1. Role of p38MAPK in regulating expression of NF-κB and COX-2 in rat mesangial cell

Objective: To investigate the relationship between p38 mitogen-activated protein kinase (p38MAPK), nuclear factor-κB (NE-κB) and cyclooxygenase-2 (COX-2), so as to study the roles of p38MAPK, NF-κB nd COX-2 in diabetic nephropathy. Methods: Rat mesangial cell line HBZY-1 was incubated with certain concentrations of glucose(25 mmol/L), insulin (100 mmol/L), H2O2 (100 mmol/L) and glycation endproducts (AGEs) (100 mg/L) with or without pretreatment with SB203580, a specific inhibitor of p38MAPK, then the expression of p38MAPK, NF-κB and COX-2 in the HBZY-1 cells was examined. Results: High glucose, high insulin, H2O2 and AGEs independently activated p38MAPK, increased p38MAPK phosphorylation, and significantly up-regulated the expression of NF-κB and COX-2 compared with the control group (P<0.01). Pretreatment with SB203580 significantly decreased expression of NF-κB and COX-2 compared with the corresponding stimulating group (P<0.01). Conclusion: p38MAPK may induce renal damage during diabetic nephropathy by activating NF-κB and COX-2, indicating that p38MAPK, NF-κB and COX-2 may play important roles in the development of diabetic nephropathy.
2. Influence of sodium selenite on expression of p38 mitogen-activated protein kinase and peroxisome proliferator-activated receptor γ in rat mesangial cells line HBZY-1

Objective: To observe the influences of sodium selenite on expression of p38 mitogen-activated protein kinase (p38MAPK) and peroxisome proliferator-activated receptor γ (PPARγ) in rat mesangial cells line HBZY-1, so as to study the role of p38MAPK and PPARγ in diabetic nephropathy and the mechanism by which sodium selenite prevents diabetic nephropathy. Methods: Rat mesangial cell line HBZY-1 was incubated with high glucose, high insulin, H2O2 and advanced glycosylation end products (AGEs) separately before and after HBZY-1 cells were pre-treated with SB203580 (p38MAPK special inhibitor)or sodium selenite. Cells receiving no stimulation were taken as control. The expression of p38MAPK protein and PPARγ mRNA was detected respectively by immunohistochemistry assay and RT-PCR in all groups and the results were compared. Results: High gluco se, high insulin, H2O2 and AGEs all activated p38MAPK, increased phospho-p38MAPK expression and decreased the expression of PPARγ mRNA in rat mesangial cells line HBZY-1. The expressions of phospho-p38MAPK protein was markedly inhibited by sodium selenite, while the expression of PPARγ mRNA was significantly increased by SB203580 or sodium selenite in rat mesangial cells lines HBZY-1(P<0.01). Conclusion: p38MAPK may antagonize the expression of PP ARγ in rat mesangial cells lines HBZY-1. Sodium selenite, with a similar effect to the agonist of PPARγ, can obviously increase the expression of PPARγ.

4.The relationship between Fc? receptor ⅢB genotype and severe periodontitis in Han Nationality population

Objective:To study the relationship between Fc? receptor ⅢB(Fc? RⅢB) genotype and severe chronic periodontitis or generalized aggressive periodontitis.Methods:DNA from 35 subjects with severe chronic periodontitis,25 with generalized aggressive periodontitis and 95 of periodontal healthy controls were sampled by buccal swab and extracted using Chlex-100 method.Polymerase chain reaction with sequence specific primers (PCR-SSP)was carried out to determine the Fc?RIIIB genotype.The data were analyzed with ? 2 test. Results: NA1/NA2 heterozygote was the predominant genotype in the 3 groups.No statistically differences of Fc?RIIIB genotype distribution were found between any two of the stuied groups.The gene frequencies of NA1 and NA2 were 0.6 and 0.4 respectively.Conclusion:No direct relationship exists between Fc?RⅢB genotype and the occurrence of severe chronic periodontitis and generalized aggressive periodontitis according to the present study.
5.Effect of rAAV2-TGFβ3 on collagen degradation of liver fibrosis in rats

