1.Relationship between Injury Time and Expressions of EPO and Its Receptors in Rats Brain after Cerebral Injury

Objective To explore the relationship betw een injury age and expressions of erythropoietin (EPO ) and its receptor EPO R in the brain tissue of rats after cerebral injury. Methods Seventy-tw o rats w ere random ly divided into control group (36 rats) and cerebral injury group (36 rats). The rats w ere sac-rificed at 1, 2, 4, 8, 12, 24 h after cerebral injury (6 rats at each tim e point) and the brain tissues w ere extracted. The expressions of m RNA and protein of EPO and EPO R at different tim e points w ere de-tected by real-tim e fluorescent quantitative PC R and W estern bloting. Results The expressions of EPO and EPO R increased w ithin 24 h after injury. The expressions of m RNA and protein of EPO w ere relat-ed to the injury age, and the correlations w ere 0.875, 0.911, respectively (P<0.05). OBJECTIVE:

CONCLUSION
The expressions of EPO and EPOR increase gradually in the early stage of the rat's cerebral injury, which are associated with the injury age and could be a useful value for estimating injury age.

Experimental results showed Zn(Ⅱ)-ammonium thiocyanate-hexadocyltrimethylammonium bromide complex was floated in the presence of sodium chloride. There fore Zn(Ⅱ) could be completely separated from Fe(Ⅲ), Co(Ⅱ),Ni(Ⅱ), Mn(ⅡD, Al(Ⅲ) by floation. The method is non-toxic, simple, rapid and economical.
4.Comprehensive analysis of the ischemic times of main artery injury in the limbs.

OBJECTIVETo study the ischemic times of the main artery injury in the limbs and the influence on the limbs survival rate, and to analyze the reasons for the formation of the ischemia time.
METHODSFrom June 1996 to November 2012, 83 patients with completely severed limb main artery treated in our hospital were retrospectively studied. There were 77 males and 6 females, including 81 adults (ranging in age from 16 to 52 years old, with a median age of 35 years old) and 2 children (4 and 5 years old respectively). Seventy-five patients were treated with end to end anastomosis, 7 patients were treated with great saphenous vein transplantation, and 1 patient was treated with artificial blood vessel transplantation. The prior to admission ischemia time, after admission ischemia time and total ischemia time for successful and unsuccessful patients, as well as the causes of the formation of the ischemia time were studied. Limb survival rate of different ischemic time was counted. Sharpness injury or blunt injury ischemia time and amputation rate were researched statistically. All data were analyzed using SPSS statistical software.
RESULTSThere were 72 limbs survived, 11 amputated. The average ischemia time was (7.45±5.94) h for limb-salvage group and (13.73±14.00) h for amputation group. Prior to admission ischemia time between amputation and limb-salvage group had no significant difference. After admission the ischemia time in amputation group was longer than limb-salvage group. The amputation rate for ischemia time 21 to 44 h group was higher than other three groups (≤ 5 h, 6 to 10 h, 11 to 20 h) (P=0.023, 0.038, 0.044). Amputation reasons can be divided into anastomosis failure in the operation, vascular thrombosis 2 to 4 days after operation and late infection.
CONCLUSIONVascular injury limbs can tolerate longer ischemia time and the limb salvage succeed. Only too long ischemia time (> 20 h), limb amputation rate increases significantly. The main cause of long time ischemia is delayed diagnosis in the hospital. Whether limb-salvage success mainly depends on the degree of trauma and the quality of the anastomosis than ischemia time.

5.Judgment of defect length of extremities artery trauma and reconstruction.

