1.Anticancer Effects of Xiaotansanjie Recipe on Microsatellite Instability in MKN-45 Human Gastric Cancer Orthotopic Transplantation Tumor

Objective To explore the mechanism of Xiaotansanjie recipe on MKN-45 human gastric cancer orthotopic transplantation tumor with microsatellite instability. Methods MKN-45 human gastric cancer cell line was used to establish subcutaneous tumor model in nude mice as an experimental model which passed three generations of tumor source. Then OB glue was used to establish human gastric cancer orthotopic xenograft model. Twenty model nude were divided into model group and Xiaotansanjie recipe group (treated with Xiaotansanjie recipe). After 6 weeks of treatment intervention, tumor weight was measured, inhibitory rate was calculated and microsatellite instability (D17S250, D2S123, D5S346, Bat-25 and Bat-26) was detected. Results The tumor weight in Xiaotansanjie recipe group was significantly lower than that in model group (P<0.01). The tumor-inhibition rate was 40.84%. Five microsatellite markers (D17S250, D2S123, D5S346, Bat-25 and Bat-26) were amplified in the tumor tissue of model group, and microsatellite markers D2S123, D5S346 appeared some prolong and reduction. Microsatellite markers (D2S123, D5S346) mutation was stabilized after 6 weeks treatment of Xiaotansanjie recipe. Conclusion Xiaotansanjie recipe can inhibit the MKN-45 human gastric cancer orthotopic transplantation tumor by down-regulating the microsatellite instability.

Objective To evaluate the effect of ImmunoCyt on the early diagnosis of bladder cancer by comparing the result of cystoscopy with that of cytology. Methods Eighty-six consecutive patients with bladder cancer (mean age,63 years) were included in this group.Among them 38 had undergone transurethral resection of superficial transitional cell carcinoma and had been followed up and 48 had hematuria.These patients were evaluated with standard cytology and ImmunoCyt test.Then all the patients underwent cystoscopy and suspected lesions were biopsied. Objective To observe the effects ofXiaotan Sanjie Decoction on ultramicrostructure in MKN-45 human gastric cancer orthotopic xenograft;To discuss its mechanism for gastric cancer. Methods MKN-45 human gastric cancer cell line was used to establish subcutaneous tumor model in nude mice as an experimental model which passed three generations of tumor source. Then OB glue was used to establish human gastric cancer orthotopic xenograft model. 40 model nude mice were divided into control group, model group (received gavage with saline),Xiaotan Sanjie Decoction group (received garage withXiaotan Sanjie Decoction) and Tegafur group (received gavage with Tegafur), 10 mice in each group. Mice in the control group were under normal diet, without medicine intervention. After 6 weeks of treatment intervention, tumor weight was detected, inhibitory rate was calculated, the ultrastructural changes of gastric cancer cells were detected through transmission electron microscopy.Results The tumor weight inXiaotan Sanjie Decoction group was significantly lower than that in control group and model group (P<0.05,P<0.01). The tumor inhibition rate was 46.2%. Under the electron microscope, MKN-45 gastric cancer cells strain changed to typical apoptosis.ConclusionXiaotan Sanjie Decoction can lead to apoptosis effects on MKN-45 human gastric cancer orthotopic xenografts. All evidences indicate that inducing apoptosis may be one of the most important mechanisms ofXiaotan Sanjie Decoction in treating gastric cancer.
4.In Vitro Study on Morusin Regulating Cell Cycle Arrest and Inhibiting Cell Proliferation of ;HCT116 Cells in Colorectal Cancer

