Objective Based on the result that a gene coding for human HSS had been identified and prokaryotically expressed, this study is aiming to investigate the proliferative and protective effect of recombinant HSS on hepatoma cells. Methods hHSS protein was produced in BL\|21 strain of E.Coli containing pET\|42a vector and purified with His\5Tag affinity chromatography. A various dosages of 80\|400??g/L were administrated into cell culture. The cell proliferation was analyzed by MTT, cell\|count and flow cytometry methods. Furthermore, cellular growth signaling as indexed by phosphorylation of mitogen\|activated protein kinase (MAPK) was determined with Western blot. In addition to cell growth, protection of hHSS on the cells against H\-2O\-2 was also observed. Results It was showed that recombinaht HSS of 400??g/L was able to promote DNA synthesis by 21\^5% as compared to non\|treated cells. After 24?h of HSS action, the cell division measured by MTT method as well as cell\|count was significantly enhanced. Meantime, it was indicated that the phosphorylation of MAPK Thr202/Tyr204 was increased in 79\^0% as compared with that of the non\|treated cells. Pretreatment of the cells with hHSS for 12?h prior to H 2O 2 injury preserved the survival of them.Conclusion It is postulated that hHSS is an active protein to stimulate liver proliferation. [

G protein- coupled receptors (GPCRs), also known as seven- transmembrane domain receptors, constitute the largest superfamily of cell surface receptors. By coupling to heterotrimeric G proteins, arrestins and other signaling molecules, GPCRs modulate diverse signal transduction pathways under physiological and pathological conditions. Recent studies have revealed crucial roles of GPCRs in tumorigenesis and development of cancer metastasis. This review summarizes roles of GPCRs, particularly the roles of those coupled to chemokines, prostaglandin, lysophosphatidic acid, endothelin, catecholamine and angiotensin in proliferation, invasion, metastasis and angiogenesis of hepatoma cells and development of hepatocellular carcinoma. The potential of GPCRs- based therapeutics being used for hepatocellular carcinoma is also highlighted.

Objective To explore the curative effect of coronary artery bypass graft (CABG) in the treatment of acute coronary syndrome (ACS).Methods Clinical data of 64 patients with ACS were retrospectively analyzed,41 patients received CABG (CABG group) and 23 patients received percutaneous coronary intervention (PCI,PCI group).Mainly cardiac events and secondary cardiac events were compared.Results The incidence rate of cadiovascular and cerebrovascular events during hospitalization in CABG group was 7.32％(3/41),in PCI group was 8.70％ (2/23),there was no significant difference (P ＞ 0.05).All patients accomplished follow-up and follow-up time was 12-48 months.The meta follow-up time in CABG group was 35.26 months,in PCI group was 33.19 months.The incidence rate of cadiovascular and cerebrovascular events during follow-up in CABG group was 19.51％ (8/41),in PCI group was 30.43％(7/23),there was no significant difference (P＞ 0.05).The incidence rate of death,myocardial infarction,cerebrovascular events,angina pectoris and total cadiovascular and cerebrovascular events between two groups had no significant difference(P＞ 0.05).Conclusions The surgery treatment in ACS is feasible,the curative effect of CABG and PCI is therapeutic equivalence.The choice of operation should be based on patients' condition and individual choice.Appropriate revascularization can get the best treatment efficiency.

BACKGROUND: As the result of estrogen shortage due to ovariectomy,osteoporosis occursin the general and local bones, displaying bone loss and changes in bone microstructure and growth factor mRNA expression,which definitely has an important effect on fracture healing. OBJECTIVE: To probe into the effect of estrogen on bone microstructure and bone morphogenetic protein-4 mRNA expression and location during fracture healing. DESIGN: Randomized controlled study. SETTING:Laboratory of the Department of Molecular Biology of Yunnan University. MATERIALS: This study was conducted at the laboratory of Molecular Biology Department of Yunnan University between July 1999 and July 2002. We recruited 96 healthy female SD rats of 2 months old and with the mean body mass of 160 to 200 g. METHODS:①Of the 96 rats that received intraperitoneal anesthesia, 48rats were subjected to bilateral ovariectomy(osteoporotic group), and the other 48 rats received sham operation (control group). One month later, bilateral tibia fracture at the middle segment was artificially made on all rats under anesthesia, and no treatment was given so as to prepare fracture healing model. Then rats of both groups were put to death for collecting callus and the surrounding parenchyma at postoperative 1, 3, 5, 7, 14, 28,56 and 112 days, with 6 rats in each time point. ② The tibia bone microstructure was observed under electron microscope. ③ The mRNA expression of bone morphogenetic protein-4 at callus and the surrounding paranchyma was detected by RT-PCR method; no probe in hybrid fluid was used as negative controls. Six in situ hybridization slices with positive expression were selected from both groups at postoperative 1 and 3 days time points, and 3 visual fields were randomly selected from each slice for observing the positive granules under 25 times field lens. MAIN OUTCOME MEASURES: ①The tibia bone microstructure;② bone morphogenetic protein-4 mRNA expression at callus and the surrounding parenchyma. RESULTS: All the 96 rats entered the final results analysis. ① Observation of the tibia bone microstructure: at 28 days after tibia fracture, osseous callus and ostein fibers were found arranged densely in control group. Osteocytes, small with fewer cytoplasts, were observed in osseous lacuna, but osteoclasts were found surrounding small-sized bone trabecula. In osteoporotic group, fibrous callus and collagenous fibers in bone matrix were arranged loosely, lots of big osteoblasts could be observed with osteocytes easily seen. ② Bone morphogenetic protein-4 mRNA expression at callus and the surrounding parenchyma: it was less expressed in control group than in osteoporotic group at postoperative 1 to 3 days (23.714 3±5.056 8,21.714 3±5.023 8 vs 51.285 7±8.138 7,49.571 4±9.071 1, P ＜ 0.01) and the expression was mainly observed in parenchyma surrounding the fracture where callus was formed. CONCLUSION: Bone morphogenetic protein-4 mRNA expression is increased in osteoporotic group and is mainly observed in parenchyma surrounding the fracture, displaying a manner of regional expression. However,the formation and quality of callus matrix during fracture are obviously poorer than in control group.

