4.Significance of HIF-1α, MVD and EGFR expression in detection of nasal inverted papilloma and clinical effect of taxol

Objective:To explore the significance of the expression of HIF-1α,MVD and EGFR on diagnostic of nasal inverted papilloma and the clinical effect of taxol .Methods:The expression of HIF-1α,MVD and EGFR were detected by immunohistochemistry in nasal inverted papilloma .Results:The expression of HIF-1α,MVD and EGFR were enhanced with the degree of atypical hyperplasia in epithelium.The research also showed that the expression of HIF-1α,MVD and EGFR were increased dramatically in severe atypical hyperplasia with a significant statistical differences ( P<0.01 ) .The expression of EGFR and MVD showed a positive correlation with HIF-1α.The administration of Taxol alleviated the enhanced expression of HIF-1α, MVD and EGFR with a significant statistical difference(P<0.01),but with on statistical difference with moderately severe tissue (P>0.05).Conclusion:HIF-1α,MVD and EGFR are involved in the pathogenesis and deteriorate of nasal inverted papilloma and can be taken as biomarkers for the diagnosis of the dete -rioration of nasal inverted papilloma .
5.Effect of sequoyitol on expression of NOX4 and eNOS in aortas of type 2 diabetic rats.

The aim of the present study is to investigate the effects of sequoyitol (Seq) on expression of eNOS and NOX4 in aortas of type 2 diabetic rats. Type 2 diabetic rats induced by high fat and high sugar diet and low dose of streptozotocin (STZ, 35 mg x kg(-1)) and were administered Seq (12.5, 25 and 50 mg x kg(-1) x d(-1)) for 6 weeks. The fasting blood glucose (FBG) and body weight were tested. Acetylcholine (Ach) induced endothelium-dependent relaxation and sodium nitroprusside (SNP) induced endothelium-independent relaxation were measured in aortas for estimating endothelial function. Aortic morphological change was observed with HE staining. The level of serum insulin was measured by radioimmunoassay. The total antioxidative capacity (T-AOC), malondialdehyde (MDA) and NO levels in aortas were determined according to the manufacturer's instructions. In addition, the expressions of eNOS and NOX4 in aortas were measured by immunohistochemisty, real-time PCR or Western blotting. The results showed that Seq significantly decreased FBG and insulin resistance, and improved aortic endothelium-dependent vasorelaxation function. The expressions of NOX4 and MDA content were obviously decreased, while the expression of eNOS, the levels of NO and T-AOC increased significantly in aortas of diabetic rats with Seq treatment. In conclusion, Seq protects against aortic endothelial dysfunction of type 2 diabetic rats through down-regulating expression of NOX4 and up-regulating eNOS expression.

6.Effect of sequoyitol on expression of NOX4 and eNOS induced with glucose in human umbilical vein endothelial cells.

OBJECTIVETo investigate the protective effect and mechanism of sequoyitol (Sep) on high glucose-induced human umbilical vein endothelial cells (HUVECs) injury.
METHODSHUVECs were cultured with high glucose (30 mmol/L) in the presence or absence of sequoyitol (0.1, 1 and 10 micromol/L) for 24 h. Cell proliferation was measured by BrdU marking and cell cycle was detected by flow cytometry. 2', 7'-dichlorofluorescein diacetate was used to evaluate intracellular reactive oxygen species (ROS) levels. The NO, malonydialdehyde (MDA) and H2O2 levels were determined by colorimetric method according to the manufacturer's instructions. The expression of endothelial nitric oxide synthase (eNOS) and NADPH oxidase 4 (NOX4) were measured by real-time PCR and Western blot.
RESULTSIn the present study, we found that sequoyitol pretreatment for 1 h significantly decreased cell injury, promoted cell proliferation. Meanwhile sequoyitol significantly down-regulated NOX4 expression and decreased the level of ROS, MDA and H2O2 and obviously increased NO levels and up-regulated eNOS expression.
CONCLUSIONSequoyitol alleviates high glucose-induced cell injuries in HUVECs via inhibiting oxidative stress and up-regulating eNOS expression.

