1.The experimental study on the relationship between the hepatic glycogen content and the liver transplantation of non-heart-beating donor

Objective To study whether the warm ischemia-reperfusion injury can be alleviated if the hepatic glycogen content were increased before liver transplantation.Methods The male SD rats as donors and recipients were divided into groups A,B and C randomly.In group A,the rats were al- lowed access to solid and water ad libitum;In group B,the rats drank glucose liquor for 4 days prior to liver harvesting;In group C,the rats were intravenously injected with 50% glucose besides the protocol in group B 3-4 h prior to liver harvesting.According to the non-heart-beating time(the non- heart-beating time were set into 60,90,120 and 150 min),the rats in groups A,B and C were divided into 4 subgroups,and the orthotopic liver transplantation was performed and the hepatic glycogen and ATP contents were measured.The one-week survival rate and the serum levels of MDA and SOD in the recipients were determined after liver transplantation.Results The hepatic glycogen and ATP con- tents in groups B and C were more than those in group A significantly(P
2.Enzymatic degradation of marine polysaccharide YCP and analysis of YCP-derived fragments

Aim: To explore the relationship between structure and immunological activity of marine polysaccha-ride YCP,the physicochemical property and immunological activity of the YCP-derived fragment were studied.Methods: YCP was hydrolyzed by α-amylase from human saliva.The hydrolysate was purified to obtain an polysaccharide fragment by gel filtration chromatography.The physicochemical properties of this YCP-derived fragment was characterized by HPLC,FT-IR and TLC.In addition,changes of phagocytic activity,production of reactive nitrogen and macrophage binding were investigated.Results: The relative molecular weight of YCP-de-rived fragment was approximately 6.6 × 10~3.The monosaccharide composition and FT-IR of the YCP-derived frag-ment were identical to YCP.No significant effect of the YCP-derived fragment on NO production and murine mac-rophage phagocyte were observed.And this fragment was not able to compete the binding between YCP and mac-rophages.Conclusion: The remarkable decrease of immunological activity of YCP-derived fragments degraded byα-amylase of human saliva suggests that the complete structure and high molecule weight of YCP are essential for its immuno-modulatory activity.

Long non-coding RNA (lncRNA) is a newly identified non-coding RNA subfamily with various regulatory functions.Recent evidence has shown the fundamental role of long noncoding RNAs in affecting the development of diseases at different levels,from gene modification,transcriptional regulation to protein transla tion.The study on lncRNA has made great progress in the studies of genomic imprinting,cancer diseases and neurodegenerative disorders,while the research relevant to autoimmune diseases has just staaed recently,However,many lncRNAs have been identified to involve in immune cells proliferation,differentiation and maturation,acting as the key regulators in immune homeostasis and autoimmune diseases.This review is focused on the advances of lncRNA in immune cells and autoimmune diseases.
6.Inhibitory effects of blocking voltage-dependent potassium channel 1.3 on human monocyte-derived macrophage differentiation into foam cells

Objective: To investigate the expression of voltage-dependent potassium channel 1.3(Kv1.3) mRNA and protein during human monocyte-derived macrophage differentiation into foam cells and its function in foam cell formation. Methods: Human peripheral blood monocytes were isolated from healthy male volunteers by density gradient centrifugation and then by adherent method. The obtained monocytes were cultured for 5 days to differentiate into macrophages. Based on establishment of the human macrophage-derived foam cell model, the expression of Kv1.3 channel was investigated by immunocytochemical staining, reverse transcription-polymerase chain raction (RT-PCR) and Western blot. Furthermore, the effects of rMargatoxin, a Kv 1.3 channel-specific inhibitor, on cholesterol metabolism in macrophages incepting oxidized low density lipoprotein (OxLDL) were studied. Results: After the macrophages co-incubated with 30 mg/L OxLDL at 37 ℃ for 60 hours, the cellular volume obviously enlarged and many red lipid granules were deposited in cytoplasm. The total amount of cholesterol (TC),free cholesterol ( FC ) and cholesterol ester ( CE ) in cells markedly increased and the ratio of CE/TC rose from ( 14.4±6.8) % to (57.9±3.5) % (n=7,P<0.05). However, the expression of Kv1.3 channel had no significant change. r Margatoxin (0.1 nmol/L and 10 nmol/L) markedly reduced the contents of TC, FC and CE in macrophages and the ratios of CE/TC decreased to (42.8±11.6) % and (22.6±8.0)% , respectively (n=7, P<0.05). Meanwhile, the red lipid granules deposited in the cytoplasm of macrophages also decreased. Conclusion: These data clearly show that the expression of Ky1.3 channel does not change obviously during human monocyte-derived macrophage differentiation into foam cells and the blocking of it would prevent foam cell formation.
7.Targeted contrast-enhanced ultrasound in the assessment of renal cell carcinoma angiogenesis in nude mice

