1.Effects of estrogen on behavior and norepinephrine content in habenular nucleus of depressive rats

Objective To observe the effects of estrogen on the behavior and the norepinephrine level in the habenular nucleus of depressive rats,and explore the mechanism of estrogen involved in the process of antidepression.Methods Thirty rats were chosen from forty male Wistar rats,which had the similar scores in open field test.The depression models of rats were established by chronic mild stress.Open field test and forced swimming test were performed to detect the behavioral changes between rats with ovary excision(OVX) and with estradiol-treated ovary excision(OV/ER).High performance liquid chromatography(HPLC) was used to observe the changes of norepinephrine level in habenular nucleus.Results After 21-day chronic mild stress,the open field test showed that the vertical motion and cross motion in OV/ER group were increased(P
3.Expressions of Bile Acid Receptor FXR and TGR5 in Patients with Ulcerative Colitis

Background:The incidence of ulcerative colitis (UC)in developed countries is higher than that in developing countries,which may be related with westernized lifestyle,especially high animal protein and low complex carbohydrate diet. With the increased high fat and meat intake,synthesis and secretion of bile acid in liver is also increased,which may have an impact on the occurrence of UC. Aims:To investigate the expressions of farnesoid X receptor (FXR)and G protein-coupled bile acid receptor 5 (TGR5)in patients with UC. Methods:Thirty patients with active UC from January 2013 to June 2016 at the Affiliated Jiangning Hospital of Nanjing Medical University were enrolled,and 30 healthy subjects were served as controls. Expressions of FXR and TGR5 were determined by immunohistochemistry. Results:Compared with control group,expression of FXR was significantly decreased in UC patients (4. 63 ± 2. 07 vs. 6. 91 ± 2. 62,P =0. 00),however,no significant difference in expression of TGR5 was found between the two groups (6. 70 ± 2. 90 vs. 6. 11 ± 2. 44,P = 0. 40). Expression of FXR was significantly increased in right hemicolon colitis than in left hemicolon colitis (P < 0. 05). Conclusions:There is a significant decrease in FXR in active UC patients,indicating that FXR may have some role in the pathogenesis of UC,however,TGR5 may have no obvious effect in the pathogenesis of UC.
4.Association between rheumatoid arthritis and shared epitope of HLA-DRB1 gene in Han nationality Neimenggu population

Objective To investigate the assoiation between rheumatoid arthritis(RA)and the pres- ence of the shared epitope(SE)of HLA-DRBI gene in Han nationality of Neimenggu population.Methods The method of DNA amplification with sequence-specific primers(PCR-SSP)was used to determine 17 alleles of HLA-DRB1*01,*04,*10 genotypes in 80 RA patients and 110 healthy controls from the Han nationality population in Neimenggu.Results The frequencies of SE were significantly increased in RA patiens com- pared with controls(48.8%:20%,P<0.01).Epitope analysis revealed that the most predominant allele subtype of DR4(*0405)was usceptible sequence in Neimenggu patients with RA(28.8%:12%,P<0.01).No statisticall significant difference of other subtypes of DR1,DR4 nd DR10 was noted including DRB1*0101(2.5%:0.9%), *0102(2.5%:0),*0103(1.25%:0.9%),*0104(2.5%:0),*0401(6.25:1.8%),*0402(3.75%:0.9%),*0403 (1.25%:1.8%),*0404(2.5%:1.8%),*0406(2.5%:2.7%),*0407(1.25%:0.9%),*0408(3.75 %:0.9%),*0409 (1.25%:0),*0410(2.5%:0.9%),*0411(0:0)and *1001(8.75%:4.5%)respectively.Logistic regression analy- sis showed that the disease of patients with SE homozgote was more severe than that of patients with heterozy- gote(P<0.01).Conclusion The results suggest that there is an association between SE of HLA-DRBI gene and susceptibility and severity of RA,especially,HLA-DR4 subtypes are strongly associated with RA in Han nationality in Neimenggu population.
5.Effects of agarwood formation induced by Fusarium sp. A2 on distribution and community composition of endophytic fungi in leaves of Aquilaria sinensis.

The aim of the paper is to observe the distribution of the endophytic fungi in leaves of Aquilaria sinensis by using permanent paraffin-cut section, optical microscope photography and histochemistry. Total DNA was extracted with modified CTAB method and rDNA ITS regions of plant and endophytic fungi were amplified with eukaryotic universal primers. The rDNA ITS amplicon was characterized by RFLP analysis, sequencing of rDNA ITS library and phylogenetic analyses using PAUP by maximum parsimony. Fusarium sp. A2 was used to induce the formation of resinous in A. sinensis trees. As a result, endophytic fungi mainly distributed in spongy and phloem in leaf. Endophytic fungi distributed in the phloem in agarwood-producing tree and had a relatively high abundance. Phoma sp. and Collectrotrichum sp. were the absolute advantage species in the leaf tissues of non-resinous and agarwood-producing tree, respectively. Collectrotrichum sp. was the only fungal species detected both in the two types of A. sinensis with different levels of abundance. The culture-independent molecular method can be used to identify fungal species directly and rapidly from the plant tissues. Endophytic fungal communities in non-resinous and agarwood-producing A. sinensis leaf tissues were quite different.

