1.Small intestinal bacterial overgrowth and endotoxinemia in patients with chronic liver disease

Objective To study the correlation between small intestinal bacterial overgrowth (SIBO) and plasma endotoxin,platelet-derived growth factor(PDGF)level,and levels of serum hyaluronic acid (HA),laminin(LN),N-terminal procollagen Ⅲ peptide(PⅢNP)and type Ⅳ collagen(Ⅳ.C). Methods Lactulose hydrogen breath test (LHBT) was performed in 64 patients with chronic liver disease. Plasma endotoxin was measured with Limulus test, plasma PDGF was tested with ABC-enzyme-linked immunosorbent assay,serum HA,LN,PⅢNP and Ⅳ.C were tested by radioimmunoassay(RIA) in the patients. The relationship between SIBO and plasma endotoxin, PDGF and HA,LN,PⅢNP and Ⅳ.C levels were analyzed. Results Positive LHBT suggesting the existence of SIBO were observed in 22 patients(34.4%). 2.Comparison of the clinical results between living related and cadaveric donor renal transplantation

Objective To compare the clinical results between living related and cadaveric donor renal transplantation in the same immunosuppressive regimen and HLA match. Methods Twelve cases of living related donor renal transplantation and 22 cases of cadaveric donor renal transplantation were analyzed retrospectively. The 1-, 3-year patient/graft survival rate, renal function and 1-year acute rejection rate were analyzed respectively.Results No renal function injury and surgical complications were found for living donors during the follow-up period. In living donor group and cadaveric donor group, 1-year acute rejection rate was 16.7 % and 22.7 % respectively (P 0.05). Germplasm Resources Information System for Chinese Medicines (GRIS-CM) is designed and realized for improving the management efficiency of the germplasm resources for Chinese materia medica. The system integrates and analyzes the germplasm resources data, realizes information management of the germplasm resources for Chinese materia medica, and provides better services for scientific research institutions, governments, enterprises and medicinal herb growers. It contains 3 databases and 13 function modules, including the basic information base, literature library and gene bank. GRIS-CM can help manage the related data of germplasm resources for Chinese materia medica, and is better for data integration, analysis and statistics to find the rules and patterns. GRIS-CM provides flexible data comparison and visualization functions, and has significant practical value and research value.
4.Prolongation of murine cardiac allograft survival by dendritic cells treated with NF-?B decoy

Objective To investigate the function of donor-derived dendritic cells (DCs) treated with NF-?B decoy in prolonging murine cardiac allograft survival time.Methods Donor bone marrow- derived DCs were treated with NF-?B decoy in vitro.Heterotopic abdominal heart transplantation was performed from BALB/c to C57BL/6 mice.Recipients were grouped according to different pretreat ments as follows:(1) Control group,infusion of PBS (0.2 ml) alone intravenously via the portal vein 7 days before heart transplantation;(2) CsA group,treated with sub-therapeutic CsA only for 7 days (10 mg?kg~(-1)?day~(-1)) through intraperitoneal injection after transplantation,and the same as control group before transplantation;(3) Control DCs group,infusion of only cultured 5th-day recipient DCs untreated with NF-?B ODN decoy;(4) Treated DCs group,infusion of recipient DCs pretreated with NF-~cB ODN decoy;(5) Combined treatment group,infusion of recipient DCs treated with NF-~cB ODN decoy before transplantation and intraperitoneal injection of sub-therapeutic CsA for 7 days (10 mg?kg~(-1)?day~(-1)) after transplantation;(6) Third party donor group,C3H/HeJ mice used as donor, and recipient (C57BL/6) was treated the same as combined treatment group.Every group had 8 mice and graft survival time was observed.Cytokines (IL-2,INF-?,IL-4 and IL-10) in recipient serums were analyzed by ELISA at 7th day after transplantation.Results The graft mean survival time (MST) in control group,CsA group,Control DCs group,treated DCs group,combined treatment group and third party donor group was 7 days,10.3 days,7.6 days,21.4 days,53.6 days and 9 days,respectively.There was significant difference in MST between treated DCs group and control group or control DCs group (P