Objective To investigate the value of contrast enhanced ultrasound(CEUS) with microbubbles targeted to vascular endothelial growth factor receptor type 2 (VEGFR2) for imaging tumor angiogenesis in murine tumor models.Methods Established human renal cell carcinomas(RCC) subcutaneous xenograft tumor model in nude mice,microbubbles targeted to VEGFR2(MBV) was prepared,control microbubbles(MBC) and MBV were injected respectively in each mouse,the intensity of each microbubble was compared,the expressions of VEGFR2 in tumors were tested by immunohistochemistry.Results CEUS imaging showed the intensity of MBV and MBC was (6.50±1.43)dB,(2.59±0.99)dB,respectively.There was significantly higher intensity when using MBV compared with MBC (P<0.01).The time to wash out was longer in MBV contrast group compared with MBC group.Immunohistochemistry showed VEGFR2 was highly expressed in novel vessel walls.Conclusions Contrast microbubbles targeted to VEGFR2 can specially enhance the images of RCC and has tremendous significance in the assessment of angiogenesis.
8.Expression and clinical significance of c-kit and PCNA in renal cell carcinoma (RCC) subtypes

Objective To investigate the expression of c-kit and analyze its relationship with proliferating cell nuclear antigen (PCNA) in RCC subtypes and its clinical progression. Methods Expression of c-kit protein was retrospectively studied with immunohistochemistry in paraffin sections from 137 cases of clear renal cell carcinoma (CCRCC), 82 papillary renal cell carcinoma (PRCC), 51 chromophobe renal cell carcinoma (ChRCC). Results The positive rate of c-kit in ChRCC was 94.1%(48/51), it was statistically higher than that in CCRCC (16. 1%, 22/137) and PRCC (28.1 %, 23/82)(P=0. 001 ). In ChRCC, the positive expression of c-kit was related with TNM stages. The positive expression of PCNA was related with the grade in CCRCC and PRCC. But there was no relationship between PCNA expression and grade of ChRCC. It also had the relationship with the metastasis in CCRCC. Conclusions The expression of c-kit in ChRCC is higher than in other subtype of RCC, and associated with tumor local progression. That makes c-kit as a helpful marker to discriminate different subtypes of kidney cancer.

10.Study on cognitive function of the patients with bipolar depression and unipolar depression and their first-degree relatives

Objective To explore the cognitive function of the patients with bipolar depression (BP), unipolar depression (UP) and their first-degree relatives.Methods 52 patients with BP and 49 their first-degree relatives ,67 patients with UP and 59 their fi~t-degree relatives, 97 normal controls were tested by information, arithmetic, digital symbol, digital span test, block design,logical memory (included immediately and delayed) , visual memory(included immediately and delayed), Stroop Test (included Stroop C and Stroop CW), trail making test (included Part A and Part B) ,verbal fluency and modified Wisconsin card sorting test(M-WCST).Results The scores of digital symbol, block design, immediately logical memory, delayed logical memory, immediately visual memory,delayed visual memory,trail making test B and WCST in the patients with BP and UP were worse than those in normal control(P<0.01).The score of WCST in the patients with BP was lower than that in the patients with UP ((3.02± 1.96), (4.40± 1.13), P<0.01).The score of WCST in the relatives of the patients with bipolar depression was worse than that in the normal control((4.32±1.66), (5.15±1.27), P< 0.05).Conclusion The patterns of impaired cognitive function on sustained attention,concept of visual space,visual memory,verbal memory and executive function are similar in the patients with BP and UP.The executive function of the patients with BP is worse than the patients with UP.