7.Design of heart sound processing analog circuit based on the electret capacitor microphone

This paper designs a heart sound processing analog circuit, in which the amplification circuit, the filter circuit and the lifting of electrical level circuit are involved. It also gives a safety design problem analysis using an electret capacitor microphone to pick-up the heart sound signals. It is proved that the circuit is reliable and effective.
8.Efficacy Observation for Treating Ankylosing Spondylitis by Chinese Herbs and Recombinant Hu- man Tumor Necrosis Factor Receptor II-Antibody Fusion Protein.

OBJECTIVETo observe the clinical effect of Chinese medical (CM) syndrome differentiation based Chinese herbs and recombinant human tumor necrosis factor receptor II-antibody fusion protein (etanercept) for treating ankylosing spondylitis (AS) patients.
METHODSTotally 35 AS patients were treated with syndrome differentiation based Chinese herbs and etanercept. Reinforcing Shen and strengthening Du channel, activating meridians to stop pain was principle used in syndrome differentiation based treatment. Etanercept was subcutaneously injected, 25 mg each time; twice per week for the first three months and once a week for the latter three months. The clinical efficacy was evaluated after 3 and 6 months of treatment. Meanwhile, ASAS20 and ASAS50 standards arriving rates were also observed. Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), Bath Ankylosing Spondylitis Functional Index (BASFI), visual analog score (VAS) for spine pain, VAS for night pain, patient global assessment (PGA), VAS for physician global assessment, CM syndrome score, finger-ground distance, thoracic activity, tragus-wall distance, lumbar scoliosis, cervical rotation, Schober improved test, erythrocyte sedimentation rate (ESR), and C-reactive protein (CRP) were observed before treatment, 3 and 6 months after treatment.
RESULTSCompared with before treatment, BASDAI, BASFI, VAS for spine pain, night pain, physician global assessment, PGA, CM syndrome score, finger-ground distance, thoracic activity, tragus-wall distance, lumbar scoliosis, Schober improved test, ESR, and CRP all decreased after 3 and 6 months of treatment, with statistical difference (P < 0.05). Cervical rotation also decreased after 6 months of treatment, with statistical difference (P < 0.05). Compared with 3 months of treatment, total effective rate of CM syndrome, ASAS20 and ASAS50 standards arriving rates increased after 6 months of treatment, with statistical difference (P < 0.05). There were statistical differences in all indices mentioned above between after 3 months of treatment and after 6 months of treatment (P < 0.05).
CONCLUSIONSyndrome differentiation based Chinese herbs combined etanercept could alleviate inflammatory reaction favorably, control the progression of active AS, and improve joint functions.

10.Impact of adding folic acid and vitamin B12 combined with standard antihypertensive medication on therapeutic effects

Objective To investigate the impact of adding folic acid and vitamin B12 combined with standard antihypertensive medication on plasma homocysteine (Hcy) and asymmetric dimethylarginine (ADMA),serum NO and endothelial nitric oxide synthase (eNOS) of essential hypertensive patients.Methods One hundred and fifty-six patients with essential hypertension were divided into control group and experimental group with 78 cases by random digits table method.Control group received conventional antihypertensive therapy,while experimental group were treated with folic acid and vitamin B12 based on the treatment of control group.Plasma Hcy and ADMA,serum NO and eNOS levels were observed at baseline 2 and 12 weeks after treatment.Results In control group,the level of Hcy and ADMA showed no significant difference before and 2,12 weeks after treatment (P > 0.05).The level of Hcy and ADMA before treatment was significantly higher than that 2,12 weeks after treatment in experimental group (P< 0.05).The level of NO and eNOS 12 weeks after treatment in experimental group was significantly higher than that in control group[(77.35 ± 9.55)μmol/L vs.(64.26 ± 11.49) μ mol/L,(30.05 ±6.88) μ mol/L vs.(19.92 ± 10.24) μ mol/L,P < 0.05].The level of ADMA was positively correlated with Hcy at baseline(r =0.612,P < 0.05),and was negatively correlated with NO and eNOS (r =-0.557,-0.529,P <0.05).Conclusions Supplementation of folic acid and vitamin B12 helps to improve endothelial function and reduce plasma Hcy and ADMA levels as well as improve serum NO and eNOS levels in patients with essential hypertension,which helps to prevent from cardiovascular and cerebrovascular accidents.
