2.Preventive effects of sodium butyrate on gastric epithelial cells damaged by N-methyl-N’-nitro-N-nitrosoguanidine

Objective To investigate the effects of sodium butyrate (SB) on fetal gastric epithelial cells (GECs) in vitro damaged by N-methyl-N’-nitro-N-nitrosoguanidine (MNNG). Methods The fetal gastric epithelial cells were isolated, cultured and identified. Then the cells of the second generation were cocultured with 10 -5 mol/L, 10 -6 mol/L, 10 -7 mol/L SB for 4 h, then added with 10 -5 mol/L MNNG culturing for another 24 h. After the treated cells were finally cultured in normal culture medium for 40 d, the unscheduled DNA synthesis (UDS), lipid peroxidation (LPO) and ras p21 were detected. The cells were only cultured with 10 -5 mol/L MNNG served as negative control and without MNNG or SB as normal control. Results The level of UDS, the contents of lipid peroxidation products and the concentrations of ras p21 in GECs treated with 10 -5 mol/L and 10 -6 mol/L SB decreased obviously, and had significant differences with that of GECs treated without SB. Conclusion SB can effectively prevent the damage of GECs induced from MNNG.
3.Reparative effect of BPI on damaged mucosa of rats with experimental otitis media with effusion

Objective To explore the reparative effect of bactericidal/permeability-increasing protein(BPI) on the damaged mucosa of rats with otitis media with effusion (OME),and state the pathogenesis of OME.Methods Wistar rats(40 ears) were randomly divided into six groups:normal control group (n=4),BPI control group(n=4),eustachian tube obstruction (ETO) group (n=8),lipopolysaccharide (LPS) injection group (n=8),ETO+LPS group (n=8),ETO+LPS+BPI group (n=8).The experimental OME model was made through eustachian tube obstruction and LPS injection.The rats were killed after 1,2 and 4 weeks and the changes of mucosa of middle ear were observed under light and scanning electron microscope.Results The rats in normal control group and BPI control group had the normal mucosa in the tympanic orifice of the eustachian tube.It consisted of pseudostratified ciliated cubical or columnar epithelium which contained an abundant number of ciliated cells and a few goblet cells,these were the mucociliary clearance system of the middle ear.The hypotympanum consisted of thin,squamous epithelium with few microvillus.Middle ear mucosa was obviouly thickened in LPS injection,ETO and ETO+LPS groups.An increase in goblet cells and a decrease in ciliated cells were observed in the tympanic orifice of the eustachian tube.The epithelial layer in the hypotympanum had become more pseudostratified ciliated cubical epithelium.In ETO+LPS+BPI group,there was thin squamous epithelium in the hypotympanum near normal,which was not thickened and contained few microvillus. Conclusion LPS and ETO can result in the occurrence and protracted courses of OME by mimosa's inflammatory reaction which can reduce the activity of ciliary cells and weaken the function of mucociliary clearance system.BPI could bind avidly to LPS,reduce inflammatory reaction,and break the inflammatory cycle and reestablish an effective mucocillary clearance system.The results suggest that BPI treatment is a potential effective drug for prevention and therapy of chronic OME.
4.An experimental study on the expression of heme oxygenase-2 mRNA in Hirschsprung's disease.

In order to investigate the relationship between the expression of heme oxygenase-2 (HO-2) mRNA and the pathogenesis of Hirschsprung's disease (HD), total ribonucleic acid (RNA) was extracted in the aganglionic and ganglionic segments of colon respectively from 15 cases of HD. The single-stranded cDNA of HO-2 was synthesized and further amplified by reverse transcription-polymerase chain reaction (RT-PCR). The expression of HO-2 mRNA was normal in ganglionic segments, but absent in aganglionic segments. It is concluded that the absence of HO-2 mRNA expression may be an important mechanism responsible for HD.

5.Relationship between Oxfordshire community stroke project classification and imaging classification in acute cerebral infarction

Objective To investigate the relationship between Oxfordshire community stroke project(OCSP) classification and imaging classification in acute cerebral infarction. Methods Two hundred and thirty-six patients with acute cerebral infarction were retrospectively evaluated with OCSP classification and imaging characteristics. Results According to OCSP classification,of all the 236 patients with acute cerebral infarction,28(11.9%) experienced total anterior circulation infarction(TACI),71(30.1%) partial anterior circulation infarction(PACI),94(39.8%) lacunar infarction(LACI),and 43(18.2%) posterior circulation infarction(POCI).The consistency was found in 171 cases(72.5%) between the OCSP classification and imaging classification,with the accuracy of 76%(25/33) for TACI,81%(34/42) for PACI,71%(81/114) for LACI and 66%(31/47) for POCI. Conclusion OCSP classification can predict the location and size of cerebral infarction with a high accuracy,and is well consisted with the imaging findings.
6.Astragalus Polysaccharide Prevent Type 1 Diabetes in Nonobese Diabetic Mice

