Along with the progressing of micro-electronics technology and the improving of FPGA's performance, it's more and more common to process digital signals using FPGA. In most cases, FPGA is used to do the preprocessing or postprocessing besides processor in order to get a higher processing speed. This article introduces how to construct the signal processing unit of Doppler measurement system with FPGA and universal digital signal processor.

Objective To establish an expression system of TCR?9/?2-Fc protein by baculovirus vector expression system and identify biological function of expressed TCR?9/?2-Fc protein. Methods ?9Fc and ?2(OT3)Fc gene fragments were amplified by overlap PCR and inserted into expression vector pBACp10ph.The recombinant plasmid pBACp10ph-?9/?2(OT3)-Fc and the baculovirus DNA were co-transfected into sf9 cells. The expression position of TCR?9/?2(OT3)-Fc was identified by Western blot and the expression efficiency of ?9Fc and ?2(OT3)Fc was tested by flow cytometry(FCM).Furthermore, the binding activity of TCR?9/?2(OT3)-Fc protein with SKOV3 cells and MNS protein was evaluated with laser scanning confocal microslopy and surface plasmon resonance(SPR).Results The recombinant vector pBACp10ph-?9/?2(OT3)-Fc was constructed and TCR?9/?2(OT3)-Fc protein was expressed in sf9 cells. However, the expression efficiency of ?9Fc and ?2(OT3)Fc was quite different. It was proved that purified TCR?9/?2(OT3)-Fc protein can bind with SKOV3 cell and MNS protein. Conclusion TCR?9/?2-Fc protein is successfully expressed in baculovirus vector expression system and TCR?9/?2-Fc protein can simulate the binding activity of TCR in vitro.

ObjectiveTo determine the dispersion of atrial contraction time(A) and electromechanical time (P A) in different parts of atria among paroxysmal atrial fibrillation (PAF) patients by Doppler tissue imaging (DTI). MethodsDTI data were acquired in 58 PAF patients and 31 normal subjects. The DTI images of various parts were obtained by using apical 4 chamber view and sample volume was placed at the interventricular septum atrioventricular annulus, left lateral mitral annulus and right lateral tricuspid annulus, then P A and A were detected. The difference of P A and A between various parts of atria were calculated. ResultsThe dimension of left atria, right atria, systolic blood pressure and age in PAF group were much greater than those in controls. The difference of P A and A between left lateral mitral annulus and right lateral tricuspid annulus (T3,TT3) were significantly larger in PAF patients than in normal subjects[T3:( 24.27 ? 18.29 )ms vs ( 16.76 ? 10.96 )ms, P

Journal of International Oncology 2016;43(8):593-596
Objective To explore the expression of ErbB3 in osteosarcoma cell lines Saos-2 and its significance. Methods Real-time quantitative PCR was used to detected ErbB3 mRNA expression in osteosarcoma cell lines Saos-2 and normal human osteoblasts cell lines N704,and Western blotting was used to detected ErbB3 protein expression.Short hairpin RNA was used to construct ErbB3 knockdown cells and the cell count in 0-3 d was detected. The 48 h survival rates of ErbB3 knockdown cells and normal Saos-2 cells were detected in the presence of 0-100 μmol/L paclitaxel. Results Real-time quantitative PCR showed significant enhanced ErbB3 mRNA expression in Saos-2 cells compared with N704 [(4.15 ± 0.04) times,t =7.31,P <0.05],and Western blotting showed significant enhanced ErbB3 expression in Saos-2 cells compared with N704.As compared with normal Saos-2 cells,ErbB3 knockdown could reduce the Saos-2 cells proliferation,at the first three days in culture, the Saos-2 cell number of ErbB3 silent was (22.2 ± 2.9) thousand, and the control was (45.8 ± 4.1) thousand, with statistical significance (t =8.23,P < 0.05).Moreover,ErbB3 knockdown could reduce the Saos-2 cells tolerance to paclitaxel, when treated with 20 μmol/L paclitaxel, surviving cells in ErbB3 silent group was (43.2 ± 4.7) %,and the control was (61.4 ± 5.9) %,with statistical significance (t =6.74,P < 0.05).Conclusion ErbB3 highly expresses in Saos-2 cells,ErbB3 expression can enhance the proliferation of Saos-2 cells and the tolerance of Saos-2 cells to paclitaxel.

Objective To establish an expression system of TCRγ9/δ2-Fc protein by baculovirus vector ex-pression system and identify biological function of expressed TCRγ9/δ2-Fc protein. Methods γ9Fc and 82 (OT3) Fc gene fragments were amplified by overlap PCR and inserted into expression vector pBACp10ph. The recombinant plasmid pBACp10ph-γ9/δ2(OT3)-Fc and the baculovirus DNA were co-transfected into st9 cells. The expression position of TCRγ9/δ2 (OT3)-Fc was identified by Western blot and the expression efficiency of γ9Fc and δ2 (OT3) Fc was tested by flow cytometry (FCM). Furthermore, the binding activity of TCRγ9/δ2 (OT3)-Fc protein with SKOV3 ceils and MNS protein was evaluated with laser scanning confocal microslopy and surface plasmon resonance (SPR). Results The recombinant vector pBACp10ph-γ9/δ2(OT3)-Fc was constructed and TCRγ9/δ2(OT3)-Fc protein was expressed in sf9 ceils. However, the expression efficiency of γ9Fc and 82 (0T3) Fc was quite differ-ent. It was proved that purified TCRγ9/δ2 (OT3)-Fc protein can bind with SKOV3 cell and MNS protein. Conclu-sion TCRγ9/δ2-Fc protein is successfully expressed in baculovirus vector expression system and TCRγ9/δ2-Fc protein can simulate the binding activity of TCR in vitro.

OBJECTIVETo analyze the clinical characteristics of elderly patients with spinal tuberculosis and explore its clinical effects with anti-TB drugs alone.
METHODSFrom January 2008 to July 2010, the data of 36 patients with spinal tuberculosis underwent conservative treatment of anti-TB drugs alone were analyzed. There were 19 males and 17 females with an average age of 73.5 years (ranged, 60 to 85). All patients were in the active phase with high ESR and CRP levels and were treated with 3HRZE/6-9HRE (course from 9 to 12 months). According to clinical symptoms, chemical examination, radiological image to adjust drug and depending on VAS score to evaluate pain.
RESULTSAll the patients were followed up from 8 to 24 months with an average of 15 months. Tuberculose of 31 patients healed after chemotherapy from 9 to 12 months and ESR and CRP recovered normally. Levofloxacin and para-amino salicylic acid were used in 4 cases because of 4 cases occurred drug fast for RFP or INH, after 15 months, their obtained healing. Symptom of 1 case got worse during chemotherapy, and surgical treatment were performed, after 3 months, ESR and CRP recovered normally, X-ray and CT showed spinal osteosclerosis and fusion without significant kyphosis and internal fixation loosening. Cobb angle was respectively(17.6+/-2.3) degrees, (18.1+/-2.7) degrees before treatment and last follow-up (P>0.05). MRI showed abscess was absorbed and spinal inflammation subsidised. VAS score was respectively 6.5+/-1.7, 1.4+/-0.5 before treatment and last follow-up (P<0.05). Seven patients had complications relating with drug adverse reaction, after discontinuation and treated with clinical symptom, the patients recovered normally.
CONCLUSIONAnti-TB drugs alone can obtain satisfactory effects in treating early senile spinal tuberculosis, but strict supervision and individual administration should not be disregardful.
