1.Study of differentially expressed cDNA on hepatocellular carcinoma

Objective:To search for gene related to hepatocellular carcinoma ( HCC), and study its, potential significance in liver oncogenesis. Methods:The difference in mRNA expression among HCC, nontumorous liver tissue, fetal liver tissue and normal liver tissue was investigated by differential display technique. Ten hepatic tissues of HCC versus surrounding noncancerous liver tissues were examined with MRG98.2 as a probe in Dot blot analysis. Expression of vascular endothelial growth factor (VEGF)mRNA in above mentioned samples was examined with reverse transcription- polymerase chain reaction(RT~ PCR). Results:Fifty five differentially expressed cDNA fragments were isolated from HCC samples, 24 of them were present in HCC and fetal liver tissues simultaneously. Dot blot analysis showed that differential display fragment MRG98.2 expressed in 7 cases of HCC samples (7/10)but only slightly in 2 of the surrounding noncancerous hepatic tissues (2/10). The expression of VEGFmRNA was upregulation in 6/7 of HCC samples expressed MRG98. 2. Gmclusian :The activation of some genes expressed in fetal period may have an important role in the genesis of HCC. MRG98.2 may be a gene related to HCC. Its expression in HCC correlates with VEGFmRNA. MRG98.2 expression is helpful in predicting a tendency toward invasion and metastasis of HCC.
2.Construction and practice of psychological sitcom teaching model in Psychiatric Nursing

Objective To construct psychological sitcom teaching model and explore the effect of its application in Psychiatric Nursing teaching.Methods Through literature review,study of the theories,summary of clinical practice and educational experience,we constructed a theoretical framework of psychological sitcom teaching model for Psychiatric Nursing.This model is composed of four contents (teaching aim,teaching modules,teaching process,teaching basic program) and four sessions (initial calculation,acting scripts,classroom observation and sharing,writing experience and internalization of knowledge).A total of 192 nursing students of grade 2012 from the nursing department were selected as the research objects.At the end of the course,questionnaires were employed to evaluate the effects of the students.Results The questionnaires demonstrated that 99.0% studentsprefer this teaching model and more than 97.9% students considered it helpful to improve four aspects of comprehensive abilities including theoretical learning abilities,leadership and decision making skills,nurse-patient communication skills and psychological nursing abilities.Conclusions Psychological sitcom teaching model is helpful to improve learning effect of Psychiatric Nursing for nursing students and should be widely spread.

6.Quantitative detection of the expression level of transforming growth factor-β1 and-β2 in rat retina with real time PCR

AIM: To quantitatively detect the expression level of transforming growth factor-β1 (TGF-β1) and transforming growth factor-β2 (TGF-β2) genes in the retina of normal rat in order to determine the expression difference of TGF-β1 and TGF-β2 in retina.METHODS: The total RNA was isolated from which the first strand of cDNA was prepared. The mRNA levels of TGF-β1 and TGF-β2 were detected quantitatively by real time polymerase chain reaction (PCR).RESULTS: The mRNA levels of TGF-β1 and TGF-β2 were 0.0008±0.0003 and 0.0378±0.009, respectively. Expression of TGF-β2 was obviously higher than that of TGF-β1 in rat retina with statistical significance (t=12.37, P<0.001). The ratio of TGF-β2/TG-β1 was 55.00±26.61.CONCLUSION: QRT-PCR could specifically and accurately detect gene expression level in rat retina. In retina the TGF-β2 gene was expressed more abundantly than TGF-β1. It is suggested that TGF-β2 play an important role in retina diseases.
7.Expression of transforming growth factor-β type Ⅰ receptor and transforming growth factor-β type Ⅱ receptor in rat retina

AIM: To quantitatively investigate transforming growth factor-β type Ⅰ receptor (TβRⅠ) and transforming growth factor-β type Ⅱ receptor (TβRⅡ) gene expressions in rat retina.METHODS: Sprague-Dawley rats were chosen in this research. Gene expression was detected quantitatively by reverse transcription polymerase chain reaction (RT-PCR) analysis. RESULTS: The expression level of TβRⅠ and TβRⅡ were 0.00034±0.00013 and 0.0001±0.00005, respectively. The expression level of TβRⅠ was obviously higher than that of TβRⅡ in the rat retina with statistical significance (P<0.01). The ratio of TβRⅠ/TβRⅡ was 3.9±1.7.CONCLUSION: Real time quantitative RT-PCR is an effective method to detect differential expression genes in retina. The change of TβRⅠ/TβRⅡ expression may play an important role in the pathogenesis of retinopathy, which could be further investigated in its significance in the development of proliferation retinopathy.
8.Gene expression of transforming growth factor-β2 in retina of diabetic rats

