4.Effect of valsartan on vascular endothelial function and brachial-ankle pulse wave velocity in patients with essential hypertension

Objective To investigate the effect of valsartan on vascular endothelial function and brachial-ankle pulse wave velocity(baPWV)in patients with essential hypertension(EH).Methods Blood pressure,NO value and brachial-ankle pulse wave velocity were determined and analyzed in 30 patients with essential hypertension before and after therapy of valsartan.Results Both blood pressure and brachial-ankle pulse wave velocity wefe re-duced greafly(P<0.05 and P<0.01),while serum NO increased significantly(P<0.01)following valsartan thera-Py,and brachial-ankle pulse wave velocity was well correlated to serum NO(r=-0.71,P<0.01).Conclusions Valsartan is not onlv effective in the control of blood pressure,but also effective in reveming the impaired endothelial function and artery elasticity in patients with essential hypertension,at the same time brachial-ankle pulse wave ve-locity is also good surrogate of endlothelial functional improvement induced by Valsartan therapy.
5.CT measured parameters of upper airway in OSAHS patients

OBJECTIVE To explore the value of spiral CT in detecting narrow level of obstructive sleep apnea hypopnea syndrome(OSAHS) through investigating measured parameters of upper airway. METHODS The upper airway levels from the roof of nasopharynx to the inferior border of cricoid cartilage were scanned with spiral CT in 127 OSAHS patients and 52 controls.The coronal and sagittal diameter and the cross sectional areas of palate,uvula,tongue, epiglottis level were measured using image handling workstation.The measured values of each level were divided by the measured values of the cricoid cartilage level,and get relative values.The relative values of the OSAHS patients and controls were compared. RESULTS The relative values of posterior soft palate and uvula region,lingual region and epiglottis regions were statistically different.The difference also existed between genders.The relative values of 46 controls and 17 OSAHS patients were in the normal range. Among 127 OSAHS,the relative values of 110 cases with OSAHS(86.6%) were below the normal range in every region,while in control group,the relative values were found below the normal range in only 6 persons (11.2%).CONCLUSION The relative values of each regions is useful to judge the narrow level of upper airway in OSAHS patients.
6.Effects of nuclear factor-?B on cell apoptosis and left ventricular segmental function in acute myocardial ischemia-reperfusion

Objective To probe the effects of nuclear factor kappa B (NF-?B) on cell apoptosis and left ventricular segmental function in acute ischemia repeffusion in dogs.Method Twenty-four dogs were randomly divided into three groups:without left anterior artery (lAD) ligation group (C group),LAD was occluded 30 min following reperfusion 120 minutes in isehemical reperfusion group (IR group),and dogs were administered with PDTC before LAD ligation in ischemical reperfusion plus pyorrole dithitocarbamate group (PDTC group).The left ventricular segmental function was detected by echo cardiography using strain rate anlysis software.EF measured by Simpson's method.Cardiac myocyte apoptosis numbers were determined by terminal deoxynudeotidy transferease-mediated biotinylated deoxyuridine triphosphate nick end labeling (TUNEL).lmmunohistochemistry and western-blot anylysis of NF-?B protein expression.Results NF-?B was obviously expression on injury myocardium of IR group,and increased significantly in contrast to control group (P0.05)Conclusions NF-?B might play an important role in acute myocardial ischemia reperfusion.PDTC reduces myocardial iscbemia/repeffasion injury by preventing expression of factor NF-?B.
7.Anti-oxidative effect of total extract of astragalus(TEA) on aging mice

Aim To study the mechanism of the anti-aging effect of total extract of astragalus(TEA) in mice. Methods MDA content, the activities of Mn-SOD and GSHpx and the reduced to oxidized glutathione (GSH/GSSG) ratio in mitochondria of D-galactose(D-gal) treated mice and 17-month-old mice were measured. Results Treatment with TEA(40 mg?kg-1?d-1 ig) for 10 wk would lower the content of MDA and restore activities of Mn-SOD,GSHpx and GSH/GSSG ratio in mitochondria of D-gal treated mice.Treatment with TEA (40 mg?kg-1 ?d-1 ig) for 3 mon had the same effect on 17-month-old mice.Conclusion TEA has anti-aging effect on D-gal treated mice and 17-month-old mice significantly, probably being related to its anti-oxidative effect.
8.A new photodynamic therapy drug toward gastric cancer MGC803 cell.

