1.Changes of Malondialdehyde,Myeloperoxidase and Intracellular Free Calcium Contents in Brain Tissue of Fetal Rats Induced by Intrauterine Infection

Objective To establish a model, fetal rat brain damage made by lipopolysaccharide (LPS)-induced intrauterine infection and explore content change and significance of malondialdehyde(MDA),myeloperoxidase(MPO),intracellular free calcium in the development of fetal brain damage.Methods Twenty-eight pregnant rats were injected with LPS(500 ?g/kg)on gestation d20 intraperitoneally in experimental group rats;the same volume of normal saline was injected to 28 rats as control group;there were 8 pregnant rats as blank control rat. Fetal brain water,MDA,MPO,Ca~(2+) contents were measured, fetal brain,placenta for pathological check after injection 2,6,12,24 hours,respectively.Results Brain water,MDA,MPO contents of LPS group were higher than those of controls at 2 h,and Ca~(2+) content of LPS group was higher than that of controls at 6 h,they were continued to be on high level until 24 h.There were histopathological inflammation changes of placenta, edema,inflammation in fetal brain at 6 h ,degeneration and necrosis of nerve cell at 12 h.Conclusions Fetal rat brain damage is made by LPS-induced intrauterine infection.MDA,MPO,Ca~(2+) content increasing in fetal brain tissue that indicates there is lipid peroxidation damage in fetal rat brain damage due to intrauterine infection; damage of free radical,persistent activation of polymorphonuclear,calcium overlord are mechanisms of fetal brain damage .
2.Anti-inflammation, Analgesic and Hemostatic Effects of Veronicastrum sibiricum

Objective To explore the anti-inflammation, pain-relieving and bleeding-stopping effects of Veronicastrum sibiricum and its potential to be a medicinal resource of Radix clematidis.Methods Total of 120 mice were randomly divided into 6 groups, including the Veronicastrum sibiricum water extract (1.5, 1.0, 0.5 g·mL-1) groups, Radix clematidis water extract (1.0 g·mL-1) group, the 0.9% sodium chloride solution group and 2% aspirin solution group.The mouse hot-plate test and writhing test were carried out to evaluate the analgesic effect.The anti-inflammation effect was observed by the xylene-attracted swelling of the mouse's ears.The bleeding time of the breaking mouse's tails was used for studying hemostatic effect.Results The Veronicastrum sibiricum could significantly increase the pain threshold of mice in hot plate.At the same dosage, the Veronicastrum sibiricum reducing the writhing times of mice was similar to Radix clematidis.Auricle swelling inhibition rate was 5.72% higher than Radix clematidis (P<0.05);The mouse tail bleeding time was 7.6% shorter than Radix clematidis (P<0.05).Conclusion The Veronicastrum sibiricum shows similar analgesic effect to Radix clematidis,and more strong hemostatic and anti-inflammation effects than Radix clematidis.
3.Basic research and clinical application of musculocutaneous perforator flaps

The use of flaps for plastic and reconstructive surgery is very popular today.More and more surgeons have realized that the blood supply plays an important role in the design and survival of flaps.After Ian McGregor and Ian Jackson first introduced the axial flaps into clinics,Mathes divided the flaps into two types in 1981,the fasciocutaneous perforator flaps and musculocutaneous perforator flaps.In 1989,the concept of perforator flap was first brought into clinical practice.With the development of microsurgery and anatomy,the epidermis of human skin is divided into areas based on the vascular anatomy.A new century of musculocutaneous perforator flaps has been established.The terminology,classification,characteristics and application of musculocutaneous perforator flaps are described in this paper.
4.Proliferation of desmoglein 3-stimulated T lymphocytes from patients with pemphigus vulgaris

Objective To investigate the impact of desmoglein 3 (Dsg3) on the proliferation of peripheral T lymphocytes from patients with pemphigus vulgaris (PV).Methods Peripheral blood mononuclear cells (PBMCs) were obtained from 12 patients with PV and 22 normal human controls,cultured with or without the presence of Dsg3 or phytohemagglutinin for 3 days.Flow cytometry was performed to detect the changes of T-lymphocyte subsets in PBMCs stimulated with Dsg3 and proliferation of T-lymphocytes.Results In patients with PV,the percentage of Th2 and Th1 cells was 12.17%±5.32% and 4.08%±1.50%,respectively in Dsg3stimulated PBMCs,9.84%±5.41% and 3.91%±1.38%,respectively in non-stimulated PBMCs.Increased percentage of Th2 cells was observed in Dsg3-stimulated and non-stimulated PBMCs from patients with PV compared with those from normal human controls (both P<0.05).After stimulation with Dsg3,there was a significant proliferation of T cells from patients,and the proliferation rate of CD4+T cells was 4.65%±3.28%,which was significantly higher than that from normal controls(P<0.05).Conclusion Dsg3 can induce the specific proliferation of CD4+T cells,especially Th2 type CD+4 T cells,from patients with PV.
5.Blockage of PD-L1/PD-1 pathway for enhancing the chemotherapeutic efficacy of cisplatin