Objective To investigate the effects of Morusin on cell line proliferation, cell cycle and cell apoptosis of colorectal cancer HCT116 cell;To discuss its mechanism. Methods HCT116 cells were treated with different concentrations of Morusin for 72 h. Cell proliferation was detected by CCK-8 assay, and cell growth inhibition rate and IC50 value were calculated. HCT116 cells were treated with 25.4, 50.8 μmol/L Morusin for 24 h, 48 h and 72 h. Cell morphology was observed under the microscope, and cell proliferation was detected by CCK-8 assay. After HCT116 cell line was treated with 25.4μmol/L Morusin for 24 h, cell cycle phase distribution was detected by flow cytometry and the cell apoptosis was detected by TUNEL assay under the fluorescence microscope. Post-intervention protein expressions were detected by Western Blot. Results The inhibitory effects of Morusin on the proliferation of HCT116 cell line was concentration/time dependent and IC50 value at 72 h was 25.4μmol/L;Morusin induced the cell cycle arrest at G0/G1 phase and G2/M phase, but there was no induction of cell apoptosis;Morusin significantly decreased the expression ofβ-catenin and its target c-Myc, and downregulated the expressions of cyclinD1 and cyclinB1, which were involved in cell cycle regulation. Conclusion Morusin can inhibit HCT116 cell cycle and the proliferation of colorectal cancer cells. Its mechanism might be realized by suppressing the activity ofβ-catenin pathway and reducing the protein expressions of cyclinD1 and cyclinB1.
5.Effect of Xiaotan Sanjie Recipe on Proliferation and Apoptosis of HCT116 Cell and Expression of Caspase3 Protein

Objective To investigate the effects of Xiaotan Sanjie Recipe (XTSJR) on the proliferation and apoptosis of colorectal cancer cell;To study its relevant mechanism. Methods The original XTSJR aqueous solution was lyophilized, weighted, and dissolved in cell culture and stored. HCT116 cell line was treated with different concentrations of XTSJR for 72 h, and the cell proliferation was detected by CCK-8 assay. XTSJR treated HCT116 cell line with the concentration of 0.94 mg/mL and 1.88 mg/mL for 24 h, 48 h, 72 h, and the cell proliferation was detected by CCK-8 assay. HCT116 cell line was treated with different concentrations (0.24, 0.47, 0.94, 1.88 mg/mL) of XTSJR for 72 h. The cell apoptosis was detected by TUNEL assay under the fluorescence microscope, and the post-intervention expression of Caspase3 protein was detected by Western blot. Results The inhibitory effects of XTSJR on the proliferation of HCT116 cell line was concentration-time dependent. The IC50 value at 72 h-time point was 0.94 mg/mL. Medicine concentration and the treated time have interactive contribution to the inhibitory effect. XTSJR induced the cell apoptosis. The apoptosis rate increased with the increasing medicine concentration. There was statistically significant difference compared with the control group ( P<0.01). After treatment the expression of Pro-Caspase3 decreased, while the expression Cleaved Caspase3 protein increased. Conclusion XTSJR inhibits the proliferation of HCT116 cell line and induce apoptosis. Its mechanism might be related to the activity of Caspase3 protein.
6.Inhibitory effect of Xiaotan Sanjie Recipe on the microsatellite instability of orthotopic transplantation tumor in MKN-45 human gastric cancer nude mice.

OBJECTIVETo study the inhibitory effect of Xiaotan Sanjie Recipe (XSR) on the microsatellite instability of orthotopic transplantation tumor in MKN-45 human gastric cancer nude mice.
METHODSThe 3rd passage subcutaneous transplantation tumor was taken as the origin of the model by using MKN-45 human gastric cancer cell lines. MKN-45 human gastric cancer nude mouse model was established using OB glue adhesive method. Then 30 nude mice were divided into the model group, the XSR group, and the chemotherapy group. Mice in the XSR group were intragastrically given XSR at the daily dose of 0.4 mL. Mice in the chemotherapy group were intragastrically given Fluorouracil at the daily dose of 0.4 mL. No intervention was given to mice in the model group. After 6 weeks of medication, the tumor weight was measured, and the tumor inhibition rate calculated. The size, the peak height, and the peak area of 5 microsatellite instability sites were detected.
RESULTSThe tumor inhibition rate was 40. 84% in the XSR group. The tumor weight was significantly lower in the XSR group than in the model group (P < 0.01), showing no statistical difference when compared with the chemotherapy group (P >0.05). The incidence of high microsatellite instability (MSI-H) in the model group was 70%, and the incidence of low microsatellite instability (MSI-L) was 30%. Microsatellite stable site tended be stable after 6 weeks of XSR treatment.
CONCLUSIONXSR showed inhibition on microsatellite instable orthotopic transplantation tumor in MKN-45 human gastric cancer nude mice.

