[Objective] To compare the effects of Radix Astragali (RA) and triperygium glucosides (TG) on glomerular mesangial cells (GMC) proliferation and interleukin-6 (IL-6) secretion. [Methods] GMC were cultured with in-vitro cell culture technology. Serum containing RA and serum containing TG were prepared with serum pharmacological methods. GMC proliferation was detected by methylthiazolyltetrazolium (MTT) assay and IL-6 level by enzyme-linked immunosorbent assay (ELISA) to evaluate the effect of low-, moderate- and high-dosages of RA-containing serum and TG-containing serum under the condition of GMC over-proliferation. [ Results] Different dosages of RA-containing serum and TG-containing serum inhibited GMC proliferation and IL-6 secretion in a dose-effect manner, and the inhibitory effect of high- and moderate-dosage TG and high-dosage RA was stronger. [Conclusion] Under the condition of in-vitro GMC over-proliferation, RA has the similar inhibitory effects on GMC proliferation and IL-6 secretion with TG. The combined treatment of RA and TG can reduce the dose of TG and probably has an action in increasing effect and reducing toxicity.

Objective To study effects of fluid resuscitation on thoracoabdominal injury combined with hemorrhagic traumatic shock.Method A total of 98 patients,who were treated in Affiliated Zhongshan Hospital of Dalian University from November 2004 to December 2006,were retrospectively analyzed.The patients were diagnozed according to Surgery(fifth edition).Patients were divided into delayed fluid resuscitation group(n= 51)and immediate fluid resuscitation group(n=47).Patients in delayed fluid resuscitation group were given with balanced salt solution for the body to maintain basic requirements.Patients in immediate fluid resuscitation group were rapidly administered with a lot of isotonic crystaUoid and(or)colloid solution after admission. Hemoglobin,platelet count,hematocrit,blood lactic acid,basedeficit,preoperative resuscitation time and mortality were compared between the two groups.Paired t test and variance analysis or x~2 test were used.Results The transfusion fluid volume of delayed group and immediate group was(1586?346)ml,(3520?575)ml, respectively,with P value

OBJECTIVETo explore factors related to pandemic influenza A (H1N1) virus infection among healthcare workers.

METHODSH1N1 influenza confirmed cases of health workers in hospital of Beijing from Aug 30th 2009 to Jan 31st 2010 were included. A 1:4 matched case-control study was conducted with 54 healthcare workers who were infected by influenza A(H1N1) virus and 216 matched controls who were not infected. Face-to-face interview with questionnaires was used to collect information of work and family aspects of the study participants. Conditional logistic regression was used to analyze the H1N1 infection factors.

RESULTSThe age was 29.6 ± 7.4 years old and male subjects accounted for 17.4% (47/270). There were 5.6% (3/54) and 34.4% (74/216) of the cases get the influenza A (H1N1) vaccine immunization separately in the case group and control group. Among 18.5% (10/54) and 40.1% (88/216) of the cases and controls used high protection level masks during the epidemic. Besides, 33 (61.1%)and 161 (74.5%) cases lengthening the time of mask wearing separately. There were 13 (24.1%) and 85 (39.4%) cases using disposable tissue to clean hands in the case group and control group, 24 (44.4%) and 46 (21.3%) cases feel they were much easier to get infected in respiratory disease than others based on experience in the case and control group. Univariate analysis showed that factors such as using disposable tissue to clean hands (OR = 0.15, 95%CI = 0.04 - 0.57), lengthening the time of mask wearing (OR = 0.43, 95%CI = 0.20 - 0.92), using high protection level masks (OR = 0.26, 95%CI = 0.11 - 0.58), getting influenza A (H1N1) vaccine immunization (OR = 0.04, 95%CI = 0.01 - 0.32), much easier to get infected in respiratory disease than others based on experience (OR = 2.85, 95%CI = 1.44 - 5.62), were all associated with influenza A (H1N1) infection. Results of multivariate logistic regression analysis showed that variables such as history of influenza A (H1N1) virus immunization (OR = 0.18, 95%CI = 0.06 - 0.51), using high protection level masks (OR = 0.05, 95%CI = 0.01 - 0.35), much easier to get infected in respiratory disease than others based on experience (OR = 3.69, 95%CI = 1.58 - 8.63) were all correlated to influenza A (H1N1) infection.

