1.Correlation between Gab2 expression in ovarian cancer and resistance to platinum drugs chemotherapy

Objective To investigate the correlation between Gab2 expression and platinum drugs chemotherapy resistance by detecting the expression of Gab2 in ovarian cancer.Methods Immunohistochemical and Western blotting techniques were used to detect the expression of Gab2 in 107 specimens of serous ovarian tumor tissue and 12 specimens of normal ovarian tissues resected during bladder cancer radical operation in the department of gynaecology of our hospital from January 2011 to June 2015.The correlation between Gab2 expression with clinical stage and effect of combined chemotherapy based on platinum drugs was analyzed.Results The Gab2 expression level in ovarian cancer tissue was significantly higher than that in normal ovarian tissue,moreover had a relation with the FIGO clinical stage,the expression level of Gab2 increased with the increase of clinical stage,the difference was statistically significant (P<0.05);61 cases were sensitive to chemotherapy,46 cases were resistant to chemotherapy,the Gab2 expression level in the patients with chemotherapy resistance was significantly higher than that in the patients with chemotherapy sensitivity,the difference was statistically significant (P<0.05).Conclusion Gab2 may become one of effective indexes for predicting platinum drugs chemotherapy sensitivity.

OBJECTIVETo study the clinical efficacy of needle-knife to cut off the medial branch of the lumbar posterior ramus under C-arm guiding to treat low back pain caused by lumbar facet osteoarthritis.
METHODSFrom July 2009 to June 2011, 60 patients with low back pain caused by lumbar facet osteoarthritis were reviewed,including 34 males and 26 females, ranging in age from 39 to 73 years old,averaged 61.9 years old; the duration of the disease ranged from 6 to 120 months, with a mean of 18.9 months. All the patients were divided into two groups, 30 patients (18 males and 12 females, ranging in age from 39 to 71 years old, needle-knife group) were treated with needle-knife to cut off medial branch of the lumbar posterior ramus under C -arm guiding and the other 30 patients(16 males and 14 females, ranging in age from 41 to 73 years old, hormone injection group) were treated with hormone injection in lumbar facet joint under C-arm guiding. The preoperative JOA scores and the scores at the 1st, 12th and 26th weeks after treatment were analyzed.
RESULTSBefore treatment,the JOA scores between the two groups had no significant difference (P= 0.479); after 1 week of treatment, the JOA scores between the two groups had significant difference (P= 0.040), the improvement rate of hormone injection group was superior than that of the needle-knife group,which were (58.73+/-18.20)% in needle-knife group and (71.10+/-22.19)% in hormone injection group; after 12 weeks of treatment, the JOA scores between the two groups had no significant difference(P=0.569), and the improvement rate between the two groups had no significant difference,which were (50.09+/-19.33)% in the needle-knife group and (48.70+/-18.36)%) in the hormone injection group; after 26 weeks of treatment,the JOA scores between the two groups had significant difference (P=0.000), the improvement rate of hormone injection group was superior than that of the needle-knife group,which were (48.56+/-28.24)% in needle-knife group and (15.62+/-11.23 )% in hormone injection group.
CONCLUSIONUsing needle-knife to cut off the medial branch of the lumbar posterior ramus could get longer efficacy than hormone injection in the treatment of lumbar facet osteoarthritis.

4.Pentosan polysulfide sodium for chronic non-bacterial prostatitis in the rat model.

OBJECTIVETo investigate the therapeutic effect of pentosan polysulfide sodium (PPS) on chronic non-bacterial prostatitis (CNP) in rats.
METHODSBased on Robinette's method, we established a CNP model in 80 male SD rats, aged 6 months and weighing 315 - 450 g, by castration followed by subcutaneous injection of estradiol at 0.25 mg / (kg x d) for 30 consecutive days. Then we randomly allocated the model rats into a placebo group (n = 40) and a PPS group (n = 40) to receive intragastric administration of normal saline and PPS, respectively. After 8 weeks of treatment, the pathological changes in the rat prostatic tissue were observed by HE staining.
RESULTSVaried degrees of chronic inflammation and inflammatory cell infiltration were seen in the prostatic tissues of both groups of rats before the treatment. The inflammation was significantly improved after the treatment in the PPS group but not in the placebo group.
CONCLUSIONPPS has some therapeutic effect on CNP in the rat, and its mechanism may be associated with the abilities of PPS to repair the damaged glycosaminoglycan layer and inhibit inflammation in the prostate.