8.Protective effects of Jingqi Capsules on damage to rat reproductive system

AIM To study the protective effects of Jingqi Capsules (Cordycps Militaris,Rehmanniae Radix Praeparata,Lycii Fructus,) on the damage to rat reproductive system induced by cyclophosphamide.METHODS After low and high dose groups of Jingqi Capsules and positive group were administered for two weeks in advance,cyclophosphamide [25 mg/(kg · d)] modeling was conducted consecutively by intraperitoneal injection for five days except the normal group,while the treatment groups were administered consecutively.All the rats were executed the day after the end of the modeling.Serum testosterone content was measured by radioimmunoassay;body weight,testis,epididymis and prostate of rats were weighed and organ coefficient was calculated;morphology of testis was observed by HE;sperm count and sperm motility were detected by CASA;apoptosis of cells was observed by TUNEL.RESULTS Compared with the model group,each dose group of Jingqi Capsules increased in the level of serum testosterone and organ coefficient of testis,epididymis and prostate.The damage to seminiferous epithelium relieved and the number of all spermatogenic cells increased.Sperm count and sperm motility increased.Decrease in apoptosis of cells was demonstrated by TUNEL.CONCLUSION Jingqi Capsules has protective effects on damage to rat reproductive system induced by cyclophosphamide.
9.Relationship between expressions of estrogen and progesterone receptors and syndrome types of gastric carcinoma.

Objective: To explore the relationship between expressions of estrogen (ER) and progesterone (PR) receptors and syndromes of traditional Chinese medicine (TCM) in gastric carcinoma and to establish prognostic indicators for gastric carcinoma. Methods: A total of 72 patients with gastric carcinoma were divided into six groups according to TCM syndrome differentiation. Specimens were collected after operation and ER and PR protein expressions were detected by EnVision immunohistochemical method. Results: The common syndromes in female patients with gastric carcinoma were disharmony between liver and stomach, yin impairment due to stomach heat, and insufficiency of both qi and blood; while in males, interior retention of stagnant toxin, interior retention of phlegm and dampness, and deficiency-cold in spleen and stomach were common. Different TCM syndromes were related with gender (P<0.01), pathology (P<0.01), cell differentiation (P<0.05), infiltration depth (P<0.01), lymphaden metastasis (P<0.05), distant metastasis (P<0.05), and TNM stage (P<0.01). Deficiency and excess syndromes were associated with gender (P<0.05), pathology (P<0.05), tumor location (P<0.01) and TNM stage (P<0.05). The deficiency syndromes were common in female patients. The total positive rates of ER and PR expressions were 8.33% and 37.5% respectively. There was a significant difference in PR expression among different TCM syndromes (P<0.01). PR expression was significantly higher in the syndrome of yin deficiency due to stomach heat than in the other syndromes. The PR expressions in deficiency syndromes were significantly higher than those in excess syndromes (P<0.01). No correlation was found between ER expression and different TCM syndromes. Conclusion: There is a correlation between PR expression and different TCM syndromes in gastric carcinoma.
10.Diagnosis and treatment of urothelial tumors in multiple organs

Objective To explore the experiences of the diagnosis and treatment of urothelial tumor in multiple organs.Methods Clinical data of 10 patients with urothelial tumor in multiple organs were retrospectively reviewed.Urothelial tumors were found in two or more organs at the same time by B ultrasound,IVU,R-P,CTU,MRU,cystoscopy,ureteroscopy and so on before operation.Results 6 cases were operated by radical total nephroureterectomy and partial cystectomy,3 cases were operated by radical total nephroureterectomy and cystectomy with urinary diversion,1 case was operated by partial ureterectomy and total cystectomy.8 of them were alive,1 case was operated by total urethrectomy because of tumor recurrence in the posterior urethra,one died of metastasis tumor 18 months after operation,and the other died 32 month after operation.Conclusions Combined use of various kinds of the diagnostic means (ultrasound,IVU,R-P,CTU,MRU,cystoscopy,ureteroscopy) are important for the diagnosis of urothelial tumor in multiple organs.It needs to select the operate mode according to the tumor staging and grade and the patient's condition.Reinforcement surveillance and close follow up is required after operation.
