1.Research progress of fluorometholone on dry eye

?Dry eye is a common eye disease around the world, which has highly incidence. Although there are many treatment measures for dry eye, the clinical effect is not satisfactory. There is a close relationship between inflammation and dry eye, so the researchers pay more attention on anti -inflammatory therapy, such as fluorometholone. To explore the effect of anti -inflammatory medicine on dry eye, put forward the problems and shortcomings as well, lay the foundation for further study, and provide a strong theoretical basis for the clinical use of anti-inflammatory drugs in the treatment of dry eye, the author summarizes the clinical and experimental research progress of fluorometholone on dry eye in China and abroad.

4.Treatment with Chinese botulinum toxin type A in 16 cases of masticatory spasm

Objective To study the therapeutic efficacy of Chinese botulinum toxin type A (CBTX- A) in masticatory spasm patients.Methods 16 patients with masticatory spasm were treated with CBTX-A local injection.12 patients showed jaw clench with masseter and temporalis affected (type Ⅰ).Four showed jaw clench and deviation to one side with pterygoid muscle affected unilaterally or bilaterally (type Ⅱ).All patients showed paroxysmal clenched jaw with difficulty in opening their mouths.There were no other clinical manifestations.CT and MRI did not reveal any intracerebral abnormalities.The efficacy and adverse effects were observed.Results CBTX-A were injected into 16 patients,resulting in a significant improvement of symptoms in 13 cases (4 cases of unilateral type Ⅰ,7 of 8 cases bilateral type Ⅰ,2 of 4 type Ⅱ).The spasms ceased within 3-10 days after the injection,and the effects lasted for 8-26 weeks.Four patients were observed to have slight masseter weakness after the injections,which recovered within a few weeks.The benefit persisted after identical repeated injection.Conclusion CBTX-A injection is an effective and safe treatment for masticatory spasm.
5.Effects of Salinity and pH on the Growth and Active Products-secreting of Aspergillus sp. F3 from the Mangrove Rhizosphere

An Aspergillus sp. strain F3 was isolated and identified from the rhizosphere soil of mangrove plant, Rhizophora stylosa Griff in Dongzhai harbor mangrove forest conservation in China. In this study, the effects of media salinity and pH on the mycelial biomass and the ability of producing antibacterial metabo- lites from this isolate were carefully analyzed. Results showed that this isolate can grow well on the SDA medium with higher salinity (3%~9%) and higher pH (8~10). Under the modified culturing conditions, this isolate can secret the antibacterial and antitumor metabolites. The extracts of acetic ether were about 448 mg/L of the fermentation broth. The antibacterial activities of the acetic ether extract were analyzed with bacteria and fungus. Results showed this extract can suppress the growth of Staphylococcus aureus、S. epi-dermidis、Sarcina lutea、Bacillus subtilis and Escherichia coli with MIC of 31.3 ?g/mL, 31.3 ?g/mL, 7.8 ?g/mL, 7.8 ?g/mL and 125.0 ?g/mL, respectively. It can also suppress the growth of Candida albicans with MIC of 125.0 ?g/mL. Further studies uncovered the cytotoxicity of this extract against the tumor cells, such as ECV304, Lovo and HepG2 with IC50 of 3.45 ?g/mL, 4.88 ?g/mL and 14.31 ?g/mL respectively.
6.The reverse flow island flap nourished by sural nerve nutrition blood vessel for repair of severe frostbite of feet.

OBJECTIVEThe study was to verify the feasibility of an improved method using reverse flow island flap nourished by the sural nerve nutrition vessel to repair severe frostbite of feet.
METHODSAt the proximal end of the principal flap, an auxiliary triangular skin flap of 6.5 - 7 cm in length was designed in order to cover the pedicle of the principal flap. This operation was performed on 13 patients (21 feet) with frostbite.
RESULTSAll the flaps survived well. Postoperative follow-up for 5 - 18 months demonstrated satisfactory results in all the cases. No ulcer happened.
CONCLUSIONSThe method is helpful to prevent constriction of the pedicle and ensure blood supply of the flap. It is an ideal treatment for severe frostbite of feet.

7.Application of FLOW-PRA detection in clinical organ transplantation

Objective To apply flow cytometry-panel reactive antibody (FLOW-PRA) and compare the application of traditional (enzyme-linked imrnunosorbent assay) ELISA-PRA in clinical organ transplantarion,so as to evaluate the concordance,sensitivity,accuracy and practicability of FLOW-PRA.Methods PRA was detected in 212 serum samples from 185 patients awaiting organ transplantation using FLOW-PRA and ELISA-PRA.Results It took 1.5 h and 3 h for FLOW-PRA vs ELISA-PRA.Concordance correlation coefficient for the results of the two methods was 94% (class Ⅰ)and 89% (class Ⅱ),respectively.Of all sera,24.5% (in comparison to ELISA-PRA,P<0.005)were class I positive,18.4% (P<0.05) class Ⅱ positive by flow cytometry,and 17.9% and 14.6% by ELISA,respectively.The positive incidence in Flow group was higher than in ELISA group.Low titer of antibodies was detected positively only by flow eytometry,furthermore,the antigen specificity of PRA could only be discriminated by FLOW-PRA.Conclusion Flow cytometry is more sensitive and more accurate than ELISA in PRA detection.FLOW-PRA is easy to operate and time-effective,and suitable for clinical application.
8.Mild cognitive impairment in elderly hypertensive patients:a retrospective case series study

