Chinese Pharmaceutical Journal 2013;48(4):289-292
OBJECTIVE: To study the effect of rabeprazole on the steady-state pharmacokinetics of clopidogrel in rats. METHODS: Eighteen healthy SD rats were divided into three groups, which were intragastricly administered clopidogrel (30 mg · kg-1 middot; d-1), rabeprazole (8 mg · kg-1 middot; d-1) + clopidogrel (30 mg · kg-1 middot; d-1), omeprazole (8 mg · kg-1middot; d-1) + clopidogrel (30 mg · kg-1 middot; d-1) respectively for 7 d. The plasma concentrations of the metabolite of clopidogrel, SR26334, were determined by HPLC-DAD. The pharmacokinetic parameters of SR26334 were obtained with statistical analysis by DAS 3.0. RESULTS: There was no significant difference between the single-drug group and the rabeprazole combination group in the main pharmacokinetic parameters of SR26334, such as AUC0-t, AUC0-∞, MRT0-t, t1/2, CLz/F, ρmax and ρss (P≤0.05), except that the tmax significantly decreased from (1.17±0.41) h in the single-drug group to (0.58±0.20) h in the combination group (P<0.01). Compared with single-drug group, the AUC0-t, and AUC0-∞ of the omeprazole combination group increased for about 20% (P<0.05), MRT0-t significantly prolonged (P<0.01), and CLz/F decreased for about 20% (P<0.05). CONCLUSION: Long-term combinaton treatment with rabeprazole can accelerate the in vivo metabolism of clopidogrel to SR26334, but there is no significant influence on the degree of metabolism.

Tianjin Medical Journal 2014;(2):101-104
Objective To construct the luciferase reporter gene vector of cell division cycle 2 (Cdc2) gene promoter and determine its transcriptional activity. Methods Primers were designed based on human Cdc2 promoter sequence from UCSC software. Then Cdc2 promoter from human genome DNA was replicated. After pGL3-Basic vector and Cdc2 promoter were digested with restriction enzymes SacⅠand XhoⅠseparately, Cdc2 promoter was inserted into pGL3-Basic vector. The recombinant plasmid named pGL3-Cdc2-promoter was transiently co-transfected into U2OS cells with control vector pRL-SV40, and then the activity of dual luciferase was detected. Results pGL3-Cdc2-promoter was constructed successfully. The restriction analysis and sequencing proved the entirely correct sequencing results. The luciferase activity was higher in pGL3-Cdc2-promoter/pRL-SV40 group than that of pGL3-Basic/pRL-SV40 group (1.591 5±0.199 8 vs 0.049 9±0.010 4). Conclusion pGL3-Cdc2-promoter can be transcribed and activated in U2OS cells. This study provided an important basis for screening and evaluation of anticancer drugs.

Carthamus tinctorius L. is a traditional Chinese medicine with the effect of promoting blood circulation and removing blood stasis. HSYA (hydroxysafflor yellow A) is the main effective component of Carthamus tinctorius L. In order to study the inhibitory effects of HSYA against PMN (polymorphonuclear) activation induced by LPS (lipopolysaccharide), rabbit PMN adhesion potency which was activated by LPS through colorimetry method was observed. Cellular free calcium concentration was determined by fluorescence spectrophotometry. RT-PCR was applied to study the effect of HSYA on PMN TNF-alpha and IL-6 mRNA expression; The inhibition of HSYA on NF-kappaB activation was monitored with immunofluorescence. The results showed that after treated with HSYA, the increase of adhesion potency (HSYA dose 1.01 x 10(-4) mol x L(-1)), free calcium concentration (HSYA dose 3.1 x 10(-5) mol x L(-1)), TNF-alpha and IL-6 mRNA expression elevation (HSYA dose 5.2 x 10(-1) mol x L(-1)) induced by LPS were inhibited. HSYA can inhibit NF-kappaB p65 subgroup nuclear translocation (HSYA dose 5.2 x 10(-5) mol x L(-1)). It is suggested that HSYA is effective in PMN activation induced by LPS.

Chinese Journal of Orthopaedic Trauma 2004;0(09):-
Objective To evaluate the feasibility and principle of direct reduction and fixation through the pedicle of fractured vertebra in treatment of thoracolumbar burst fractures. Methods Between June 2001 and November 2005,24 cases of one level thoracolumbar burst fracture,16 males and eight females, were treated in our department.Their average age was 32.5 years old.The fractured vertebrae were as follows:T11 in two cases,T12 in nine cases,L1 in 11 cases and L2 in two cases.According to the ASIA(American Spinal Injury Association) neurological function grading system, there were one grade A,two grade B,five grade C,three grade D anti 13 grade E.The vertebral canal area blocked was rated as gradeⅠ(<1/4)in three cases,gradeⅡ(1/4~1/2)in nine cases,gradeⅢ(1/2~3/4)in 10 cases and gradeⅣ(>3/4)in two cases. The pedicle-screws were im- planted into the pedicles of the fractured vertebra and its adjacent ones. The connective rod was pre-bent according to the normal sagittal curvature of the fixation segment. After necessary decompression of the vertebral canal, the pedicle-screws stabilized the rod which was rotated to prop open upper and lower pedicle screws and push the burst vertebra so that the kyphosis and lateral dislocation were corrected. The graft bone was implanted between the transverse processes. Results On average, the height of fractured vertebra was restored from 48.3% of the normal value before operation to 93.6% after operation, the lateral dislocation from 17.1% to 0.6%,the Cobb's angle on the sagittal plane from 26.5?to 3.1?,and the Cobb's angle on the frontal plane from 9.5?to 0?.The area of vertebral canal was enlarged from 44.6% of the normal value to 92.1%.There were no complications. For in- complete neurological injuries, improvement of one to two grades was made in neurological functions. Conclusion Direct reduction and fixation through pedicles of fractured vertebrae is a safe and effective therapeutic option to treat the thoracolumbar burst fracture, for it can improve the stabilization and stress distribution.
