1.The Clinical Use of Double-paper Synergy Test in the Detection of Bacteria Producing Extended-spectrum β-lactamase

The optimal condition of double-paper synergy test in the detection of bacteria producing extended-spectrum β-lactamase (ESBLS) was studied. In 244 isolates of Enteric bacilli,Klebsiella pneumonia,Enteric cloacae,there were 49 ESBLS-producing strains with the positive rate being 19.67 %. Five ESBLS-producing strains were detected in Pseudomonas maltophilla. One ESBLS-producing strains was detected in Pseudomonas aeruginose,Enteric aerogenes and Citrobacter,respectively. The optimal substrates of double-paper synergy test were cefotaxime,aztreonam and cefepime,and the interspace was 25 mm. This method was still a simple,economical and appropriate method in the detection of ESBLS-producing isolates.
2.Detecting chlamydia psittaci DNA in patients with ocular adnexal lymphomas

Objective:To detect the chlamydia psittaci DNA in patients with ocular adnexal lymphomas,and find out the relationship between chlamydia psittaci infection and ocular adnexal lymphomas.Methods:Tissue samples from 36 patients with ocular adnexal lymphomas were collected,as well as samples from patients with benign lymph disease and patients with eye trauma as controls.Chlamydia psittaci DNA were detected by PCR.Results:The chlamydia psittaci DNA positive rates in patients with ocular adnexal lymphomas,benign lymph diseases and ocular trauma organizations were 47.2%,5.0%,0.0%(?2 = 24.578,P=0.000 1),respectively.The DNA positive rates in patients withⅠ A period were 52.0%,36.4% of Ⅱ A period(?2 =0.749,P=0.387).5,8-year survival rates of all the cases were 90.1%,78.5%,which were the same in Chlamydia psittaci DNA positive group and negative group(?2 = 0.163,P= 0.687).Conclusion:The incidence of ocular adnexal lymphomas may be related with Chlamydia psittaci infection.But there is no evidence of the relationship with Chlamydia psittaci infection and of the ocular adnexal lymphomas stage and prognosis.
3.Preparation of total isomalabaricanes and determination of Stelletin B and Stellettin D in Stelletta tenuis Lindgren by high pressure liquid chromatography

Objective:To establish a method for determining the contents of Stellettin B and Stellettin D in total isomalabaricanes in different batches of Stelletta tenuis Lindgren.Methods: A high pressure liquid chromatography(HPLC) method was applied.The column YMC Pack-SIL(250 mm?4.6 mm,5 ?m) was used at room temperature,and the mobile phase was n-hexane∶ethyl acetate(75∶25) with a flow rate at 1.0 ml?min~(-1) for 30 min.UV detector wave length was at 360 nm.Results: The regression equation were:Y=52 181 633 X+957 495(r=0.996 9)for Stellettin B and Y=62 636 828 X-451 022((r=0.998 0)) for Stellettin D,with the linear range being 2.4-30 ?g and 2.0-32 ?g,correspondingly.The RSD values for accuracy test,stability test and repeatability test were 3.07%-4.76%.The average contents of Stellettin B and Stellettin D in total isomalabaricanes in different batches of Stelletta tenuis Lindgren were 59.93% and 30.98%,respectively.Conclusion: This HPLC method provides a fast,accurate and effective way to determine the active contents in total isomalabaricanes from Stelletta tenuis Lindgren.
4.Inhibitory effect of recombinant fibronectin polypeptide CH50 on invasion and metastasis of melanoma B16 cells.

In order to investigate the inhibitory effect and mechanism of recombinant polypeptide CH50 on invasion and metastasis of melanoma B16 cells, the recombinant polypeptide CH50 was separated and purified by ion exchange chromatographic technique. The melanoma B16 cells treated with purified CH50 were cultured in vitro, the number was counted at 4, 24, 48 and 72 h and their morphological changes were observed in order to detect their adhesion and spreading abilities. In in vivo study, the melanoma B16 cells were labeled with CFSE and treated with CH50 and then they were injected into mice via mouse-tail veins. After 5 h, the lung tissues were fixed by frozen section. Accumulation and invasion abilities of B16 cells on lung tissues were observed under the fluorescent microscopy. The results showed that the morphological character of B16 cells treated with CH50 changed greatly and the number of B16 cells treated with CH50 decreased significantly (P<0.05). The adhesion and spreading abilities of B16 cells treated with CH50 were weakened obviously and the metastasis foci on lung tissues reduced. It was concluded that the recombinant polypeptide CH50 inhibited invasion and metastasis of melanoma B16 cells on tissues and could be a prospective bio-product in tumor general therapy.
5.Training of cardiopulmonary resuscitation skill should be implemented from primary and middle school

In view of the increased incidences of cardiac arrest outside hospital year by year and younger tendency of the patients,foreign medical education practice showed that school-age children accepting cardiopulmonary resuscitation (CPR) skill training will benefit most from this survival skill in the future.This article elaborated on the meaning and methods of promoting CPR training in primary and middle school based on the present situation of CPR in order to promote its further development.
6.Analysis of chromosome variation of lymphocytes and comparative genomic hybridization of lung carcinoma in patients with long survival after operation

