Objective To evaluate the diagnostic value of MRI in hepatic echinococcosis.Methods MRI features of 37 cases with hepatic echinococcosis proved by operation and pathology were analysed in comparison with that of CT.Results The hepatic echinococcosis could be divided into five types according to the manifestations of CT and MRI:simple cyst in 8 cases,multiple daughter cyst in 13 cases,ruptured cyst in 7 cases,consolidated and calcified cyst in 4 cases and mixed cyst in 5 cases.Conclusion MRI can clearly show and determine the location,size,form,number and types of hepatic echinococcosis,that are helpful for the plan of operation.
2.The Application of the Low Field MRCP in Obstructive Jaundice and theComparative Analysis with US and CT

Objective To evaluate the diagnostic value of the low field MRCP in obstructive jaundice by comparing its result with that of US and CT. Methods 55 cases of obstruction jaundice patients proved by clinical diagnosis were studied and compared the collected original diagnosis results by low filde MRCP with US and CT.Results Of 55 cases of obstructive jaundice, 30 were cholelithiasis ,21 were tumors and 4 belonged to the other type. The accuracy of MRCP for defining the site of obstruction was 100%. The accuracy for identifying the cause of obstruction was 81.8%.Conclusion The low filde MRCP can accurately define the obstructive site in biliary obstructive disease .When used together with 2D single-shot breath-hold MRCP , T 2WI, T 1WI and 3D-FSE MRCP shows high accuracy and sensitivity in diagnosing obstructive jaundice.
3.Predictors of femoral head avascular necrosis after internal fixation of femoral neck fractures

Objective To identify the independent prognostic factors for femoral head avascular necrosis after internal fixation of femoral neck fractures.Methods A retrospective analysis was made on 246 patients (246 hips) with femoral neck fractures treated by internal fixation between January 2002 and January 2010.There were 111 male and 135 female patients aged 19-59 years.A total of 101 patients were injured in traffic accidents,79 patients in falls from the height and 66 patients in ground-level falls.Eight parameters were identified as the prognostic factors of femoral head avascular necrosis,including gender,fracture type,preoperative traction,interval from injury to surgery,reduction methods,quality of fracture reduction,time of weight bearing,and fixation removal.Multivariate Logistic regression analysis was used to explore the risk factors of postoperative femoral head avascular necrosis.Results Period of follow-up was 1-13 years (mean,7.5 years).There were 38 patients (15.4%) with avascular necrosis of the femoral head.Multivariate analysis identified fracture type (95% CI-3.41--2.43,P < 0.01),preoperative traction (95% CI-3.77--0.6,P < 0.05) and quality of fracture reduction (95% CI -3.33--0.94,P < 0.01) as the independent predictors of avascular necrosis of the femoral head.Conclusion Fracture type,preoperative traction and quality of fracture reduction are the independent risk factors for avascular necrosis of the femoral head after internal fixation of femoral neck fractures.
4.Changes in expression of sodium channel?subunit mRNA in dorsal root ganglion in a rat model of neuropathic pain

Objective To examine the changes in the expression of voltage-gated sodium channel?subunit mRNA in the dorsal root ganglion(DRG)and the role it plays in the neuropathie pain.Methods Thirty- two male SD rats weighing 250-400g were randomly divided into 2 groups:groupⅠneuropathic pain(SNL,n= 20)and groupⅡsham operation(n=12).Neuropathic pain was produced by ligation of right sciatic nerve according to Seltzer.Paw withdrawal latency to noxious thermal(PWHL)and mechanical(PWML)stimulation were measured before(baseline)and 1,2,3,5,8,11,14,28 day after sciatic nerve ligation(SNL).DRG at L_(4,5) was isolated on the 14th day after SNL in 8 SNL and 4 sham-operated animals for determination of sodium channel?subunit mRNA expression(by in-situ hybridization).Results PWHL and PWML were significantly decreased on the 2nd-28th day after SNL as compared to the baseline in SNL group.There was no significant difference in?_1 subunit mRNA expression between the 2 groups.The?_2 subunit mRNA expression in DRG was hardly detectable.The?_3 subunit mRNA expression in DRG on the operated side was significantly higher in SNL group than in sham-operation group.Conclusion The up-regulation of sodium channel?_3 subunit mRNA expression in DRG may play an important role in neuropathic pain.
5.Preliminary Investigation of Imaging Parameters on Optimizing Low Field MRCP

