Bacillus subtilis is a good producer of mannanase.This is the first study to investigate mannanase production under solid state fermentation(SSF) by Bacillus subtilis.Bacillus subtilis WY34 was used for the production of extracellular mannanase in the SSF of wheat bran.The highest mannanase activity was 7,650 U/g dry matter when the following conditions were used: wheat bran 5g,initial moisture content 71%,Tween-80 1%,konjac powder 0.4 g,initial pH 7.0,incubation temperature 50 ℃.Enzyme production was increased 178% after optimization.
2. The association between breast carcinoma and columnar alteration with prominent apical snouts and secretions

Objective: To study the pathological characteristics of columnar alteration with prominent snouts and secretions (CAPSS) and its relationship with incidence of breast carcinoma. Methods: One hundred forty-four CAPSS specimens identified by breast core needle biopsies(CNB) were retrospectively evaluated. The CAPSS specimens were subdivided into lesions with (n=108) and without (n=36) atypical ductal hyperplasia and their pathological findings were compared. Results: In situ and invasive carcinomas were found in 21.5% (31/144) of the specimens, including those with and without atypical hyperplasia. We also found that specimens with atypical hyperplasia had a higher malignant incidence than those without atypical hyperplasia (P=0.001). Conclusion: CAPSS, especially when with atypical ductal hyperplasia, are associated with breast carcinoma.

Bioreactor is a key equipment in bioengineering.The bioprocessing efficiency using bioreactors is significantly affected by the reactor configuration.It is apparent that research on the bioreactor structure is one of key issues in bioengineering.Along with industrialization of penicillin mechanically stirred bioreactor was developed,after that large quantity of bioreactors was invented for the sake of the development of mammal and plant cell cultivation,epiphyte cultivation and algae cultivation.Among these bioreactors mechanical stirred bioreactors and airlift bioreactors are very popular.Recent trends of studies on mechanically stirred bioreactors and airlift ones were reviewed.The focus is on the summarization and analysis of 11 kinds of novel bioreactors developed around the world.

5.Clinical effects of gabapentin on the treatment of pruritus of scar resulting from deep partial-thickness burn.

OBJECTIVETo study the clinical effects of gabapentin on the treatment of pruritus of scar resulting from deep partial-thickness burn.
METHODSA total of fifty-eight patients suffering from pruritus of scar after deep partial-thickness burn were hospitalized from January 2013 to January 2014. Patients were divided into placebo group (n =18, treated with oral vitamin C in the dose of 100 mg for 4 weeks, twice per day) , cetirizine group (n = 20, treated with oral cetirizine in the dose of 10 mg for 4 weeks, twice per day) , and gabapentin group (n = 20, treated with oral gabapentin in the dose of 300 mg for 4 weeks, twice per day) . Before treatment and on post treatment day (PTD) 3 and 28, the Visual Analog Scale (VAS) was used to assess the itching degree, and the mean scores were recorded. The remission rates of pruritus on PTD 3 and 28 were calculated. The adverse effects were observed during treatment. Data were processed with analysis of variance, q test, and chi-square test.
RESULTSCompared with that before treatment, the itching degree of patients with light, moderate, and severe itching in placebo group was not relieved after treatment; the itching degree of patients with moderate or severe itching in cetirizine group was alleviated after treatment, but not in patients with light itching; itching degree of all patients in gabapentin group was significantly relieved after treatment. There were no obvious differences in VAS scores among the 3 groups before treatment (F = 2.78, P > 0.05). On PTD 3 and 28, the VAS scores of patients in both gabapentin group [(2.3 ± 0.8) and (0.6 ± 0.3) points] and cetirizine group [(4.2 ± 1.7) and (2.8 ± 1.2) points] were lower than those in placebo group [(5.7 ± 2.0) and (5.7 ± 1.9) points, with q values from 6.70 to 7.75, P values below 0.05]. The VAS scores of patients in gabapentin group on PTD 3 and 28 were lower than those in cetirizine group (with q values respectively 6.30 and 6.90, P values below 0.05). The remission rates of pruritus of patients in gabapentin group on PTD 3 and 28 were respectively (66 ± 20)% and (91 ± 17)%, and they were higher than those in cetirizine group [(33 ± 8)% and (56 ± 14)%, with q values respectively 4.70 and 3.82, P values below 0.05]. The remission rate of pruritus of patients in placebo group on PTD 3 and 28 was 0, which was lower than that of the other 2 groups each (with q values from 3.94 to 6.76, P values below 0.05). During the course of treatment, 5 patients in gabapentin group suffered from adverse effects including mild-to-moderate drowsiness and dizziness, but they disappeared one week later. No adverse effects were observed in patients of the other two groups.
CONCLUSIONSFor patients with deep partial-thickness burn, gabapentin can effectively alleviate scar itching after wound healing with safety.

