1.Research on the antibacterial effect of fresh pepper leaves

ObjectiveTo explore whether fresh pepper leaves have antibacterial activity or not in vitro test and to make clear their inhibition degree.MethodsTo detect the inhibitory effect and the minimal inhibitory concentration of fresh pepper leaves in vitro by agar disk diffusion method and tube dilution method.ResultsThere was bacterial growth phenomenon in all of the reaction tubes of staphylococcus saprophyticus and no inhibitory effects on agar disk.Significant inhibitory phenomenon both on agar disks and in more than two reaction tubes of Staphylococcus aureus,Staphylococcus epidermidis,Escherichia coli,Proteus vulgaris,Shigella and Haemophilus influenzae was observed.ConclusionFresh pepper leaves had obvious inhibitory effect for the growth of common intestinal bacteria,pyogenic cocci,especially for Haemophilus influenzae.
2.Antimicrobial resistance in a class A tertiary hospital in Baise, Guangxi Province: 2015 surveillance report

Objective? To?understand?the?distribution?and?antibiotic?resistance?profile?of?clinical?isolates?in?the?Affiliated?Hospital?of Youjiang Medical University during 2015. Methods The pathogenic microorganisms were isolated from inpatients from January to?December,?2015?in?the?Affiliated?Hospital?of?Youjiang?Medical?University.?Antimicrobial?susceptibility?testing?was?carried?out?according?to?a?unified?protocol?using?Kirby-Bauer?method?or?VITEK?2-Compact?systems.?The?results?were?interpreted?according?to CLSL breakpoints released in 2014, and analyzed by WHONET 5.6 software. Results A total of 4004 strains of bacteria were collected, including 1146 (28.6%) strains of gram-positive bacteria, and 2858 (71.4%) strains of gram-negative organisms. The?prevalence?of?MRSA?and?MRCNS?was?25.8?%?and?79.4?%,?respectively.?MRSA?and?MRCNS?strains?were?significantly?more?resistant to most antibiotics than MSSA and MSCNS except trimethoprim-sulfamethoxazole. Most of the S. pneumoniae isolates were from non-meningitis patients, showing high resistance rate to macrolides and tetracycline, but very low resistance rate to quinolones. Enterococcus isolates were mainly E. faecium and E. faecalis. More E. faecium were resistant to high-level gentamicin and high-level streptomycin than E. faecalis. E. faecium isolates were generally more resistant than E. faecalis to most of the antimicrobial agents tested except clindamycin and tetracyclines. But no gram-positive cocci were found resistant to vancomycin, linezolid or tigecycline. ESBLs-producing strains accounted for 53.1% of the E. coli strains and 28.5% of K. pneumoniae isolates. Enterobacteriaceae isolates were still very susceptible to carbapenems. E. coli isolates were more resistant to most of the antimicrobial agents tested than other Enterobacteriaceae except to piperacillin-tazobactam and carbapenems. Enterobacteriaceae showed higher resistance to trimethoprim-sulfamethoxazole than the other antibiotics tested. Majority of P. aeruginosa isolates were susceptible to all the antibiotics tested (<10% resistant). A. baumannii?strains?showed?significantly?higher?resistance rate than P. aeruginosa to all the antibiotics tested. Conclusions Most of the data in this report are consistent with the national?data?in?terms?of?antimicrobial?resistance?profile.?These?data?are?useful?for?rational?use?of?antibiotics.
4.Effects of exogenous transforming growth factor-β3 on the expression of endogenous transforming growth factor-β3 and proliferation of hepatic stellate cells

