1.Effect of ginseng-sanqi extract on the Ras associated signal proteins.

OBJECTIVETo explore the effect of ginseng-sanqi extract (GSE) on the vascular endothelium growth factor receptor-2 (VEGFR-2), Ras and mitogen-activated protein kinase (MAPK) in VEGFR-2-Ras-MAPK signal pathway of angiogenesis in cultured human umbilical vascular endothelial cells (HUVECs).
METHODSThe protein expressions of VEGFR-2, Ras and MAPK in HUVEC and the effects of GSE (at different doses) and basic fibrin growth factor (bFGF) on them were detected by Western-Blot test.
RESULTSGSE can enhance the expression of angiogenesis signaling proteins (VEGFR-2, Ras, MAPK) to different extents, especially in the groups treated by bFGF or higher dose GSE, the levels showed significant differences to those in the blank group (P<0.05).
CONCLUSIONGSE can promote the expressions of VEGFR-2, Ras and MAPK in the angiogenesis signaling pathway, which may be the foundation of Chinese medicine for tonifying qi and activating blood circulation in promoting endothelial proliferation and angiogenesis.

2.The Screening and Study of Arctic Marine Psychrophilic Bacterial for Anti-tumor and Antibacterial Activity

In this study, 101 strains of bacteria were isolated from arct ic water and sediment samples. The methanol extracts of the fermented broth prod uced by these strains were screened in vitro for anti-tumor activity on mou se tsFT210 cells using the method of flow cytometry, and screened for antibacter ial activity by the method of paper disk diffusion. The result showed that one strain exhibited anti-tumor activity and eight strains had antibacterial activ ity. The stability of the antibacterial components produced by strain AR084 an d its optimum medium were also studied. The research indicated that arctic bac teria had potential application in pharmaceutics.
3.Antitumor screening of deep ocean water and sediment derived fungi and primary investigation of their secondary metabolites

Objective The cytotoxic microbial strains isolated from the deep ocean water and sediments were screened,and the secondary metabolites of bioactive fungus c2b were investi- gated.Methods Active bioactive microbial strains were screened using brine shrimp and chro- nic medulla leucocythemia leukocythemia(K562)cell line.The cytotoxic components of fun- gus c2b were isolated by bioassay-guided fractionation and solvent extraction,silica gel col- umn chromatography and preparative HPLC.Their structures were established by pbysico- chemical properties and spectral analyses.The cytotoxicities of compounds were evaluated by SRB method.Results and Conclusion Twenty-nine strains of fungi were isolated.Among them,seven strains showed cytotoxic activities.Six compounds(1~6)were isolated and i- dentified as N-acetyl histamine(1),chrysogine(2),ergosterol peroxide(3),5,8-epidioxy- 24-methylcholesta-6,22-dien-3?-ol(4)cerevisterol(5)and(4E,8E)-N-[(2'R,3'E)-2'-hy- droxy-3'-hexadecenoyl]-1-O-?-D-glycopyranosyl-9-methyl-4,8-sphingadiene(6),respective- ly.Compound 3 and 4 showed median cytotoxicity.
4.Analysis of Th1/Th2 response pattern for erythrodermic psoriasis.

As one of the most serious types of psoriasis, pathogenesis of erythrodermic psoriasis (EP) is unclear so far. In this study, we aimed to detect the levels of Th1/Th2 cytokine-associated transcription factors and T-lymphocyte clone in peripheral blood mononuclear cells (PBMCs) derived from EP patients, and gene expression level of T-bet/GATA-3 in skin lesion. The potential role of Th1/Th2 reaction pattern played in the pathogenesis of EP was also discussed. Serum levels of IFN-γ, IL-2, IL-4 and IL-10 were quantified by ELISA among 16 EP patients, 20 psoriasis vulgaris (PV) patients and 15 healthy controls. The expression levels of T-bet/GATA-3 in the skin lesion and PBMCs were examined by real-time qPCR. The ratio of Th1/Th2 was measured by flow cytometry. The levels of IFN-γ, IL-2, IL-4 and IL-10 were higher in EP patients than in the healthy controls. The levels of IL-4 and IL-10 were 69.44±11.45 and 12.62±4.57 pg/mL, respectively, in EP patients, significantly higher than those in PV patients and healthy controls (P<0.05). Flow cytometry revealed the levels of both Th1 and Th2 in PBMCs from EP patients were higher than those in healthy controls, and the Th1/Th2 ratio was dramatically lower than in PV patients (P<0.01). The ratios of IFN-γ/IL-4 and T-bet/GATA-3 in EP patients were both less than 1.0, suggesting a reversal when compared with the other two groups. Our study indicated that the EP patients exerted a Th1/Th2 bidirectional response pattern, and the balance of Th cell subsets inclines to Th2, which might be one of the important mechanisms of EP pathogenesis.
5.CT and CTA 3D-reconstruction of keyhole operation approaches in the saddle area

