1.Promotion of resveratrol on psoriasis like skin damage by down-regulating expression of Keratin 17

Objective To observe the therapeutic effect of resveratrol (Rsv) on psoriasis like skin damage induced by imiquimod (IMQ) and study its mechanism.Methods BALB/c mice were randomly divided into four groups:control group,Rsv group,model (IMQ) group and Rsv treatment (IMQ + Rsv) group with six mice in each group.Psoriasis like skin model induced by smearing IMQ in the mice's ear for 8 d,20 mg Rsv cream were gave at the same time for treatment.Observe erythema and psoriasis degree,and measure ear thickness with vemier caliper.HE staining was used to observe tissue change,real time quantitative PCR (qRT-PCR) was used to detect K17,IL-17A,IL-19 and IL-23 mRNA and Western blotting were used to detected K17 protein expression in tissues.Results Compared with the model group,mice erythema area and the thickness of epidermal scale decreased significantly in Rsv treatment group;HE staining results showed that,Rsv significantly reduced ear marginal tissue epidermal thickness,and diminished severity of the psoriasis-like skin inflammation.qRT-PCR and Western blotting confirmed a Rsv dependent decrease of in K17,IL-17A and IL-19 in mRNA level and protein levels of K17.Conclusion Rsv improves psoriasis like skin damage by down-regulating expression of Keratinl 7.
2.Low-dose nonylphenol promotes the proliferation of DU-145 cells and expression of membrane estrogen receptor GPR30 in DU-145 cells.

OBJECTIVETo observe the effects of low-dose exogenous estrogen nonylphenol (NP) on the proliferation of human prostate cancer cell lines DU-145 and the expression of the membrane estrogen receptor GPR30 in the DU-145 cells.
METHODSWe exposed DU-145 cells to different concentrations of NP for 24 hours, followed by measurement of the half maximal inhibitory concentration (IC50) of the cells by cell proliferation assay and determination of the concentration of exposure to low-dose NP. We also observed the expressions of 3 estrogen receptors (ER), including ER-alpha, ER-beta and membrane estrogen receptor GPR30, in the DU-145 cells exposed to low-dose NP by RT-PCR.
RESULTSCell proliferation assay showed that within a certain range of doses, NP inhibited the proliferation of the DU-145 cells with an IC50 of 46 micromol/L, a much lower dose of NP than IC50, 0.01, 0.1.1 micromol/l NP, that can promote the proliferation of DU-145 cells. The results of RT-PCR indicated that the expressions of the three ERs in the DU-145 cells were similar to those in prostate epithelial cells, and that low-dose NP promoted the expression of GPR30.
CONCLUSIONMembrane estrogen receptor GPR30 may play a role in low-dose NP promoting the proliferation of DU-145 cells.

4.Finite element analysis of cemented long-and short-stem prosthetic replacement in elderly patients with partial marrow type Ⅰ intertrochanteric fractures

Objective To investigate the stress distribution in a cemented femoral component in elderly patients with a partial marrow type Ⅰ intertrochanteric fractures and to compare differences in the stress distribution after a long-and short-stem prosthetic replacement.Methods Spiral CT images of right femur of the volunteer were obtained and processed with Mimics software and modeling software to reconstruct a three-dimensional model of the femur.Based on this,a three-dimensional physical model for partial marrow type Ⅰ intertrochanteric fractures,long-and short-stem femoral prostheses,and cement mantle was established.Subsequently,a three-dimensional finite element model of long-and short-stem prosthetic replacement for intertrochanteric fractures was established using the finite element analysis software,and biomechanical analysis was implemented for the model.Results No significant changes in stress distribution of the femur occurred after long-or short-stem prosthetic replacement.Stress on the femur still presented a gradual increase from the proximal end to the distal end,peaked at the lower 1/3 of the medial and lateral junction and then decreased at the end.A stress concentration zone formed in the medial and lateral end of cement-stem interface in short-stem prosthetic replacement.Besides,the maximum value of stress in the lateral interface reached 15.3 MPa,but without surpassing the fatigue strength of the bone cement.Whereas,a stress concentration zone formed in the distal medial and lateral part of cement-stem interface and the medial middle part of the interface in long-stem prosthetic replacement and the maximum value was also lower than the fatigue strength of bone cement.No significant stress concentration zones were found in the femoral calcar reconstructed using bone cement.Conclusions Cemented long-or short-stem prosthetic replacement achieves no significant alteration in stress distribution of the femur.Loosening probability of the cemented long-or short-stem prosthesis is almost the same,but the latter has advantages of shorter surgery time,minor trauma,and fewer complications and may be more suitable for treatment of the elderly patients with a partial marrow type Ⅰ intertrochanteric fractures.
6.Seroepidemiology of varicella-zoster virus infection measured by the fluorescent antibody to membrane antigen test

Objective Adopting serology assay-fluorescent antibody to membrane antigen (FAMA) as "gold standard" , sero-prevalence status of varicella-zoster virus (VZV) infection was investigated, in Guangzhou. Methods FAMA test was established with VZV infected human embryo fibroblasts as antigens and fluorescein isothiocyanate(FITC) labeled goat anti-human IgG as the secondary antibody. Sensitivity and specificity of the assay were evaluated. The sero-prevalence of anti-VZV IgG in 592 serum specimens randomly collected from a clinical laboratory, was analyzed with FAMA. Results Data from FAMA test showed no cross-reaction with other Herpesviruses when it was used to detect VZV antibodies. The overall prevalence of VZV antibody was 76.52%. Age-specific prevalence rates of VZV antibody in different age groups as: 1-3, 4-6, 7-13, 14-19, 20-29, 30-39, 40-49, ≥50, were found to be 14.67%, 51.56%, 73.91%, 91.26%, 92.78%, 95.65%, 98.11% and 100%, respectively.The sero-prevalence of 1-3 age group appeared the lowest but rose sharply with the increase of age but showing no association with gender. Conclusion Our data indicated that VZV infection occurred in early childhood, in Guangzhou, suggesting that the primary recipients of VZV vaccine should be under the 1-3 age group. Additional subjects for vaccination would be children above 3 years old with no history of VZV infection, and serology test negative for VZV. The assay was validated by its excellent specificity and could be used as the first choice in the detection of protective antibodies against VZV infection.
7.Serum vancomycin concentration distribution and its clinical therapeutic effect on children with severe Gram-positive cocci pneumonia