AIM: To detect the gene expression of TGF-β2 in retinas of diabetic rats at different stages, to observe and analyze the effect of TGF-β2 on the retinas of diabetic rats, to explore the role of TGF-β2 in pathogenesis of diabetic retinopathy (DR), and to provide experiment data and experience for further clinic studies.METHODS: Sprague-Dawley (SD) rats were used and retinas were dissected. The total RNA was isolated from which the first strand of cDNA was prepared. Diabetes mellitus was induced by a single intraperitoneal injection of 60mg/kg streptozotocin (STZ) and the rats were held without insulin treatment until sacrifice. Besides, agematched rats treated with saline were used as controls. Tail vein blood glucose was measured after 2 days and rats were considered hyperglycemic if blood glucose reading>16.7mmol/L. Animals with blood glucose level<16.7mmol/L were excluded from the study. The rats were killed at the 4th, 8th, 12th, 16th, 20th and 24th week respectively after hyperglycemic models were established. The retinas were separated and preserved in liquid nitrogen. The expressions of TGF-β2 gene mRNA were detected by reverse transcription PCR (RT-PCR).RESULTS: The RNA of rat retina was integrative enough to be used to further carry out PCR analysis. Compared with control groups, the expression of TGF-β2 mRNA in retinas of diabetic rats was up-regulated at the 4th week, but there was no statistical difference (P>0.05); it was down-regulated at the 8th week, and there was statistical difference (P<0.05); it was also down-regulated at the 12th week, and there was statistical difference (P<0.05); at the 16th week there was no statistical difference (P>0.05); it was up-regulated at the 20th week, but there was no statistical difference (P>0.05); it continued to be up-regulated at the 24th week, and there was statistical difference (P<0.05).CONCLUSION: Since the expression of TGF-β2 mRNA in retinas of diabetic rats was down-regulated at the 8th week and 12th week statistically, up-regulated at the 24th week statistically, it has obviously shown that TGF-β2 was down-and up-regulated through the period of DR. That is, its changes are diphasic with time. It may confirm that TGF-β2, with the characteristic of diphasic regulation, played an important role in DR. It is necessary to study it furthermore.
9.The placement of Y-shaped titanium-nickel memory alloy stent in the carina: a fundamental study in experimental canines

Objective To investigate the pathological changes of local airway tissue after inserting Y-shaped titanium-nickel memory alloy stent in carina, and to evaluate CT three-dimensional reconstruction technique in postoperative and follow-up observation. Methods Twelve healthy adult canines were enrolled in this study. Based on the information of the carina obtained from CT three-dimensional reconstruction images, the preparation of Y-shape netlike stent was made by knitting method with single Ni-Ti memory alloy wire. The stent was then inserted in canine's carina with the help of a releasing system. After the operation fiberbronchoscopy and CT three-dimensional reconstruction were performed. The animals were sacrificed 12 weeks after the procedure and the bronchus of the stenting segment was removed and sent for histopathologic examination. Results Technical success was achieved in all canines with the stent fight in the carina. The airway remained unobstructed 12 weeks after the procedure. Histopathologic examination revealed that the stent became partial epithelialization. Conclusion The Y-shaped titanium-nickel memory alloy stent has good histecompatibility and physicochemical stability and no re-stenosis of the airway develops in the follow-up period of three months. The stent-releasing technique is easy and simple. As a non-invasive and convenient exam, CT three-dimensional reconstruction technique is of great value in postoperative follow-up observation.
10.Biomarkers of oxidative stress in stroke

Oxidative stress has been demonstrated to be one of the important mechanisms for brain injury. Free radicals are one of the most important products following cerebral ischemia/reperfusion and intracerebral hemorrhage. They are the important markers for evaluating the 2 types of stroke injuries. At present, some relevant biomarkers of oxidative stress, such as lipid, DNA, protein peroxidation, antioxidant enzymes, and non-enzymatic antioxidant molecules as well as inflammatory and anti-inflammatory factors, are increasingly receiving attention. This article reviews and evaluates the biomarkers of oxidative stress in stroke.