OBJECTIVETo study the influence of actual defect length and gap width of the limbs main artery on the method selection of repairing and reconstruction.
METHODSRetrospective study was carried out for 32 patients with extremity main artery injury from 1996 to 2009, including 30 males and 2 females; 30 adults with an average age of 36 years old ranging from 18 to 51 years, 2 children of 4 and 5 years old respectively. Injured body parts involved axillary artery in 4 cases,brachial artery in 7 cases,radial artery in 2 cases, femoral artery in 4 cases, popliteal artery in 13 cases, posterior tibial artery in 2 cases. Main arterial injury defect gap width of all cases were observed and the reasons were analyzed. All cases were repaired by the method of end to end anastomosis after vessels stretch.
RESULTSThe artery defect width was 3 cm to 7 cm with an average of (4.375 +/- 1.200) cm. Defect width of the upper extremity brachial artery and axillary artery group was (5.73 +/- 0.63) cm,the lower extremity femoral and popliteal artery group (3.80 +/- 0.73) cm, the posterior tibial artery group (3.25 +/- 0.35) cm, the radial artery group (3.00 +/- 0.00) cm. Defect width of upper extremity brachial artery and axillary artery group was larger than that of the other three groups (P < 0.01). End to end anastomosis was performed successfully in all cases. Blood supply recovered well. Because of the severe limb infection 2 patients had amputation in the late. All patients received follow-up. The patients without fracture were followed up to 2 weeks postoperatively, all patients with fractures were followed up to 1 year at least. Limb blood supply was good in all patients during the follow-up.
CONCLUSIONBlood vessel defect gap width is different from the actual vessel defect, but is larger than the actual vessel defect. Misjudgment of the vascular defect length will lead to more vascular transplantation. The vast majority of vascular defect can be directly repaired by the method of end to end anastomosis after the vessel free and stretch.

6.A clinical research about predicting the changing of malignant tumor patients serum glucose after chemotherapy by SELDI technology

Objective By surface-enhanced laser desorption / ionization time-of-flight mass spectrometry(SELDI-TOF-MS), the serum pmteomic fingerprints related with the changing of malignant tumor patients' serum glucose after chemotherapy was selected and constructed as an predictive model. Methods By SELDI-TOF-MS, the serum of 182 malignant tumor patients who had received chemotherapy were tested, and the pmteomic fingerprints were received. After 2 years follow-up, all the patients were divided into 3 groups: the euglycemia group(136 people), the carbohydrate tolerance abnormality group(27 people), and the diabetes mellitus group (19 people). The proteomic fingerprints were analyzed by Biomarker Wizard Software and the idio-proteomic fingerprint of protective models were constructed by BPS (biomarker pattern software). Results The diagnosis model composed with 2 proteins (M/Z values were 5298 and 9608) could classify the carbohydrate tolerance abnormality group, and the diabetes mellitus group correctly. In the test model, the sensitivity and specificity were 81.48 %(22/27) and 100.00 %(17/17) respectively, the accuracy was 88.64 % (39/44). The diagnosis model composed with 3 proteins (M/Z values were 10324, 2761 and 4084) could classify the diabetes mellitus group and the euglycemia group correctly. In the test model, the sensitivity and specificity were 62.35 %(53/85) and 88.24 %(15/17) respectively, the accuracy was 66.67 %(68/102). The diagnosis model composed with 6 proteins (M/Z values were 5895,6010,6099,3930,5430 and 2495) could classify the diabetes mellitus group and the the carbohydrate tolerance abnormality group correctly. In the test model, the sensitivity and specificity were 77.65 %(66/85) and 96.30 %(26/27) respectively, the accuracy was 82.14 %(92/112). Conclusion SELDI-TOF-MS could be utilized to analyze protein profiling in screening serum glucose changing-related biomarkers and developing diagnostic and predictive patterns, and the developed patterns may be used to predict the changing of serum glucose after chemotherapy in malignant tumor patients.
7.Study on Screening the High-Efficiency Predominant Bacteria for Decompounding the Organic Wastes in Rubber Industry Wastewater

Objective To screen the high-efficiency predominant bacteria which can decompound 2-mercaptobenzothiazole, the accelerant for producing latex, in the organic wastewater. Methods Sampling from manufacturing environment, we got the predominant bacteria by primary screening, isolating and functional tests, and performed simulated test ground decompounding tests by using all bacteria. The enrichment of the predominant bacteria was followed by screen and identification to select the high-efficiency bacteria. Results 75 strains of predominant bacteria were obtained by primary screening. The simulated decompounding tests were performed after the mixed bacteria were tamed. The ratio of elimination for chemical oxygen demand (COD) was about 60.8%-97.7%, and the average was 77.2%. The predominant bacteria adhered to the surface of the active carbon, the carrier, and formed the biological film. Through screening and identification the Bacillus cereus showed to be predominant (90%). Conclusion The technology of high-efficiency predominant bacteria can be used for decompounding 2-mercaptobenzothiazole in the organic wastewater.