Objective To investigate the relationship between hypertension,other vascular risk factors and mild cognitive impairment and its subtype.Methods A total of 297 outpatients and inpatients were collected from the Departments of Neurology in 4 municipal hospitals in Qingdao from April 2011 to September 2011.The unified questionnaires of cognitive impairment status in the departments of neurology in Qingdao city were developed.The risk factors for mild cognitive impairment and its subtype were investigated.Results Univariate analysis showed that low levels of education (odds ratio [ OR] 0.239,95% confidence interval [ CI] 0.129 -0.442; P =0.000),hypertension (OR 1.928,95% CI 1.107 - 3.358; P =0.019) and hyperlipidemia (OR 1.812,95% CI 1.041 -3.155; P =0.034) were all the risk factors for mild cognitive impairment; Multivariate logistic regression analysis showed that low levels of education (OR 0.807,95% CI 0.742 - 0.878; P =0.000) and hypertension (OR 1.788,95% CI 1.004 -3.146; P =0.048 ) were the independent risk factors for mild cognitive impairment; and hypertension (OR 2.091,95% CI 1.030 -4.242; P=0.041) was an independent risk factor for non-amnestic mild cognitive impairment,and was mainly impaired visuospatial and executive abilities (P =0.026).Conclusions Hypertension is an independent risk factor for mild cognitive impairment and its subtype-non-amnestic mild cognitive impairment,and it mainly impairs executive ability.
9.Effect of Sox2 on invasion and migration of cervical cancer via Wnt signaling pathway

ABSTARCT:Objective To investigate the effects of the transcription factor SRY-related high-mobility-group box 2 (Sox2)related to stem cells on the invasion and migration of cervical squamous carcinoma cell line SiHa and its mechanism.Methods The expression of Sox2 was detected in Sox2 stably over-expressed cell line SiHa-Sox2 and negative control SiHa-EGFP cells by RT-PCR and Western blotting,respectively.The effects of Sox2 on the invasion and migration capacities of SiHa cells were detected by wound-healing assay and transwell assay.The expression of β-catenin was detected by Western blot.Results Compared with that of SiHa-EGFP cells,the expression of Sox2 at both mRNA and protein levels was obviously upregulated in SiHa-Sox2 cells.The migration and invasion capacities of SiHa-Sox2 cells were increased significantly (P <0.01 ),and the expression of β-catenin increased dramatically compared with that of the control cells.Conclusion Sox2 promotes the invasion and migration capacities of SiHa cells by increasing the expression of β-catenin and activating Wnt signaling pathway, which contributes to the development of cervical cancer.
10.Comparative study of computer tomography presentations and pathological results of chronic intestinal schistosomiasis

ObjectiveTo retrospectively analyze the abdominal computer tomography (CT)findings and pathological results of chronic intestinal schistosomiasis in order to improve the diagnostic accuracy of the disease.MethodsThe plain plus enhanced CT scanning was performed in 56 cases (male 37,female 19; mean age 69.2 years).All cases confirmed by pathological examination.The location,shape,size,calcification and pattern of enhancement of lesion were analyzed by two radiological physicians independently.ResultsThe colon wall of all 56 cases was presented with thickening (0.3-1.2 cm) by CT scanning,among which,43 (76.8%) were presented with welldistributed thickening.The calcification of colon wall included linear calcification (n=42),tram-like calcification (n=19) and spot calcification (n=8).The margin of calcification was clear.The locations of calcification in colon were as follows:whole colon (n =5 ),ascending colon (n =9 ),transverse colon (n=12),descending colon (n =24),rectosigmoid (n=34) and rectum (n=30).There were 17 cases presented with severe calcification.The pathological examinations confirmed that linear and tram-like calcification resulted from calcified ova deposited in submucous,subserosa. In 8 cases with spot calcification and 17 with severe calcification,there were calcified ova deposited in all layers of colon wall,accompanied by chronic inflammation,polyp and schistosomiasis granuloma.There were 5 cases complicated by adenocarcinoma.ConclusionsCT scanning is an important imaging method in the diagnosis of chronic intestinal schistosomiasis,with the distinguishing presentation of well-distributed thickening and calcification in the colon wall.When irregular thickening,mass or nodular are found in the colon wall of patients with chronic intestinal schistosomiasis,colorectal carcinoma should be highly suspected.
