Objective To investigate the relation between the expression of toll receptor-4 (TLR-4) and the increase of blood brain barrier (BBB) permeability in the rats with acute pancreatitis.Methods Male Sprague-Dawley rats were randomly divided into 8 groups:control group;acute edematous pancreatitis (AEP) 2 h,6 h groups;acute necrotizing pancreatitis (ANP) 2 h,6 h,12 h,24 h,48 h groups with 8 rats in each group.The BBB permeability,pathological score of pancreas,TLR-4 expression were determined and the relationships between them were analyzed.Results The BBB permeability in control group;acute edematous pancreatitis (AEP) 2 h,6 h groups;ANP 2 h,6 h,12 h,24 h,48 h groups were 1.55±0.29,1.64±0.17,1.69±0.24,1.89±0.12,2.66±0.32,2.91±0.29,2.89±0.69 and 1.84±0.07,respectively;the pathological scores of pancreas were 0,2.38±0.92,3.13±0.64,8.50±1.07,9.75±0.71,10.25±1.28,11.13±1.25 and 10.13±1.13,respectively;there was no significant difference between AEP groups and control group,while there was significant difference between AEP groups and ANP groups (P<0.05 orP <0.01).BBB permeability was correlated with pancreatic injury ( r = 0.626,P <0.01).There was no TLR-4 mRNA and protein expression in the control and AEP group,while they were significantly expressed in ANP groups,and the expression were positively related with BBB permeability ( r =0.208,P = 0.027 ).Conclusions BBB permeability was present in the course of ANP.Activation of TLR-4 signal pathway may be involved in the BBB permeability increase caused by ANP.
4.Effects of probiotics on the gut flora and mucosal tumor necrosis factor?expression in rats with experimental colitis

Objective To evaluate the effects of probioties on inflammation,intestinal flora and mucosal tumor necrosis factor alpha(TNF-?)expression in experimental colitis rats.Methods Thirty Wistar rats were randomly divided into normal control group(NC),model control group(UC),and pro- biotics treatment group(PC).The experimental colitis were induced by trinitrobenzene sulfonie acid (TNBS)/ethanol enema.Rats in PC group were fed with Bifico[live probiotics of combined bifidobacte- rium(Bif),lactobacillus (Lac) and enterococcus]by 2.2?10~9 colony-forming unit (CFU)/d for 4 weeks.Inflammatory scores and mucosal morphological changes under light microscopy were studied. Plasma endotoxin levels,expressions of TNF-?and interleukin-6 (IL-6) in mueosa were measured. Enteric microorganisms in cecum were fostered with standard methods.Results Inflammatory scores in PC group decreased compared with those in UC group(7.94?0.85 vs 10.25?1.36,P<0.05),but still higher than those in NC group(7.94?0.85 vs 4.35?0.88,P<0.01).There was a significant alteration in the enteric microbial flora,Bif and Lac were decreased significantly in UC group,Bifico supplement ameliorated this imbalance.Plasma endotoxin and mucosal TNF-?levels in PC were lower than those in UC group[endotoxin:(93.33?21.22)pg/ml vs (121.25?39.07)pg/ml;TNF-?:67.51?14.63 vs 85.99?18.17,P<0.05],but higher than those in NC group[endotoxin:(93.33?21.22) pg/ml vs(35.20?15.12)pg/ml;TNF+?:67.51?14.63 vs 43.28 4?19.98,P<0.01].There was no statistically significant difference in mucosal IL-6 levels between PC group and UC group,though its level in PC group was lower than that in UC group(155.22?34.01 vs 184.09?29.11,P>0.05),but higher than that in NC group(155.22?34.01 vs 108.73?37.35,P<0.01).Conclusions Probiotics can attenuate inflammation of experimental colitis of rats through reinstituting gut flora bal- anee.Improving gut flora imbalance,reducing endotoxin and pro-inflammatory cytokines may be the effects of probiotics.