Purpose To observe the effects of the intervention or prevention of astragalus polysaccharide(APS)on type 1 diabetes in non-obese diabetic(NOD) mice. Methods the APS group was compared withthe normal solution(NS)group by the incidence of diabetes, the serum C-peptide levels and GAD-Ab levels,the proportion of CD4 or CD8 T subsets in splencytes, pancreatic histopathology and immunocyto-chemistry.Results It shows that the APS group has lower incidence of diabetes, higher serum C-P levels, decreaseddegree of the lymphocytic inflammation of pancreatic islets, stronger proliferation of CD8 T subsets and lowerratio of CD4/CD8 subgroup in splencytes than those of the NS group. Conclusions It proves thepreventive effects of APS on the onset of type 1 diabetes in NOD mice.

Objective To study the expressions of TGF-?1 and p21WAF1 in middle ear cholesteatoma and discuses their effects on cholesteatoma.Methods The expressions of TGF-?1 and p21WAF1 were determined by immunohistochemical S-P technology and computer image analysis in 20 specimens of cholesteatoma epithelium and 10 specimens of normal external canal skin.Results ① p21WAF1 and TGF-?1 could be detected in middle ear cholesteatoma and in normal ear canal skin.p21WAF1 was located at karyon,taking on yellow-brown pellet;TGF-?1 was located at cytoplasmic,taking on yellow-brown pellet.②The p21WAF1 positive rate in middle ear cholesteatoma was 65%;and there were almost no positive cells in normal external ear canal skin.p21WAF1 positive cells were detected in each layers,its LI was 28.9%?6.7%.Compared with normal ear canal skin,the expression level of p21WAF1 in middle ear cholesteatoma epithelium was higher than that in normal ear canal skin(P
9.Analysis of related factors of recurrence after myomectomy

Objective To study the related factors of recurrence after myomectomy. Methods The clinical data of 441 cases with myomectomy were collected. The age at the time of operation, body mass index(BMI), preoperative parity, and the location, size and number of myomas were recorded, and the postoperative recurrence rate was calculated. Univariate and multivariate analysis were performed for possible factors related to recurrence. Results The average time of follow-up was(37.79?18.64) months. Recurrence was found in 125 cases, and 5-year cumulative rate of recurrence was 38.9%. Background In patients with syphilis,one of the common manifestations is uveitis.However,the lack of understanding on syphilic posterior uveitis may result in misdiagnosis and delayed treatment.Objective This study was to assess ocular clinical features,treatment and prognosis in patients who had latent syphilis and presented with posterior uveitis as the initial symptom.Methods A serial cases-observational study was designed.Twelve patients(19 eyes)who were diagnosed as syphilis from September 2011 to January 2013 were retrospectively analyzed.All patients were characterized by initial posterior uveitis before the final diagnosis.The clinical manifestation,management and prognosis were evaluated and analyzed.All patients were informed of this study and consented with the use of their data for this retrospective study.Results The patients presented with initial clinical signs of acute or chronic posterior uveitis and lacked systemic syphilis symptom.Bilateral posterior uveitis were found in 7 patients and unilateral in 5 patients.The dust-like and grey-white opacification in vitreous was seen by threemirror contact lens in all the patients.Mild hyperemia of the optical disc was found in 10 eyes with the unclear peripapillary boundary.Yellow-white lesions were observed in retinas of 7 eyes and retinal punctuate hemorrhage was in 1 eye.Fundus fluorescein angiography demonstrated fluorescine staining or hyperfluorescence of optic disc in all of the 19 eyes,and fluorescein leakage from vein in 18 eyes,retinal pigment epithelium dye pooling in 6 eyes and cystoid macular edema in 11 eyes.Rapid plasma reagin (RPR) test and treponema pallidum hemagglutination assay (TPHA) were performed and presented positive results in all patients.The visual acuity improved in 19 eyes and partly elevated in 2 eyes following a standard therapy for neurosyphilis was used,including penicillin and/or small doses of glucocorticoid.Conclusions The serological examination for syphilis should be considered for patients with unexplained uveitis.Early diagnosis and prompt treatment are important for the improvement of prognosis in syphilitic posterior uveitis.