Objective To comprehend the anatomical image data related to the keyhole microsurgery operation approaches in patients with saddle lesions.Methods Fifteen healthy volunteers and 10 patients with intracranial aneurysm were selected in the experiment and 16-slice spiral CT, CT angiography (CTA) and 3D-reconstruction were performed on them. The anatomitical distance and angle were measured through the ways from supraorbital keyhole approach, frontolateral keyhole approach, pterional keyhole approach, and subtemporal keyhole approach to the anterior/posterior clinoid process, the anterior communicating artery and the bifurcation of internal carotid. Results Of the distances between keyhole centre and anterior/posterior clinoid process, the shortest one was the way from subtemporal keyhole approach (4.98±0.54 em; 5.35±0.65 cm) and the longest one was the way from supraorbital keyhole approach (6.44±0.36 cm; 37.68±1.06 cm). Of the angles between the skull sagittal plane and the operation plane from keyhole centre to anterior/posterior clinoid process, the way from subtemporal keyhole approach was better than the way from frontolateral keyhole approach. Of the angles between skull transverse plane and the operation plane from keyhole centre to anterior/posterior clinoid process, that of subtemporal keyhole approach was better than pterional keyhole approach. Distances between keyhole centre and bifurcation of intemal carotid ordered as follows: the way from frontolateral keyhole approach, that from supraorbital keyhole approach, that from subtemporal keyhole approach and that from pterional keyhole approach. Of the angles between cranial sagittal plane and the operation plane from keyhole centre to the anterior communicating artery or the bifurcation of ipsilateral internal carotid,the way of frontolateral keyhole approach was better than that of supraorbital keyhole approach.Conclusion CT and CTA 3D-reconstruction, by demonstrating the distances and angles of each approach,play a significant role in choosing the right keyhole operation approach in different saddle lesions.
6.The study of EEG Higher Order Spectral Analysis technology.

The basic theory of Higher Order Spectral Analysis and the most generally used Bispectrum are introduced in the paper. By certain experiments of EEG signal acquisition and bispectrum analysis, it is showed that the Higher Order Spectrum has an advantage over power spectrum, which is based on Second Order Statistics, in processing non-linear signal and restraining Gauss noise signal.

7.Methods of extracting AEP for monitoring the anesthetic depth.

Many researches have proved that auditory evoked potentials (AEP) can indicate the depth of anesthesia in the clinical practice. This paper introduces two methods of extracting AEP: the moving time average (MTA) model and the autoregressive model with exogenous input (ARX). In our study we used both the MTA model and the ARX model to extract AEP and the results show that using the ARX model can get the results significantly faster than using the MTA model, and the ARX model is more suitable for the real-time monitoring of the anesthetic depth.

8.Changes of morphology and erectile function of the cavernous tissues in spontaneously hypertensive rats.

OBJECTIVETo study the changes of morphology and erectile function of the cavernous tissues in spontaneously hypertensive rats.
METHODSSpontaneously hypertensive male rats (SHR) (n = 15) and normotensive Wistar-Kyoto rats (WKY) (n = 15) were studied for 20 weeks. Systolic blood pressure (SBP) was measured weekly by the tail/cuff method. Erectile function was tested by injecting apomorphine (APO). The expression alpha-smooth muscle actin (alpha-SMA) and collagen III was examined by immunohistochemistry.
RESULTSSHR showed a higher systolic blood pressure (205.7 +/- 11.9 vs 114.3 +/- 10.2 mm Hg) and a lower erection frequency (0.6 +/- 0.5 vs 2.4 +/- 0.6). The expression of alpha-smooth muscle actin and collagen III in the cavernous tissues in the SHR was significantly higher than in the WKY.
CONCLUSIONThe erectile function of the penis is markedly affected by hypertension, and the pathological changes may be one of the most important mechanisms of decreased erectile function in SHR.

