Objective To study the effects of propofol pretreatment on S100? and neurosecretion enzyme (NSE) of brain tissues with global cerebral ischemia-reperfusion in rat, and to evaluate the effects of propofol in protection of brain. Methods 30 SD male rats were randomly divided into 3 groups: sham operation group (group A, n=10); single cerebral ischemia-reperfusion group (group B, n=10); propofol pretreatment at 2h before ischemia group (group C, n=10), in which propofol (100mg/kg) was given intraperitoneally (ip) before ischemia. 24h after ischemia-reperfusion, the neuroethology scores were recorded and evaluated, and S100? and NSE in rat brain were determined. 2.Effects of Microfiltration Membrane Adsorption on Detection of Pesticides in Water by Direct Injection Liquid Chromatography-Tandem Mass Spectrometry

Effects of microfiltration membrane adsorption for eight types of pesticides with diverse physicochemical properties on direct injection liquid chromatography-tandem mass spectrometric method ( DI-LC-MS/MS) were investigated. The results showed that the adsorption effects ( AE,%) on the loss of signal detection at concentration of 2. 5 μg/L ranged from 8%-92%, 0%-81% and 2%-59% for nylon (NYL), polyethersulfone ( PES) and hydrophilic polypropylene ( GHP) membrane, respectively ( accumulated volume of feed is 1 mL). The pesticides with higher octanol-water partition coefficient (lgKow) values appeared to adsorb onto the membrane surface more readily than those with lower lgKow. The AE was found to be alleviated as the feed volume of samples increased to 10 mL, except the case of profenofos adsorption on PES membrane ( AE=9%) . The effects of PES membrane on the eight pesticides at initial individual concentration of 0 . 25 , 2. 5 and 25 μg/L were also studied. The results showed that, the lower the initial concentration of pesticides, the stronger adsorption effect under the same filtration conditions was obtained. Besides, it was effective in eliminating the membrane adsorption effects by adding 40% methanol ( V/V) into water samples as a modifier, which could consequently improve the accuracy of DI-LC-MS/MS.
3.Research of oxaliplatin inhibit growth in glioma U87 cells by regulating PI3K/Akt signal pathway

Objective:To investigate the effect of oxaliplatin inhibit the growth in glioma U 87 cells by regulating PI3K/Akt sig-nal pathway.Methods:The glioma U87 cells were cultivated in vitro ,using 0,20,40,80 μg/ml oxaliplatin treated U87 cells 24,36, 48,72 h respectively,MTT was used to detect cell proliferation.Using 40 μg/ml oxaliplatin treated U87 cells 48 h,flow cytometry was used to detect cell cycle and apoptosis .Cells apoptosis protein and PI 3K/Akt pathway protein expression after 40 μg/ml oxaliplatin treated U87 cells 48 h was detected by Western blot .Results:MTT assay showed that compared with the 0μg/ml treatment ,oxaliplatin treatment could significantly inhibited U 87 cell survival ( P<0.01 ) ,40μg/ml oxaliplatin treatment 48 h ,the survival inhibitory was the most obvious.U87 cell cycle was arrested in S phase after 40 μg/ml oxaliplatin treatment 48 h.After 40μg/ml oxaliplatin treatment 48 h,compared with the 0 μg/ml treatment,U87 cell apoptosis rate significantly increased (P<0.01).Western blot showed that after 40μg/ml oxaliplatin treatment , anti-apoptotic factor Bcl-2 expression had significant decreased , pro-apoptotic factors Bax , Cleaved-caspase3 protein expression had significantly increased (P<0.01).PI3K,p-Akt expression was significantly decreased (P<0.01),and Akt expression had no significant change in Akt pathway (P>0.05).Conclusion:Oxaliplatin may suppress U87 cell proliferation,ar-rest cell cycle,and promote apoptosis by inhibit PI3K/Akt signaling pathway.
4.Analysis of dual energy CT imaging in the diagnosis of gouty tophus deposition value

Objective To explore the value of dual energy CT imaging in the diagnosis of gouty tophus deposition.Methods Sixty-five cases of patients with gout treated in this hospital between December 2012 to December 2015 were selected.Another 65 cases of patients(rheumatoid arthritis,degenerative osteoarthritis and unexplained leg pain) were selected as control group.The re sults of dual energy CT imaging between two group were compared.Three experienced clinicians who didn't understand the imaging results were selected.They had a medical history and physical examination in patients with gout.The differences between clinical evaluation and imaging results were compared.Results Dual energy CT imaging showed that there were no tophi deposition in con trol group;the topi deposition in wrist joint,knee joint and ankle joint were 6.06%oo (33/34),100.00% (22/22) and 96.49% (55/57) in gout group.Dual energy CT imaging detected a total of 545 parts of tophus deposition.It was 4.36 times of the result of clinical evaluation(125 parts).Conclusion Dual energy CT imaging shows the advantages of simple noninvasive,high accuracy and low rate of misdiagnosis in diagnosing gout nodules.
5.Value of Multi-slice CT in Diagnosis of Papillary Renal Cell Carcinoma

