1.Changes of psychological state and family relationship in process of treatment for depression and anxiety

Objective To investigate the changes of psychological state and family environment of the patients with neurosis (including depression and anxiety) in the process of treatment. Methods The Hamilton depression scale (HAMD), Hamilton anxiety scale(HAMA), self-rating depression scale(SDS), self-rating anxiety scale(SAS), family environment scale(FES), and family adaptability and cohesion scale(FACESⅡ-CV) were used to evaluate the changes of psychological state and family relationship in the process of treatment among patients with depression and anxiety hospitalized in general hospitals (n=45) and their family members (n=45). Results Before intervention, values of HAMD, HAMA, SDS and SAS in patients were higher than normal ones, and were in line with the corresponding diagnostic criteria. After psychological intervention and medication for 2 to 3 months, symptoms such as depression and anxiety were improved significantly, and values of the above scales were within normal ranges. Besides, there was also significant improvement in FES and FACESⅡ-CV after intervention among the patients. All the values of the family members were in the normal ranges before intervention, however, significantly positive changes were observed after intervention. Conclusion With the improvement in depression and anxiety after intervention among patients, the psychological pressure of family members is released, and family relationship is partially restored. The rehabilitation of patients plays an important role in the restoration of family function.
2.Efficacy of Domperidone Combined with Pantoprazole and Hydrotalcite Tablets in the Treatment of Bile Reflux Gastritis

Objective:To observe the curative effects of the combined application of domperidone, pantoprazole and hydrotalcite tablets in the treatment of bile reflux gastritis ( BRG) and explore the underlying mechanism. Methods:Totally 80 patients with BRG were randomly divided into the treatment group and the control group with 40 cases in each. The treatment group was received panto-prazole, hydrotalcite tablets and domperidone, and the control group was treated by hydrotalcite tablets and domperidone. The efficacy was observed in both groups after 4 weeks. Results:The effective rate of symptom relief in the treatment group and the control group was respectively 92. 5% and 62. 5% (P<0. 05). After the 4 week treatment, the gastritis healing rate in the treatment group was higher than that in the control group (P<0. 05). The improvement of gastric mucosal hyperemia, edema and erosion in the treatment group was better than that in the control group with significant difference (P<0. 05). Cholic acid in gastric juice and gastric acid se-cretion were significantly reduced in both groups after the treatment, while the reduction of gastric acid secretion in the treatment group was more notable than that in the control (P<0. 05). Conclusion:Pantoprazole can be used to control BRG, and the combination of the three drugs shows better efficacy in the treatment of BRG.
3.Current progress in structure and function of angiopoietin family

Angiopoietin family is a recently discovered type of cellular factors that specifically bind to the TIE-2 receptors located exclusively in endothelial cell membrane. The protein structures of this family members are similar. They can be structurally divided into three domains: an N-terminal region lacking homology to any known structures, an alpha-helical rich coiled-coil segment, and a fibrinogen-like domain. The distribution and biological activity of these factors are different in organism. Angiopoietin-1 as a agonist, mostly locates in close proximity with vascular endothelial cells, keeps the stability of blood vessels, enhances the affinity of vascular endothelial cells with surrounding cells and matrix, decreases the leakage of vessel. Ang-2 is a naturally occurring antagonist of Ang-1, exists in the angiogenic remodeling region and is related to the decrement of the stability of vessel. Ang-3 is widely distributed in multiple mouse tissues, while Ang-4 is expressed only in lung. Although Ang-3 and Ang-4 are structurally diverged from each other, they appear to represent the mouse and human counterparts of the same gene locus. Biological functions of Ang-3 and Ang-4 have not been elucidated yet. Angiopoietin family has potentially clinical applications for incurring illnesses which lead to vessel wound and vascular abnormal development.
5.Gene expression of mitogen-activated protein kinases in fetal skin at different developmental stages and its potentially biological siginificance

Objective To explore the change in gene expression of extracellular signal regulated protein kinase1 (erk1),erk2,p38MAPK and 3 c-Jun N- terminal kinases (jnk1,jnk2,jnk3) in fetal skin at different developmental stages and children skin. Methods After morphological characteristics of fetal skin at different developmental stages were examined histologically,gene expressions of erk1,erk2,p38MAPK,jnk1,jnk2 and jnk3 were determined with reverse transcription-polymerase chain reaction analysis (RT-PCR). Results Compared with early gestational fetal skin,the levels of gene expression of erk1 and erk2 showed no substantial change in late gestational fetal skin,while the contents of transcripts of p38MAPK and jnk1 were significantly decreased,the expressions of mRNA of jnk2 and jnk3 were obviously elevated. In children skin,gene expressions of erk2,p38MAPK and jnk1 were even more remarkably lowered. In contrary,gene expressions of jnk2 and jnk3 were marked enhanced. Conclusion The relative elevation of gene transcription of erk2 and p38MAPK and the inhibition of gene expression of jnk2 and jnk3 in fetal skin of earlier developmental stage might be related to fetal scarless healing.
6.Analysis of differentially expressed genes in keloids and normal control skin

