1.Effects of tacrolimus on the expression of nuclear factor-κB and glucocorticoid receptor by HaCaT cells in vitro

Objective To investigate the effect of tacrolimus on the expression of nuclear factor-κB (NF-κB) in HaCaT cells stimulated by tumor necrosis factor-α(TNF-α),and on the expression of glucocorticoid receptor (GR)α and β in untreated HaCaT cells in vitro.Methods Cultured Ha CaT cells were treated with TNF-α(10μg/L) only,combination of TNF-α(10μg/L) and various concentrations (10-8mol/L, 10-7mol/L,10-6moL/L) of tacrolimus or tacrolimus of different concentrations only.After additional 12-,24-, 36- or 48-hour cnlture, Westem blot and immunofluorescenee-confocal laser scanning microscopy were used to detect the expressions of NF-κB,GRα and GRβ in HaCaT cells.Those untreated HaCaT cells served as the control.Results The relative protein expression level of NF-κB was increased in HaCaT cells after treatment with TNF-α for 24 and 48 hours zompared with untreated ceils (0.73±0.0316 and 0.8925±0.0171 vs 0.4988±0.03506,both P<0.05);however,the increase in NF-κB expression was inhibited by the combination treatment with tacrolimus,and the relative expression level of NF-κB protein was 0.6825±0.0263.0.6200±0.0163 and 0.5575±0.0299 in HaCaT cells treated with TNF-α plus tacrolimus of 10-8mol/L 10-7mol/L and 10-6mol/L,respectively;the difference was significant etween TNF-α-treated cells and those dealt with the combination of NF-α and tacrolimus of 10-7 or 10-6 mol/L (both P<0.05).No significant difference was observed in the expression of NF-κB by HaCaT cells between different time oints treated with tacrolimus of 10-8,10-7 or 10-6 mol/L.Also,there was no zignificant difference in the expression of GRα or GRβ between untreated HaCaT cells and those treated with tacrolimus of 10-8, 10-7 or 10-6 mol/L at any time point.Conclusions Tacrolimus ould inhibit the expression of NF-κB by TNF-α-stimulated HaCaT cells,but does not affect the expression of GRα or GRβ,in untreated HaCaT cells.
2.Maternal and neonatal perinatal outcomes in pregnancies after in vitro fertilization and natural pregnancy: a systematic: a meta analysis

Objective To systematically evaluate maternal perinatal complications and neonatal outcomes after in vitro fertili zation and embryo transplantation(IVF-ET).Methods The computer retrieval was performed on the databases of PubMed,Cochrane library,EMbase,CNKI,Wanfang and Chinese biomedical literature database.The articles on the comparison between IVF and natural pregnancy were included.The RevMan5.3 software was adopted to conduct the meta analysis.Results Twenty-three articles conforming to the requirements were included,including 1 312 651 cases,among them 113 044 cases were IVF and 1 199 607 cases were naturalpregnancy.IVF mother's age was larger than that of natural pregnancy,while the gestational weeks were smaller than those of natural pregnancy,the newborn birth weight was lighter than that of natural pregnancy(P<0.05);the probability of pregnancy-induced hypertension (PIH),placenta previa,postpartum hemorrhage and gestational diabetes mellitus (GDM) occurrence in IVF mother were higher than that of natural pregnancy (P<0.05);the rate of multiple and cesarean section in IVF mothers were higher than that of natural pregnancy (P<0.05);the occurrence rates of small for gestational age(SGA),low birth weight infants,stillbirth and premature in IVF were higher than those of natural pregnancy (P<0.05).Conclusion The maternal perinatal complications in IVF are higher than those of natural pregnancy,and the birth outcomes in IVF are worse than those of natural pregnancy.
3.Relationship of lens advanced glycosylation end products with diabetic retinopathy

Background The generation of advanced glycosylation end products (AGEs) is one of the most important characteristics in diabetes mellitus (DM).Its role in diabetic eye disease is still unclear.Objective This study was to investigate the changes of AGE levels in human cataractous lens in diabetic patients and further explore the relationship of lens AGEs with early diabetic retinopathy (DR).Methods This study protocol was approved by Ethic Committee of He Eye Hospital.A cases controlled clinical study was carried out.Forty human nuclei were obtained from 40 cataract patients during the phacoemulcification in He Eye Hospital from August to December 2014,with the nuclear hardness of Ⅲ-Ⅳ grades (Emery grading).The eyes were divided into non-DM (NDM) group and DM group,and the DM group was further divided into non-DR (NDR) group,non-proliferative DR (NPDR) group and proliferative DR (PDR) group.Lens AGE level was quantitatively detected by ELISA.Results The content of pentosidine was (31.90±5.00),(35.83±4.22) and (38.59±5.11)pg/ml,and that of carboxymethyl lysine (CML) was (18.82±3.84),(19.65±2.12) and (23.77±4.73) μg/L,and that of imidazolone was (13.12±2.71),(14.48±2.01) and (15.37 ±2.74) pg/ml in the NDR group,NPDR group and PDR group,respectively,which was significantly higher than (27.69 ± 1.53) pg/ml,(11.36 ±1.47) μg/L and (10.21 ± 0.54) pg/ml in the NDM group (all at P<0.01).The contents of pentosidine and CML in the lens were significantly higher in the PDR group than those in the NDR group (all at P<0.05);while there was no significant differences in the pentosidine content in the lens among the NDR group,NPDR group and PDR group (all at P>0.05).The positive correlations were found between CML content and pentosidine content (r =0.623,P =0.000),between CML content and imidazolone content (r =0.717,P =0.000) as well as between pentosidine content and imidazolone content (r=0.669,P =0.000).Conclusions The levels of pentosidine,CML and imidazolone in cataractous lens are obviously elevated in the DM patients.The levels of pentosidine and CML in the lens are gradually raised as the aggravation of DR.AGEs might be a potential maker for the assessment of diabetes.