Purpose To evaluate the value of multi-slice spiral computed tomography (MSCT) scan in the diagnosis of papillary renal cell carcinoma. Materials and Methods A retrospective study was conducted on the findings of plain CT scan and triphasic dynamic contrast enhanced CT scan of 23 patients with papillary renal cell confirmed pathologically, and further contrasted with the findings of 96 patients with clear cell renal cell carcinoma at corresponding phases in such aspects as cystic degeneration, enhancement degree and pattern (homogeneous, peripheral or heterogeneous) and tumor spreading pattern (perinephric invasion, lymphadenopathy or venous invasion). Results The degree of enhancement CT value for both diseases showed statistical significance in corticomedullary phase, parenchymal phase and excretory phase (t=9.70, 8.08, 5.92;P<0.01), but papillary renal cell carcinoma was lower than clear cell renal cell carcinoma. In terms of cystic degeneration, papillary renal cell carcinoma was significantly lower than clear cell renal cell carcinoma (21.7%vs 54.2%,χ2=7.54, P<0.01). Papillary renal cell carcinoma mainly showed homogeneous enhancement (60.9%) whereas clear cell renal cell carcinoma mainly presented heterogeneous enhancement (68.8%) with statistical difference (χ2=30.40, P<0.01). The tumor spreading patterns like perinephric invasion, lymphadenopathy and venous invasion had no significant difference (χ2=0.51, 0.11, 0.03;P>0.05). Conclusion Papillary renal cell carcinoma shows homogeneous enhancement, seldom cystic degeneration or necrosis on MSCT scan, which is helpful for the differential diagnosis from clear cell renal cell carcinoma.
6.Wuling Capsule promotes hippocampal neurogenesis by improving expression of connexin 43 in rats exposed to chronic unpredictable mild stress.

Objective: To investigate the effects of Wuling Capsule, a compound traditional Chinese herbal medicine, on hippocampal neurogenesis by examining the expressions of brain-derived neurotrophic factor (BDNF) and connexin 43 (Cx43) in rats with depression induced by chronic unpredictable mild stress (CMS), and thereby to explore its antidepressant mechanism. Methods: Forty-five adult male Sprague-Dawley rats were randomly divided into three groups: control group (n=15), untreated group (n=15) and Wuling group (n=15). All rats except those in the control group were subjected to 3-week CMS to induce depression. At the same time Wuling Capsule was daily added to the diet of the rats in the Wuling group at a dose of 100 mg/kg body weight for 21 days. The degree of depression was determined by sucrose preference test. BDNF expression and neurogenesis were tested by using immunohistochemical staining with BDNF and 5-bromodeoxyuridine (BrdU) antibodies; and the mRNA and protein expression levels of Cx43 in hippocampus were examined by semi-quantitative reverse transcription-polymerase chain reaction and Western blotting. Results: The numbers of BDNF-positive neurons and BrdU-positive particles in dentate gyrus (DG) were significantly decreased in CMS rats as compared with the normal rats, and the same changes were found in Cx43 mRNA and protein expressions. After Wuling Capsule treatment, the depressed behaviors were improved. Moreover, the reduced expression levels of Cx43 mRNA and protein and fewer newborn neurons induced by CMS were recovered to the normal levels. However, BDNF-positive cells remained low in DG. Conclusion: Wuling Capsule can improve the low hippocampal neurogenesis in rats subjected to CMS and the antidepressant effects are related to enhancing the Cx43 expression but not through BDNF mediation.
7.Nardosinone reduces neuronal injury induced by oxygen-glucose deprivation in primary cortical cultures.

The aim of the study is to investigate the effect of nardosinone (Nar) on neuronal injury induced by oxygen-glucose deprivation (OGD) in primary cortical cultures isolated from embryos at gestational day 14. MTT method was used to determine the dosage regimen of Nar in primary neuronal cultures and observe the influence of Nar on the neurons suffering OGD; Western blotting analysis was used to detect expressions of protein kinase A (PKA), Ras related protein 1 (Rap1), mitogen-activated protein kinase kinase 1 (MEK1) and phospho-extracellular signal-regulated kinase 1/2 (p-ERK1/2) of OGD-injured or uninjured primary cultured neurons after Nar treatment. Results showed that Nar (50 and 100 micromol x L(-1)) improved the cell viability during OGD damage (P < 0.01) and increased the expression of PKA, Rap1, MEK1 and p-ERK1/2 in injured neurons. Additionally, elevations of PKA, Rapl, MEK1 and p-ERK1/2 in uninjured neurons were caused by Nar (50, 100 and 200 micromol x L(-1)) with a dose-dependent tenclency as well (P < 0.01). In conclusion, Nar could protect against the neuronal injury exposed to OGD, which may be relevant to the promotion of PKA and ERK signaling pathway.
8.Determination of Trace Metal Impurities in High Pure Tin Oxide Electrode Material by Inductively Coupled Plasma Mass Spectrometry