OBJECTIVETo investigate the treatment efficiency of a new photodynamic therapeutic(PDT) drug synthesized by our laboratory toward MGC803 cells and related mechanisms.
METHODSBleaching method was used to evaluate the photostability of drug upon repetitive illumination. MTT assay was used to determine the ability of new drug killing MGC803 cells after PDT. Laser scanning confocal microscopy (LSCM) was applied to investigate the subcellular localization of drug in MGC803 cells (mitochondria and/or lysosomes). Hoechst staining and flow cytometry(Annexin V/PI double-staining) were performed to detect the death mode of MGC803 cells after PDT.
RESULTSThis new PDT drug had good stability to light irradiation after repetitive illumination. MTT assay showed no cytotoxicity towards MGC803 cells only by drug or only by irradiation(P>0.05), but intense lethal effect was observed with drug and light combination(P<0.05). The phototoxicity of medicine increased with the elevation of concentration, the LD50 was 1.74 μmol/L, and reaching plateau at the concentration of 3.12 μmol/L, even increasing the concentration. LSCM found that drug localized in lysosomes of MGC803 cells. Hoechst staining showed that the death mode of cells was mainly necrosis and Annexin V/PI double-staining proved this result further.
CONCLUSIONThis new PDT drug is an effective PDT sensitizer for MGC803 cells and the death mode of cells is mainly necrosis.

9.Pleuropulmonary blastoma in an adult women with pleurorrhea as the major clinical manifestation: report of a case.

Pleuropulmonary blastoma (PPB) is a rare dysontogenetic neoplasm in children and has been recognized as a distinct clinicopathological entity different from the ordinary pulmonary blastoma of adulthood. We report a very rare adult case of PPB. A 43-year-old female patient presented with massive pleural infusion, and a misdiagnosis of pleural tuberculosis (TB) was made on the basis of ultrasound scan and CT images, for which therapy with TB drugs was administered for 3 weeks. Subsequent operation and pathologic examination of the mass revealed a multicystic neoplasm consisting of malignant mesenchymal cells immunohistochemically positive for vimentin and actin. Local recurrence occurred in the left pleural 20 months after the surgical tumor resection and 4 cycles of adjuvant chemotherapy.

10.Modified effect of modified danshou decoction on teratogenicity of bisphenol A intoxicated pregnant rats.

OBJECTIVETo explore the protective effect of modified danshou decoction (MDD) on teratogenicity of bisphenol A intoxicated pregnant rats.
METHODSForty-four successfully mated rats were randomly divided into 4 groups, 10 in the blank group and 10 in the model group, 12 in the MDD group and 12 in the positive control group. Bisphenol A (BPA) at the dose of 600 mg/kg was given to rats by gastrogavage in the latter three groups from the 1st day of mating to the 20th day, while the soybean oil was given to rats by gastrogavage in the blank group. No intervention was given to rats in the model group, but the normal saline, MDD condensed decoction, and shoutai pill (STP) condensed decoction was respectively given to rats in the rest three groups during the experimental period. All rats were sacrificed by the 20th pregnancy day.
RESULTSCompared with the model group, the body weight of pregnant rats and fetal rats, body length and tail length of the fetal rats significantly increased in the MDD group (P < 0.05). But the effect of MDD was superior to that of STP (P < 0.05). Moreover, the teratogenic rate was significantly lowered in the MDD group (P < 0.05).
CONCLUSIONMDD could promote the weight gaining of pregnant rats and fetal rats, improve the body length and tail length of fetal rats, and lower the teratogenic rate in fetal mice.