9.Effect of L-methionine on trace elements in lead-intoxicated mice.

OBJECTIVETo investigate the effect of L-methionine (L-Met) on the content of Zn, Cu, Mn, Fe in liver, brain, spleen and kidney of lead intoxicated mice.
METHODSDistilled water was given to 10 mice (normal control group) and lead acetate solution of 400 micro g/ml Pb(2+) to 20 mice to serve as drinking water for 10 days. The lead administration was then withdrawn and lead exposed mice were randomly divided into two groups: the lead control group took distilled water as drinking water for 4 weeks to serve as positive control, the other one took L-Met solution (0.5 mg/ml) as drinking water for 4 weeks (Pb + L-Met group) to serve as the treatment group. All the animals were sacrificed on the 1st day after 4 weeks, and the contents of Zn, Cu, Fe, Mn, Pb in liver, brain, spleen and kidney were measured by Inductively Coupled Plasma (ICP) Emission Spectrometry.
RESULTSLead contents in liver, brain, spleen and kidney of Pb control group [(1.490 +/- 1.654) micro g/g, (3.470 +/- 2.757) micro g/g, (4.975 +/- 2.993) micro g/g, (0.066 +/- 0.001) micro g/g respectively], were higher than those in normal control group [(0.015 +/- 0.001) micro g/g, (0.009 +/- 0.007) micro g/g, (0.027 +/- 0.002) micro g/g, (0.006 +/- 0.015) micro g/g, P < 0.05] while Zn contents in liver, brain, spleen and Fe and Mn content in liver, brain, spleen and kidney in Pb control group were lower than those in normal control group (P < 0.05). Pb contents of brain, spleen and Cu content of kidney in Pb + L-Met group were higher than those in normal control group (P < 0.05). Zn contents of liver, brain, spleen, Fe contents of liver, brain, spleen, kidney, and Mn contents of brain, spleen in Pb + L-Met group were lower than those in normal control group (P < 0.05). Fe contents of liver, brain, Zn content of spleen, Cu content of kidney and Mn contents of liver, brain, spleen in the Pb + L-Met group were higher than those in the Pb control group (P < 0.05). The lead levels of four organs in the Pb + L-Met group were lower than those in the Pb control group (P < 0.05).
CONCLUSIONLead could be eliminated by L-Met, which may affect the distribution and metabolism of trace elements in mice.

10.Temporal regulation of transcription factor Mef2c by histone acetylases during cardiogenesis.

OBJECTIVETo observe the temporal modification of transcription factor Mef2c by histone acetylases (HATs) P300, PCAF, and SRC1 during cardiogenesis and to provide a basis for investigating the pathogenesis of congenital heart disease.
METHODSThe normal heart tissues from embryonic mice (embryonic days 14.5 and 16.5) and neonatal mice (postnatal days 0.5 and 7) were collected. The binding of P300, PCAF, and SRC1 to Mef2c gene and level of histone H3 acetylation in the promoter region of Mef2c were evaluated by chromatin immunoprecipitation assays. Meanwhile, real-time PCR was used to measure the mRNA expression of Mef2c.
RESULTSP300, PCAF, SRC1 were involved in histone acetylation in the promoter region of Mef2c during cardiogenesis in mice, and binding of P300, PCAF, and SRC1 to the promoter of Mef2c varied significantly in different stages of cardiogenesis (P<0.01). The level of histone H3 acetylation and mRNA expression of Mef2c in the promoter region of Mef2c also varied significantly in different stages of cardiac development (P<0.01). The levels of acetylated H3, Mef2c mRNA, and HATs (P300, PCAF, SRC1) changed over time. They were highest on embryonic day 14.5 (P<0.01), decreased gradually with cardiac development, and were maintained at low levels after birth.
CONCLUSIONSThe mRNA expression of Mef2c varies during cardiogenesis in mice, which indicates that Mef2c plays an important role in the process of cardiac development. Meanwhile, histone acetylation in the promoter region of Mef2c is regulated temporally by HATs P300, PCAF, and SRC1.
