1.Effects of Sancaoanqian Decoction on Expression of IL-1?, IL-6 and TNF RⅡ in Experimental Autoimmune Prostatitis Rats

Objective To observe the effects of Sancaoanqian decoction on expression of cytokine IL-1?, IL-6 and TNF RⅡ in experimental autoimmune prostatitis rats, and explore the target protein of cytokine of Chinese herbal compound in treating chronic nonbacterial prostatitis. Methods Seventy-two male Wistar rats were divided randomly into six groups and 12 rats in each group. All groups were made experimental autoimmune prostatitis rat model by injecting SC purified prostate protein with FCA except normal group. The normal and model group were given saline, western medicine group was given indomethacin, large, medium and small doses decoction groups were treated with large, medium and small doses of Sancaoanqian decoction. All rats were sacrificed after 30 days. The mRNA and protein expression of cytokines were tested by methods of immunohistochemistry and fluorescent quantitation RT-PCR. Results Sancaoanqian decoction could reduce the mRNA and protein expression of IL-1?, IL-6 in experimental autoimmune prostatitis rats. There were no significant difference between large and medium doses decoction group, but had a better activity than small dose. The expression of TNF RⅡ were reduced in all doses groups and no difference among three doses of Sancaoanqian decoction. Conclusion Sancaoanqian decoction can regulate the immune function in chronic prostatitis rat model. Down-regulation of cytokines maybe one of important mechanisms in treating chronic prostatitis.
2.Analysis of Chinese medicine on differentiation of symptoms and treatment of 36 cases of Sjāgre's syndrome

BACKGROUND: Sjogre's syndrome is a common disease of aged people,in which, dry mouth and throat have disturbed chawing, swallowing and speech and have seriously affected living quality of patients. OBJECTIVE: To probe into the improvement of oral function treated with Chinese medicine based on differentiation of symptoms. DESIGN: Retrospective analysis on cases was designed. SETTING: Department of Periodontal Disease, College of Stomatology,Fourth Military Medical University of Chinese PLA. PARTICIPANTS: Thirty-six cases of Sjogre's syndrome were selected from clinic of Department of Periodontal Disease of Stomatology College of Fourth Military Medical University of Chinese PLA from May 2000 to February 2004, with agreement and in the know of patients. Of those, 5cases were male, 31 cases were female, aged varied from 27 to 70 years,with duration of sickness in range from 0.5 to 10 years. METHODS: ① On basis of moistening, drying up heat, producing fluid and stopping thirst, the drugs for activating blood circulation, resolving stasis, clearing away heat and detoxification were supplemented. Formula:shashen (Radix Glehniae), maidong (Radix Ophiopogonis), shihu (Herba Dendrobii), yuzhu (Rhizoma Polygonati Odorati), huafen (Radix Trichosanthis), xuanshen (Radix Scrophulariae), shengdi (Radix Rehmanniae), chenpi (Pericarpium Cirri Reticulatae), danshen (Radix Salviae Miltiorrhizae), banzhilian (herba Scutellariae Barbatae), bahuasheshecao (Herba Hedyotis Dissusae), etc. ②Treatment of vitamins and transfer factor were associated.③ Artificial saliva in local was applied to prevent from secondary infection. ④ It was to test 99Tc m parotid function (After 99Tc m was injected intravenously on elbow, the parotid uptaking and concentrated 99Tc m curve were traced immediately. When radioactivity intensity was increased to maximum, the patients were required to chaw Vit. C tablet, 200 mg. In following, it was to trace the minimum of 99Tc m radioactivity on decreased curve. Normal imaging was illustrated clearly, the development after Vit. C stimulationwas declined swiftly. Time-radioactivity curve illustrated obvious uptaking and excreting segments. Positive results were unclear parotid development or the development after Vit. C stimulation was not declined obviously. Time-radioactivity curve was of horizontal type, low-horizontal type, parabola type or persistent rising type), erythrocyte sedimentation (ERS) (Normal value: 0-15 mm/h for male adult, 0-20 mm/h for female adult) and a part of immunology, such as rheumatoid factor, immunoglobulin (I gG normal value: 7.6-16.6 g/L), immune complex, etc. The effect was determined as increased saliva secretion and improved oral function. MAIN OUTCOME MEASURES: 99Tc m parotid function test, rheumatoid factor, ERS, IgG and immune complex were applied to determine the therapeutic effects of Chinese herbs. RESULTS:Thirty-six cases all entered result analysis. ① After treatment, the positive rate of rheumatoid factor was lower significantly compared with that before treatment [36.7%, 73.3%,(x2=6.796, P < 0.01)]. The levels of ERS, IgG and immune complex were all decreased significantly compared with those before treatment [(35.50±5.65) mm/h,(13.47±1.58) g/L,(8.30±1.24) mg/L;(55.20±8.85)mm/h,(23.57±2.37) g/L,(14.96±2.21) mg/L; (t=3.549-4.962,P < 0.01)]. The result of 99Tc m parotid function test and main determined indexes had been improved for all. ② After treatment, dry mouth, dry eye and general symptoms were all improved of various degrees and saliva secretion was increased. Total effective rate reached 80.7%. CONCLUSION: Treatment based on differentiation of symptoms is applied in Chinese medicine. On basis of moistening, drying up heat, producing fluid and stopping thirst, the drugs for activating blood circulation, resolving stasis, clearing away heat and detoxification are supplemented and effectively promote rehabilitation of oral function in Sjogre's syndrome,such as chawing, swallowing, speech, etc.
3. Experimental studies on anti-tumor angiogenesis effect of phosphodiesterase (PDE2, PDE4) inhibitors

