular endothelial cells (BRECs).DESIGN: A completely randomized controlled opening study.SETTING and MATERIALS: The research was conducted in the Laboratory for patch-clamp, Department of Clinical Aerospace Medicine, Fourth Military Medical University of Chinese PLA. Experimental subjects were BRECs cultured.INTERVENTIONS: The cultured BRECs were exposed to the infrasound of 8 Hz, 130 dB for 30 minutes.MAIN OUTCOME MEASURES: The activity of BKCa in BRECs was observed.RESULTS: The activity of BKCa channel in BRECs increased after the exposure of infrasound of 8 Hz, 130 dB for 30 minutes. BRECs were cultured in an incubation cabin for 30 minutes after exposure for another detection by patch-clamp ionic current, and the increased BKCa channel activity slightly decresed.CONCLUSION: The breakdown of blood-retinal barrier under infrasound is partly due to the stimulatory effect of infrasound on Ca2 +-activated-K +channel, which will cause membranous depolarization providing the driving force of Ca2+ influx on BRECs.
