Objective To investigate the effects of Simo decoction on ghrelin and calcitonin gene-related peptide (CGRP) in diabetic rats.Methods A total of 48 SD rats were divided into healthy control group,diabetes group (DM),DM treated with low dose Simo decoction group (DMTL),DM treated with medium dose Simo decoction group (DM-TM),DM treated with high dose Simo decoction group (DM-TH) and DM prevented with medium dose Simo decoction group (DMPM) ; there were eight rats in each group.The rats of DM group and other intervention groups were intraperitoneally injected with 1％ streptozotoein (STZ) to establish the model.The rats of healthy control group were injected with solvent.At eight weeks after the model successfully established,the rats of each intervention group were gavaged with low dose of Simo decoction (0.15 ml · kg-1 · d-1 ),medium dose of Simo decoction (1.5 ml · kg-1 · d-1 ) and high dose of Simo decoction (3.0 ml · kg-1 ·d-1 ) for two weeks; the rats of control group and DM groups were gavaged with equal volume of 0.9％NaCl.The rats of DM-PM group were gavaged with Simo decoction at 1.5 ml · kg-1 · d-1 for 10 weeks after the model successfully established.Rats were sacrificed at the end of drug intervention and gastric antrum tissues were dissociated for detecting the expression of ghrelin and CGRP by real time polymerase chain reaction (PCR).The data were analyzed by one way ANOVA and t test.Results There was no significant difference in ghrelin expression at mRNA level in gastric antrum tissues between diabetic rats and control group.However after intervened by low dose and medium dose of Simo decoction,the ghrelin expression increased (1.45 ± 0.34,1.87 ± 0.68 compared with 0.87 ±0.28,Dunnett's T3 test,P＜0.05).There was no effects of high dose Simo decoction on the expression of ghrelin in diabetic rats.There was no significant difference in ghrelin between DM-TM and DM-PM.The expression of CGRP at mRNA level in gastric antrum tissues of diabetic rats was significantly higher than that of control group (4.61 ± 1.67 compared with 1.06 ± 0.40,t =5.843,P＜0.01).There was no effects of low dose Simo decoction on CGRP expression of diabetic rats.The expression of CGRP of diabetic rats was down-regulated by medium and high dose of Simo decoction intervention.There was no significant difference in CGRP expression between DM-TM and DM-PM.Conclusions Simo decoction can promote ghrelin expression in gastric antrum of diabetic rats.The expression of CGRP in gastric antrum of diabetic rats significantly increased,but Simo decoction may down-regulate the expression of CGRP in gastric antrum of diabetic rats.

Objective:To seek differentially expressed plasma proteins of unstable angina blood-stasis syndrome.Methods:2-dimensional di rence gel electrophoresis and mass spectrometry were used to screen the di erentially expressed plasma proteins among unstable angina blood-stasis syndrome and healthy volunteers.Results:Compared with healthy people,the express of Clusterin,ApoA-I of unstable angina blood-stasis syndrome patient decreased while the expression of Fibrinogen ? chain,Vitamin D-Binding Protein,Haptoglobin ? chain,Haptoglobin ?2 chain increased.Conclusion:Fibrinogen ? chain,Fibrinogen ? chain,Haptoglobin ? chain,Haptoglobin ?2 chain,Clusterin,ApoA-I may be correlative with blood-stasis syndrome.