Objective To study the differentially expressed genes between keloids and normal control skin using cDNA microarray. Methods The PCR products of 8400 human genes were spotted on a chemical-material-coated-glass plate in array. Total RNAs were isolated from the freshly excised human keloids and normal control skin,and then were purified to mRNAs,which were reversely transcribed to cDNAs with the incorporations of fluorescent dUTP,for preparing the hybridization probes. The mixed probes were hybridized to the cDNA microarray. According to the results of cDNA microarray,three genes (NGF,TGF-?_ 1 and c-myc) were chosen to study their differential expression in keloids and normal control skin by RT-PCR. Results Among the 8400 target genes,there were 402 genes with different expression (4.75%),and they were mainly associated with extracellular matrix genes,cellular signal molecule genes and cellular skeleton molecule genes. Analysis of collagen related molecule and growth factor gene expression confirmed that our molecular data obtained by cDNA microarray were consistent with published biochemical and clinical observations of keloids. RT-PCR showed that the levels of gene expression of NGF,TGF-?_ 1 and c-myc were all higher in keloids than those in normal control skin. Conclusion Many genes were found to be involved in the formation of keloids. Further analysis of the obtained genes might contribute to reveal the molecular mechanism of keloids formation,in which NGF,TGF-?_ 1 and c-myc might play important roles.

To investigate the different expression of Fas, Fas L and Caspase 3 in hypertrophic scars and normal skins in order to explore their influences on scar formation, the expression intensity and distribution of Fas, Fas L and Caspase 3 were detected by immunohistochemistry and pathological methods in 8 cases of hypertrophic scars and 8 cases of normal skins. In normal skins, Fas and Fas L were mainly located in the cellular membranes and cytoplasms of epidermal cells and some fibroblasts. Caspase 3 was mostly distributed in epidermal basal cells and some fibroblasts. The purpose of this study was to investigate the localization and expression characteristics of phosphorylated form of extracellular-signal regulated-protein kinasel/2 (p-ERKl/2), Ras and C-fos in skin at different development stages and to explore their potential biological significance. Immunohistochemistry and pathological methods were used to detect the expression intensity and distribution of p-ERKl/2, Ras and C-fos in skin of 8 fetuses with different gestational ages (13 to 31 weeks) and 8 adults. Positive immunohistochemical signals of p-ERK1/2, Ras and C-fos. could be found in fetal and postnatal skins. Along with the increment in gestational age, the positive cell rates of p-ERK1/2, Ras and C-fos in the skin elevated progressively. 9.CHARACTERISTICS OF INSULIN-LIKE GROWTH FACTOR-Ⅰ RECEPTOR EXPRESSION IN FETAL AND ADULT SKINS

Localization and expression of two subunits (IGF ⅠR? and IGF ⅠR?) of insulin like growth factor Ⅰ receptor and phosphorylated tyrosine proteins (P Tyr) in skins at different developmental stages were studied in order to explore their potential biological significance. Immunohistochemistry and pathological methods were used to detect the expression intensity and distribution of IGF ⅠR?, IGF ⅠR? and P Tyr in skins of 12 fetuses with different gestational ages and 8 adults. The results showed that positive immunohistochemical signals of IGF ⅠR?, IGF ⅠR? and P Tyr could be found in fetal and adult skins. Along with growth and development of the fetus, the positive cell rates of IGF ⅠR?, IGF ⅠR? and P Tyr in skins elevated progressively. In adult skins, IGF ⅠR was mainly located in the cell membrane of epidermal cells, while P Tyr was chiefly distributed in epidermal cells and some fibroblasts. These results suggested that IGF ⅠR and its mediating signaling pathway might be involved in cutaneous development at embryonic stage, in cutaneous structure and function maintenance, and in wound healing at postnatal stage.
10.GENE EXPRESSION OF p38 MITOGEN-ACTIVATED PROTEIN KINASE AND ITS MAPKKs IN HYPERTROPHIC SCARS

To explore the change in gene expressions of p38 mitogen-activated protein kinase (p38MAPK) and its upstream signal-regulated molecule (mkk3 and mkk6) in normal skin versus hypertrophic scars underlying its potentially biological significance. Total RNAs were isolated from 8 specimens of hypertrophic scars and 8 specimens of normal skin, then they were purified to mRNAs, and the gene expression of mkk3, mkk6 and p38MAPK was examined with reverse transcription-polymerase chain reaction (RT-PCR). The results showed that the intensities of gene expression of mkk6 and p38MAPK were weak in normal skins, while the expression of these 2 genes was significantly elevated in hypertrophic scars compared with normal skins (P0.05). In hypertrophic scar, the elevation of mkk6 and p38MAPK gene expression, which plays pilot roles in cell proliferation, may be one of the mechanisms controlling the formation of hypertrophic scars.
