Many microbial pathogens,including those responsible for major enteric infections,exploit oligosaccharides that are displayed on the surface of host cells as receptors for toxins and adhesins. Blocking crucial ligand receptor interactions is therefore a promising therapeutic strategy. One approach is to express molecular mimics of host receptors on the surface of harmless recombinant bacteria that can survive in the gut. These recombinant probiotics bind bacterial toxins in the gut lumen with very high avidity,thereby preventing disease.
2.Kinetic Analysis of the Inhibition of in vitro Growth of Pathogenic Escherichia coli K88 and O138 by Lactobacillus Isolates from Porcine Intestine

Fermentation property and kinetic analysis of the inhibition of in vitro growth of pathogenic Escherichia coli K88, O138 by Lactobacillus intestinalis isolated from porcine intestine mucus were investi-gated. The results of fermentation showed that L1 result in a pH of 3.90 within 12 h and a large amount of lactic acid production, with the concentration of 104.08 mmol/L. Kinetic analysis of the antibacterial activity of L1 metabolic product toward K88, O138 showed that CFUS display strong antibacterial activity toward K88, O138; the antibacterial activity of L1 CFUS was higher than the lactic acid control samples and was mainly due to the production of lactic acid; K88, O138 had the tolerance property toward pH 4.5.
3.The effects of antibody against heat shock protein B of Helicobacter pylori on the associated gastric diseases

Objective To investigate the effects of antibody against heat shock protein (HSP) B of Helicobactor pylori (H. pylori) on H.pylori associated gastric disease for the study of the relationship between serum antibody to HSP B of H.pylori and the pathologic changes of the gastric mucosa. Methods 1 036 patients from Shandong province were collected. Six hundred patients were infected with H.pylori. Of those, three hundred received triple therapy for eradication of H.pylori and the other received placebo. Five years later, patients with H.pylori positive chronic superficial gastritis, chronic atrophic gastritis, peptic ulcer, and H.pylori negative chronic superficial gastritis underwent gastroscopy and collected serum specimens again, each group had 20 patients. The Western blot techniques were used to assay antibody to HSP B of H.pylori in sera of these patients. 5.Imaging manifestations and its clinical significance in patients with synovitis acne pustulosis hyperostosis osteomyelitis syndrome

Objective To describe the clinical and imaging manifestations of patients with synovitis acne pustulosis hyperostosis osteomyelitis (SAPHO) syndrome,and to analyze the diagnostic importance of different clinical and imaging manifestations for SAPHO syndrome. Methods Seventeen patients (7 males and 10 females) with SAPHO syndrome were recruited in this study.Age ranged from 36 to 67 years with a mean age of (48 ± 8) years. All patients fulfilled the diagnostic criteria of Benhamou. Serum HLA B27 antigen records were reviewed for all patients. Imaging data of the abnormal bone sites were collected by conventional radiograph in all patients,CT in 13 patients as well as MR in 3 patients.Average time to take for a definite diagnosis of the syndrome was 3.7 years (ranged from O.5 to 13 years).Results Serum HLA B27 antigen was positive in all patients. Both skin and bone abnornalities were found in all patients.Ten patients had skin palmoplantar pustulosis and two patients had acne. Involving sites of bone and joints include sacroiliac joints,anterior chest and limbs.Sacroiliac joints were asymmetrically involved with imaging features in all patients.Eight patients exhibited anterior chest wall involvement. Five patients had osteomyelitis at limbs. For all images of 17 patients,CT was superior to conventional radiography in detecting abnormal changes of bone erosion and soft tissue swelling.MR imaging was able to depict edema changes that was not detectable by CT and radiography.Conclusion SAPHO syndrome is a rare disease,but for patients with skin and bone-joint abnormalities,especially with skin palmoplantar pustulosis,acne as well as with imaging features at the sacroiliac joint and anterior chest wall,SAPHO syndrome should be taken into a diagnostic consideration.
6.Cause analysis of missing diagnosis for vertebral fracture on lateral chest radiography