Background and purpose:Human epidermal growth factor receptor-2 (HER-2), a member of epidermal growth factor receptor family, initiates a diverse set of signaling pathways that ultimately affect such fun-damental processes as cell proliferation, cell motility and cell apoptosis. It is reported that HER-2 was associated with epithelial-mesenchymal transition (EMT) process. However, the mechanism needs further investigation. The purpose of this study was to investigate the mechanism of HER-2 on regulating EMT process.Methods:Transwell assay was used to determine the motility of breast cancer cells; Real-time lfuorescence quantitative polymerase chain reaction (RT-FQ-PCR) was employed to determine the expression of genes of interest, and reactive oxygen species production was measured by reactive oxygen species detection kit.Results:HER-2 overexpression in breast cancer cells could promote cell migration and invasion. Mechanistic study showed that HER-2 overexpression could upregulate ZEB1 expression. ZEB1 silencing by siRNA reduced cell motility of HER-2-overexpressing breast cancer cells. Furthermore, reactive oxygen species produced in HER-2-overexpressing breast cancer cells were less than those produced in corresponding control cells.Conclusion:Our study demonstrated that HER-2 overexpression endowed breast cancer cells with EMT related properties by upregulating ZEB1 expression. ZEB1 could be a candidate target for further study of the relation-ship between HER-2 and EMT.

Objective To evaluate the reliability of autologous blood withdrawal during cesarean section. Methods Fifteen patients preoperatively diagnosed with pernicious placenta previa and∕or accrete by using ultrasound and magnetic resonance imaging, aged 20-35 yr, weighing 55-75 kg, at≥36 weeks of gestation, were enrolled in the study. Blood containing amniotic fluid from the surgical field was collected, and the washed blood was processed using cell?salvage machine and then filtered using a leukocyte depletion filter during cesarean section. The 20 ml blood samples collected included maternal central venous blood after delivery of fetus, unwashed blood, washed blood and filtered blood. The fetal squamous cells were counted using papanicolaou staining. The concentrations of a?fetoprotein, tissue factor, endothelin?1 and histamine were measured by enzyme linked immunosorbent assay. The fetal red blood cells were counted using the acid elution method and HE staining. Results Compared with unwashed samples, the tissue factor concentrations were significantly increased, and the fetal squamous cell count, concentrations of a?fetoprotein and endothelial?1, and fetal red blood cells were decreased in the washed samples. Compared with washed samples, the fetal squamous cell count, concentrations of a?fetoprotein and fetal red blood cells were significantly decreased in filtered samples. Compared with maternal venous blood samples, the tissue factor concentrations were significantly increased, and the fetal squamous cell count and concentrations of a?fetoprotein and endothelial?1 were decreased in filtered samples. Conclusion Autologous blood withdrawn during cesarean section can be used for reinfusion in cesarean section.

OBJECTIVETo seek the special plasma molecule in unstable angina (UA) patients of qi deficiency and blood stasis syndrome (QDBS) and method for explore the proteomic specificity of the disease.

METHODSLC-MS(E) analysis was performed in UA patients of QDBS or non-QDBS and in healthy persons after the 6 proteins with optimal abundance in plasma being removed by polyclonal antibody affinity column (product of Agilent Co. USA).

RESULTSActin were found only expressed, and FN, ApoH and ANXA6 were found highly expressed in plasma of patients with UA-QDBS, suggesting they might be the special molecules for the disease. Moreover, as compared with health persons, SAA, CP, MYH11 and C6 showed high expression, and the 6 proteins, e.g. A1BG, ApoA4, GSN, HBB, HBD and TF, showed low expression in the plasma of UA-QDBS patients.

CONCLUSIONUA-QDBS might belong to a kind inflammatory reaction. There are simultaneous existence of myocardial injury, blood coagulation factor abnormality, lipid metabolic disorder and oxygen transport obstacle in patients of UA-QDBS, they influence and interact mutually. The newly discovered differential proteins might provide clues for studying or discovering new protein targets of angina relieving drugs. The new technique of label free quantitative proteomics is an efficient method for bio-marker research of diseases and syndromes.