5.A preliminary exploration of the evaluation standard and the problems for graduation thesis in higher vocational college students of pharmaceutical majors

The writing of vocational college thesis is the maln way to test whether the students are able to analyze and solve practical problems with the professional knowledge and skills they've learnt, as well as improve the capability of production and practice. This paper analyzes the present situation of the thesis written by higher vocational college students majoring in pharmaceutical sci-ences, and figures out the four kinds of evaluation standards about graduation thesis for students in various pharmaceutical fields with different problems. The four kinds of evaluation standards includes key point analysis on Standard Operation Procedure (SOP), planning strategy for pharmaceutical mar-keting, investigation of rational use of drugs in hospitals and subject research. The author puts forward the solving method for the problem, and formulate the evaluation requirements.

Methods The mRNA and protein expressions of GR? and GR? in HaCaT cells after the treatment of Dex ( 10 -6mol/L) for 24 and 48 h and in 24,48 and 72 h after withdrawal of Dex were detected by real-time PCR and Western blot analysis respectively. Abstract: Objective To explore the value of real-time shear wave elastography in evaluating the severity of liver fibrosis in hepatitis B, and to analyze the factors that affecting its accuracy. Methods A total of 196 chronic hepatitis B patients, who admitted to the Third Affiliated Hospital of Chengdu Medical College from February 2018 to October 2020, were selected for retrospective analysis. 9.Aeromonas hydrophila bloodstream infection in patients with hematologic diseases: a report of 4 cases

Abstract: To report the diagnosis, treatment and outcome of 4 patients with hematological diseases complicated with Aeromonas hydrophila bloodstream infection in the Second Affiliated Hospital of Kunming Medical University, further clarify the importance of blood culture and deepen the clinical understanding of the disease. Four patients with hematological diseases complicated with Aeromonas hydrophila bloodstream infection treated in the Second Affiliated Hospital of Kunming Medical University from 2017 to 2021 were recruited as the study objects. The clinical manifestations, blood culture collection, detection time of Aeromonas hydrophila, laboratory examination, treatment and prognosis of the patients were retrospectively analyzed. In this study, 4 cases were male patients with hematological diseases, who were in myelosuppression after chemotherapy. After fever, blood culture was collected and Aeromonas hydrophila was detected. The positive time of blood culture in 4 cases ranged from 4 to 11 hours. The results of antibiotic sensitivity showed that it was highly sensitive to the second, third and fourth generation cephalosporins, quinolones and carbapenems. Four patients were treated with imipenem cilastatin sodium in the early stage, and one patient recovered after active anti infection and leukocyte raising treatment. One patient did not complete chemotherapy due to a request for discharged, and the follow-up was unknown. Two patients developed rapidly into necrotizing fasciitis and died later. Hematological diseases complicated with Aeromonas hydrophila bloodstream infection are rare, but the mortality rate is high. For patients with repeated fever and considering infection, blood culture should be carried out as soon as possible to confirm the pathogen and drug sensitivity test. During clinical treatment, the treatment should be adjusted in time in combination with the patient's situation. In addition to anti-infection treatment, the patient's immunity should be improved and the development of necrotizing fasciitis should be vigilant.

10.Effect of fibrous root extract of Coptis chinensis on soil microbes and enzyme activities.

Coptis chinensis is widely used as Chinese medicine herbs and serious soil problems occur after continual cultivation of this medicinal plant. In the preset experiment, fibrous root extract of C. chinensis (REC) was added into soil to study the effect of REC on microbes and enzyme activity in soil. The results showed that both bacteria and actinomycetes decreased by about 2 times in contrast to fungi, which increased by about 3 folds. Phosphorus bacteria, potassium bacteria, azotobacter, ammonia bacteria, and nitrifying bacteria were also reduced significantly by REC, suggesting the inhibition of nitrogen biofixation and supply, mobilization of phosphorus and potassium, ad plant growth promotion as REC added into soil. There were multiple influences of REC on soil enzyme activities. Invertase activity was stimulated, while urease was inhibited and dehydrogenase unchanged by REC, indicating the interference of biochemical reactions in soil. In addition, type and total content of phosphorus lipid fatty acids (PLFAs) , the signature of microbes, decreased while the ratio of bacterium to fungus PLFAs increased as REC increased in soil, which suggested that fungi increased relatively with bacteria decreased thereby leading to easy occurrence of crop fungus diseases following cultivation of C. chinensis. The decrease in diversity and evenness indexes of microbial community in soil by REC indicated soil ecosystem deterioration and reduction of microbial groups and densities in soil. Therefore, allelopathic chemicals released from the roots of C. chinensis could change microbial community structure and resulted in serious soil problems by continual cropping of this medicinal plant.
