Objective:To locate the region of antigenic epitopes recognized by monoclonal antibody(Z8) against human tumor necrosis factor ?(hTNF ?).Methods:Several recombinant fusion expression vectors were constructed which contained hTNF ? mutant genes deleted different parts respectively. After induction with IPTG, the fusion proteins were expressed and analyzed by SDS PAGE and Western blot.Results:Z8 specially recognized the fusion products which contained amino acids from 92 to 157 of hTNF ?, but not GST and its fusion protein with amino acids from 1 to 91 of hTNF ?.Conclusion:The antigenic epitopes recognized by Z8 were located in the sequence region 92 157 of hTNF ?.

3.Relation between animal models of RA and clinic

Animal models of rheumatoid arthritis(RA)especially adjuvant arthritis(AA)in rats and collagen induced arthritis(CIA)in mice have much character similar to human RA in pathological mechanism,clinical representation and so on. The models are more ideal in investigating RA and selecting drugs of anti inflammation and immunity. To discuss the pathological mechanism and the relation between animal models of RA and clinic will be important not only to research RA in pathology,physiology,nervous endocrine immunity and so on,but also to find out safety and effective drugs of treatment RA. The articl summarizes several animal models of RA in establishment,pathological and clinical changes,inflammatory immune regulation and so on.
4.Effects of selenium-enriched yeast on DTH in immunosuppressive mice

The effects of selenium-enriched yeast (Sey) on DTH in immunosuppressive mice have been investigated. It was found that Sey (30, 60, 90 mg?kg-l?d-1?6 d, ig) was able to antagonize lowered DTH by cyclophospha-mide (Cy); Meanwhile, the thymus and spleen weights were increased by Sey (30, or 60 mg?kg-1?d-1) in immunosuppressive mice. Sey also potentiated the lowered ConA and LPS-indu-ced proliferation of splenocytes in DTH mice at the doses of 30, 60, or 90 mg ?kg-1 and 60 or 90 mg?kg-1 respectively. These results suggested that Sey could enhance the functions of lymphocytes.
5.Effect of positive pressure ventilation on radiosensitivity of human pr imary lung cancers

Objective To investigate the effect of positive pressure ventilation on radiosensitivity of the patients suffering from primary lung cancer. Methods Thirty cases of lung cancer were randomly divided into two groups: ① combining therapy group: patients treated with posi tive pressure ventilation using BiPAP respirator and radiotherapy;② simple radi otherapy group. The changes of PaO2, SaO2, blood white cells and the cellula r immunological function were observed before and after treatment. Resul ts Nasal or naso-facial positive pressure ventilation with BiPAP respi rator increased PaO2 significantly with the maximum of 7.5 kPa, SaO2 was ma inteined at above 95%. No significant change for the cellular immunological func tion was found in the combining therapy group (P>0.05)and only one patient with leukopenia(6.7%). But in the radiotherapy group, the lymphocyte transfor mation efficiency and the ratio of CD4/CD8 were obviously decreased(P<0 .01), and leukopenia was found in 5 cases(33.3%). Conclusion Nasal or naso-facial positive pressure ventilation with BiPAP respirator could improve the effect of radiotherapy for lung cancers. It could also increase PaO 2 significantly and has protective rote to the marrow and cellular immunologic al function of the patients.
6.Clinical observation of intervention of group B streptococcus infection in gestation period

ObjectiveTo observe the effection of different interventions of group B streptococcus (GBS) infection in gestation period.MethodsOne hundred and seventeen cases with GBS infection were obtained from 1885 pregnant women,who were got routine prenatal examination at 34 to 37 weeks of pregnancy,and drug seeitivity test of secretions which were taken from under paragraph 1/3 of vagina,and divided into treatment group (91 cases) and untreatment group (26 cases).The treatment group was divided into treatment group one (47 cases) and treatment group two (44 cases).Treatment group one was treated with oral antibiotics for 7 days after diagnosis,treatment group one and two were treated with postpartum antibiotics intravenous infusion once every 4 hours in labor.Comparison of maternal and fetal outcomes.ResultsThe GBS infection rate was 6.2% (117/1885).The morbidity of premature delivery,premature rupture of membrane and neonatal infection of treatment group [ 5.5% ( 5/91 ),13.2% ( 12/91 ),5.5% (5/91 ) ]were lower than those of untreatment group [ 19.2%(5/26),30.8%(8/26),23.1%(6/26) ](P < 0.05).The morbidity of premature dehvery and premature rupture of membrane of treatment group one[ 0,6.4% (3/47)]were lower than those of treatment group two[ 11.4% (5/44),20.5% (9/44)](P < 0.05).Conclusion Anti-GBS treatment can improve the outcomes of mothers and infants,especially early anti-GBS treatment during the period of pregnancy.
8.Ancient LiteratureAnalysis of Point Shenmai Indications and the Characteristics of Acupuncture and Moxibustion