5.Energy expenditure in reading and watching TV studied on 30 young females.

OBJECTIVETo measure the energy expenditure of reading and watching TV for young women.
METHODSAll 30 girls in the college were selected according to questionnaire, medical examination, blood test, liver function and thyroid gland level. Energy expenditure of reading and watching TV was measured respectively by the Cosmid K4b2 portable gas analysis system from Italy. Basic metabolic rate (BMR) and resting metabolic rate (RMR) were measured as to analyzing results conveniently.
RESULTSThe energy expenditure in reading was (226.35 +/- 56.07) kJ/h and the heart rate was (69 +/- 5) bmp. For watching TV it was (220.79 +/- 65.69) kJ/h and the heart rate was (68 +/- 5) bmp. BMR was (178.53 +/- 53.51) kJ/h and the heart rate was (62 +/- 6) bmp. RMR was (214.76 +/- 44.56) kJ/h and the heart rate was (65 +/- 6) bpm.
CONCLUSIONSThe energy expenditure in reading should be higher than that of watching TV, however, the difference was not obvious. The expenditure rates might get closer to those of RMR. It might be regarded as an expenditure of very light physical active in Chinese.

6.Multiple viral transmission chains of enterovirus 71 co-circulated in Jilin Province during 2009-2010

Objective To analyze the genetic characterization of enterovirus type71 ( EV71 )associated with hand foot and mouth disease (HFMD) epidemics in Jilin province,during 2009-2010.Methods Randomly selected 31 representative EV71 strains from the eases of 8 prefectures to amplify and sequences of VP1 genes of EV71 strains,and analyzed with Bioedit and Mega4.0 program.Results All representative 31 EV71 strains belong to C4a subgenotype,the homology of nucleotide in VP1 region among the 31 EV71 strains were 94.5%-100.0%,and were clustered into 5 transmission chains respectively.25 strains out of 31 strains were associated with a predominant transmission chain,and circulating in 8 prefectures,while other 6 strains clustered into 4 lineages.Conclusion Multiple transmission chains of EV71 C4a subgenotype were co-circulating in Jilin province during 2009-2010,and a predominant transmission chain was circulating in 8 prefectures,associated with HFMD outbreaks of Jilin province.
7.Effect of anaphylatoxin C3a, C5a on the tubular epithelial-myofibroblast transdifferentiation in vitro.

BACKGROUNDTubulointerstitial renal fibrosis is the common end point of progressive kidney diseases, and tubular epithelial-myofibroblast transdifferentiation (TEMT) plays a key role in the progress of tubulointerstitial renal fibrosis. Anaphylatoxin C3a and C5a are identified as novel profibrotic factors in renal disease and as potential new therapeutic targets. The aim of this study was to investigate whether C3a, C5a can regulate TEMT by transforming growth factor-β1 (TGF-β)/connective tissue growth factor (CTGF) signaling pathway and the effects of C3a and C5a receptor antagonists (C3aRA and C5aRA) on C3a- and C5a-induced TEMT.
METHODSHK-2 cells were divided into C3a and C5a groups which were subdivided into four subgroups: control group, 10 ng/ml TGF-β1 group, 50 nmol/L C3a group, 50 nmol/L C3a plus 1 µmol/L C3aRA group; control group, 10 ng/ml TGF-β1 group, 50 nmol/L C5a group, 50 nmol/L C5a plus 2.5 µmol/L C5aRA group. TGF-β1 receptor antagonist (TGF-β1RA) 10 µg/ml was used to investigate the mechanism of C3a- and C5a-induced TEMT. Electron microscopy was used to observe the morphological changes. Immunocytochemistry staining, real-time PCR and Western blotting were used to detect the expressions of a smooth muscle actin (α-SMA), E-cadherin, Col-I, C3a receptor (C3aR), C5aR, CTGF and TGF-β1.
RESULTSHK-2 cells cultured with C3a and C5a for 72 hours exhibited strong staining of α-SMA, lost the positive staining of E-cadherin, and showed a slightly spindle-like shape and loss of microvilli on the cell surface. The expressions of α-SMA, E-cadherin, Col-I, C3aR, C5aR, TGF-β1 and CTGF in C3a- and C5a-treated groups were higher than normal control group (P < 0.05). C3aRA and C5aRA inhibited the expressions of α-SMA, Col-I, C3aR, C5aR, and up-regulated the expression of E-cadherin (P < 0.05). TGF-β1 and CTGF mRNA expressions induced by C3a and C5a were partly blocked by TGF-β1RA (P < 0.05).
CONCLUSIONC3a and C5a can induce TEMT via the up-regulations of C3aR and C5aR mRNA and the activation of TGF-β1/CTGF signaling pathway in vitro.

