1.Effect of uroacitides on the expression of cyclin D1 of breast cancer cells

Objective To investigate the effect of CDA-Ⅱ on t he expression of cyclin D1 of breast cancer cells. Methods The br east carcinoma cell lines MCF-7 and MDA-MB-231 were cultured in vitro tog ether with CDA-Ⅱ, with retinoic acid as positive control, and the effect of CD A-Ⅱ on the growth curve as well as the expression of cyclin D1 of breast carci noma cell lines was observed. Results CDA-Ⅱ may reduce the gr owth of MCF-7 and MDA-MB-231, cripple their proliferative ability and inhibit the cyclin D1 expression of breast cancer cell lines. Conclusion CDA-Ⅱ may inhibit the growth and proliferation of MCF-7 and MDA-MB-231, reduce the expression of breast carcinoma cell cyclin D1, hence showed a good a pplied prospect for the treatment of breast carcinoma.
2.Effects of ELOVL4 gene overexpress on the synthesis efficiency of n3 and n6 very long chain polyunsaturated fatty acids

AIM:To compare the synthesis efficiency of n3 and n6 very long chain polyunsaturated fatty acid ( VLC-PUFA ) by overexpressing ELOVL4 protein, providing guidance for treating Stargardt-like macular dystrophy (STGD3).
METHODS:To establish recombinant adenovirus with the ELOVL4 protein and green fluorescent protein, transferred into cultured PC12 cells. The cells were divided into 3 groups: PC12, PC12 + Ad- GFP and PC12 + Ad-ELOVL4, former two groups serve as controls. ELOVL4 gene expression was quantified by qRT-PCRs. ELOVL4 protein was analyzed by Western - Blot ( WB ) . The transduced cells were treated with both EPA and AA (1:1). After 48h of incubation, cells were collected, total lipids extracted and fatty acid methyl esters prepared and analyzed by gas chromatography-mass spectrometry ( GC-MS) .
RESULTS:When supplemented together, 20:5n3 (EPA) and 20:4n6 ( AA) were efficiently taken up at almost the same amounts in the PC12 cells regardless of ELOVL4 expression. The ELOVL4-expressing cells elongated both EPA and AA to a series of n3 and n6 VLC-PUFAs. From 20:5n3/EPA, 34:5n3 and 36:5n3 account for 0. 71% and 1.6%, respectively. From 20:4n6/DHA, 34:4n6 and 36:4n6 were only 0. 46% and 0. 61%, respectively. The total relative mol% of n3 VLC-PUFAs synthesized from EPA was almost two times that of n6 VLC-PUFAs synthesized from AA.
CONCLUSION: ELOVL4 protein preferentially elongates n3 PUFA to VLC - PUFAs over n6 PUFA. Dietary supplementation of appropriate n3/n6 PUFAs may provide STGD3 patients with some therapeutic benefits.
3.Effect of Uroacitides on Cell Cycle Progression of Breast Cancer Cells

Objective To investigate the effect of CDA-Ⅱ on the cell cycle progression of breast cancer cells.Methods The effects of CDA-Ⅱ on growth curve, cell cycle progression and morphology of breast cancer cell lines MCF-7 and MDA-MB-231 were observed when CDA-Ⅱ and MCF-7 or CDA-Ⅱ and MDA-MB-231 were blended to cultivate in vitro, in comparison with the classical cell differentiation inducer ATRA. Results CDA-Ⅱ decreased the growth speed and inhibit proliferation ability in breast cancer cell lines MCF-7 and MDA-MB-231.It caused G0/G1 phase block of cell cycle and reduced the rate of S phase of breast cancer cells. Conclusion CDA-Ⅱ has remarkable effect of anti-cell-proliferation and can induce cell cycle block of G0/G1 on breast cancer cells. This results provide experimental bases for the treatment of breast cancer with CDA-Ⅱ.
5.Determination and pattern recognition of trace elements in serum samples from patients with renal cell carcinoma by ICP-MS

