2.Hope for the patients with retinitis pigmentosa

Retinitis pigmentosa (RP) is a hereditary disease characterized by the progressive loss of photoreceptor cells. Disease pathology primarily affects rod photoreceptor cells first but light-insensitive cone photoreceptor bodies can survivor longer. Delivered by adeno-associated viral vector or lentiviral vector, expression of microbial-type rhodopsin, channelrhodopsin-2 or archaebacterial halorhodopsin in the survived retinal cells can substitute for the native phototransduction cascade in mouse models of retinitis pigmentosa. It can also restore light sensitivity and activate all retinal cone pathways and drive sophisticated retinal circuit functions. Thus, expression of channelrhodopsins or halorhodopsin might be a potential strategy for the restoration vision of patients with RP.

5.Preoperative oral pregabalin for postoperative pain relief after laparoscopic cholecystectomy: a randomized controlled double-blind study

ObjectiveTo evaluate the efficacy of preoperative oral pregabalin for attenuating postoperative pain after laparoscopic cholecystectomy.MethodsIn this prospective,randomized controlled double-blind study,sixty ASA Ⅰ or Ⅱ patients aged 19-72 yr weighing 46-86 kg undergoing laparoscopic cholecystectomy were randomly divided into 2 groups ( n =30 each):control group (group C) received placebo,and pregabalin group (group P) received oral pregabalin 150 mg 1 h before surgery.Anesthesia was induced with propofol,fentanyl and rocuronium and maintained with sevoflurane inhalation and intermittent iv boluses of fentanyl and rocuronium.The patients were intubated and mechanically ventilated.BIS value was maintained at 40-50 during operation.Static and dynamic VAS score,Ramsay score and consumption of morphine were recorded at 6,12,and 24 h after surgery.Side-effects including nausea,vomiting,headache and dizziness were also recorded.ResultsStatic and dynamic VAS scores and morphine consumption were significantly lower during the first 24 h after surgery while Ramsay scores were higher at 6 h after operation in group P than in group C.There was no significant difference in the incidence of side-effects between the 2 groups.No over-sedation occurred in group P.ConclusionPreoperative oral pregabalin 150 mg is safe and effective in reducing postoperative pain after laparoscopic cholecystectomy.
6.Resting state functional magnetic resonance imaging changes in spasmodic torticollis patients based on fractional amplitude of low frequency fluctuation

Objective To observe the differences of cerebral activation pattern with resting state functional magnetic resonance imaging (rs-fMRI) between patients with spasmodic torticollis (ST) and healthy controls,thus to investigate the pathogenesis of ST.Methods Nineteen ST patients and 21 age,sex and education-matched healthy controls,recruited from the Department of Neurology,First Affiliated Hospital of Guangxi Medical University between November 2012 and January 2016,were included in this study.rs-fMRI and factional amplitude of low frequency fluctuation (fALFF) were used to obtain differences between patients with ST and healthy controls,and correlative analysis was made on fALFF values of abnormal brain regions and ST patients' symptom severity (Tsui scores).Results Compared with healthy controls,patients with ST had significantly increased fALFF in the left cerebellum and significantly decreased fALFF in the left posterior cingulate cortex/precuneus,right posterior cingulate cortex/precuneus,left middle temporal gyrus,right angular gyrus,left post-central gyrus,right supplementary motor area (t =-5.714-5.920,P <0.01),and abnormal brain regions' fALFF values had no correlation with patients' age of onset,disease course,symptom severity (P > 0.05).Conclusion Abnormal sensorimotor area,default mode network and cerebellum dysfunction may play a role in the pathophysiology of ST.
7.An optimized voxel-based morphometry study in the evaluation of brain structural abnormalities in anisometropic amblyopia patients