Objective To investigate imaging parameters on low field MRCP and to optimize the quality of images.Methods MRCP in 77 cases were performed using Airis-Ⅱcomfort open 0.3T low field MR imaging system made by HITACHI company.3D-FSE combined with 2D single-shot breath-hold FSE sequences were used.The 3%~5% compound solution of Gd-DTPA was taken to restrain the hige signals from remaining liquid in stomach.Results MRCP examination was succeeded in all 77 cases and the images in all cases but 2 were fine for diagnosis.The total diagnostic accuracy was 79.2%.The accuracy of MRCP in the detection of the degree and location of pancreaticobiliary duct obstruction was 100%. Conclusion The distinct MRCP images can be obtained with low field MR system using combination of several scanning techniques and proper scanning parameters,and also patients have to prepare fully.

Objective To investigate the depressing effect of antigene peptide nucleic acid (AGPNA)on the k-ras gene expression of human pancreatic cancer Patu8988 cells, the inducing apoptosis effect on Patu8988 cells, and the biodistribution characteristics in nude mice bearing xenografts using 188Re-k-ras-AGPNA.Methods The expression level of k-ras mRNA and the expression ratio of k-ras protein in Patu8988 cells transfected with AGPNA was measured by RT-PCR and flow cytometry ,respectively. The degree of cellular apoptosis 3 to 5 d after treating Patu8988 cells with 188Re-k-ras-AGPNA or 188ReO4- was determined by flow cytometry. For biodistribution study, 58 nude mice bearing Patu8988 cell xenografts were divided into two groups: intratumoral injection of 188Re-k-ras-AGPNA (Group A) and 188ReO4- (Group B). At different time points, the mice were sacrificed and organs of interest were excised, weighted and counted by a gamma counter. The organ uptake was calculated as a % ID/g and the absorbed doses of organs were calculated. One-way analysis of variance was used. Results After transfected with 1 nmol/ml AGPNA, the k-ras mRNA gray scale ratio and the expression ratio of k-ras protein were 1.00 ± 0.39 and (15.05 ± 5.07)%, respectively. They were significantly lower than those of the control group with 1.86 ± 0.07 and (24. 38 ± 5.40) % (F = 2. 545, 5. 327, P<0. 05). At 4 and 5 d after treatment in Group A, float cells' apoptosis ratios were (26.30 ± 7.45) % and (27.90 ± 10. 38) %, respectively. Tumors were the major distribution site in Group A with uptake of (37.47 ±21.31), (35.96 ±7.80) and (15.46 ±4.93) %lD/g at 1 h, 1 d and 7 d after intra-tumor injection, respectively. The absorbed dose of tumor was 15 569 mGy/MBq. Condusions Transfection with k-ras-AGPNA on Patu8988 cells may inhibit k-ras expression at mRNA and protein expression level, and 188Re-k-ras-AGPNA can induce apoptosis of Patu8988 cells.Tumor is the major distribution site in nude mice bearing human pancreatic cancer xenografts after intratumoral injection of 188Re-k-ras-AGPNA.
8.Analysis of the monitoring results of iodized salt consumed by residents in Chenzhou city of Hunan province from 2008 to 2011