Protein fulfilling the their roles, one of important ways is through protein-protein interaction. In functional genomic era, identifying all of protein-protein interaction in proteome and mapping the protein interactions that have been attracting many scientists' attention , of which large-scale yeast two-hybrid system is one strategy of most widely used. In recent two years, ambitious projects have launched to examine all of the protein-protein interaction in Saccharomyces cer-evisiae using large-scale yeast two-hybrid system. Nevertheless, huge protein network is larger than that we predict and single yeast two-hybrid system cannot solve all the problems, which need be complemented by other wags.
9.Experimental study of repairing damaged human amniotic epithelial cells with formulated fibrin clot and cell growth factor

Objective To investigate whether damaged human amniotic epithelial cells(HAEC)could be repaired on the matrix of formulated fibrin clot in vitro and the effects of epidermal growth factor(EGF),basic fibroblast growth factor(bFGF)and transforming growth factor ?_1(TGF-?_1)on the proliferation of HAEC. Methods Ring drill was used to drill the HAEC layer on culture sheets to make quantified models of damaged HAEC, on which the lacks were then covered with fibrin clot. Subsequently, EGF (EGF group), bFGF(bFGF group) and TGF-?_1 (TGF-?_1 group) of different concentration were added into the sheets respectively. After the predesigned culturing time, the growing and transiting conditions of HAEC were observed under inverted microscope after Giemsa stain. Also, the proliferating conditions of HAEC were detected by using 5-bromodeoxyuridine(BrdU). Results In all groups, HAEC could transit toward damaged area on fibrin clot and grow there. Higher transiting speed and larger cell numbers were observed in the EGF and bFGF groups followed by the control group, while the TGF-?_1 group showed the relatively poorer results. Proliferating rates of HAEC were 17.8%, 28.0%, 35.3%, 51.6%, 34.1%, 34.2% and 26.0% respectively by EGF of different cultured concentration (1.0ng/ml, 5.0ng/ml, 10.0ng/ml, 20.0ng/ml, 40.0ng/ml, 80.0ng/ml and 160.0ng/ml). Proliferating rates of HAEC were 18.0%, 35.7%, 43.0%, 52.7%, 67.4%, 43.6% and 30.5% respectively by bFGF of different cultured concentration (1.0ng/ml, 5.0ng/ml, 10.0ng/ml, 20.0 ng/ml, 40.0 ng/ml, 80.0 ng/ml and 160.0ng/ml). Compared with the control group, EGF groups (EGF concentration ranging from 10ng/ml to 80ng/ml) and bFGF groups (bFGF concentration ranging from 5ng/ml to 80ng/ml ) showed better proliferating effects of HAEC(P
10.Effect of transforming growth factor beta 1 on vein transplantation

BACKGROUND:Many previous experiments have confirmed that transforming growth factor?1(TGF-?1) plasmids can inhibit or decrease the injection in nerve transplantation,but there has rare report about their effects on vein transplantation.OBJECTIVE:To investigate the action pathway of immune tolerance of allogenic femoral vein transplantation in rats induced by local injection of TGF-?1.DESIGN,TIME AND SETTING:A randomized block animal observational experiment was performed at Animal Experimental Center of Harbin Medical University from March 2007 to April 2008.MATERIALS:A total of 18 Wistar rats were selected to assign as donors.A total of 48 Sprague Dawley rats were selected to assign as recipients and randomly divided into four groups with 12 rats in each group:autograft,allograft,immunosuppressant,and TGF-?1 groups.METHODS:The rats in the immunosuppressant group were administated(i.p.) cyclosporin A(10 mg/kg) one time per day from three days before transplantation till they were sacrificed.Each rat in the TGF-?1 group was injected 40 ?g TGF-?1 plasmids into two ends of femoral vein.Vein transplantation was performed in the autograft and allograft groups.MAIN OUTCOME MEASURES:Imaging,histology and immunological assays of these groups were conducted at two weeks after transplantation.RESULTS:In the autograft group,the endothelial cells of vein were flat,and nuclear membrane thickened.In the allograft group,the exfoliated endothelial cells and fragments,as well as subendothelium were observed.In the immunosuppressant group,the nuclear membrane of endothelial cells thickened,and rough endoplasmic reticulum expanded widely.In the TGF-?1 group,the expand of rough endoplasmic reticulum and mitochondrial vacuolation were observed.Tight junction could be seen between adjacent cells.Histologic and immunologic parameters in the TGF-?1 group were better than those in the immunosuppressant and allograft groups.The absorbance value of lymphocyte culture in the autograft,allograft,immunosuppressant,and TGF-?1 groups were 1.07?0.14,4.15?0.67,1.77?0.23 and 1.38?0.23,respectively.The absorbance value of lymphocyte culture in the TGF-?1 group was significantly lower than that in the immunosuppressant and allograft groups(P