7.Effect of umbilical cord mesenchymal stem cell transplantation on cognitive function in endotoxemic rats

Objective To evaluate the effect of umbilical cord mesenchymal stem cell (UC-MSC) transplantation on cognitive function in endotoxemic rats.Methods Seventy-two pathogen-free male Wistar rats,aged 180-250 g,were randomly divided into 4 groups (n=18 each) using a random number table:control group (group C),endotoxemia group (group E),UC-MSC group (group M),and endotoxemia+ MSC group (group PM).Cognitive function was assessed using Morris water maze test.At 4 h before training on 2nd day of place navigation test,lipopolysaccharide 100 μg/kg was injected intraperitoneally in group E,the equal volume of normal saline was given in group C,UC-MSC suspension 300 μl (5× 105cells,the fluid was normal saline) was injected via the tail vein immediately after intraperitoneal lipopolysaccharide in group PM,and UC-MSCs were injected via the tail vein in group M.At 4 h after injection,Morris water maze test was continued.At 4,24 h and 5 days after intraperitoneal injection or injection via the tail vein (T1-3),6 rats were selected,and blood samples were collected from hearts.The rats were then sacrificed,and the hippocampus was removed.The levels of interleukin-lbeta (IL-1β),IL-6 and tumor necrosis factor-alpha (TNF-α) in serum and hippocampus were determined.Results Compared with group C,the escape latency was significantly prolonged on 2nd and 3rd days of Morris water maze test,the number of crossing the platform was significantly decreased,the percentage of swimming distance in 1 st quadrant in the total swimming distance was significantly decreased,and the levels of IL-1 β,IL-6 and TNF-α in serum and hippocampus were significantly increased at T1,2 in group E (P<0.05).Compared with group E,the escape latency was significantly shortened on 2nd and 3rd days of Morris water maze test,the number of crossing the platform was significantly increased,the percentage of swimming distance in 1st quadrant in the total swimming distance was significantly increased,and the levels of IL-1β,IL-6 and TNF-α in serum and hippocampus were significantly decreased at T1,2 in group PM (P<0.05).Conclusion UC-MSC transplantation can improve the cognitive decline in endotoxemic rats.
8.Effect of ulinastatin on postoperative delirium in elderly patients receiving hip fracture surgery

Objective To explore the effect of ulinastatin treatment on postoperative delirium (POD) in elderly patients receiving hip fracture surgery.Methods Ninety-six elderly patients (38 males,58 females,aged 70-93 years,ASA grade Ⅱ or Ⅲ) undergoing elective hip fracture surgery were randomly divided into two groups using a random number table: ulinastatin group (group U) and control group (group C),48 cases in each group.After spinal anesthesia and fascia iliaca compartment block,ulinastatin 5 000 U/kg diluted with normal saline to a volume of 50 ml (group U) was administered intravenously over 10 min before surgical incision and the equal doses on post-operative days 1,2.The equal volume of normal saline was administered intravenously in group C at the same time.POD was assessed by using the Confusion Assessment Method (CAM) on post-operative days 1-3.Serum samples were collected to measure the levels of IL-6,IL-10 and S100β before the anesthesia (T0),at the end of surgery (T1) and three days after surgery (T2) by ELISA.Results The incidence of POD in group U was significantly lower than that in group C (4.3% vs.28.2%) (P<0.05).Compared with T0,the levels of serum IL-6 and IL-10 in group C at both T1 and T2 significantly increased (P<0.05).Compared with group C,serum IL-6 levels in group U decreased at both T1 and T2 (P<0.05).Compared with T0,the levels of serum S100β in group C at T1 significantly increased (P<0.05).Compared with group C,ulinastatin significantly inhibited the release of serum S100β at T1 (P<0.05).Conclusion Ulinastatin can significantly reduce the incidence of POD in elderly patients undergoing hip fracture surgery.The mechanism may involve inhibition of IL-6 and S100β in serum.

10.Evaluation of levetiracetam on the gray matter structure remodeling in benign epilepsy children with-central temporal spikes

Objective At present, there is no study on effect of levetiracetam(LEV) on the gray matter structure remodeling in benign epilepsy children with central temporal spikes(BECTS).The purpose of this study was to study the influence of LEV on the gray matter structure in BECTS and to evaluate the mechanism of LEV on the brain structure of BECTS through using voxel-based MRI morphological(VBM) methods.Methods From January 2014 to September 2016, twenty-four BECTS treated with LEV(LEV group), twenty-four drug-na?ve BECTS(untreated group) and twenty-four normal children(normal group) consulted in department of Neurology, Nanjing Children′s Hospital and the Nanjing Military Region, Nanjing General Hospital were continuously included to receive three-dimensional T1-weighted imaging with 3T MRI and the gray matter volume was calculated by VBM.We compared the difference of grey matter volumes of the three groups and analyzed their correlation with epilepsy duration, age of onset and medication time and other clinical index.Results Compared with the normal group, the grey matter volume of bilateral thalamus were decreased, and the volume of bilateral Rolandic areas, anterior insula/frontal operculum/frontal triangle, left supplementary motor area, paracentral lobule, precentral gyrus, superior frontal gyrus and right middle frontal gyrus were increased in the untreated group, but the grey matter volume of the bilateral Rolandic areas, frontal operculum and left supplementary motor area were decreased in the LEV group.Compared with the untreated group, the grey matter volume of bilateral supplementary motor, left paracentral lobule, precentral gyrus, bilateral anterior insula/frontal operculum/frontal triangle, left superior frontal gyrus and right middle frontal gyrus in the LEV group were decreased.The grey matter volume of left anterior insula/frontal operculum areas was negatively correlated with the medication time in LEV group(r=-0.527, P<0.01).Conclusion T The mainly representations of BECTS are thalamic gray matter damage and epileptic-related cortical area irritation structural abnormalities, but the LEV could reshape the epilepsy-related cortical area and the gray matter in the brain area associated with clinical symptoms.