Hematoma enlargement is a common harmful event after cerebral hemorrhage, which can lead to deterioration of neurological function and poor outcome. Early detection of high-risk patients can help prevent hematoma enlargement and improve the outcome of patients. Although the exact mechanism of hematoma enlargement is unclear, more and more evidence shows that many imaging signs may be related to hematoma enlargement, such as dot sign, mixed sign, black hole sign, island sign, etc. This article mainly reviews the relevant imaging prediction factors of hematoma enlargement from the perspective of imaging.
7.Small Intestine Absorption Behavior of Collagen Peptides Microspheres in Rats

Objective To investigate small intestinal absorption behavior of oral collagen peptides microspheres in rats. Methods The everted gut sac (EGS) and Single-pass intestinal perfusion (SPIP) model were established in Wistar rats (250-350 g, 5 months).The absorption of collagen peptide in different segments of rat small intestines was detected by BCAmethod. Results The cumulative transmittance of different particle size of calcium alginate/ Chitosan (SA/ CTS) microspheresin EGS experiments was that 500 μm microspheres<200 μm microspheres<10 μm microspheres ( P<0.01, except the jejunumintestine).The cumulative transmittance of microspheres ( 10 μm) was: SA/ CTS rate ( Ka ) and permeability rate ( Peff )microspheres> SA microspheres(duodenum segment, jejunum segment P<0.05;ileum segment, P<0.01).The absorption rate ofSA/ CTS microspheres was evidently higher than that of SA microspheres in SPIP experiments. Conclusion Small intestineabsorption of collagen peptides microspheres is correlated with particle size, and the chitosan in SA/ CTS microspheres isbeneficial to the absorption of collagen peptide in small intestine segments.
8.Carriage and Antibiotic Resistance of Streptococcus pneumoniae from Respiratory Infected Children

OBJECTIVE To study the carriage,the risk factors and the antibiotic resistant pattern of Streptococcus pneumoniae isolated from hospitalized children with respiratory infection below 3 years old.METHODS A total of 453 children with respiratory infection hospitalized at Children′s Hospital of Soochow University in Suzhou Jiangsu Province from Mar 2006 to Mar 2007 were enrolled and given deep orotracheal aspiration(OTA) technique to get the sputum.S.pneumoniae strains were tested and analyzed.RESULTS The carriage rate of S.pneumoniae was 9.3% which was related with age and season.The percentage of resistance to penicillin,erythromycin,tetracycline,clindamycin,chloramphenicol and trimethoprim/sulfamethoxazole was 16.7%,100%,95.2%,95.2%,14.3% and 73.8%,respectively.No resistant strains to ceftriaxone,ofloxacin and vancomycin were detected.All S.pneumoniae strains were resistant to multiple drugs.CONCLUSIONS The carriage of S.pneumoniae in respiratory infected children below 3 years old in Suzhou area is relatively low.Age and incidence season are the risk factors.The antibiotic resistance is serious.
9.Detection of Y chromosome microdeletions in AZF region by liquid chip technology.

OBJECTIVETo establish a liquid chip technology to detect Y chromosome microdeletions in Chinese infertile males with azoospermia or oligozoospermia.
METHODSMultiplex PCR and liquid chip technology were used to detect the Y chromosome microdeletions in AZF region in 178 infertile patients with azoospermia and 134 infertile patients with oligozoospermia as well as 40 fertile control men.
RESULTSForty out of 312 patients (12.8%) were found to have deletions in AZF region. The microdeletion frequency was 14%(25/178) in the azoospermic group, 9.6%(11/114) in the oligospermic and 20%(4/20) in the severe oligospermic group.
CONCLUSIONThe authors developed a high-throughput, fast and simple assay to screen the AZF region microdeletions of Y chromosome.

10.Establishment of HPLC fingerprints of Yupingfeng Powder aqueous decoction and determination of nine constituents

AIM To establish the HPLC fingerprints of Yupingfeng Powder aqueous decoction and to determine the contents of nine constituents.METHODS The analysis of aqueous decoction was developed on a 30 ℃ thermostatic Hypersil ODS column (250 mm × 4.6 mm,5 μm),with the mobile phase comprising of acetonitrile-water flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 220 nm.RESULTS There were fifteen common peaks in the fingerprints of ten batches of samples,with the similarities of more than 0.95.Nine of them were identified as prim-O-glucosylcimifugin,calycosin-7-O-β-D-glucoside,cimifugin,4'-O-β-glucopyranosyl-5-O-methylvisamminol,psoralen,calycosin,sec-O-glucosylhamaudol,formononetin and atractylon,which showed good linear relationships within their own ranges (r ≥ 0.999 7),the average recoveries were 97.91%-99.81% with the RSDs of 0.58%-1.27%.CONCLUSION This stable and reliable method can beused for the quality control of Yupingfeng Powder aqueous decoction.