Objective: The purpose of our study was to investigate the effect of phosphodiesterase (PDE2 and PDE4) inhibitors on the proliferation and migration of endothelial cells as well as angiogenesis in vivo. Methods: Human umbilical vein endothelial cells (HUVEC) were selected for investigation of angiogenesis in vitro and the C57BL/6N mouse for in vivo study. The effects of PDE2 inhibitor [Erythro-9-(2-hydroxy-3nonyl) adenine, ENHA] and PDE4 inhibitor (RP705) on the intracellular cAMP levels in HUVEC were detected by enzyme-linked immunosorbant assay (ELISA) and their effects on the proliferation and migration of HUVEC were also determined. The tumor-inhibiting effects of combined application of ENHA and RP705 were observed. Results: EHNA (25 μmol/L) and RP705 (15 μmol/L) specifically inhibited the proliferation and migration of HUVEC stimulated by vascular endothelial growth factor (VEGF) and increased the intracellular cAMP level in HUVEC. EHNA combined with RP705 inhibited tumor angiogenesis in vivo. The tumor inhibitory rate was 45.50% in C57BL/6N mouse after combined therapy with EHNA and RP705. Conclusion: These results strongly suggested that PDE2 and PDE4 might be new potential therapeutic targets in pathological angiogenesis.
4.Protection of daidzein on myocardial hypertrophy and fibrosis in rats

Objective To investigate the protective effects of daidzein (DD) on myocardial hypertrophy and fibrosis induced by pressure overload in rats and to study its mechanism. MethodsMyocardial hypertrophy and fibrosis model of rats induced by pressure overload was prepared by constricting abdominal aorta. The operated rats were randomly divided into sham operated control group, aorta-constricted model group, and three DD groups (30, 60, and 120 mg/kg). Four weeks later, the heart-weight (HW), left ventricular weight (LVW), the ratio of HW/BW and LVW/BW (LVI), and the cardio-myocyte diameters (MD) after dying by HE color were measured. The content of collagen and nitric oxide (NO), the activity of calcineurin (CaN) and Na+, K+-ATPase, Ca2+-ATPase in the left ventricle were quantified with spectrophotometry. The angiotension Ⅱ (AngⅡ) in the left ventricle was messured with radioimmunoassay. Results In aorta-constricted model group, the ratio of HW/BW, LVI, and MD as well as the content of collagen and AngⅡ, the activity of CaN in the left ventricle was significantly increased, and Na+, K+-ATPase, Ca2+-ATPase activity and NO content in the left ventricle were obviously decreased. After treatment of the left ventricular with DD, NO content, Na+, K+-ATPase, Ca2+-ATPase activity were significantly increased, the content of collagen and of AngⅡ and the activity of CaN in the left ventricle and the ratio of HW/BW, LVI, and MD were significantly reduced. ConclusionDD has protective effects on ventricular remodeling in rats with myocardial hypertrophy and fibrosis induced by pressure overload and its mechanism may be related to raising NO content and reducing the level of AngⅡ and the activity of CaN.
5.Protective effects of total saponins of panax notoginseng on myocardial hypertrophy and fibrosis induced by isoproterenol in rats