Parkinson's disease(PD)is the most common neurodegenerative disease,and Lewy bodies(LBs)is the most typical pathological change.α-synuclein(α-syn),as the main component of LBs,is considered as the pathogenic factor of PD.According to the transmission hypothesis,pathological α-syn can be transported from damaged neurons to normal neurons,leading to pathological misfolding and aggregation of α-syn in recipient neurons,resulting in cascading neuronal damage.According to the hypothesis of gut-brain axis,pathological α-syn can be produced in the intestine,and uploaded to the central nervous system via the vagus nerve.In this paper,the role of α-syn in the pathogenesis of PD is summarized,in order to provide reference for the study of intervention targets of PD.
9.Use of variable-number tandem repeats to examine genetic diversity of Bacillus anthracis.

OBJECTIVETo study the genotyping of Bacillus anthracis based on multiple-locus variable-number tandem repeats(VNTR) in the B. anthracis genome.
METHODSWe selected 13 VNTR loci (which cited from published articles) to study 88 strains of B. anthracis isolated from China. The methods used were: (1) Selecting the primers which were at both ends of the tandem repeat locus; (2) Amplifying the sequence of the locus by PCR; (3)cDetecting the PCR products by agarose gel and polyacrylamide electrophoresis; (4)Analyzing the PCR products and computing the molecular weight by analysis software of gel images;(5) Double-checking with sequencing results; (6)Reckoning the repeat numbers and study the VNTRs loci characters.
RESULTS(1) We used multiple-locus variable-number tandem repeat analysis (MLVA) to characterize 88 B. anthracis isolates from diverse geographic locations which were divided into 45 MLVA genotypes and 3 groups through cluster analysis. The genotypes was relative to restricted geographical region. It seemed clear that the multiple isolates from the same anthrax outbreak frequently having identical genotypes. (2)Results from VNTR analysis showed that A16R vaccine strain isolated from China was having the nature of representativeness in the country.
CONCLUSIONAnalysis showed that the VNTR patterns was an appropriate study method for B. anthracis genetic diversity from different geographical areas and different time. Isolates from the same anthrax outbreak had identical

10.Evaluation of the protein requirement in Chinese young adults using the indicator amino acid oxidation technique.

OBJECTIVETo accurately calculate the protein requirements in Chinese young adults using the indicator amino acid oxidation technique.
METHODSNine women and ten men received a restricted daily level of protein intake (0.75, 0.82, 0.89, 0.97, and 1.05 g/kg), along with L-[1-13C]-leucine. Subjects' protein requirement was determined by a biphasic linear regression crossover analysis of F13CO2 data. In doing so, a breakpoint at the minimal rate of appearance of 13CO2 expiration specific to each level of dietary protein was identified. This trial was registered with the Chinese clinical trial registry as ChiCTR-ONC-11001407.
RESULTSThe Estimated Average Requirement (EAR) and the Recommended Nutrient Intake (RNI) of protein for healthy Chinese young adults were determined to be 0.87 and 0.98 g/(kg•d), respectively, based on the indicator amino acid oxidation technique.
CONCLUSIONThe EAR and RNI of mixed protein are 5% and 16% that are lower than the current proposed EAR and RNI (0.92 and 1.16 g/(kg•d), respectively), as determined by the nitrogen balance method. The respective EAR and RNI recommendations of 0.87 and 0.98 g/(kg•d) of mixed protein are estimated to be reasonable and suitable for Chinese young adults.