CONCLUSIONSFactors such as history of influenza A (H1N1) vaccine immunization, using high protection level masks and improving respiratory health can protect healthcare workers from infection of influenza A (H1N1).

Objective To investigate the relationship of CAG repeat length polymorphism in the androgen receptor(AR)gene to the development of acne.Methods A total of 238 patients with ache vulgaris and 207 healthy human controls in Northeast China were included in this study.Genomic DNA was isolated and purified from the blood of these subjects.The CAG repeat lengths in the AR gene were analyzed by somatic microsatellites (STRs).Results A significant difference was found in the CAG repeat number between the male acne Patients(22.70±3.09)and male controls(23.48±2.83,P=0.046),but not between the female cases and controls(23.41±2.87 versus 23.85±0.21.P=0.12).In order to assess the risk associated with CAG repeats,the male and female subjects were dichotomized based on the median repeat length in the corresponding control group as the arbitrary cut-off point.Those men and women with a short CAG repeat length(＜23 in men,and＜24 in women)had a significantly increased risk for agne than those with a long CAG repeat length(men:95%confidence interval,1.21-3.54,OR=2.07,P=0.008;women:95%confidence interval.1.18-3.56,OR=2.05,P=0.01).Conclusions This study strongly indicates that the CAG repeat length in AR gene is associated with the development of acne in Northeast China,and those men with a short CAG repeat length seem to have a high risk for ague.Consequently,CAG repeat length may serve as a genetic susceptibility marker.

OBJECTIVETo determine the serum level and mRNA expression of type-1/type-2 cytokines of T lymphocytes in spleens of rats after thermal injury and to investigate the effects of escharectomy during burn shock stage on IFN-gamma and IL-4.

METHODSOne hundred and sixty male Wistar rats were randomized into four groups. In group A, animals were not subjected to escharectomy. In groups B, C and D, escharectomy and skin allograft were performed at 8, 24, 96 hours postburn (PB) respectively. At 4, 12, 24, 48, 96, 120 and 168 hours PB, animals were killed and blood and spleens samples were harvested. ELISA was applied to determine the concentration of IFN-gamma and IL-4 in serum. The expression pattern of IFN-gamma and IL-4 were observed at mRNA level in T lymphocytes isolated from spleen by RT-PCR.

RESULTSThe serum level of IFN-gamma and IL-4 rose rapidly and significantly after scald injury, expression of IFN-gamma and IL-4 mRNA in rats' T lymphocyte were also up-regulated spontaneously. The serum level of IFN-gamma and its mRNA expression began to rise within 4 hours PB, peaking at 24 hours PB. Whereas IL-4 and its mRNA expression showed a persistent elevation. Thereby leading to a dominant tendency of Th2 cytokine response on 7 d PB. In group A all above parameters revealed most obvious changes compared with controls, then ranked in group D, B and C.

CONCLUSIONEscharectomy during burn shock stage is helpful to decrease the harmful over expression of Th2-type lymphocyte after severe thermal injury.

OBJECTIVETo study gene expression profiling in human type I and II endometrial carcinoma.

METHODSSix Affymetrix human genome genechips were utilized to investigate the differences in gene expression profiles between type I and II endometrial carcinoma with bioinformatic analysis.

RESULTSMany genes were highly expressed in estrogen-dependent endometrial carcinoma, and some of them were involved in the metabolism and conversion of estrogen, while some others in estrogen regulation. CYP2C9, for instance, was involved in the conversion of estrogen sulfate to 16-hydroxy sulfate metabolite, DDC in estrogen-dependent pathogenesis of endometrial carcinoma possibly by DDC interaction with AR to enhance steroid receptor transcription.

CONCLUSIONHigh expression of these genes in estrogen-dependent endometrial carcinoma may provide insights into their roles in the pathogenesis and prognosis of this malignancy.