Objective To investigate the effects of exogenous transforming growth factor-β3 (TGF-β3) on the expression of endogenous TGF-β3 and proliferation of rat hepatic stellate cells (HSC).MethodsRat HSC cells were seeded in 24-well plates and were divided into 2 groups.One group of cells were exposed to TGF-β3 of different concentrations (0.08,0.4,2,10 and 50 ng/ml) for 2 hours and then the cell culture supernatant was collected; the other group of cells were exposed to 10 ng/ml TGF-β3 and the cell culture supernatant was collected at different time point (0.25,0.5,1,2,4,8 and 13 h).The content TGF-β3 was determined by ELISA method.Some other HSC cells were seeded in 96-well plates and divided into 2 groups.One group of cells were cultured in the presence of exogenous TGF-β3 of different concentrations (0.001,0.005,0.02,0.08,0.32,1.25,5,20,100,500 ng/ml) for 24 hours and then the cell proliferation was detected; the other group of cells were treated with 5 ng/ml TGF-β3 for 24 and 48 hours.The cell proliferation was measured by MTT method.The HSC cell morphology of control group and TGF-β3 treated group was observed under inverted microscope. Results The endogenous TGF-β3 expression of HSC cells obviously increased after exogenous TGF β3 treated at 2 ng/ml and reached the peak at 3 hour [(0.845±0.028) ng/ml vs (0.026±0.021) ng/ml,F=210.168,P=0.00].Low concentrations of exogenous TGF-β3 did not affect the proliferation of HSC cells.Above 0.32 ng/ml,exogenous TGF-β3 could promote HSC proliferation.There was no dose-dependent relationship between HSC cell proliferation and the concentration of exogenous TGF-β3 (F=0.68,P=0.57).Under microscope,lhere was no significant difference in HSC cell morphology between control group and TGF-β3 treated group.Conclusions Exogenous TGF-β3 can promote the expression of endogenous TGF-β3 in HSC cells,and can promote HSC cell proliferation.There is no obvious effect of exogenous TGF-β3 on HSC morphology.
5.Drug Sensitivity and Identification of 393 Strains of Candida

OBJECTIVE To get the message of drug sensitivity and characteristic of Candida infecting patients in our hospital. METHODS The CHROMagar candida color medium was applied to isolate and identify Candida strains.Drug sensitivity was analyzed by the K-B slip diffusion method. RESULTS Totally 393 strains of Candida in our hospital were isolated in the three years,including Candida albicans;C.tropicalis;C.glabrata;(C.krusei);Cryptococcus neoformans and the others;and their infection rate was 63.10%,12.47%,9.41%,8.40%,1.53%,and 5.09%,respectively.Among all strains,156(39.69%) strains were isolated from sputum,(66(16.79%))from stool,45(11.45%) from urine,38(9.67%)from leucorrhea,and 34(8.65%)from throat swabs.Sensitivity results to nine antifungals showed that 5-flucytosine,fluconazole,clotrimazole,amphotericin B,and nystatin had higher drug sensitivity to these fungi. CONCLUSIONS Isolated rate of Candida is higher from clinical samples.Drug sensitivity of different Candida to various antibiotics is different.Culturing and drug sensitivity of Candida should be put on important place and antibiotics should be used reasonably based on drug sensitive test to reduce drug resistance.
6.Changes in expression of sodium channel?subunit mRNA in dorsal root ganglion in a rat model of neuropathic pain

Objective To examine the changes in the expression of voltage-gated sodium channel?subunit mRNA in the dorsal root ganglion(DRG)and the role it plays in the neuropathie pain.Methods Thirty- two male SD rats weighing 250-400g were randomly divided into 2 groups:groupⅠneuropathic pain(SNL,n= 20)and groupⅡsham operation(n=12).Neuropathic pain was produced by ligation of right sciatic nerve according to Seltzer.Paw withdrawal latency to noxious thermal(PWHL)and mechanical(PWML)stimulation were measured before(baseline)and 1,2,3,5,8,11,14,28 day after sciatic nerve ligation(SNL).DRG at L_(4,5) was isolated on the 14th day after SNL in 8 SNL and 4 sham-operated animals for determination of sodium channel?subunit mRNA expression(by in-situ hybridization).Results PWHL and PWML were significantly decreased on the 2nd-28th day after SNL as compared to the baseline in SNL group.There was no significant difference in?_1 subunit mRNA expression between the 2 groups.The?_2 subunit mRNA expression in DRG was hardly detectable.The?_3 subunit mRNA expression in DRG on the operated side was significantly higher in SNL group than in sham-operation group.Conclusion The up-regulation of sodium channel?_3 subunit mRNA expression in DRG may play an important role in neuropathic pain.
7.Subclinical atherosclerosis in patients with rheumatoid arthritis

