Objective To establish the blood glucose fluctuation diabetic rat model by subcutaneous injecting insulin. Method The male Sprague-Dawley (SD) rats were randomly assigned into two groups, normal group ( n =20) and model group ( n =40). Normal group rats were fed with normal diet, model group rats were fed with a high-sucrose-high-fat diet. After 6 weeks, a low dose of streptozotocin ( STZ ,30mg/kg) was injected to induce hyperglycemia in model group rats. And then the model group rats were ran-dom divided into two subgroups, diabetic group ( n = 20) and blood glucose fluctuation group ( n = 20). Blood glucose fluctuation group rats were induced by subcutaneous injecting insulin twice daily. All rats were executed after intervening for 3 months, metabolic cages were used to collect 24-hour urine one week before all rats were executed. Results The average blood glucose levels (MBG), the standard deviation of daily aver-age blood glucose (SDBG) and the largest amplitude of glycemic excursions (LAGE) were significant differ-ent (P<0. 05) among normal group, diabetic group and blood glucose fluctuation group. The 24 hours, pro-teinuria of diabetic group and blood glucose fluctuation group was higher than that in normal group[( 100. 38±7. 99)mg/24h vs (18. 13±1.41)mg/24h, (123. 71±11.23) mg/24h vs (18.13 ± 1.41 ) mg/24h, P <0.01] , and blood glucose fluctuation group was higher than diabetic group[(123. 71 ± 11.23 )mg/24h vs(100.38±7.99)mg/24h, P <0.05] . Conclusions The blood glucose fluctuation diabetic rats model was successfully induced by the method of subcutaneous insulin injection in diabetic rats twice daily.

Objective To observe the influence of N-acetylcysteine(NAC)on the formation of Pseudomonas aeruginosa biofilm(BF),and to study the synergism of bactericidal activities of NAC combined with levofloxacinon to the mature Pseudomonas aeruginosa biofilm. Methods The experiment was done in experiment centre of Xiangya Hospital.Scanning electronic microcopy(SEM)were utilized to demonstrate the effect of NAC on the formation of biofilm. Then biofilm cultured without NAC were detected by optical microscopy after being stained with AgNO3.The minimal inhibitory concentration were detected by double test tube diluted method. The mature biofilm was treated by NAC and/or different concentrations of levofloxacin for 24 hours. The influence of levofloxacin combined with NAC were examined by MTT method on the bacterial quantity in biofilms. Results Biofilm was achieved on the silicon slides in all groups 7 days after the incubation of Pseudomonas aeruginosa.Scanning electronic microcopy showed that the mucoid materials among bacteria was significantly reduced and the thickness of biofilm was decreased in NAC groups.Levofloxacin beyond 1 MIC could significantly decline the viable cells on biofilms(P

Objective:To observe the effect of blood glucose lfuctuation and the sustained high blood glucose on renal pathological change and collagen IV (Col IV) expression in diabetic rats.
Methods:hTe 60 male Sprague-Dawley (SD) rats were randomly assigned into a normal control group (NC) and a model group (DM). hTe rats in the normal control group were fed with normal diet, while the rats in the model group were fed with high-sucrose-high-fat diet for 6 weeks. Atfer that, streptozocin (STZ, 30 mg/kg) was injected to induce diabetic model. The model group was then randomly divided into 2 subgroups:a sustained high blood glucose group and a fluctuation blood glucose group (animals in the latter group were subcutaneously injected with insulin twice daily). Rats were sacriifced atfer 3 months and kidney tissues were dissected for HE and PAS staining, Col IV immunohistochemistry and Western blot.
Results:Compared with the normal control group, the renal glomeruli and capillary basal membrane in the diabetic rats was getting larger and thicker, respectively;the capsular space and ground substance was extended and increased, respectively;the volume of renal tubule, kidney hypertrophy index, glomeruler sclerosis index and Col IV content were all increased in the diabetic rats (P<0.01). Compared with the sustained high blood glucose group, the above mentioned pathological changes were more serious in the blood glucose lfuctuation group.
Conclusion:The capillary basal membrane of kidney in diabetic rats is thicker and the ground substance is increased. The degree of glomeruler sclerosis is more serious in the blood glucose lfuctuation group compared with the sustained high blood glucose group, which is conifrmed by the increased level of Col IV.

Objective To investigate the impact of desmoglein 3 (Dsg3) on the proliferation of peripheral T lymphocytes from patients with pemphigus vulgaris (PV).Methods Peripheral blood mononuclear cells (PBMCs) were obtained from 12 patients with PV and 22 normal human controls, cultured with or without the presence of Dsg3 or phytohemagglutinin for 3 days.Flow cytometry was performed to detect the changes of T-lymphocyte subsets in PBMCs stimulated with Dsg3 and proliferation of T-lymphocytes.Results In patients with PV,the percentage of Th2 and Th1 cells was 12.17%±5.32% and 4.08%±1.50%,respectively in Dsg3stimulated PBMCs,9.84%±5.41% and 3.91%±1.38%,respectively in non-stimulated PBMCs.Increased percentage of Th2 cells was observed in Dsg3-stimulated and non-stimulated PBMCs from patients with PV compared with those from normal human controls (both P<0.05).After stimulation with Dsg3,there was a significant proliferation of T cells from patients, and the proliferation rate of CD4+T cells was 4.65%±3.28%,which was significantly higher than that from normal controls(P<0.05).Conclusion Dsg3 can induce the specific proliferation of CD4+T cells, especially Th2 type CD+4 T cells, from patients with PV.

Programmed cell death 5(PDCD5)is a new kind of programmed cell death regulation and control gene. PDCD5′s expression is widespread, and evolution is conservative. PDCD5 has the effect of promo-ting a wide variety of tumor cells to apoptosis and inhibiting proliferation, such as lung cancer, liver cancer et al. Several laboratories′ discoveries show that PDCD5′s expression is significantly decreased when in some dis-ease cases, especially in tumor cases. PDCD5,which is related to the clinical relevance of the tumor, has a great clinical value of discovering, diagnosis and treatment of the tumor.

Microbiology 1992;0(03):-
In order to improve the metabolism yield of cordycepin and adenosine,we studied nitrogen sources ,the levels of nitrogen sources and carbon sources, dynamic development in liquid culture ,as well as the total yield of cordycepin and adenosine in culture system through determining the contents of cordycepin and adenosine by HPLC .The results are as follows: not only animal protein but also some plant protein is very good nitrogen sources; in culture solution the suitable levels of nitrogen sources and carbon sources is respectively 3% and 4%; the content of adenosine in culture solution is very low, while it is quite high in mycelia; it is much higher that the total yield of cordycepin in culture solution than in mycelia; when the culture system is oscillated and cultured after 7~9 days, the total yield of cordycepin and adenosine are both high.