Objective In order to study the effect on chondrocyte proliferation and the mechanism of treating osteoarthritis(OA) by XBL,the experimant is carried out.Method The first- subcuiture chondrocytes are implanted into two 96- foramen culture plates and sixteen culture flasks.After 24 hours in cubation 8 groups are divided into as follows:No 1 group(control)5% serum DMEM No,2~ 7 groups(XBL)5% serum,0.5% 、 1% 、 2% 、 4% 、 8% 、 16% XBL- DMEM are added respectively.NO 8 group no cell incubation.After culture of 24 and 48 hours ,two plates are detected for viablilty and proliferation respectively by MTT method(Light absorption value A),cell total protein is assessed by comassie Brilliatnt G- 250 colormetric assay.Result 2% ~ 8% XBL can promote the proliferation and protein synthesis of chondrocyte.Conclusion XBL can protec degradation from Matrix metallo protzinase and decrease the impairment of chondrocyte.
6.The anterior chamber depth measurement of primary angle-closure glaucoma with UBM and pentacam

Background Nowadays,glaucoma is the major cause of irreversible blindness worldwide and the second cause of global visual loss.To accurately obtain the anterior chamber depth (ACD)is of helpful for screening primary angle-closure glaucoma (PACG).ObjectiveThe aim of this study is to explore the difference and agreement in the measurement of ACD between Scheimplug Imaging System (Pentacam)and ultra-sonographic biomicroscope (UBM).MethodsSeventy-seven eyes of 77 PACG patients aged (69.96±7.87)were divided into two groups.Thirty-seven eyes with acute primary angle-closure glaucoma (APACG)were assigned to group 1,and forty eyes with chronic primary angle-closure glaucoma (CPACG)were assigned to group 2.Central ACD was measured with Pentacam and UBM,respectively.The Bland-Altman method was used to evaluate the agreement of the two methods.ResultsThe ACD values measured by Pentacam and UBM were (1.6467±0.2687)mm and (1.5601±0.2677)mm respectively in APACG,indicating a significant difference (t=-7.259,P<0.01)and a positive linear correlation between the results of these two methods (r=0.939,P<0.01).The ACD values measured by Pentacam and UBM were (2.0622±0.2317)mm and (1.9648±0.2176)mm respectively in CPACG eyes with a significant difference between them(t=-10.433,P<0.01)and a significant linear correlation between these two outcomes (r=0.967,P<0.01).The Bland-Altman method showed that the two modalities had comparable results for ACD.ConclusionPentacam,as a new 3-dimensional mathematical model of the anterior segment,presents some different results from UBM in the measurement of ACD,but it is not clinically significant.Combination of Pentacam and UBM may be available for the clinical measurement of ACD.
7.Effects of T20N Site-directed Mutation on GTPase Activities of OsRacD from Oryza sativa

OsRacD belonging to rice Rho family of the small GTP binding proteins,is a pivotal gene involved in rice photoperiod fertility conversion of photoperiod sensitive genic male sterile rice,which influences rice fertility via controlling the pollen tube growth.T20N site-directed mutation was introduced into its highly conserved G1 motif by PCR-mediated method to mimic its GDP-binding state based on the sequences alignment and conserved domains analysis.The prokaryotic expression vector of OsRacD and T20N-OsRacD were constructed,and the His6 tag fused proteins were expressed and purified from E.coli.After identified by Western blot,the GTP hydrolysis activities were detected.The results showed that the GTPase activities of T20N-OsRacD were significantly reduced comparing with that of OsRacD,suggested that OsRacD and T20N-OsRacD have different biochemical characteristics.
8.Changes of Malondialdehyde,Myeloperoxidase and Intracellular Free Calcium Contents in Brain Tissue of Fetal Rats Induced by Intrauterine Infection

Objective To establish a model, fetal rat brain damage made by lipopolysaccharide (LPS)-induced intrauterine infection and explore content change and significance of malondialdehyde(MDA),myeloperoxidase(MPO),intracellular free calcium in the development of fetal brain damage.Methods Twenty-eight pregnant rats were injected with LPS(500 ?g/kg)on gestation d20 intraperitoneally in experimental group rats;the same volume of normal saline was injected to 28 rats as control group;there were 8 pregnant rats as blank control rat. Fetal brain water,MDA,MPO,Ca~(2+) contents were measured, fetal brain,placenta for pathological check after injection 2,6,12,24 hours,respectively.Results Brain water,MDA,MPO contents of LPS group were higher than those of controls at 2 h,and Ca~(2+) content of LPS group was higher than that of controls at 6 h,they were continued to be on high level until 24 h.There were histopathological inflammation changes of placenta, edema,inflammation in fetal brain at 6 h ,degeneration and necrosis of nerve cell at 12 h.Conclusions Fetal rat brain damage is made by LPS-induced intrauterine infection.MDA,MPO,Ca~(2+) content increasing in fetal brain tissue that indicates there is lipid peroxidation damage in fetal rat brain damage due to intrauterine infection; damage of free radical,persistent activation of polymorphonuclear,calcium overlord are mechanisms of fetal brain damage .
9.Clinical and Radiographic Features of Short Stature Combined with Skeletal Deformity in Children