A new method for determination of trace Cu, Cr, Mn, Co, Ni, Cd, Fe and Pb impurities in high pure tin oxide material was proposed. 0. 1 g of sample was successfully digested at 230℃ in 3 h when autoclaves with high pressure and high temperature were used. After that, over 99. 9% of the matrix Sn was removed by chloride volatilization at 150℃ for 1. 5 h, which eliminated the matrix effect and spectra interference in the following ICP-MS detection. The recoveries for all the analytes of the spiked test were 100%±5%, the RSDs for 6 runs of sample analysis were from 0. 09% to 3. 68%, and the limits of detection (LOD, 3σ) were from 0. 002 mg/kg to 0. 034 mg/kg, which indicated that the established method were of both accuracy and high sensitivity, and could be applied for the analysis of samples with purity up to 6N. Finally, three samples specified with 4N purity were analyzed by this method.
9.Curative analysis of suture anchors in repairing delayed Achilles tendon rupture

Objective To summarize the clinical efficacy of suture anchors in repairing the delayed Achilles tendon rupture.Methods From January 2010 to December 2013,23 patients with delayed Achilles tendon rupture were treated using the suture anchors.There were 19 males and 4 females,at mean age of 43 years (range,35-53 years).Injury on the left side occurred in 5 patients and right side in 18 patients.Mean time from injury to operation was 26.3 days (range,21-40 days).Ten patients were diaguosed by ultrasound and the other by MRI.All patients underwent suture anchor fixation without external support.Functional training started at the early stage.Incision complications were detected.Ankle flextiou-extension range,American Orthopedic Foot and Ankle Society (AOFAS) score,maximum bilateral leg circumference,tendon rerupture and walking posture were recorded at postoperative 6,12,18 and 24 months.Achilles tendon muscular strength was measured with the Lunsford-Perry heelrise test.Results Follow-up was 24 months.All incisions healed by first intention without sural nerve injury,adherence with skin and deep infection.Six months after operation,the ankle range of motion was dorsiflexion 10.8° (range,9-15°) and plantar flexion 43.8° (range,40-48°),with no significant difference in comparison to the health side (P > 0.05).AOFAS score differed significantly before and after operation (P < 0.01).Maximal leg circumference was 38.2 cm in the health side versus 35.8 cm in the injury side (P < 0.05),but the difference was no more than 3 cm.All patients completed 25 times heel raising without difficulty.Four patients walked with a slight limp and recovered from the limp 12 months after operation.No Achilles tendon ruptured again during the follow-up time.Conclusions Repairing the old Achilles tendon rupture with suture anchors can supply strongly strain between broken ends of the tendon,and the outer cast is not needed after operation.Complications are less and functional practice can be commenced in the early postoperative period for better function restoration.
10.Histological analysis of chondrons isolated from age-related normal rabbit knee

Objective To explore the histological properties of isolated chondrons and chondrocytes from rabbit knee cartilage,and to determine if these properties vary with age.Methods Three groups of rabbit knees were evaluated according to different age:(1) young (2 months,n=10);(2) adult (8 months,n=10);and (3) old (31 months,n=10).The cartilage structure,proteoglycan,collagen-2,and collagen-6 content were determined by light microscopic using hematoxylin-eosin (HE),Toluidine Blue,and col-2,6 staining.The chondrons were enzymatically isolated using 0.3 g/L dispase and 0.2 g/L collagenase-2 by shaking for 3 hours.The morphology and composition of isolated chondrons were observed by HE and collagen-6 immunostaining staining after overnight coverslip monolayer culture under a microscopy.Results The chondrocytes became sparser and the total content of proteoglycans and collagen-2 were decreased in the articular cartilage with age.Compared to the chondrocytes,the surrounding rim or capsule was more obvious in the isolated chondrons,and they exhibited obvious differences in shape.The cells within one cluster from different age groups were similar to the morphology observed in cartilage in situ.The adult and old chondrons generally possessed a thicker pericellular matrix with more enclosed cells,and the chondrons contained more cells can reach 47%.Conclusion These findings further suggest that the properties of the chondrons and pericellular matrix have an important influence on the biomechanical microenvironment of the knee joint cartilage degeneration that occurs with age.