6.Genetic determination of fast plasma glucose concentration and correlation with anthropometric indices.

OBJECTIVETo study the genetic determination of fast plasma glucose (FPG) and correlation with its potential correlated traits, anthropometric measures and blood pressure.
METHODSTwo hundred and eighteen Type 2 diabetes mellitus (T2DM) pedigrees composed of 1383 Chinese Han individuals residing in the East and South-East China were analyzed. Univariate variance decomposition analyses were used to estimate the narrow-sense heritability (h(2)) of FPG, anthropometric indices and blood pressure, and bivariate quantitative genetic analyses were used to estimate the genetic and environmental correlations between FPG and anthropometric measures or blood pressure.
RESULTSWe found that FPG, blood pressure and all anthropometric indices except for waist to hip ratio were under significant genetic determination, and the h(2) was from 0.28 to 0.43. We did not find significant genetic and environmental correlation between FPG and anthropometric indices and blood pressure.
CONCLUSIONThe present study demonstrated that T2DM, obesity and hypertension were controlled by some genetic factors, and FPG shares little common genetic and environmental factors with obesity-related anthropometric indices and blood pressure in our Chinese sample population.

7.Paraganglioma of urinary bladder (two cases reports and review of literatures)

Objective To study the clinical,histopathological,immunohistochemical features and the diagnosis and treatment of paraganglioma of urinary bladder. Methods Two cases of paraganglioma of urinary bladder were treated. The first case was a male with painless haematuria. The abdominal ultrasonography and CT scan showed a 2.5 cm× 2.0 cm mass in the right anterior wall of the bladder, and urine vanillylmandelic acid elevated to 17. 9- 31. 3 μmol/24 h (normal range 10- 35 μmol/24 h). The second case was a female who presented with 8 years history of headache and palpitation after voiding. Abdominal ultrasonography and CT scan showed a 2. 6 cm× 1.5 cm mass in the left wall of the bladder, and her urine vanillylmandelic acid was 35.3-43.3 μmol/24 h. Results One patient underwent transurethral resection (TURBT) and the other underwent partial cystectomy.The two cases were diagnosed as bladder paraganglioma by pathological examination. Immunohistochemically, the tumor cells were positive for CgA, Syn, NSE and S-100. No evidence of recurrence was detected during follow-up at 3 months and 3 years. Conclusions Paraganglioma of urinary bladder should be considered as a low grade malignancy. Partial cystectomy should be recommended. The diagnosis depends on clinical symptoms, pathological and immunohistochemical results.

Objective To assess and compare the effectiveness of gadolinium diethylenetriamine pentacetic acid(Gd-DTPA) solution and barium sulfate(BaSO4) suspension with different concentrations in improving the imaging quality of magnetic resonance cholangiopancreatography(MRCP). Methods The phantom study was carried out to determine the optimal concentration of Gd-DTPA and BaSO4 suspension used as an oral negative contrast agent in MRCP.The patients were grouped randomly and performed MRCP before and after using oral contrast agents in combination with intravenous injection of contrast agents.A comparison of the influence of BaSO4 suspension and Gd-DTPA with different concentrations on the signal intensity of the fluid in gastrointestinal tract on MRCP images was made.Results The phantom study showed that the dilution ratio 1∶10 of Gd-DTPA solution and 100%(W/V)BaSO4 suspension were the optimal concentrations in decreasing the signal intensity.In all patients the high signal intensity of the gastrointestinal fluid was completely suppressed after oral administration of Gd-DTPA diluted solution(P
10.Inhibitory effect of chailing decoction on renal tubular epithelial phenotype transformation in rats with cyclosporine A-induced nephropathy.

OBJECTIVETo observe the effect of Chailing Decoction (CLD) on alpha-smooth muscular actin (alpha-SMA, a marker of renal tubular epithelial phenotype transformation) in rats with cyclosporine A (CsA)-induced nephropathy for investigate its mechanism of action in inhibiting tubulo-interstitial fibrosis.
METHODSForty Sprague-Dawley rats were equally randomized into four groups: the normal group, the model group, the positive control group and the Chinese medicine (CM) group. Excepting those in the normal group received gastrogavage with olive oil, rats in the other three groups were made into nephropathy model by oral infusion of CsA 30 mg/ (kg x d) for 28 days. At the same time of modeling, rats in the positive control and CM groups were treated respectively with Valsartan 10 mg/(kg x d) and CLD 3 g/(kg x d). The mRNA and protein expressions of alpha-SMA in rats' kidney were detected by RT-PCR, Western blot, immunohistochemical and flow cytometry, and the mRNA expressions of transforming growth factor-beta1 (TGF-beta1) and collagen type III (Col III) were determined by RT-PCR.
RESULTSLevels of (alpha-SMA, TGF-beta1, and Col III were significantly higher in the model group than those in the normal group respectively (P < 0.01 or P < 0.05), and the high levels were down-regulated in the positive control and CM groups after treatment (P < 0.01 or P < 0.05).
CONCLUSIONCLD could retard the progress of renal interstitial fibrosis by way of inhibiting renal tubular epithelial phenotype transformation.