AIM:To investigate the protective effects of total saponins of panax notoginseng (PNS) on myocardial hypertrophy and fibrosis induced by isoproterenol (ISO) in rats. METHODS: Myocardial hypertrophy and fibrosis model of rats were induced by injection of ISO (5 mg?kg-1?d-1, sc) for 7 days. From day 2, the rats were administered with PNS at dose of 25 and 50 mg?kg-1?d-1, ip for 14 days, the control and ISO model group were received saline injection. Then, the heart-weight (HW), left ventricular weight (LVW), the ratio of HW/BW and LVW/BW (LVI) were measured; the hydroxyproline and malondialdehyde (MDA) and angiotensin (AngII) content of left ventricle. The level of nitric oxide (NO), nitric oxide synthase (NOS), superoxide disrnutase (SOD) and glutathione peroxidase (GSH-Px) activities in left ventricle were determined by spectrophotemetry and radioimmunoassay, respectively. RESULTS: Compared with NS control group, the ratio of HW/BW, LVW/BW and the content of hydroxyproline, AngII, MDA and iNOS activity in the left ventricle were significantly increased. The cNOS, SOD, GSH-Px activities and NO content were obriously decreased in the ISO model group. After treatment with PNS, the left ventricular NO content, cNOS, SOD and GSH-Px activities were markedly higher than those in ISO model group. The content of MDA, AngII and iNOS activities and the ratio of HW/BW, LVI were significantly lower than those in ISO model group. CONCLUSION: PNS reverses the myocardial hypertrophy and fibrosis induced by isoproterenol in rats. This effect may be related to eliminating the oxygen free radicals and raising NO level.
6.Changed accumulation of active ingredient in different localities and growth period of Hemsleya zhejiangensis (Cucurbitaceae).

In this paper, the content of moisture, ethanol-soluble extractives, total saponins and polysaccharide of different tuber samples of Hemsleya zhejiangensis, from different localities, years and seasons, were detected based upon Chinese Pharmacopoeia 2010 version. The samples of roots, stems and leaves in summer were detected as well. The results are mainly as follows. (1)With tuber quality increasing, the content of total saponins increased and then decreased. The individual quality of tubers getting 594.06 g, the content of total saponins reached the peak. (2) The content of active ingredients in different localities was significantly different, and the population of Wuyanling had the maximum content of total saponins and polysaccharide. (3) The content of active ingredients revealed stability between the years 2012 and 2013, but the content of polysaccharide was significantly different. The content in 2012 was higher than that of 2013. (4) The content of active ingredients reached the peak in autumn, which was the best harvest season. (5) Among different component content detection of nutritional organs, tubers had the maximum content of ethanol-soluble extractives, total saponins and polysaccharide. Leaves also contained higher content of ethanol-soluble extractives and total saponins than roots and stems. All of these provide theoretical basis for plant, harvest and production of H. zhejiangensis, which is an endemic, rare, and endangered medicinal plants.

This article introduced professor WANG Shu-chen's experience in picture treating bronchiolitis obliterans syndrome (BOS) after hematopoietic stem cell transplantation with tougue picture as the key link. Patients with BOS showed deficiency of essence of tongue crack, which should be treated by warming kidney yang, replenishing essence and marrow; fester tongue for qi disorder, inflammation caused by fire, which should be treated by regulating qi, and clearing heat; exfoliative fur accumulation, which should be treated with blood stasis and toxin, removing blood stasis and toxin; thick and greasy fur, which should be treated by warming spleen and activating spleen.
8.All-Trans Retinoic Acid Attenuates Interleukin-23/Interleukin-17 Pathway and Promotes Skin Allograft Survival in Mice

Objective To investigate the effects of all-trans retinoic acid (ATRA)-intragastric-administration on the survival time of mouse skin allografts and the role of interleukin (IL)-23 and IL-17 thereof. Methods The skin trans-plantation of mice was done by DBA/2 as donors and Balb/c as recipients. The recipients were divided randomly into three groups:control group, low-dose group and high-dose group. Mice of the corresponding groups were intragastrically adminis-tered corn oil, 10 mg/kg ATRA and 30 mg/kg ATRA respectively from 1 day before the transplantation to the 14th day after the transplantation. The survival time of transplanted skin was observed after the operations. Skin grafts of mice were harvested for histopathological examination in three groups. The serum levels of IL-23 and IL-17 were measured by enzyme-linked im-munosorbent assay (ELISA). The expression levels of IL-23, RORγt and IL-17 mRNA in skin allografts were detected by re-al-time fluorescent quantitative reverse transcriptase polymerase chain reaction (RT-PCR). Results Compared with con-trol group, the average survival time of mouse skin allografts was significantly prolonged in low-dose group and high-dose group (P<0.05). The less lymphocyte infiltration and destruction of architecture were found in the skin biopsies. The serum expression of IL-23 protein was lower (P<0.05), but no significant difference was found in two treatment groups. The serum expression levels of IL-17 protein were reduced in turn in receptors of control group, low-dose group and high-dose group (P < 0.05). The expression levels of IL-23, RORγt and IL-17 mRNA in skin grafts were significantly lower in low-dose group and high-dose group than those of control group (P<0.05), but no significant difference was found in two treatment groups. Conclusion ATRA can effectively prolong the survival time of skin allografts, which may related with the inhibi-tion of the expression of IL-23, RORγt and IL-17 mRNA and the development of IL-23 and IL-17 protein.
9.Distribution and antibacterial resistance of multidrug-resistant pathogens in tertiary grade A comprehensive hospital