Objective To analyze the characteristics of serum vancomycin concentrations and its clinical therapeutic effects. Methods Serum vancomycin concentrations of 59 children diagnosed with severe Gram positive cocci pneumonia and treated with vancomycin were retrospectively analyzed. Vancomycin concentrations, biochemical values and disease status of patients were analyzed. Results The serum vancomycin concentrations of severe Gram positive cocci pneumonia children accompanied by acyanotic congenital heart disease was significantly higher than those without congenital heart disease, ( 12 . 12 mg/L vs 7 . 76 mg/L, P=0 . 008 ). The therapeutic effect of 40-60 mg/(kg·d) dosage group was signiifcantly higher than that of40 mg/(kg·d) group (89.47% vs 46.15%, P=0.004), while the therapeutic effect was similar between 40-60 mg/(kg·d) and >?60 mg/(kg·d) dosage group. Acute liver function damage and moderate/severe anemia may be risk factors for poor therapeutic effects to severe Gram positive cocci pneumonia children (P?0 . 05 ). Conclusions Severe Gram positive cocci pneumonia children accompanied by acyanotic congenital heart disease may lead to a high serum vancomycin concentration. The 40-60 mg/(kg·d) dosage group may reach a satisfactory therapeutic effect. For children with acute liver function damage and moderate/severe anemia, a close monitoring to the state of illness is recommended to prevent poor prognosis.

9.A murine model with murine cytomegalovirus infection resulting in colon inflammation after allogeneic skin transplantation.

UNLABELLEDTo provide a reliable animal model for study of human CMV disease in gastrointestinal track, we tried to infect with murine cytomegalovirus (MCMV) in mice that were received allogenetic skin transplantation under immunosuppression. (1) Skin transplantation was performed between 18 donor C57BL/6 mice and 72 recipient BALB/c mice. (2) All recipient mice were then given Cyclosporine at 12 mg/kg daily for 2 weeks by intraperitoneal injection. Mice were randomly divided into 3 groups. Two experimental groups were received MCMV-infected mouse embryonic fibroblasts (MEF) at 10(4) PFU and 10(5) PFU respectively, and the control group received MEF only. We observed any possibly pathophysiological behavior changes and recorded the changes in body weight. The mice were sacrificed at 5d, 9d, 14d, 21d post infection and colon tissue was collected for analysis.
RESULTSMice infected with MCMV at 10(5) PFU group showed anorexia, lethargy and degression in locomotor activity. This group of mice showed significant decrease in body weight than that of other groups. Colon tissues were collected 14 days after infection. Histological examination revealed that the mucous layer became thinner in the proximal colon and increased number of lymphoid follicles in distal colon in infected animals. The changes in the mucosal structure was most prominent in the group 10(5) PFU MCMV. Viral DNA was present in the colon by in situ hybridization for IE1 gene, and viral gB transcript was positive by RT-PCR. One of the viral major proteins, pp65, was widely distributed in the colon by immunohistochemistry. These data demonstrated that MCMV established infection in colon of the mice after allogenetic skin transplantation. Electron microscopy showed that there were herpes virus particles in the colon tissue.
CONCLUSIONInfection with MCMV in mouse after allogenetic skin transplantation by nasal cavity inoculation resulted in the pathological changes in colon tissue similar to that of inflammation in human colon. The small animal model of colon inflammation may provide a platform for further study of pathogenesis as well as medical intervention of HCMV involved inflammation of human bowel.

10.Effect of noise on the auditory system and the intelligence development of premature infants treated in the neonatal intensive care unit.

OBJECTIVEHigh noise levels (>70 dB) in the neonatal intensive care unit (NICU) are common in some primary hospitals. This study aimed to investigate the noise in the NICU on auditory system and intelligence development in premature infants.
METHODSOne hundred premature infants with respiratory distress syndrome who needed mechanical ventilation therapy were randomly divided into observation and control groups according to the use of earmuffs. The duration of mechanical ventilation therapy lasted for 2 to 15 days in the two groups. After weaning from mechanical ventilator, the auditory brainstem response, cranial B-ultrasonography, and the intelligence development assessment were performed.
RESULTSThe percentage of total (23% vs 47%) and mild hearing loss (15% vs 35%) in the observation group was significantly lower than that in the control group (p<0.05) 2 to 3 days after weaning from mechanical ventilator. The incidence of periventricular hemorrhage intraventricular hemorrhage (PVH-IVH) or periventricular leukomalacia (PVL) in the observation group was significantly lower than that in the control group (21% vs 42%; p<0.05). The intelligence development assessment performed in the first 6 and 12 months of life showed that the mental development index and the psychomotor development index in the observation group were much higher than those in the control group (p<0.05).
CONCLUSIONSThe noise in the NICU can result in mild hearing loss and retardation of intelligence development and increase the incidence of PVH-IVH and PVL in premature infants. The use of earmuff may reduce the adverse events.