Objective To explore the risk factors for septicemia after cesarean section,and provide reference for clinical prevention of postoperative septicemia.Methods Clinical data of patients who underwent cesarean section in a maternal and child health hospital between January 1,2013 and October 31,2016 were collected by retrospective survey method,risk factors were analyzed by multivariate logistic regression model.Results A total of 4 604 cases of cesarean section were selected,32 cases of septicemia occurred,incidence was 0.70％.Multivariate logistic regression analysis showed that there were seven independent risk factors for septicemia:gestational diabetes mellitus (OR =4.03),trying vaginal delivery(OR =15.86),No.of vaginal examination ≥3 times(OR =6.77),premature rupture of membrane≥12 hours(OR =3.47),intra-operative bleeding≥1 000 mL(OR =4.66),postoperative duration of indwelling urinary catheter≥24 hours(OR =2.83),and antimicrobial use within a week(OR =3.20).Four factors were protective factors:gestational weeks≥34 weeks(OR =0.20),hemoglobin≥100 g/L(OR =0.40),albumin≥35 g/L(OR-0.28),and amniotic fluid volume at a normal level(OR =0.22).Conclusion It is possible to prevent and control the occurrence of septicemia after cesarean section through strict management of independent risk factors and intervention in protective factors of pregnant women during peri-operative period.

Objective: To compare different specimen types of lung adenocarcinoma in the detection of epidermal growth factor receptor (EGFR) gene and to correlate EGFR mutations with patient clinical features. Methods: One hundred lung adenocarcinoma cases were collected from June to December in 2015, at the First Affiliated Hospital of Xinjiang Medical University.Of the 100 lung adenocarcinoma samples, 43 were male and 57 were female. The age was from 40 to 88 years old, and the average age was 66 years. One hundred lung adenocarcinoma cases were divided equally into two groups. Mutation analysis of EGFR gene by real-time PCR was performed using biopsied tissue and paired blood samples in one group (n=50) and using pleural effusion and paired blood samples in the other group (n=50). Results: The mutation rate of EGFR gene in biopsy samples was 54% (27/50) , higher than that of blood samples (46%, 23/50), but without statistical differences (χ²=0.640, P=0.424). In contrast, mutation rate of EGFR gene in pleural effusion samples (42%, 21/50) was higher than that of blood samples (34%, 17/50), but without statistical differences(χ²=0.679, P=0.409). Two patients had EGFR mutation detected in paired blood samples but not in the corresponding biopsy samples, and four patients had EGFR mutation detected in pleural effusion samples but not in their paired blood samples. The mean progression-free survival of patients with detectable EGFR mutation were 9.5 months (tissue samples), 8.6 months (pleural effusion) and 8.5 months (blood). However, there was no statistical difference. Conclusions Blood samples may be used to assess EGFR mutations for patients with lung adenocarcinoma. However, further studies are needed to improve the sensitivity and accuracy in the detection of EGFR mutations using blood samples.

Persistent baculovirus infection is observed frequently in insect populations. Persistent infection can be transformed to a replicative and infective state caused by stress factors and plays an important role in regulating the size of insect population and in epizoology of baculoviruses. The aim of this study is to establish a persistently baculovirus-infected cell system to explore the molecular mechanisms of baculoviral persistence. Spodoptera exigua nucleopolyhedrovirus (SeMNPV) was serially undiluted passaged in Se301 cells to reduce virulence. Upon infection of Se301 cells with the SeMNPV up to passage 8, a few cells survived even if most of cells died due to virus infection. The surviving cells were passaged and designated as P8-Se301 cell strain. P8-Se301 cells had a population doubling time of 58-65 hours and grew slower than Se301 cells. Light microscopy and electron microscopy observation showed symptom of baculovirus infection, such as virogenic stroma, viral particles and occlusion bodies, in some of P8-Se301 cells. End-point dilution assay and infectious center assay showed that 4.14% +/- 0.99% cells continually released infectious progeny virus which replicated slower than SeMNPV in Se301 cells. The result indicated that P8-Se301 cells show a typical character trait of baculovirus persistent infection.