Objective To explore the anti-tumor effect and the influence of antitumor immunity of PD-L1/PD-1 blocked by PD-1 antibody combined with cisplatin. Methods Tumor models were established by injecting TC-1 cells into C57BL/6 mice, and the mice were divided into four groups (n = 4). The tumor growth curves and survival curves were drawn to observe the anti-tumor effect. The tumors were then removed; and the PD-L1 and CD8+ T cells were analyzed by immunohistochemical method. Results The anti-tumor effect was greater in the cisplatin group , PD-1 antibody group , and PD-1 antibody plus cisplatin group than in the control group (P < 0.05). Expression of PD-L1 in the tumor tissues was markedly increased in the cisplatin group and it was obviously decreased in the combination group (P < 0.05). CD8+ T cells decreased in the cisplatin group; and expression of CD8+ T cells was significantly increased the combination group (P < 0.05). Conclusion The anti-tumor effect and anti-tumor immunity of cisplatin are enhanced by blocking PD-L1/PD-1 pathway with PD-1 antibody.
6.Curative analysis of suture anchors in repairing delayed Achilles tendon rupture

Objective To summarize the clinical efficacy of suture anchors in repairing the delayed Achilles tendon rupture.Methods From January 2010 to December 2013,23 patients with delayed Achilles tendon rupture were treated using the suture anchors.There were 19 males and 4 females,at mean age of 43 years (range,35-53 years).Injury on the left side occurred in 5 patients and right side in 18 patients.Mean time from injury to operation was 26.3 days (range,21-40 days).Ten patients were diaguosed by ultrasound and the other by MRI.All patients underwent suture anchor fixation without external support.Functional training started at the early stage.Incision complications were detected.Ankle flextiou-extension range,American Orthopedic Foot and Ankle Society (AOFAS) score,maximum bilateral leg circumference,tendon rerupture and walking posture were recorded at postoperative 6,12,18 and 24 months.Achilles tendon muscular strength was measured with the Lunsford-Perry heelrise test.Results Follow-up was 24 months.All incisions healed by first intention without sural nerve injury,adherence with skin and deep infection.Six months after operation,the ankle range of motion was dorsiflexion 10.8° (range,9-15°) and plantar flexion 43.8° (range,40-48°),with no significant difference in comparison to the health side (P > 0.05).AOFAS score differed significantly before and after operation (P < 0.01).Maximal leg circumference was 38.2 cm in the health side versus 35.8 cm in the injury side (P < 0.05),but the difference was no more than 3 cm.All patients completed 25 times heel raising without difficulty.Four patients walked with a slight limp and recovered from the limp 12 months after operation.No Achilles tendon ruptured again during the follow-up time.Conclusions Repairing the old Achilles tendon rupture with suture anchors can supply strongly strain between broken ends of the tendon,and the outer cast is not needed after operation.Complications are less and functional practice can be commenced in the early postoperative period for better function restoration.
7.Diagnostic value of contrast-enhanced CT in mesenteric fibromatosis

Objective To evaluate the value of contrast-enhanced CT in diagnosis of mesenteric fibromatosis (MF).Methods CT images of 6 patients with pathologically proved mesenteric fibromatosis were retrospectively analyzed.The imaging characteristics analysis in-cluded the number,size,density of the lesion,the degree and quality of enhancement,as well as the relationship with the surrounding tissues.Results Singular lesion located in the mesentery was found in all the 6 patients (100%).The size of all cases ranges from 2.0 cm to 8.5 cm and the mean diameter was 6.5 cm.CT scan showed 5 tumors were homogeneous (83.3%),while one tumor had mixed den-sity (1 6.7%).On contrast-enhanced CT images,5 cases showed mild enhancement (83.3%),while 1 case showed moderate en-hancement (1 6.7%).And 4 cases appeared as heterogeneous enhancement (66.7%).4 cases of lesions infiltrated the surrounding tissue(66.7%)and the peripheral intestines were compressed in 2 cases(33.3%).5 masses were surrounded by mesenteric vessels (83.3%).Conclusion MF has certain CT manifestations,especially when mesenteric vascular surrounded the lesion.CT can provide important information for the diagnosis of this disease.
8.Application of DNA-image cytometry in the diagnose of urothelial cell carcinomas

