2.The effect of leukoreduction on the IL-2 and IL-8 leveIs in blood storage

Investigate the effect of prestorage white cell filtration on the cytokines content in blood. Methods: Ten units of whole blood was divided into two parts just after blood collection:one was used as control, the other was filtrated by leukoreduction filters. All units were stored at 4℃ for 6 weeks. Immediately after filtration and every week subsequently, samples for analses of IL-2, IL-8 were obtained. Results:The amounts of IL-2 and IL-8 increased during the storage period. In filtered units, the amounts of IL-8 did not increase during the study period, IL-2 still increased but was still lower than that of unfiltered units at the end of storage. Conclusion: Many cytokines, such as IL-2 and IL-8, were generated and accumulated in blood during blood storage. Leukocyte filtration may prevent this effect to some extend. Prestorage filtration may decrease FNHTRs by preventing the accumulation of cytokines in blood.
3.Screen of human single chain antibody against N protein of SARS-CoV

Objective To prepare the single chain antibody against N protein of SARS-CoV. Methods With N protein of SARS-CoV expressed in E.coli as antigen, we obtained the single chain antibody against N protein by screening the phage display library of human single chain antibodies. Results The anti-N protein antibody didn’t cross-interacte with BSA and the short peptide containing 6 histidines. The specific interaction between the antibody and N protein was inhibited by the anti-N protein monoclone antibody from immunized mice. ConclusionThe single chain antibody we got is specific to N protein of SARS-CoV,it can be a candidate antibody for fast detection of N protein of SARS-CoV and SARS virus particles in clinical trial study of SARS pathogenesis.
4.The analysis of blindness and low vision by routine physical examination in the elderly

ObjectiveTo investigate the visual condition and causes of blindness and low vision of ov er 60 years old people found by routine physical examination. Methods A total of 892 cases over 60 years old people took a yearly routine examinatio n, such as far and near vision in daily life, anterior segment and fundus of the eye and intraocular pressure. If the vision were under 0 3, the refractive vis ion needs to be corrected. The blindness and low vision patients were selected a nd taken further related examinations to analyse statistically the causes of bli ndness. Results Among 892 cases investigated, blindness together with low vision were 42 cases (4 71%). The morbidity rate of blindness and low vision were 1 01% (9 cases) a nd 3 70% (33 cases) respectively. Diseases caused blindness were macular lesio ns (57 14%), glaucoma(11 91%), cataract (9 52%), retinal diseases, optic nerv e atrophy, keratopathy etc. Refractory eye diseases were obviously more than t he eye diseases which can be treated. The morbidity rate of over 80 years old pe ople were obviously more than that of under 80 years group. Conclusions With ageing, the old people easily suffer from eye diseases and results in bli ndness. For the prevention and treatment of blindness for the elderly, much atte ntion to treatment of eye diseases should not only paid, active training is also important.
5.The investigation of the eye condition and analyses of the diseases in the longevous aged 90 years or over

Objective To investigate the vision and the eye conditions of the longevous aged 90 years or over Methods The vision was examined by Snellen chart, and the outer eye, anterior segment, fundus and intraocular pressure were examined by slit lamp, funduscope and non contact tonometer in 135 longevous cases Some related examinations were adopted on suspective cases The refractive errors were corrected in 53 cases who were cooperative to examination Results The visions of right eyes were 0 01~1 0 with the average of 0 46?0 50, and the left eyes were 0 03~1 0, with the average of 0 49?0 50. The probabilities of blindness and low vision were 5 93% and 10 27% respectively. 7.Effect of umbilical sticking therapy with qitou xiaogu plaster in treating liver cirrhosis patients with portal hypertension.

OBJECTIVETo observe the effect of umbilical sticking therapy (UST) with Qitou Xiaugu Plaster (QXP) on hemodynamics of portal system in patients with liver cirrhosis.
METHODSOne hundred and twenty patients of liver cirrhosis with portal hypertension were assigned to two groups. On the basis of conventional therapy, UST was applied in the 66 patients in treated group, which was exchanged once every 3 days with an interval of 1-day rest. The 54 patients in the control group were orally administered with propanolol. The therapeutic course for both groups was 1 month. Before and after treatment, the hemodynamic changes in portal or splenic veins were observed by color Doppler ultrasonograph, and the changes of liver function, blood coagulation and patients' subjective symptoms were observed as well.
RESULTSAfter treatment, portal vein diameter and splenic vein diameter significantly decreased (P < 0.05, portal venous flow velocity and splenic venous flow velocity apparently increased (P < 0.05), and portal venous flow apparently decreased in both groups (P < 0.05), while no significant change was found in the splenic venous flow (P > 0.05). The liver function and blood coagulation indexes in both groups were improved. The improvement of clinical symptoms in the treated group was superior to that in the control group.
CONCLUSIONUST with QXP could decrease the portal vein pressure in a short time, with the therapeutic effect comparable to propanolol, and with no adverse reaction.

8.Papillary cystadenoma of the epididymis: a report of 2 cases and review of the literature.

OBJECTIVETo study the clinicopathological characteristics of papillary cystadenoma of the epididymis.
METHODSUsing routine pathology and immunohistochemistry, we observed the surgically obtained samples from 2 cases of papillary cystadenoma of the epididymis, analyzed their pathological features and clinical presentations, and reviewed the related literature.
RESULTSThe 2 patients were both adult males. The tumors typically manifested as painless swelling in the epididymis, with occasionally dull pain and tenesmus in 1 of the cases. Pathologically, the lesions exhibited three morphological features, i. e., dilated ducts and small cysts surrounded by fibrous connective tissue, adenoid papillary hyperplasia into the cysts embraced by fibrovascular stroma, and acidophil substance present in the cysts. Immunohistochemistry showed that the tumors were strongly positive for CK8/18, CK7, and EMA, but negative for CK20, CEA, MC, Calretenin, P53, P63, SMA, VHL, and CD10, with the positive rate of Ki-67 <1%. Follow-up visits revealed good prognosis in both cases.
CONCLUSIONPapillary cystadenoma of the epididymis is a rare benign tumor in the male urogenital system, which may be accompanied by the VHL syndrome. Surgery is the first choice for its treatment.

10.The Screening and Study of Arctic Marine Psychrophilic Bacterial for Anti-tumor and Antibacterial Activity

In this study, 101 strains of bacteria were isolated from arct ic water and sediment samples. The methanol extracts of the fermented broth prod uced by these strains were screened in vitro for anti-tumor activity on mou se tsFT210 cells using the method of flow cytometry, and screened for antibacter ial activity by the method of paper disk diffusion. The result showed that one strain exhibited anti-tumor activity and eight strains had antibacterial activ ity. The stability of the antibacterial components produced by strain AR084 an d its optimum medium were also studied. The research indicated that arctic bac teria had potential application in pharmaceutics.