Objective To investigate effect of transforming growth factor β3(TGFβ3)on expression of MMP-9,MMP-2,TIMP-1 and collagen I in rats with liver fibrosis through transduction by recombinant adeno-associated virus 2(rAAV2).Methods Rats were randomly divided into 4 groups:normal control group,model group,negative control group and TGFβ3 group.Liver fibrosis model was induced by hypodermic injection of 40% CCl4.rAAV2-TGFβ3,was injected via vena caudalis of the rats one week before CCl4 Was given.All rats were sacrificed and the liver tissues were taken 8 weeks afler injection of CCl4.The histopathological changes were observed on HE sections;the expressions of MMP-9,MMP-2,TIMP-1 and collagen I were examined by histochemistry and the positive area rates were semi-quantitatively analyzed.Results Compared with the model group and negative control group,the decreases of inflammatory infiltration and collagen fibers hyperplasia were observed in the TGFβ3 group,and the expression of MMP-9 increased(q=23.664,27.746,P<0.01),the expression of collagen I(q=5.503,5.251,P<0.01)and TIMP-1(q=5.800,8.608,P<0.01)decreased,but that of MMP-2 was not changed(q=2.1 08,0.996,P>0.05).Conclusion rAAV2-TGFβ3 can reduce the histological damage and degree of liver fibrosis in rats by inhibiting expression of TIMP-1 and collagen I,and upregulating expression of MMP-9.
6.Diagnostic values of serum free fatty acids and urine podocalyxin in the patients with type 2 diabetic nephropathy

Objective To investigate the diagnostic values of serum free fatty acids (FFA) and urine podocalyxin (PCX) in the patients with type 2 diabetic nephropathy(DN).Methods A total of 120 patients with type 2 diabetes mellitus (DM) were enrolled,and they were divided into three groups,normal albumin group(NA),microalbuminuria group(MA) and heavy albuminuria group(HA),based on the urinary albumin-creatinine ratio(UACR).In addition,40 healthy persons participated in medical examination were selected as the control group.Serum FFA levels were detected by a biochemical analyzer and urine PCX levels by ELISA.The correlation between them was analyzed by Pearson correlation,and their diagnostic values in type 2 DN were evaluated by the receiver operating characteristic curve (ROC).Results Serum FFA levels in the NA,MA,HA and control groups were (0.61 ± 0.14),(0.81 ± 0.13),(0.95 ± 0.18) and (0.49 ± 0.11) mmol/L,respectively,and urine PCX levels were (1.86 ± 0.45),(4.47 ± 1.48),(6.72 ± 1.40)and(1.38 ±0.24) ng/mL,respectively.The levels of serum FFA and urine PCX in type 2 DM patients were significantly higher than those in the control group(P < 0.05),and increased with the progression of type 2 DM.Pearson correlation analysis showed that serum FFA levels were positively correlated with urine PCX levels(r =0.73,P < 0.05).The sensitivity of serum FFA,urine PCX and combined detection in the diagnosis of type 2 DN were 74.1%,80.6% and 89.5%,respectively,and the latter significantly higher than the former two (P < 0.05).Conclusion Serum FFA and urine PCX levels increase significantly in diabetic patients with renal injury,and both of them have important clinical values in the diagnosis of type 2 DN.
7.Study on Immunoregulation Effect of Exopolysaccharide Produced by Bifidobacterium spp

The immunoregulation effect to polysaccharides from Bifidobacterium spp. was investigated on the base of functional assessment standards of health food. Effects of the EPS on immunity were investigated by promoted the proliferation of spleen lymph cells, delayed type hypersensitivity reaction, the HC50 value and macrophage function assay in mice. Data showed that the EPS could obviously increase the ratio of swallowed chicken red blood cell by macrophage and the HC50 value in mice. However, no significant effect was found on the delayed type hypersensitive induced by sheep red blood cell, for only the low dose of 100 mg/(kg?d) EPS promoted the proliferation of spleen lymph cells. Bifidobacterium spp. EPS can certain immunomodulating function.
8.Effects of Couplet Medicines (Astragalus Membranaceus and Jiaozhen) on Intestinal Barrier in Postoperative Colorectal Cancer Patients.