ObjectiveTo summarize point Shenmai(BL62)indications and the characteristics of acupuncture and moxibustion by sorting out literature on point Shenmaiin ancient literature.MethodShenmai, Yangqiao and Guilu were used as search words to search Chinese Medical Classics to sift out items on indications for point Shenmai and its combination and the characteristics of point combination and acupuncture and moxibustion and establish the relevant database. Statistical analysis was made using the SPSS19.0 software.ResultTwo hundred and eighty-seven items on point Shenmai and 274 items on the indications met the inclusion criteria. There were 47 indications for the single acupoint and 19 kinds of diseases on which the single acupoint had a marked therapeutic effect. Lumbago, ophthalmalgia, vertigo, headache and mania were ranked in the top five. There were 64 indications for its combination and 22 kinds of diseases on which its combination had a marked therapeutic effect. Epilepsy, Lumbago, stroke, podalgia and headache were ranked in the top five. The commonly used depth of needle insertion was 3 fen, acupuncture time was 6-7 respiratory cycles and stimulation intensity of moxibustion was 3 moxa cones.ConclusionSingle point Shenmai and its combination are both good at treating internal medicaldiseases and especially meridian diseases in the limb. Acupuncture and moxibustion are the commonly used methods.
9.Clinical application of the modified open-door laminoplasty perserving semispinalis cervicis insert into axis

[Objective]To verify whether or not the modified open-door laminoplasy preserving semispinalis cervicis insert into axis is effective in preventing postoperative axial symptom and maintaining postoperative cervical sagittal alignment.[Method]Since March 2002 to December 2003,forty-eight patients underwent modified open-door laminoplasy preserving semispinalis cervicis insert into Axis,among them,thirty-two patients who were followed up for more than 2 years were included in this study.Preoperative and postoperative JOA score,degree of axial symptom,ranges of neck motion,cervical curvature index were recorded and compared.[Result]Preoperative and postoperative JOA score was(9.04?2.9)and(12.5?3.1)respectively,with recovery rate as(44.9?26.9).There was significant difference between preoperative evident axil symptom rate(46.8%)and postoperative evident axil symptom rate(18.7%),while no significant difference between preoperative and postoperative ranges of neck motion,cervical curvature index was seen.[Conclusion]Modified open-door laminoplasy is less invasive to the posterior extensor mechanism especially the semispinalis cervicis than conventional open-door laminoplasy.This new procedure is effective in preventing postoperative morbidities often seen after conventional laminoplasy with adequate decompression of the spinal cord.
10.Impact on cervical axial symptom of modified open-door laminoplasty with C_3 laminectomy

Objective To verify whether the modified open-door laminoplasty with C3 laminectomy is effectve in preventing postoperative axial symptom. Methods Since March 2001 to March 2005, 44 patients who underwent modified open-door laminoplasty with semispinalis cervicis insertion intact were included in this study(group A). 50 patients who underwent conventional open-door laminoplasty in the authors' department during the same period were served as control(group B). Preoperative and postoperative Japanese Orthopedic Association(JOA) scores, the degree of axial symptom, ranges of neck motion, cervical curvature index were recorded and compared. Results The average JOA recovery rate was 59.2%?11.3% for group A and 60.1%?19.5% for group B. There was no significant difference in JOA recovery rate between two groups. The rate of patients with evident axial symptoms was 22.7% in group A and 54.0% in group B, and the difference was statistically significant(P