Objective To evaluate the prevalence of subclinical atherosclerosis in rheumatoid arthritis (RA) and the related risk factors.Methods Fifty RA patients without overt atherosclerotic disease and 121 control subjects matched for age and sex were recruited.Duplex carotid sonography was used to measure intima-media thickness (IMT) and plaque formation assessment.Differences between RA and the control group were compared, and the risk factors were explored.RA patients then were divided into two subgroups according to IMT and the comparison between the two subgroups were completed.T test, Mann-Whitney U test, chi-square test, Pearson's correlation and Logistic regression analysis were used for statistical analysis.Results Although RA patients had lower level of serum lipids and body mass index than the control group, the mean IMT value was significantly higher in the RA group than that in the control group [(0.78±0.18) mm vs (0.62±0.14) mm, t=5.853, P=0.000], and plaque formation was more prevalent [56.0%(28/50) vs 36.4% (44/121),x2=5.596, P=0.018].The difference was especially significant in the younger groups (<50 years old group and 50-60 years old group).Logistic regression showed that the presence of RA [OR=7.34, 95%CI (2.53, 21.25)], male [OR=2.0, 95%CI (91.25, 3.17)] and old age [OR=1.1, 95%CI (1.04, 21.15)] were the independent risk factors for abnormal IMT (thickened or the presence of carotid plaques).The RA patients were divided into two subgroups according to IMT.Compared with patients with normal IMT, patients with abnormal IMT were older and more common in postmenopausal, and had longer RA duration and higher cholesterol level.In treatment, less patients with abnormal IMT had been taking methotrexate (MTX) for more than 3 months compared with patients with normal IMT.Among these parameters, old age [OR=1.13, 95%CI (1.03, 1.23)] was shown to be the independent risk factor for abnormal IMT in RA patients, and more than 3 months of MTX treatment [OR=0.12, 95%CI (0.02, 0.71)] was the protective factor.Conclusion Atherosclerosis occurs frequently and prematurely in patients with RA and the presence of RA is an independent risk factor for atherosclerosis.Early primary prevention for atherosclerosis should be recommended.MTX probably has a positive effect on preventing atherosclerosis for RA patients, which needs to be confirmed by further study.
8.Investigation on the current status of oral health care in the disease controls system in Sichuan Province.

OBJECTIVETo analyze the status and characteristics of dental manpower in the center for disease controls (CDC) in Sichuan Province and to provide more evidence for strengthening the oral healthcare workforce in the CDC system.
METHODSA mass survey on dental manpower was made in CDCs in Sichuan Province through questionnaire investigation. Data were collected and entered with the Epidemiological Dynamic Data Collection (EDDC) platform and analyzed with SPSS 13.0 software.
RESULTSSichuan Province had 0.15 hospitals providing oral health services and 0.38 dentists on average per 10,000 people. About 65.53% (135/206) of the CDCs had one department responsible for the oral health service. However, oral health care personnel comprised only 2.23% (237/10,624) of the personnel of the whole CDC system. About 64.67% (119/184) of county CDCs and 47.62% (10/21) of city CDCs knew well the dental health status of local residents. Less than 5% of the CDCs used the data and assisted in the policy making of public health administrators.
CONCLUSIONThe dental care personal deficit exists in the CDC system in Sichuan Province. The distribution and composition of dental manpower are not reasonable. The oral health service ability of CDCs in Sichuan Province should be strengthened and improved.

9.EXPRESSION OF L-N-CARBAMOYLASE GENE IN PICHIA PASTORIS

N-carbamoylase is a part of hydantoinase operon which can transform N-carbamoylamino acid to corresponding ammo acids. The L-N-carbamoylase of Arthrobacter BT801, codied by the HyuC gene, is the rate-limiting and the only stereoselective enzyme. HyuC DNA fragment was amplified by PCR from the plasmid of pUC18-169. The target fragment was introduced into pPIC3. 5K plasmid to construct the pPIC3. 5K-hyuC expressing vector which was then transduced into Pichia pastoris GS115 cells after being linearized by BglⅡ digestion. Multi-copies insertion transformants were screened on G418 plates. The recombinant protein was proved to have biological activity of hydrolyzing N-carbamoylphenylalanine into phenylalanine through enzyme activity determination.

Objective To study the mRNA expression of HS1-associated protein X-1(Hax-1),an an- ti-apoptosis genc,in the peripheral blood mononuclear cells(PBMC)of patients with systemic lupus erythe- matosus(SLE),and further investigate the roles and significance of Hax-1 in the pathogenesis of SLE.Meth- ods Generation of longer cDNA fragments from serial analysis of gene expression(SAGE)tags for gene identi- fication(GLGI)was applied to identify the gene Hax-1 according to the Long SAGE tag.Then reverse tran- scription-polymerase chain reaction(RT-PCR)technique was used to semiquantitatively analyze mRNA ex- pressions of Hax-1 in PBMC from 34 active SLE patients and 25 healthy subjects.Results Compared with healthy controls,there was significant difference between SLE patients in the active stage and the normal controls(Z=-4.556,P<0.01).The average level of mRNA expression in active SLE group was higher than that in healthy controls.Significant difference was found between the group with mild SLE and either the moderate or the severe one(P<0.01).Conclusion The mRNA expression level of Hax-1 in active SLE group increase markedly,and to some extent,it is related to the activity of SLE.This provides a valuable basis for the further study on the role of apoptosis in SLE.