Objective To explore the changes and clinical value of serum insulin like growth factor-1 (IGF-1) and adiponectin in newborns with hypoxic ischemic encephalopathy (HIE). Methods Fifty-two HIE newborns were recruited in this study, including 15 severe, 20 moderate and 17 mild HIE newborns. Twenty healthy newborns were selected as controls. Serum levels of IGF-1 and adiponectin were detected 3-to-5 days (acute period) and 10-to-14 days (recovery period) after birth. Results Serum levels of IGF-1 and adiponectin in the acute period differed signiifcantly among groups of different severity (P<0.05). Serum levels of IGF-1 and adiponectin were decreased with the increase of HIE severity. The level of adiponectin in moderate and severe HIE was lower than that in mild HIE (P<0.05). In recovery period, the level of IGF-1 in severe HIE was lower than that in control (P<0.05). Serum levels of IGF-1 and adiponectin in the acute period was positivity correlated with those in umbilical cord blood of HIE newborns (r=0.531, r=0.611, P<0.01). Conclusions Decreased levels of IGF-l and adiponectin in HIE newborns are correlated with the pathological process of HIE, and can be regarded as indices for severity of HIE. IGF-l and adiponectin is of signiifcance in the clinical diagnosis and prognosis of HIE.

Objective To study the clinical feasibility of the percutaneous sclerosis treatment for limb and maxillofacial region low flux vascular malformations. Methods 17 cases with limb and maxillofacial region low flux vascular malformation were collected from 1998 to 2002 accepting interventional treatment in our hospital with application of direct percutaneous sclerosis or percutaneous sclerosis through arteriography or arterial embolism three methods, using tardener's alcohol method of Hardener with alcohol or iodinated oil accompanied by pingyangmycin. Results Altogether 17 cases included with once cured in 11 cases, multple times of treatment in 5 and the last case needed necrotic tissue resection with skin graft due to localized muscular necrosis caused by inproper embolization. Conclusions The interventional treatment for limb and maxillofacial region low flux vascular malformation is an effective and safety method.

AIM:Although there are many evidences showing positive effects of dehydroepiandrosterone sulfate(DHEA-S) in bone metabolism,the reasonable and thorough experiments are still needed to clear the mechanism of DHEA-S on bone metabolism by excluding the biological effect of its derivative hormone.This experiment is to study the efficacy and safety of DHEA-S in treatment of osteoporosis in elderly males.METHODS:①From October 2005 to October 2006,120 out-and in-patients with primary osteoporosis who were admitted in Shanghai 6th People's Hospital were selected.The diagnosis of primary osteoporosis referred to the new standard from WHO:without the application of sexual hormone,and over 2.5 standard deviation value of bone mineral density lower than the peak value of the same gender subject.All the cases were randomly divided into treatment group and control group(60 cases in every group).② DHEA-S 100 mg and Caltrate-D 600 mg were orally given to the treatment group;Caltrate-D 600 mg was orally given to the control group.③After 6 months,the bone density,serum biochemical markers,markers of bone resorption and formation as well as side effects were measured.RESULTS:120 osteoporosis patients entered into analysis.①Compared to the pretreatment and control group,the bone mineral density in lumbar spine 1,lumbar spine 2-4,femoral neck,and upper part of femoral bone were increased significantly in the treatment group(P 0.05).CONCLUSION:Treatment of osteoporosis in elderly males with DHEA-S is safe and effective without obvious side effects.

Objective To develop measles and Japanese encephalitis(JE)combined attenuated live vaccine. Methods The independent development of China′s Shanghai 191 measles vaccine strains and Japanese encephalitis SA14-14-2 vaccine strains were selected to prepare two of monovalent vaccines solution,which were used to conduct the interference test,the ratio test,the screening test of protection agent,the screening test of dilution,and the alternative of the curve of freeze-dried.Results The preparation technology of measles and Japanese encephalitis combined live attenuated vaccine was determined.The batches of the products met the quality requirement of the three kinds of monovalent vaccine in the Pharmacopoeia of the people's Republic of China(edition 2005,volume 3).The cross-interference and increased response were not found in the animal test,and the immune response of mouse was not influenced,which indicated the safety and immunogenicity of with good combined vaccine.Conclusion Measles and Japanese encephalitis combined attenuated live vaccine was developed successfully.