Objective To analyze the ophthalmologic characteristics and examinations which caused by vertebral-basilar insuffi- ciency.Design Retrospective case series.Participants 469 eases of vertebral-basilar insufficiency.Methods Retrospective analysis of 469 cases of vertebral-basilar insufficiency patients with ophthalmologic signs' features,incidence,the main cause,color Doppler flow imaging and(or)digital subtraction angiography clearly stenosis location,the degree of stenosis from July 1st 2005 to July ist 2007 in PLA General Hospital.Main Outcome Measures Ophthalmologic signs' related reasons and supplementary examination.Results 96 cases were with the corresponding eye symptoms(20.4%).Vision decreased in 83 cases,diplopia was in 56 cases,ipsilateral hemiopia quadrant blind in 12 cases and depending on deformation in 10 cases,fundus arteriosclerosis in 72 cases,nystagmus in 39 cases,the eye movement disorder in 16 cases.Among 96 patients with associated ocular symptoms,75 patients with color Doppler flow imaging (CDFI)showed unilateral vertebral artery abnormal in 66 cases(88%),among those blood flow velocity decreased in 26 cases,17 cases with vascular plaque stenosis,thinning in 20 cases,3 cases with complete occlusion;different bilateral vertebral artery abnormal in 9 cases(12%),among those velocity dropped in 4 cases,two cases with vascular plaque stenosis and thinning in 3 cases.Forty-five cases with the digital subtraction angiography(DSA),left vertebral artery was abnormal in 24 cases,18 cases with stenosis,occlusion in 5 cas- es,abnormal bending in 1 case.Right vertebral artery was abnormal in 16 cases,13 cases with stenosis,occlusion in 3 cases.Bilateral vertebral artery stenosis was found in 2 cases;vertebral artery stenosis was in 3 patients.Conclusion Vertebral-basilar insufficiency may cause eye-related symptoms,therefore,vision acuity decrease,fundus 'arteriosclerosis,diplopia and nystagmus symptoms are more likely for patients with vertebral-patients with basilar artery insufficiency performance of the eye.(Ophthalmol CHN,2002,16:406-410)

OBJECTIVETo explore the relation of the anogenital distance (AGD) with cryptorchidism in male newborns.

METHODSThis study included 350 male infants delivered in two community hospitals between September 2013 and September 2014. Within 24 hours after birth, a pediatric surgeon measured the AGD of the neonates and determined whether they had cryptorchidism. According to the testicular position, we divided the undescended testes into three types: upper scrotal, inguinal, and non-palpable.

RESULTSTotally 39 cases of cryptorchidism were found in the 350 newborns. The AGD of the cryptorchidism infants was significantly shorter than that of the normal neonates ([2.01 ± 0.22] vs [2.35 ± 0.19] cm, P < 0.01), and statistically significant differences remained even when preterm and low birth-weight infants were excluded ([2.32 ± 0.14] vs [2.06 ± 0.19] cm; (2.37 ± 0.17) cm vs (2.12 ± 0.12) cm, all P < 0.01). The newborns with higher-position cryptorchidism had a shorter AGD, though with no significant difference (F = 0.434, P > 0.05). No significant differences were observed in the AGD between unilateral and bilateral cryptorchidism ([1.96 ± 0.13] vs [2.02 ± 0.17] cm, P > 0.05).

CONCLUSIONShorter AGD is associated with a higher incidence of cryptorchidism in male newborns. AGD could serve as a potential biomarker for disruption of androgen action during the male programming window period.

ObjectiveTo examine the test-retest reliability and measurement error in using a Jamar dynamometer to determine grip strength for elder men. Methods40 elder men with a mean age of 82 years old volunteered to participate in this study. The grip strength was administered twice using a Jamar dynamometer by specially trained testers; subjects were re-tested at the same time of day a week later by the same testers. ResultsThe infraclass correlation coefficient（ICC） was 0.99. The standard error of measurement (SEM and SEM%) were 1.18～1.69 and 4.11%～6.41%. The smallest real difference (SRD and SRD%) were 3.27～4.68 and 11.39%～17.75%. The Bland-Altman analysis revealed no systematic errors between test and retest. ConclusionThe Jamar dynamometer is reliable and with acceptable measurement errors to determine grip strength for elder men.

OBJECTIVETo review the progress of cardiac differentiation and electrophysiological characteristics of bone marrow mesenchymal stem cells.