Objective To investigate the value of DNA-image cyt-ometry (DNA-ICM) in the diagnosis of urothelial cell carcinomas (UCC). Methods Totally 162 voided urine specimens (92 cases from urothel-ial car-cinomas patients and 70 cases from benign urinary system diseases patients ) were detected with DNA-ICM and liquid-based cytology (LBC), respectively. Results The sensitivity and specificity of DNA-ICM were 65.2%and 100% respectively in the diagnosis of UCC but those of LBC were 27.2% and 98.6%, respectively. The sensitivity of DNA-ICM was significantly higher than that of LBC in the diagnosis of UCC (P < 0.01). The sen-sitivity of DNA-ICM in upper urinary tract urothelial cell carcinomas (UTUC) were 77.1%, which was much higher than that in bladder urothelial cell carcinomas (57.9%) but no statistical significance was found (P >0.01). Conclusion DNA-ICM, which improves the positive rate of urinary cytology, has great application value in the diagnosis of urothelial cell carcinomas and it is an effective screening method for urothelial cancer in diag-nosis and follow-up.
9.Effects of lentiviral-mediated p38 mitogen-activated protein kinase short hair-pin RNA on cardiac function after myocardial infarction in aldosterone overload rats

Objective To investigate the effects of p38 mitogen-activated protein kinase (MAPK) short hair RNA (shRNA) delivered by lentiviral vectors (pGLV) on cardiac function after myocardial infarction (MI) in aldosterone overload rats and to explore the mechanism.Methods Aldosterone overload rat myocardial infarction model was obtained by ligating the left anterior descending coronary artery.The pGLV-shRNA was constructed,sequenced and injected into rats via tail vein.Rats were divided into 3 groups:pGLV-shRNA group (n=6),pGLV-shRNA-NC group (n=6,contained a nonsense shRNA) and the sham-operation group (n=6).Cardiac function was measured by cardiac ultrasound.Apoptosis was assessed by transferase (TdT)-mediated biotin-16-dUTP nick-end labelling (TUNEL).The p38 MAPK mRNA expression was analyzed by RT-PCR.The protein expressions of p38 MAPK and caspase-3 were detected by Western blot.Results Compared with the sham-operation group,cardiac systolic function was reduced and myocardial apoptosis index was significantly increased [(31.26 ± 4.45) % vs.(15.20 ± 2.18) %,P < 0.01] in pGLV-shRNA-NC group.The mRNA and protien expressions of p38MAPK and caspase 3 protein expression were significantly increased in pGLV-shRNA-NC group (all P<0.01).Compared with pGLV-shRNA-NC group,cardiac function was improved,myocardial cell apoptosis index was reduced [(22.35±3.59)% vs.(31.26±4.45)%,P<0.05],and the mRNA and protien expressions of p38MAPK and caspase 3 protein expression were decreased in pGLV-shRNA group (all P<0.05).Conclusions Cardiac dysfunction is associated with p38MAPK-mediated myocardial apoptosis in aldosterone overload MI rats.pGLV-shRNA may inhibit cardiomyocyte apoptosis and improve postMI cardiac function.
10.Value of histogram analysis of susceptible signal intensity in differentiating papillary from chromophobe renal cell carcinoma

Objective To evaluate the feasibility of histogram analysis of susceptible signal intensity in differentiating papillary (pRCC) from chromophobe renal cell carcinoma(ChRCC). Methods Thirteen cases with pRCC and 9 cases with ChRCC, who underwent susceptible-weighted imaging(SWI), MR common scanning and enhancement, were assessed retrospectively. The histogram parameters of susceptible signal intensity were measured, including maximum, minimum, mean, median, skewness and kurtosis. The independent samples t test(normal distribution) and Mann-Whitney rank sum test(skewed distribution) were used to compare the differences in SWI parameters between pRCCs and ChRCCs. Receiver operating characteristic curve was used to evaluate the efficiency of the whole-tumor SWI parameters in differentiating pRCCs from ChRCCs. Results The significant differences of the minimum, mean, median and skewness between pRCCs and ChRCCs were present(P<0.05), and there was no significant differences in maximum and kurtosis between between pRCCs and ChRCCs(P>0.05). In all the histogram parameters, the area of ROC curve of mean value was largest(0.80). The sensitivity of minimum value (84.62%) was the highest and and the specificity of skewness(100.00%) was the highest. Conclusion Histogram analysis of susceptible signal intensity can help differentiate pRCCs from ChRCCs.
