1.Influence of N-acetylcysteine on Pseudomonas aeruginosa biofilm

Objective To observe the influence of N-acetylcysteine(NAC)on the formation of Pseudomonas aeruginosa biofilm(BF),and to study the synergism of bactericidal activities of NAC combined with levofloxacinon to the mature Pseudomonas aeruginosa biofilm.Methods The experiment was done in experiment centre of Xiangya Hospital.Scanning electronic microcopy(SEM)were utilized to demonstrate the effect of NAC on the formation of biofilm.Then biofilm cultured without NAC were detected by optical microscopy after being stained with AgNO3.The minimal inhibitory concentration were detected by double test tube diluted method.The mature biofilm was treated by NAC and/or different concentrations of levofloxacin for 24 hours.The influence of levofloxacin combined with NAC were examined by MTT method on the bacterial quantity in biofilms.Results Biofilm was achieved on the silicon slides in all groups 7 days after the incubation of Pseudomonas aeruginosa.Scanning electronic microcopy showed that the mucoid materials among bacteria was significantly reduced and the thickness of biofilm was decreased in NAC groups.Levofloxacin beyond 1 MIC could significantly decline the viable cells on biofilms(P

The intracellular signal transduction pathways play an important role in the occurrence and development of thyroid cancer.Mitogen activated protein kinase (MAPK) signal transduction pathway,phosphatidylinositol 3-kinase (PI3K)-AKT pathway,NF-κB pathway and RASSF1-MST1-FOXO3 pathway affect the occurrence and development of thyroid cancer by modulating the activity and expression of specifical proteins,and determine the growth and apoptosis of thyroid cancer cells.
3.Establishment of blood glucose fluctuation diabetic rats model

Objective To establish the blood glucose fluctuation diabetic rat model by subcutaneous injecting insulin. Method The male Sprague-Dawley (SD) rats were randomly assigned into two groups, normal group ( n =20) and model group ( n =40). Normal group rats were fed with normal diet, model group rats were fed with a high-sucrose-high-fat diet. After 6 weeks, a low dose of streptozotocin ( STZ ,30mg/kg) was injected to induce hyperglycemia in model group rats. And then the model group rats were ran-dom divided into two subgroups, diabetic group ( n = 20) and blood glucose fluctuation group ( n = 20). Blood glucose fluctuation group rats were induced by subcutaneous injecting insulin twice daily. All rats were executed after intervening for 3 months, metabolic cages were used to collect 24-hour urine one week before all rats were executed. Results The average blood glucose levels (MBG), the standard deviation of daily aver-age blood glucose (SDBG) and the largest amplitude of glycemic excursions (LAGE) were significant differ-ent (P<0. 05) among normal group, diabetic group and blood glucose fluctuation group. The 24 hours, pro-teinuria of diabetic group and blood glucose fluctuation group was higher than that in normal group[( 100. 38±7. 99)mg/24h vs (18. 13±1.41)mg/24h, (123. 71±11.23) mg/24h vs (18.13 ± 1.41 ) mg/24h, P <0.01] , and blood glucose fluctuation group was higher than diabetic group[(123. 71 ± 11.23 )mg/24h vs(100.38±7.99)mg/24h, P <0.05] . Conclusions The blood glucose fluctuation diabetic rats model was successfully induced by the method of subcutaneous insulin injection in diabetic rats twice daily.

Objective To explore motivation alteration influencing factors of internet addiction disorder (IAD) undergraduates.Methods The study sampled randomly 793 undergraduates from China University.According to CIAS-R and Young's internet addiction diagnostic criteria,a total of 37 internet addicts were selected.Internet addiction survey was used to study the motivation alteration influencing factors of IAD undergraduates.Results (1) The negative effect of internet using on academy,personal relationship etc.(damage from internet using) had a significant positive correlation with motivation alteration (r =0.672,0.699,0.658,P < 0.01 ; r =0.653,0.673,0.628,P < 0.01 ; r =0.604,0.681,0.703,P < 0.01).(2) Benefit from psychological satisfactory level of internet using had no significant difference with motivation alteration.(3) Self-efficiency of behavioral control on using internet had a significant negative correlation with motivation alteration (r =-0.397,-0.370,P <0.05).Conclusion The motivation alteration of IAD undergraduates is influenced by the factors of damage from internet using,sense of achievement,self-efficiency of behavioral control on using internet and so on.