Objective To understand and master the situation of residents consumption of iodized salt in Chenzhou city of Hunan province,to identify problems and take appropriate interventions to ensure the residents consumption of qualified iodized salt,and to provide a scientific basis for elimination of iodine deficiency disorders (IDD).Methods According to the National Iodized Salt Monitoring Program (Amendment) and the Evaluation Scheme for Elimination of Iodine Deficiency Disorders at the County Level,the monitoring counties,towns and villages were selected in Chenzhou city from 2008 to 2011,the content of iodine in salt was detected using direct titration.The data was analyzed by SPSS 17.0 and Excel 2003.Results A total of 12700 salt samples were tested from 2008 to 2011.The iodized salt coverage rate,the qualified rate of iodized salt,the consumption rate of qualified iodized salt and the rate of non-iodized salt was 99.19% (12597/12700),96.33% (12135/12597),95.55%(12135/12700) and 0.81% (103/12700),respectively.There were significant differences between each year from 2008 to 2011 (x2 =13.99、42.35、48.45、13.99,P all < 0.01).The coefficient of variation was 21.19%.The median and average of iodine content in salt samples was 32.2 mg/kg and 31.9 mg/kg,respectively.Compared with the median and average of iodized salt content,there was no significant difference between each year from 2008 to 2011 (t =2.941,P > 0.05),while there was significant difference among the 11 counties(t =2.983,P < 0.05).Conclusions The goal of eliminating IDD has realized in the city of Chenzhou since 2010.To consolidate the IDD control results,surveillance should be strengthened in future.
9.Construction and expression of RNase-resisting virus-like particles containing PSA mRNA

Objective To construct an expression system to produce the virus-like particles containing a part of the sequence of PSA mRNA, which are ribonuclease-resistant due to the encapsulation of the mRNA by bacteriophage MS2 coat proteins. Methods The PCR products of PSA cDNA fragments were cloned to TA vector pBS-T, then the targeted segments could be obtained when the pBS-T-PSA were digested by restriction endonuclease Hind Ⅲ and cloned to prokaryocytic expression vector pNCCL1. The recombinant plasmids named PNCCL1-PSA were transfected into E. Coli BL21-DE3 and induced to express with IPTG. Results The recombinant plasmids were successfully constructed. The bacteriophage MS2 coat protein which expressed in BL21 can self- assemble to form ribonuclease resistant virus-like particles and the PSA mRNA was encapsulated into virus-like particles. Conclusions The virus-like particle containing PSA mRNA can be expressed in prokaryocyte and it can be used as standard and control in detecting PSA mRNA. It provides a new, stable and ribonuclease-resistant RNA standard in RNA detection.
10.Effect of triptolide on iNOS and SP expressions in spinal dorsal horn and dorsal root ganglion of rats with adjuvant arthritis.

OBJECTIVETo observe the analgesic effect of triptolide (TP) of high, middle and low doses on rats with adjuvant arthritis (AA), and the expressions of inducible nitric oxide synthase (iNOS) and substance P (SP) in spinal dorsal horn and dorsal root ganglion (DRG) of corresponding sections, in order to discuss the possible mechanism for the analgesic effect of TP on rats with adjuvant arthritis.
METHODFifty SD rats were selected and randomly divided into the normal group (group A), the model group (group B), and TP low (group C), middle (group D), high (group E) dose groups. Except for the group A, all of the remaining groups were injected with 0.1 mL of Freund's complete adjuvant through their right rear toes to establish the model. At 14 d after the model establishment, rats in C, D and E groups were intraperitoneally injected with different doses of TP (0.1 mg x kg(-1) for the group C, 0.2 mg x kg(-1) for the group D, 0.4 mg x kg(-1) for the group E) once a day for 9 days. Then the 50% mechanical withdraw threshold (MWT) was determined. And the expressions of iNOS and SP in lumbar5 (L5) spinal dorsal horn and DRG were detected with the immunohistochemical method.
RESULTThe 50% MWT of rats in the group B was significantly lower than that of the group A (P < 0.01). After being treated with TP, the Thermal withdrawal latencies of groups C, D and E were significantly higher than that of the group B (P < 0.01). TP could notably increase the MWT of AA rats, with a certain dose-effect relationship. The immunohistochemical results indicated that the iNOS and SP expressions significantly increased in the group B (P < 0.01), while the positive expression levels of iNOS and SP in groups C, D and E were significantly lower than that of the group B (P < 0.01), with a certain dose-effect relationship.
CONCLUSIONTP shows a good analgesic effect on AA, and could inhibit the iNOS and SP expressions in spinal dorsal horn and DRG in rats with adjuvant arthritis, which may be one of action mechanisms for the analgesic effect of TP.