Objective To investigate the characteristics of department distribution and antibacterial resistance of multidrug‐re‐sistant(MDR) pathogens in hospital ,and take specific measures for the prevention ,treatment and infection cntrol .Methods Data of 1 816 MDR isolates detected in the hospital from January to December in 2014 were analyzed retrospectively .Results Among the 1 816 MDR strains ,extended‐spectrum β lactamases(ESBLs) producing Escherichia coli ranked first(665 strains ,36 .62% ) ,fol‐lowed by methicillin‐resistant Staphylococcus aureus(387 strains ,21 .31% ) ,MDR Acinetobacter baumannii(295 strains ,16 .22% ) , MDR Pseudomonas aeruginosa(195 strains ,10 .74% ) ,carbapenem‐resistant Enterobacteriaceae(144 strains ,7 .92% ) ,ESBLs pro‐ducing Klebsiella pneumoniae(130 strains ,7 .16% ) .MDR strains were mainly distributed in general department of urology surgery (384 strains ,21 .14% ) ,burn unit(325 strains ,17 .90% ) ,intensive care unit(ICU )(266 strains ,14 .52% ) and department neurosur‐gery(110 strains ,6 .04% ) .Vancomycin and linezolid resistance were not found in methicillin‐resistant Staphylococcus aureus ;less than 2 .9% of the ESBLs producing Escherichia coli and Klebsiella pneumonia strains were resistant to carbapenems .The resistance of carbapenem‐resistant Enterobacteriaceae to amikacin ,sulfamethoxazole‐trimethoprim ,meropenem and tetracycline were relatively low (39 .6% -60 .7% ) .The resistance of MDR Acinetobacter baumannii to levofloxacin ,tetracycline ,sulfamethoxazole‐trime‐thoprim and cefoperazone‐sulbactam were relatively low (48 .9% -76 .5% ) .The resistance of MDR Pseudomonas aeruginosa to amikacin ,ceftazidime ,gentamicin and ciprofloxacin were relatively low(45 .3% -66 .7% ) .Conclusion The MDR pathogens should be monitored in high‐risk department ,preventive measures should be taken ,and antibiotics should be used according to the results of drug susceptibility tests .
10.Effects of siRNA targeting ADAMTS2 gene on hepatic stellate cells

Objective To explore the preventive and therapeutic role of silencing type Ⅰ rat platelet-binding protein motifs depolymerization protein-like metalloproteinase 2(ADAMTS2)by siRNA on experimental liver fibrosis in vitro.By studying the mechanism of siRNA silencing of ADAMTS2,we also aim to evaluate the feasibility of ADAMTS2 as a target for anti-liver fibrosis therapy.Methods Three pairs of siRNAs targeting ADAMTS2 mRNA 2237,2597 and 690 targets were designed and synthesized by utilizing RNA design software.The most effective siRNA was chosen to transfect HSC-T6 cell line to test the tendency of hepatic stellate cell(HSC)activation and ex pression of ADAMTS2,COL1α1,COL(I),α-SMA,TGF-β1,MMP-2 and TIMP-3.These were quantified using real time-PCR,Western blotting,and MTT assays.Results Of the same dosage and time of injection,siRNA 2237 inhibited ADAMTS2 gene expression significantly more than other siRNAs.siRNA-ADAMTS2 2237 markedly inhibited ADAMTS2 gene and protein expression of HSCT6 with more than 80% efficiency.Conversely,siRNA-ADAMTS2 2237 markedly reduced the gene and protein expressions of COL(I),α-SMA and TGF-β1 on HSC-T6 and inhibited the proliferation of HSC.Conclusions siRNA-ADAMTS2 2237 could effectively knockdown the gene and protein expression of ADAMTS2 in HSC-T6 cell lines.Silencing ADAMTS2 by siRNA significantly inhibited the activation,proliferation of HSC and the gene and protein expressions of COL(I),α SMA,and TGF-β1,and it may have a potential anti-fibrotic effect.ADAMTS2 might be an efficient target for anti-fibrotic therapy.
