1.Establishment and Observation of Rats Model with Acute Pyelonephritis

Objective To made the animal model which is similar to the human pyelonephritis to strengthen the experimental basis for the therapy of acute urologic infectious disease. Method Using the method of semi-ligation of the ureter on single lateral and the injection of Enterobacter into the bladder to get pathologic transformation. Simultaneously, giving the different preparation of Zishen- tongguan Capsule, Sanjin Pills to intervene the models. Result Zishentongguan capsule middle dosage, western medicine treatment team had get obviously effect on rat body weight increasing, the drug treatment were validity. Conclusion The rat model with the acute nephropyelitis is similar to the character in the break of human, which manifest on the appearance, mental condition, body weight coefficient, kidney weight coefficient and pathologic change of the kidney. Succeed in the model establishment.
2.Effect of preconditioning electrostimulation of fastigial nucleus on neuronal mitochondrion of rat early after cerebral ischemia

Objective To investigate the effect of preconditioning electrostimulation of fastigial nucleus (FNS) on the neuronal mitochondrion of rats early after cerebral ischemia. Methods Forty Wistar rats were used and divided into 5 groups: a normal control group, a sham FNS group and 3 FNS groups. The FNS of the rats in the FNS groups was conducted for 1 hr, 1 day or 7 days, respectively, before the models of middle cerebral artery occlusion(MCAO)were made with them. Three hours after the model was established, the animals were sacrificed and the water content of the brain of the ischemic side was measured. The neuronal mitochondrion was observed by electron microscope, and its Vv, specific volume (Sv) and specific surface (Ss) were analyzed. The rats in the normal control group were not given any special treatment. For the rats in the sham-FNS group, intrinsic neurons of FN were destroyed with ibotenic acid (IBO), and 5 days later, the FN was electrically stimulated, and 1 day later, MCAO models were made. Results Three hours after MCAO, the water content of the brain increased and neurological function score decreased in the sham-FNS group, while the Vv and Sv of neuronal mitochondrion increased and the Ss decreased, which were significantly different from those of the normal control group(P
4.Effect of long non-coding RNA CCAT1 on metastasis, invasion and epithelial-mesenchymal transition of endometrial carcinoma cell

Objective To investigate the effect of CCAT1 on migration,invasion and epithelial-mesenchymal transition (EMT) in endometrial carcinoma (EC) cell.Methods Using quantitative PCR assay,level of CCAT1 was detected in EC tissues to find its association with the initiation and malignancy degree of EC.Wound heal assay and transwell invasion assay were performed to study the effect of CCAT1 on migration and invasion ability of EC cell,while qPCR and western blot assay were utilized to detect the levels of related genes.Results In EC tissues,level of CCAT1 was significantly upregulated (P < 0.001) and was positively associated with the malignancy degree of EC.After CCAT1 knockout,shorter migration distance was found,fewer cell passed through the chambers,levels of metastasis-related genes (MMP2 and MMP9) and mesenchymal markers (Snail,Zeb1 and Twist1) were up-regulated,and epithelial markers (E-cadherin and ZO-1) were down-regulated.Conclusion CCAT1 was up-regulated in EC tissue and its expression level was positively associated with the malignancy degree of EC.CCAT1 knockdown inhibited the metastasis,invasion and epithelial-mesenchymal transition of EC cell.

6.Retroperitoneal laparoscopic ureterolithotomy for impacted ureteral calculi after radical cystectomy and ileal conduit (report of 5 cases)

Objective To evaluate the clinical efficacy and security of retroperitoneal laparoscopic ureterolithotomy(RLUL) for impacted ureteral calculi after radical cystectomy and ileal conduit.Methods 5 patients with unilateral impacted ureteral calculi after radical cystectomy and ileal conduit received RLUL were selected,and a retrospective study was performed for manner,duration of surgery,complications and length of stay and other indicators,and the treatment effect was evaluated.Results All procedures were successful and the mean operation time was (82.0±27.7)min,the extubation time was (5.4±1.1)d,hospitalization time was (10.0±2.9)d.Postoperative follow up for 6-48 months,there were no obvious complications.Conclusion The RLUL showed satisfactory availability and security for management ureteral calculi after radical cystectomy and ileal conduit.
7.Investigation of inhibition of liver metastasis of colon carcinoma on the nude mice models with the system of TK/GCV and CD/5-Fc

Objectives:To explore the inhibition effect of TK/GCV and CD/5-FC system on liver metastasis of colon carcinoma.Methods:Forty nude mice were divided into 4 groups randomly,which were the control group,TK group,CD group and TK-CD group with 10 nude mice in each group.The mice in the control group were injected SW480 cells in the spleen and NS into the peritoneal cavity.The mice in other groups were all injected SW480/TK-CD cells in the spleen.And GVC,5-Fc,GVC+5F-c were injected into the peritoneal cavity of the mice in the TK group,CD group and TK-CD group respectively.Liver metastasis rate,the number of liver metastasis,the pathological changes of the tumor tissues under the electronic microscope,tumor cell's apoptotic index and life span and other indexes of nude mice in four groups were analyzed.Results:Liver metastasis rate of nude mice in every treatment group was lower than that in control group.Compared with control group,average number of liver metastasis decreased,life span of nude mice was prolonged,and the apoptosis rate of cancer cell in liver metastasis increased.The combined genes in the TK-CD group worked more markedly,and there was mutual effect between TK/GCV and CD/5-Fc.Conclusions:Synergistic effect was acquired combining TK/GCV with CD/5-FC system and it inhibited the formation of liver metastasis of colon carcinoma efficiently.
8.Recent progress of targeted small molecular CDK9 degraders based on PROTAC technology

CDKs proteins are a kind of cell cycle protein-dependent kinases, which serve as important roles in controlling cell division and transcriptional stages. Among them, CDK9, as a key regulator responsible for the transcriptional elongation of cells, drives the development of various malignant cells and is considered as an important target in the field of anti-tumor drug development. However, the CDK family proteins feature high conservativeness and similarity in structure, leading to the poor selectivity and severe side effects for traditional small-molecular CDK9 inhibitors, which has limited their clinical applications. In view of this, there is an urgent need to investigate CDK9 targets through a novel strategy. The PROTAC is an emerging drug discovery strategy that the degrader could specifically recognize the target protein through indirect linkage with ubiquitin ligases and ultimately eliminate the target protein through the ubiquitination degradation system. This paper provides a brief overview of the structure and function of CDK9 protein, its relationship with the poor prognosis of clinical diseases, as well as the currently reported small molecular inhibitors. The latest research progress on the targeted degradation of CDK9 protein based on PROTAC technology is highlighted. Finally, the development prospects of this target protein in this novel technology field are summarized and prospected, aiming to provide a reference for the development of antitumor drugs in this direction.