Objective To analyze the clinical and radiographic features of short stature combined with skeletal deformity in children,and to diagnose exactly and cure promptly in children.Methods One hundred and twenty children were collected,included 69 males and 51 females.All of the children were collected detail medical history including history of birth,feeding,growth and development and family history and taken both physical and X ray examination that bone age could be evaluate by Gruelich-Pyle method.The children were taken a laboratory examination including calcium,phosphours,alkaline phosphatase,chromosome karyotype,and so on.Results Among 120 children,there were 36 cases(30.0%) of achondroplasia,20 cases(17.0%)of Turner syndrome,10 cases(8.0%)of multiple epiphyseal dysplasia,1 case(0.8%)of spondyloepiphseal dysplasia congenital,7 cases(5.8%) of spondyloepiphyseal dysplasia tarda,2 cases(1.6%) of osteogenesis imperfecta,1 case(0.8%) of cleidocranial dysplasia,5 cases(4.2%)of mucopolysaccharide Ⅳ,5 cases(4.2%) of pseudoachondroplasia,9 cases(7.5%)of hypophosphatemic ricket,24 cases(20%)of short limb dwarf.Among 120 cases,27 were regularity dwarfism(22.5%),93 cases were irregularity dwarfism(77.5%);Among 93 cases of irregularity dwarfism,80 cases were short limb dwarfism including 36 cases of short upper limb(short upper arm 35 cases,shortforearm 1 case),20 cases of short lower limb,24 cases of proportional short stature,13 cases of short trunk.X ray bone age lag 46 cases,ahead of schedule 15 cases(12.5%),normal 59 cases.Conclusions The main causes of short stature combined with skeletal deformity are genetic bone disease.To analyze the feature of clinical and radiographic is a valuble instrument for the children diagnosis of short stature combined with skeletal deformity.Part of them skeletal age were lagged.
10.Study of characteristics of three dimensional motion of cervical spine during maximal axial rotation

Objective To determine the three dimensional motion data of each segment of cervical vertebrae and analyze the characteristics of the intervertebral coupled motion during cervical axial rotation under physiological weight bearing. Methods A total of 16 healthy volunteers (ranging from 22 to 29, median age, 23 years) were recruited to our study. Any cervical spine disorder history, pain or other discomfort and malformations were excluded so as to avoid abnormal neck motion. These subjects underwent CT scans of their cervical segments in a supine position, and 3D models of C1-C7 were constructed. Next, each subject was asked to sit up straight and was positioned in the following sequence:maximal left and right twisting, while double oblique images by DFIS were taken simultaneously at each of the positions. Then, the CT models were matched to the osseous outlines of the images from the two oblique views to quantify the position of cervical vertebraes in 3D at each position. Through local coordinate systems at the center of vertebral bodies, changes of position and angle of each cephalad vertebrae relative to the cauddal one were calculated before and after the axial rotation. Results (1) In the axial rotation of the cervical spine, the contribution of C1/2 accounted for the most of the total cervical rotation range. For the lower levels, axial rotation was found to be maximal at C3/4 and C5/6, minimal at C2/3. (2) In cervical axial motion, C1/2 demonstrated a coupled lateral bending opposite to the axial rotation direction, while each segment of C2-7 demonstrated coupled lateral bending towards the same side of the axial rotation. Among these segments the lateral bending angle of C2/3 was smaller than angles of C3/4, C4/5 and C5/6. Conclusion This study investigated the cervical coupling behavior using the noninvasive 2D-3D matching technique and obtained the motion data at each cervical spinal segment. These findings will help to improve the understanding on physiological cervical spine movement and potential biomechanical mechanism and treatment of cervical spondylosis. Also our data may provide useful reference for the prosthesis design.