Objective To analyze the cause of missing diagnosis for vertebral fracture on lateral chest radiography. Methods Lateral chest radiographies of 1638 hospitalized patients (871 males and 767 females) were retrospectively reviewed for identifying vertebral fractures. Their ages ranged from 50 to 91 years with the mean of 63.5 years. Complains and application for chest radiography in all patients were not related to osteoporosis and vertebral fracture. Vertebral fracture and fracture severity were evaluated using Genant's semiquantitative visual method, taking approximately a 20%-25% vertebral height reduction as mild grade,26%-40% as moderate grade and 41% or greater as severe grade. Evaluation results of the vertebral fracture, original X-ray reports, as well as medical records were compared for further analysis. Results Eighty-four in 1638 patients showed vertebral fractures on the lateral chest radiographies. Of them,vertebral fractures were reported in 30 cases and 54 patients were not reported on their original X-ray reports. There were 63 vertebral fractures in 54 un-reported patients, most of which were single fracture (75% or 47/63). Grade Ⅰ fracture accounted for 54% (34/63), Grade Ⅱ fracture 33% (21/63), while 13% presented grade Ⅲ fracture (8/63). In all 84 patients with vertebral fractures, only 5 cases (6%)underwent dual energy X-ray absorptiometry (DXA) measurement, 5 cases ( 6% ) were recorded to have vertebral fractures on the medical papers, as well as 15 cases ( 18% ) were prescribed drugs related to the osteoporosis when discharged from hospital All drugs prescribed for the 15 patients were limited only to calcium. Conclusions More attention should be paid to osteoporosis by doctors including radiologists.Vertebral fracture on lateral chest radiography should be completely diagnosed, which is helpful for both prevention and treatment.
7.Expression of mice interlecukin-23 in pichia and preliminary studies on its biology function

Objective:To clone the mice interleukin-23(mIL-23) and express it in Pichia efficiently.Methods:Two subunits of IL-23 were amplified by PCR from pcDNA3/mIL-23,and ligated together with adaptor by over-PCR.The IL-23 cDNA confirmed by sequencing was inserted into expressing vector pPICZαA and expressed in Pichia X-33 strain.IL-23 protein expression was induced by methanol and ammonia.The recombinant IL-23 was identified by immunoblot and its biological activity was analyzed.Results:DNA sequencing confirmed that cloned cDNA was identical to the published sequence of mIL-23.The recombinant plasmid pPICZαA/mIL-23 was electroprated into X-33.An expected 72 kD protein of mIL-23 was founded both in the induced pellets and supermatants by SDS-PAGE and coomassie blue staining.The 72 kD protein could be recognized in Western blot.While we could detect bands at 72 kD in cell pellets induced more than 24 h by Western blot.Detected by Western blot using anti-His antibody,we could see positive band at 72 kD from supernatants induced 24 and 36 h.While we could detect band at 72 kD in cell pellets induced between 24 h and 96 h.Meanwhile,we could see obviously proliferation of mice peripheral blood mononuclear cells(PBMC) treated with the induced pellets and supermanants.Conclusion:We have successfully expressed mIL-23 protein in Pichia and the expressed product has its bioactivity in promoting the proliferation on PBMC.
8.Association between the Level of LDH in Gastric Cancer Patient’s Serum, the Relative Expression of LDH in Tissues and the Patient’s Prognosis