OBJECTIVETo observe the effect of the couplet medicines (Astragalus Membranaceus and Jiaozhen) on intestinal barrier functions of postoperative colorectal cancer patients.
METHODSTotally 90 inpatients with confirmed colorectal cancer by pathological diagnosis were recruited as subjects in this study. They were assigned to the Chinese medicine group (CM, treated with Astragalus Membranaceus and Jiaozhen), the Western medicine group (WM, treated with glutamine), and the blank control group (treated with normal saline) according to random digit table, 30 in each group. The treatment course consisted of eight days. Levels of blood D-lactic acid, diamine oxidase (DAO), urinary lactulose/mannitol ratio (L/M), ET, TNF-alpha, and postoperative recovery time of bowel sound were observed before surgery and after surgery. The effect of the couplet medicines (Astragalus Membranaceus and Jiaozhen) on intestinal barrier functions of postoperative colorectal cancer patients were comprehensively assessed by taking blood D-lactic acid levels, DAO levels, urinary L/M as main potency indices; ET and TNF-alpha, recovery time of bowel sound as the secondary potency indices.
RESULTSCM showed similar effect with that of WM in improving blood D-lactic acid levels and DAO levels, and urinary L/M ratio, with no statistical difference between them (P > 0.05). But they showed better effect than that of the blank control group (P < 0.05). Levels of ET and TNF-alpha were decreased more in the CM group than in the WM group (P < 0.05). The recovery time of bowel sound was shorter in the CM group than in the WM group (P < 0.05, P < 0.01). Levels of ET and TNF-alpha were decreased more in the WM group than in the blank control group (P < 0.05). There was no statistical difference in the recovery time of bowel sound between the WM group and the blank control group (P > 0.05).
CONCLUSIONSThe couplet medicines (Astragalus Membranaceus and Jiaozhen) had obvious protection for intestinal barrier dysfunction of postoperative colorectal cancer patients, showing similar efficacy to that of WM. It was even superior to glutamine in restoring bowel functions, reducing toxin absorption, and lowering levels of pro-inflammatory factors.

9.Effects of nandrolone phenylpropionate on ultrastructure of aorta in rats with or without movement training

Objective To observe the influence of nandrolone phenylpropionate(NP) on the ultrastructure of aorta in rats with or without movement training,and investigate the side effects of NP on the cardiovascular system. Methods Forty male SD rats were randomly divided into sedentary control group,sedentary+medicine group,exercise control group and exercise+medicine group.For the groups with medical treatment,NP of 10 mg/kg one time every three days was injected into the rats via gluteus for eight weeks.For the exercise groups,rats were trained to run on treadmill five days per week for eight weeks.The aortae were sampled and specimens were obtained for transmission electron microscopy. Results The ultrastructure of aorta was normal in sedentary control group.For sedentary+medicine group,mitochondrial swelling,vacuolated cytoplasm and lysis of endothelial cells were observed,disruption of intercellular conjunctions,widening of subendothelial spaces and furcation and breakage of internal elastic lamina were found,and smooth muscle cells changed into synthesis type.For exercise control group,no obvious morphologic change was observed,except that part of the internal elastic lamina disrupted.In exercise+medicine group,breakage and lysis of endothelial cells were observed,widening of subendothelial spaces and lysis of internal elastic lamina were found,and autophagosome and myelinoid body were seen in smooth muscle cells. Conclusion NP may lead to the impairment of endothelial cells and the change of smooth muscle cells into synthesis type.Exercise with NP administration may result in more severe impairment in vessel wall.
10.Effects of hyperbaric oxygen on the expression of hypoxia inducible factor-1α mRNA and vascular endothelial growth factor mRNA in colon adenocarcinoma cells

Objective To investigate the effects of hyperbaric oxygen (HBO) on the expression of hypoxia inducible factor-1α (HIF-1α) mRNA and vascular endothelial growth factor (VEGF) mRNA in colon adenocarcinoma cells. Methods SW480 colon cancer cells were divided into a control group and an HBO group, which was in turn divided into various subgroups according to the various HBO pressures (0.15 MPa, O. 20 MPa, 0.25 MPa) and exposure times (12, 24 and 36 h) tested. The expression of HIF-1α and VEGF mRNA was evaluated with RT-PCR after the various treatments. Results The expression of HIF-1α mRNA was lower in the SW480 cells than in the controls after exposure to 0.25 MPa HBO for 12 h. No expression of HIF-1α mRNA was detected after exposure to 0.25 MPa HBO for 24 h. The expression of VEGF mRNA was lower after exposure to 0.25 MPa HBO for 36 h than in the controls. Conclusion Absence of HIF-1α mRNA expression and decreased expression of VEGF mRNA in SW480 can be observed after exposure to 0.25 MPa HBO for 24 h, suggesting that HBO may inhibit the formation of tumor blood vessels through down-regulating the expression of these mRNAs.