Objective To study the relationship between serum trace elements and renal cell carcinoma.Methods The serum concentrations of multi-elements in 34 patients with renal cell carcinoma and 28 healthy volunteers were determined by inductively coupled plasma mass spectrometry(ICP-MS).The results were analyzed by partial least square discriminant analysis (PLS-DA) and Fisher discriminant.Results Compared with healthy voluteers,the levels of vanadium (5 034.56 ng/L:4 401.23 ng/L ),cobalt (211.34 ng/L:158.67 ng/L),nickel(l 850.55 ng/L:1 587.12 ng/L),manganese(1 873.35 ng/L:932.68 ng/L) and cadmium(95.63 ng/L:36.43 ng/L) were significantly higher in patients with renal cell carcinoma (P < 0.05 ).While,the concentrations of calcium( 10.83 mg/L:11.78 mg/L) and zinc(67.11 μg/L:70.92 pg/L)were significantly lower ( P < 0.05 ).Discriminant analysis showed that the serum elements levels in the patients with renal cell carcinoma were significantly different from the healthy volunteers.The scores plot showed distinct clustering between patients and controls,the points of patients were obviously offset from the controls.The classification accuracy of Fisher discriminant function was 97.61%.Conclusion Trace elements in serum are significantly different in patients with renal cell carcinoma and healthy volunteers.Discriminant analysis of serum samples based on trace element levels is possible.Thus,it is feasible for early diagnosis of renal cell carcinoma by determination of trace elements and discriminant analysis.
6.Effect of uroacitides on apoptosis of breast cancer cells

Objective To investigate the effect of CDA-II on apoptosis of breast cancer cells. Methods The effects of CDA-II on growth curve, cell apoptosis and morphology of breast carcinoma cell lines MCF-7 and MDA-MB-231 were observed by in vitro cultures, and compared with the classical cell differentiation inducer ATRA. MCF-7 and MDA-MB-231 cell lines have their different biologic characteristics. Results CDA-II can reduce growth and proliferation ability and induce cell apoptosis of breast cancer cell lines( MCF-7 and MDA-MB-231). Conclusions CDA-II has remarkable effect of anti-cell-proliferation and induction of (apoptosis) on breast cancer cells MCF-7 and MDA-MB-231 . Our results provide experimental bases for the treatment of breast carcinoma with CDA-II.

【Objective】 To investigate the effect of Weishu Powder (WP) on acute liver injury induced by carbon tetrachloride (CCl_4) in mice. 【Methods】 Fifty mice were randomized to normal control group (group A), model group (group B), bifendate pills group (group C), low-dose WP group (group D) and high-dose WP group (group E). The mice in the above groups were fed with the corresponding drugs by gastric infusion for 7 days. On the 8th day of feeding, the mice except those in group A were given 0.5% CCl_4 peanut oil solution (10 mL/kg) by intraperitoneal injection. 10.HPLC analysis of agaro-oligosaccharides profiles under diffe-rent hydrolysis modes

Objective Since the discovery of multiple bioactivities of agarobiose oligomers, the establishment of quantitative analysis of agarobiose oligomers obtained under different hydrolysis modes would be of much importance. Methods Agarobiose oligomers ranged from biose, tetraose, hexaose, octaose, to decaose were isolated and purified by gel chromatography on Sephadex column. The agarobiose oligomers were separated in RP-HPLC after introducing ?-naphthylamine into compounds. Results Each oligomer could be quantified with good linearity and the lowest detection limit reached 0.1~2 ?g?mL -1. The chromatograms profiles under different hydrolysis modes(HCl, citric acid, solid acid, and hydroxyl radical degradation)showed that the content of agarobiose could reach more than 57.8 % using solid acid hydrolysis with best yield of 33.2 % . While HCl could degrade agar into a series of agaro-oligosaccharides from biose to decaose. The yield of oligosaccharides was low if hydrated by citric acid. The method of Fenton degradation could increase the speed of hydrolysis, but the products were more complex. Conclusion The method established here could be useful for analysis of hydrolysates of oligomers under different hydrolysis modes.