Objective To explore the association between the level of lactate dehydrogenase (LDH)in gastric cancer patients’ serum or the relative expression of LDH in their tissues and the patients’prognosis.Methods 60 specimens of gastric canc-er patients who confirmed by pathological diagnosis were collected from 2012 to 2013,include the serum specimens and tis-sue specimens.Their serum level of LDH were detected,and the relative expressions level of LDH in the sample of normal tissues,gastric cancer tissues and metastatic lymph nodes were quantify by real-time fluorescence quantitative reverse tran-scription-PCR (Realtime-PCR).Finally,the relationship between the expression of LDH and clinical pathologic features were analyzed by independent t-test or anova statistics.Results The serum level of LDH was highly increased than control group (340.89±10.67 IU/ml,t=24.7,P<0.01);the relative gene expression of LDH in normal tissue,human gastric cancer and metastatic lymph node were 1.0,3.39 and 2.35.The result suggests the serum level of LDH were associated with pTNM stage and lymph node metastases (t=5.2,4.8,P<0.01).The relative gene expression of LDH in gastric canc-er tissues were associated with tumor sizes,pTNM stage and lymph node metastases (t=18.2,15.3,P<0.01 and F=7.2, P<0.01).Conclusion The serum LDH level and the expression of LDH in cancer tissue were significantly increased,and their expression were correlated with the patient’s prognosis.The serum level of LDH and the expression of LDH in gastric cancer tissue may be potential indicator to evaluated the prognosis of the patient with gastric cancer.
9.Change and significance of CRP and PCT levels in elderly patients with AECOPD

Objective To investigate the changes and significance of serum C reactive protein (CRP) and calcitonin (PCT) levels in the elderly patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD).Methods One hundred and twenty elderly patients with AECOPD in the respiration deplartment of our hospital from January 2013 to study One hundred and twenty elderly patients with AECOPD in our hospital from January 2013 to June 2015 were selected to conduct the study and performed the subgroup analysis according to the BODE index (body weight,dyspnea,airflow,motor function index),times of acute exacerbation within 1 year after treatment and recurrence time,and other 40 healthy subjects in the outpatient department were selected as the control group.The serum PCT and CRP levels were compared among various groups.Results Serum CRP and PCT levels in AECOPD patients with grade 1-4 of BODE index were significantly higher than those in the control group (P<0.05),moreover in the intra-group comparison of serum CRP and PCT,the grade l<grade 2<grade 3<grade 4,the differences were statistically significant (P<0.05).Serum CRP and PCT had the significantly positive correlation with the grades of BODE index in AECOPD patients (r=0.482,0.317,P<0.05).After treatment,serum CRP and PCT levels in AECOPD patients with recurrence occurred more than 3 months were significantly lower than those in the patients with recurrence occurred within 3 months,the difference was statistically significant (P<0.05);serum CRP and PCT levels in the patients with more than once recurrence at 1 year after discharge from hospital were significantly higher than those in the patients with recurrence ≤once,the difference were statistically significant (P<0.05).Conclusion The serum levels of CRP and PCT in the patients with AECOPD can reflect dyspnea,airflow limitation and motor function ability,and conducts the preliminary assessment on the patient's prognosis.
10.Determination of residual EDTA-2Na in Japanese encephalitis attenuated live vaccine by capillary electrophoresis with electrophoretically mediated microanalysis

Objective: To establish a capillary electrophoresis (CE) with electrophoretically mediated microanalysis (EMMA) method for the determination of EDTA-2Na in Japanese encephalitis attenuated live vaccine.
Methods: The test was performed in disodium hydrogen phosphate buffer with pH 2.5, the online metal ions complexation of 1.5 mg·mL-1 Fe3+ and incubation time of 3 min. The separation voltage was 25 kV, the detection wavelength was 257 nm, and. the column temperature was 25.0 ℃.
Results: The established method had a good linear relationship in the concentration range of 0.01-0.5 mg·mL-1 (r=0.999 9), the detection limit was 5 μg·mL-1, and the relative standard deviation (RSD) of the measured samples was less than 2.87%. The recoveries of spiked samples were between 96.49%-101.02%.
Conclusion:The optimized method was applied to the determination of EDTA-2Na in Japanese encephalitis attenuated live vaccine. The satisfactory experimental results were obtained.