7.Study of the effects of epinephrine combined with methoxamedrine on cardiopulmonary resuscitation

Objective To investigate the effect of epinephrine combined with methoxamedrine on rate of successful cardiopulmonary resuscitation (CPR) in rabbit model of sudden cardiac arrest by the method of clamping the trachea to asphyxia.Methods Twenty-seven rabbits were randomly (random number)divided into methoxamedrine group,epinephrine group and epinephrine combined with methoxamedrine group.After the modeling,rabbits in the three groups were treated methoxamedrine,epinephrine,and epinephrine combined with methoxamedrine respectively by intravenous injection during CPR.The change of heart rate (HR),mean arterial pressure (MAP) and electrocardiogram (ECG),restoration of spontaneous circulation (ROSC) and mortality within 2 hours of each experimental group were recorded before suffocation and 15 min,30 min,60 min,120 min after the success of the resuscitation.The pathological changes of myocardium were observed under optical and electronic microscope.Results The number of ROSC rabbits in methoxamedrine group,epinephrine group and epinephrine combined with methoxamedrine group were 1,6,5 respectively.The heart rate and mean arterial pressure of the rabbits after ROSC were lowered compared with those before suffocation significantly (P <0.05).The levels of mean arterial pressure at 15 minutes and 30 minutes after ROSC in combined treatment group were higher than those of epinephrine group significantly (P < 0.05).The myocardial structure of rabbits after ROSC observed by optical and electronic microscope showed an acute injury,however,the damage degree of myocardium in combined treatment group was slighter.Conclusions Epinephrine combined with methoxamedrine has no apparently additional effect on ROSC during CPR compare with epinephrine used alone,but this combination of two agents can be benefit for stabilizing hemodynamic at early post-ROSC stage,and methoxamedrine can reduce the damage of myocardium during CPR.
8.Design and Implement of the ECLSS Adaptive Learning Intelligent Decision Support System

Objective To monitor state of environment control and life support system (ECLSS), and to diagnose faults and provide decision support in manned space flight. Methods According to the principle of decision support system, neural networks and the expert system, the foundational constructional structure and work flow of the ECLSS decision support system(EDSS) were designed,fault tree analyze(FTA) method was used to sort all of the faults and to form the expert knowledge framework, expert system and neural networks were integrated to adaptive learning from experience. Results In the two-men-five-days SZ-6 manned space flight, this software system worked well, and had reduced the difficulty and intensity of ground flight control and had provided good technological support for the consummation of this flight. Conclusion For the first time, the ECLSS decision support system is designed and implemented by using database and expert system technology. It can judge parameters state and diagnose faults exactly and rapidly, and the data models are designed correctly to give the rational result of parameter prediction.
9.The nursing of circumferential mixed hemorrhoids with infection during hematopoietic stem cell transplantation

Objective To summarize the nursing experience of allogeneic hematopoietic stem cell transplantation patients with circumferential mixed hemorrhoid intercurrent infection, explore the nursing methods of severe circumferential mixed hemorrhoid before allogeneic hematopoietic stem cell transplantation. Methods The detailed nurse plan and carefully observation and assess of lesion were performed daily. Iodophor water at the concentration of 0.5‰ was used for anal bath and 2% iodine ointment was used to smear perianal to prevent of infection. And gauzes soaked iodophor were used to cover the lesions, and Chinese traditional herb for anal bath and ultraviolet exposure to control infection. Additionally, the necessary psychological support was given to overcome the fear. Results After the careful nursing, the patient passed through the agranulocytosis period and achieved the success of transplantation. Conclusions This case suggests that the methods in prophylaxis of patients with circumferential mixed hemorrhoids is effectious.
10.Analysis on Interaction between Human Serum Albumin and Zinc Ions by Circular Dichroism and Isothermal Titration Calorimetry

This article was aimed to study the mechanism of interaction between human serum albumin (HSA) and zinc ions, in order to provide the information on the secondary structure modification of HSA and the thermodynamics parameters using circular dichroism (CD) and isothermal titration calorimetry (ITC). CD and ITC were applied in the study. The concentration of HSA were 0.025 mmol·L-1, 0.05 mmol·L-1, 0.1 mmol·L-1, and 0.2 mmol·L-1, respectively. The concentration of zinc ion was 5 mmol·L-1. The CD analysis revealed that the secondary structure modification of HSA differed depending on the concentration of HAS. And the content ofα-helix was inversely proportional to the concentration of HSA. When the concentration of HSA was 0.2 mmol·L-1, the content ofα-helix was special. A series of thermodynamics parameters including association constants (Kb), stoichiometry (N), entropy (ΔS) and enthalpy (ΔH) were investigated by ITC analysis. And two types of binding sites were observed when ZnSO4(mmol·L-1)?HSA (mmol·L-1)= 5?0.2. The secondary structure modification of HSA interacting with zinc ions depended on the concentration of HSA, a dramatic reduction ofα-helix was detected when the concentration of HSA attained 0.2 mmol·L-1. And the protein hydrophobicity reduction and peptide chains expansion were equally observed at this concentration. The ITC analysis also revealed endothermic and exothermic binding sites in the ZnSO4-HSA interaction when ZnSO4 (mmol·L-1)?HSA (mmol·L-1)= 5?0.2, indicating that the endothermic sites were specific but not preferential for zinc ions interactions. These results provided theoretical supports for the application of Zn2+ to open the endothermic sites of HSA.