Objective To investigate possible neural mechanism of anisometropic amblyopia by analysing the whole brain volume changes both in grey matter and white matter using optimized voxel-based morphometry (VBM).Methods Twelve anisometropic amblyopia patients and 12 age,gender and handedness matched healthy volunteers underwent 3-dimensional (3D) fast spoiled gradient echo (FSPGR)sequence scanning on 1.5 Tesla MR system.Raw data was processed and analyzed using statistical parametric mapping (SPM)5.ResultsCompared to healthy controls,the grey matter exhibiting significantly decreased volume in patients included right cuneus,bilateral occipital gyrus,right middle frontal gyrus,left middle temporal gyrus,right superior temporal gyrus,right precuneus,and middle part of right cingulate gyrus ( clusters > 10).The grey matter showing increased volume in patients included right cerebellum,right parahippocampal gyrus,left precentral gyrus,and left superior frontal gyrus (clusters > 10).The white matter volume in bilateral optic radiation and internal capsule,especially right optic radiation,decreased significantly in patient group (clusters > 10 ).No white matter showed significantly increased volume in patient group.ConclusionVBM can be used to investigate the changes of grey matter volume and white matter volume in the whole brain of anisometropic amblyopia children,it provides a method to illustrate the presumed neuro-mechanism from a morphologic point of view.
8.Comparison of psychological health status between infertile women with and without polycystic ovary syndrome

One hundred and twenty-two infertile women with polycystic ovary syndrome (PCOS) and 112 infertile women without PCOS were interviewed with symptom checklist 90 ( SCL-90) and questionnaire of demographic and clinical characteristics to understand their psychological health status. Results demonstrated that scores of depression, interpersonal sensitivity, obsessive-compulsive and aggressive symptoms in infertile patients with PCOS were significantly higher than those in non-PCOS infertile ones (P < 0. 05 ). Acne had negative effect on their psychological health status (OR =3. 379, 95% CI = 1.096 ±10.419) . Scores of somzatization, interpersonal sensitivity, anxiety, obsessive-compulsive, aggressive and paranoid ideation symptoms were significantly higher in secondary infertile patients with PCOS than those in primary infertile ones (P<0. 05). Patients with PCOS had obviously psychological problems, especially in secondary infertile patients with PCOS ( P < 0. 05 ). Thus, it is suggested that psychological health status be evaluated in clinical diagnosis for infertile women so that they can be predicted and treated to improve its effectiveness.

10.Effects of mild hypothermia on autophagy in hippocampal CA1 neurons in rats after CPR

Objective To establish the cardiac arrest-cardiopulmonary resuscitation model in rats, and to observe the effect of mild hypothermia on autophagy in hippocampal CA1 neurons after ROSC.Methods A total of 36 Wistar rats were randomly divided into two groups: normal temperature treatment group(NT group) and mild hypothermia treatment group(HT group).To establish the cardiac arrest-cardiopulmonary resuscitation(CA-CPR) model in rats by epicardial electrical stimulation induced ventricular fibrillation, and to sacrifice 3 animals in each group to obtain the brain cortex in 2nd and 4th hours after ROSC in order to observe the expression of p-AMPK by electron microscope and LC3 granules through Western blot.The neurological deficit score(NDS) was assessed in 24、48、72 hours respectively after ROSC.To sacrifice the animals so as to take the cerebrum in 72 hours after ROSC, then calculate the apoptotic index of the hippocampal CA1 neurons, which were dyed through TUNEL method.Results The expression of p-AMPK、Beclin-1 and LC3-Ⅱ/LC3-Ⅰratio in Normothermia group were all lower than the Mild hypothermia group(P<0.05), the neurons plasma of hippocampal CA1 area in the Hypothermia group demonstrated obvious LC3 granules formation, the NDS score of the Normothermia group and the Mild hypothermia group in ROSC24h、ROSC48h、ROSC72h were 320vs205、285vs140、266vs120, respectively.The apoptotic index of the hippocampal CA1 area in the Normothemia group in ROSC72h was higher than the Mild hypothermia group,(P<0.05).Conclusions Mild hypothermia after cardiopulmonary resuscitation promotes autophagy of the hippocampal CA1 area neurons in rats and reduce neuronal apoptosis.
