0.05). The pathological findings indicated benign lesions in 63 patients(73%) and malignant lesions in 25 patients(27%)in group 1. All the lesions were found benign in group 2. There was significant difference between the two groups(P=0.0033). Conclusion Noninvasive and painless transvaginal two-dimensional ultrasonograpy and color Doppler ultrasonography may have important clinical application value in diagnosis of intrauterine anomaly in post-menopausal women.

Objective To investigate the effects of Xuebijing injection on the production of ICAM-1 and its counterreceptor(LFA-1)in ANP of rats at different time points. Methods ANP was induced by multiple points in- jection of 5% chenodeoxycholic acid into the pancreas, followed by ligation of pancreatic duct.Expressions of ICAM- 1 at different time points were assessed by immunohistochemistry method, the expressions of LAF-1 on neutrophils at different time points were detected by flow cytometer. The effect of Xuebijing injection was also evaluated by de- tecting the output of ascitic fluid, amylase level and histological changes in pancreas and lungs. Results The expres- sions of ICAM-1 in pancreas and lung of Xuebijing injection group decreased significantly than those in ANP group at most time points during the research period(P

Aim To investigate the effects of Shaoqiduogan(SQDG)on cellular immunity in mice with delayed-type hypersensitivity(DTH). Methods DTH model induced by 2, 4-dinitrochlorobenzene(DNCB)and cyclophosphamide(Cy)suppressed and potentiated DTH model were used. SQDG(30, 60 and 120 mg ? kg-1)were given intragastrically(ig)once daily for 7 consecutive days. Concanavalin A(Con A)-induced T cells proliferation was measured by MTT assay. The activity of IL-2 in culture supernatants of Con A-induced T cells proliferation was measured by testing its ability to support the proliferation of Con A stimulated thymocytes which was detected by MTT assay. Results In mice with DTH, SQDG(120 mg?kg-1)reduced ear swelling, thymus index, Con A-induced T cells proliferation and the activity of IL-2 in culture supernatants of Con A-induced T cells proliferation. The suppression and potentiation of DTH in mice were induced by given an intraperitoneal(ip)injection of Cy at the dose of 150mg?kg-1 on primary sensitization day and 250 mg?kg-1 three days before primary sensitization. SQDG(60 and 120 mg?kg-1)upregulated ear swelling, Con A-induced T cells proliferation and the activity of IL-2 in culture supernatants of T cells from mice with Cy-suppressed DTH; and downregulated levels of the aforementioned indexes in mice with Cy-potentiated DTH. Conclusion SQDG possesses a dual modulatory effect on cellular immunity in mice.

Lactate and succinate were produced by Corynebacterium acetoacidophilum from glucose under oxygen deprivation conditions. To construct knockout mutant, lactate dehydrogenase gene (ldh) of C. acetoacidophilum was deleted by double-crossover chromosome replacement with sacB gene. Comparing with the wild strain ATCC13870, ldhA-deficent mutant produced no lactate with glucose consumption rate decreased by 29.3%, while succinate and acetate concentrations were increased by 45.6% and 182%, respectively. Moreover, the NADH/NAD+ rate was less than 1 (about 0.7), and the activities of phosphoenolpyruvate carboxylase and acetate kinase of the ldhA-deficent mutant were enhanced by 84% and 12 times, respectively. Our studies show that succinicate and acetate production pathways are strengthened by blocking lactate synthesis. It also suggests that improving NADH supply and eliminating acetate generation are alternative strategies to get high succinate-producer.

Objective To investigate the effects of early rehabilitation therapy in mechanically ventilated ICU in patients. Methods A randomized controlled trial was carried out.Sixty mechanically ventilated patients, with tracheal intubation or tracheostomy more than 48 h and less than 72 h,were admitted to the intensive care unit (ICU) of the Affiliated Hospital of Medical College Qingdao University from May 2010 to May 2012.These patients were randomly (random number) divided into two groups, the rehabilitation group and the control group,30 patients in each group. In rehabilitation group, rehabilitation therapy was taken twice daily, and the training time and intensity was adjusted according to the condition of the patients.Early rehabilitation therapy included head up actively, transfer from the supine to sit,out of bed, transfer to a chair, standing bedside bed and walking bedside bed.The patient' s body mass index, days to first out of bed, duration of mechanical ventilation,ICU stay,APACHE Ⅱ score, highest FiO2,lowest PaO2/FiO2 and hospital mortality of patients were compared between rehabilitation group and control group. The differences between the groups were compared using t test. Results There was no significant difference in body mass index,APACHE Ⅱ score, highest FiO2,lowest PaO2/FiO2 and hospital mortality between rehabilitation group and control group (P > 0.05).Patients in the rehabilitation group had shorter first out of bed time [(3.8 ± 1.2) d vs.(14.9 ±4.7) d; P =0.00],duration of mechanical ventilation [(5.6±2.1) dvs.(12.7±4.1) d; P=0.005] andICUstay [(7.3±2.8) dvs.(15.2±4.5) d;P =0.01] compared with control group. Conclusions Early rehabilitation therapy was safe and effective in improving the outcomes of mechanical ventilation patient.

With the development of therapeutic antibodies over the past decade, they have become the treatment options for immunity and inflammation diseases. Major limitations of mouse antibodies as therapeutic agents - immunogenicity, lack of effectors' functions and short serum half-life -- were subsequently identified and largely overcome by the advent of humanized and fully human antibody technologies. The therapeutic antibodies for immunity and inflammatory diseases are primarily utilized in the treatment of allograft rejection, autoimmune disease, autoinflammatory syndromes, allergies and other chronic inflammation. The action mechanisms of therapeutic antibody include blocking ligands or receptors, regulating receptor activity, clearing the target cells or activating receptor. Strategies for generating the antibody drugs with high efficacy and low side effects can be realized by modulation of Fc-mediated activities and optimization of antigen-binding domains.

Acta Pharmaceutica Sinica 2013;48(1):66-70
To prepare large naive phage antibody library, the host bacteria with high transformation efficiency is used in the Cre-LoxP recombination system. The variable regions of immunoglobulin light and heavy genes were amplified from lymphocytes collected from adult peripheral blood and newborn cord blood. The genes were spliced to form the single-chain variable fragments (scFv) by overlap PCR, cloned into pDAN5a vector and then transformed into XL2-blue MRF' with the Hte gene. Compared with XL1-blue strain, the size of the primary library was increased by 3.9 times. The primary library infected Cre recombinase-expressing bacteria, and the genes between phagemids created many new VH/VL combinations. The library was calculated to have a diversity of 1.7 x 10(11) and validated by the selection of antibodies against six different protein antigens. This library provides the basis for further selection of antibody-based drugs. It is the first time to report that XL2-blue MRF' can be used to improve the diversity of the library in the recombination system.

Objective To study the related factors of recurrence after myomectomy. Methods The clinical data of 441 cases with myomectomy were collected. The age at the time of operation, body mass index(BMI), preoperative parity, and the location, size and number of myomas were recorded, and the postoperative recurrence rate was calculated. Univariate and multivariate analysis were performed for possible factors related to recurrence. Results The average time of follow-up was(37.79?18.64) months. Recurrence was found in 125 cases, and 5-year cumulative rate of recurrence was 38.9%. The age at the time of operation, BMI, number of myomas and preoperative parity were related factors of recurrence(P