Background and Purpose:To analyse the chromosome of peripheral lymphocytes and tissue samples of lung cancer for the long term survival patients after surgery,which could provide the rationale for the screening of relative genes of carcinogenesis and clinical treatment of lung cancer.Methods:Twenty patients with over 5 year survival after operation were defined as long term survival group.There were 9 patients with squamous cell carcinoma,6 with adenocarcinoma and 5 with small cell carcinoma in the long term survival group.The control group was defined as the samples from the patients with lung cancer who had surgery within one year and had to be randomly chosen,8 patients were histologically proven as squamous cell carcinoma,7 as adenocarcinoma and 5 as small cell carcinoma in the control group.The samples from healthy donors were defined as the normal group.The chromosome variations of peripheral lymphocytes in all these 3 groups were analyzed by routine methods.The comparative genomic hybridization of the embed-paraffin tissue sample of lung carcinoma was done by the DeVries methods.Results:In the long term survival group,the chromosome variations rate of lymphocytes was 3.15%,which was lower than that of the control group(3.15% vs 10.85%,P
7.Advances in the study of metastatic renal carcinoma

Renal cancer is a common malignant tumor in urology, with complicated pathogenesis and no obvious clinical symptoms. Metastatic renal cancer accounts for 20%-30% of the newly diagnosed renal cancer. The 5-year survival rate for metastatic patients is less than 10%.At present, the main non-surgical treatment means of mRCC include cytokines and targeted drug therapy, and immunotherapy has also become a research hotspot of mRCC. The current status and research progress of mRCC therapy were reviewed. The medical treatment methods of metastatic renal cancer, such as receptor tyrosine kinase inhibitors and mTOR inhibitors; immunotherapies, such as immunosuppressants for PD-1/PD-L1 were also discussed.
8.Influence of hyperthermia on cell cycle and rhythm gene Bmall expression in gastric cancer MKN28 cells

Objective To investigate the relationship between the cell cycle blocked by hyperthermia and the expression of rhythm gene Bmall in gastric cancer MKN28 cells so as to provide the academic evidence in hyperthermia therapy for gastric cancer.MethodsThe MKN28 cells were resuscitated and cultured in vitro.In control group MKN28 cells were cultivated at 37 ℃.In experimental groups MKN28 cells were heated at 43 ℃ for different durations.The cell morphology was observed by microscopy.Methylthiazdyl tetrazolium (MTT) assay was adopted to evaluate the inhibitory effect of the cell line.The flow-cytometry was adopted to observe the influence on the cell cycle.The Bmall mRNA expression was investigated by real-time quantitative reverse transcription-polymerase chain reaction ( RT-PCR).ResultsThe remarkable changes of cell morphology were observed by microscopy after exposure to heating.According to the data of MTT assay,37 ℃ heating could not inhibit the proliferation of MKN28.The inhibitive rates of cell growth after 0.5 h,1 h,l.5 h at 43 ℃ was (21.76±5.46)%,(25.30 ±4.36)% and (27.62 ± 3.78 )%,respectively.Results from flow-cytometry showed that G0/G1 phase cells in lh at 43 ℃ were remarkably less than those in the control group.However G2/M cells were significantly more than those in the control group.The mRNA expression of Bmall was the lowest when heating lh at 43 ℃ as compared to the control group.ConclusionsHyperthermia could induced the cell cycle changes and the expression of Bma11 in gastric cancer MKN28 cells.
9.Protection of melatonin against liver injuriesinduced by low-tempreture preservation

AIMTo study whether Mel has protective role in liver c old preservation and its mechanism. METHODSTo study the role of Mel at the different concentration and time in liver cold preservation, we colle cted liver washing liquid and liver tissue from male SD rats. We analysed ALT, AST, LDH, HA, ET 1, MDA and SOD to see the function of liver cell and sinusoida l lining cell and mechanism of cell injury. At the same time, we studied the eff ects of Mel at different concentration and different time on liver cold preserva tion. RESULTS① The damage to liver function and change of its histology was aggravated with the prolongation of UW co ld preservation time, and injury of the sinusoidal lining cell was also aggravated gr adually. ② Indexes of the liver function and the sinusoidal lining cell functio n in the Mel solution cold preservation group were lower than that in the UW sol ution cold preservation group. These suggest protective role of Mel against live r injury caused by cold preservation. Its mechanism may be related to powerful a nti-oxidative effect of Mel. ③ The protective effect of Mel at 1?10 -5 m ol?L -1 ,1?10 -7 mol?L -1 was better than that of Mel at 1?10 -9 mol?L -1 . We found no obvious difference between the protection of Mel 1?10 -5 mol?L -1 and 1?10 -7 mol?L -1 indicati ng a concentration-effect relationship. CONCLUSIONProtective ef fect of UW solution used in clinic weakens gradually as cold preservation time increases. Mel solution has protective effect on liver cold preservation injury and is better than the simple UW solution. Its mechanism is related to anti-oxi dative effect of Mel.
10.30 Cliaical Cases of Serious Hyponatremia From Applying Pituitrin.

Objective To summarize the clinical experience of serious hyponatremia resulted from applying pitnitrin to massive hemorrhage of gastrointestinal tract patients.Methods we used pitritrion on 2460 such patients and examined some symptoms of the digestive tract and nerve system,monitoring the dynamic state of blood sodium.Results The re- suits reveal that patients in 30 cases turned out to suffer from serious hyponatremia,whese sodium in the serum appeared less than 110 mmol/L,which was 1.22% of the rest subjects'.The relevant symptoms included nausea,vomit,spoor,coma and twitch etc.Following remedies included liquid supply with a high-concentrated sodium,ceasing using pituitrin,in- take of salute foods,and dynamic monitoring on the state of sodium in the serum.All patients recovered in 1 to 3 days without a single failure.Conclusion Thus we reach this conclusion:When applying pituitrin to massive hemorrhage of gastrointestinal tract patients,the above mentioned symptoms should be closely examined,and blood sodium should be timely monitored so that proper treatment should arrive in time.