DATA SOURCESThe databases of PubMed, Springer Link, Science Direct and CNKI were retrieved for papers published from January 2000 to January 2012 with the key words of "bone marrow mesenchymal stem cells, cardiac or heart, electrophysiology or electrophysiological characteristics".

STUDY SELECTIONThe articles concerned cardiac differentiation and electrophysiological characteristics of bone marrow mesenchymal stem cells were collected. After excluding papers that study purposes are not coincident with this review or contents duplicated, 56 papers were internalized at last.

RESULTSFor the treatment of myocardial infarction and myocardiac disease, the therapeutic effects of transplantation of bone marrow mesenchymal stem cells which have the ability to develop into functional myocardial cells by lots of methods have been proved by many researches. But the arrhythmogenic effect on ventricles after transplantation of bone marrow mesenchymal stem cells derived myocardial cells is still controversial in animal models. Certainly, the low differentiation efficiency and heterogeneous development of electrical function could be the most important risk for proarrhythmia.

CONCLUSIONMany studies of cardiac differentiation of bone marrow mesenchymal stem cells have paid attention to improve the cardiac differentiation rate, and the electrophysiology characteristics of the differentiated cells should be concerned for the risk for proarrhythmia as well.

Objective To investigate the consistency of the lymphocyte immunophenotype between labial gland and peripheral blood of patients with primary Sj(o)gren's syndrome (pSS).Methods Seveny-one pSS patients (referred as pSS group) from rheumatology department and 35 subjects patients with maxillofacial trauma (referred as control group) from department of head and neck surgery of general hospital at tianjin medical university (Tianjin,China) during August 2013 to April 2016 were included into this study.Based on the ratio of CD20 to CD3 in labial gland from pSS patients,they were divided into CD20 high proportion group and CD20 low proportion group.Lymphocyte immunophenotypes in labial glands,course of disease,erythrocyte sedimentation rate (ESR),C-reactive protein (CRP) level,immuno-globulin (Ig) and complement levels were compared among groups.Results ① The levels of serum IgG,IgA,IgM and CRP,and ESR were higher,but C4 level was lower in pSS patients than in the control group [IgG:(22 990±8 060) mg/L vs (1 1560±1 290)mg/L,t=l 1.645,P＜0.01;IgA:(3 710±1 400) mg/L vs (2 390±800) mg/L,t=6.138,P＜0.01;IgM:(1 580±1 300)mg/L vs (920±390) mg/L,t=3.893,P＜0.01;ESR:(37±14) mm/1 h vs (14±4) mm/1 h,t=12.723,P＜0.01;CRP:(5.9±8.7) mg/L vs (2.5±1.2) mg/L,t=3.199,P＜0.01;C4:(180±60) mg/L vs (250±40) mg/L,t=-6.850,P＜0.01].② The levels of IgG,IgA,IgM and C3c in labial gland were higher (IgG:Z=-6.264,P＜0.01;IgA:Z=-1.997,P＜0.05;IgM:Z=-5.459,P＜0.01;C3c:Z=-8.533,P＜0.01),but Clq was lower in pSS group than in control group (Z=-4.363,P＜0.01).③ CD20 was detected in labial gland samples of all pSS patients,in which CD3 was positive in 66 patients,and negative in 5.④ Serum IgG level,ESR were higher in CD20 high proportion group than in low proportion group [IgG:(24 970±7 510) mg/L vs (18 860±7 740) mg/L,t=3.176,P＜0.01;ESR:(40±13) mn/1 h vs (32±15) mm/1 h,t=2.148,P＜0.05].⑤ The levels of IgG and Clq in labial gland were higher in CD20 high proportion group than in low proportion group [IgG:Z=-5.387,P＜0.01;C1q:Z=-4.724,P＜0.01].Conclusion pSS patients have a higher proportion of CD20 in infiltrating lymphocytes of the labial gland,accompanied with the changes of immunoglobulins and complements in both labia gland and peripheral blood.
