3.Repairing femur defect using biomaterials and its features evaluation

OBJECTIVE: To evaluate the features and application of tissue engineered biomaterials in repairing femur defects, and to search an optimal femur substitution.METHODS: Papers published between January 1993 and October 2009 were searched using computer with key words of "tissue engineering, femur defect, mesenchymal stem cells, cytokines and materials" both in English and Chinese. Documents addressing biomaterials and tissue engineered bone in repairing femur defects were included. Repetitive research or Meta analysis was excluded. After that, 21 literatures were selected to discuss the features and application of tissue engineered biomaterials in repairing femur defects.RESULTS: Stem cells technology is obtaining seed cells from myeloid tissues, amplifying in vitro, combining with scaffold material, and in vitro constructing tissue engineered bones. The cultured cells could maintain osteoblast biological characteristics and fully fused with bone tissues at recipient sites, simultaneously, avoid immunological rejection. The combination of materials or modify the material surface could promote cells adhere to materials and increase the cellular biological activity. When transfecting seed cells to bone defect areas using certain carriers, the transplanted calls could form new bones and secret bone growth factor, induce determined osteogenic precursor cells differentiated into inducible ostegenic precursor cells, thereby, fasten the bone healing. The composite materials prepared by three-dimensional virtual surgical simulation and computer numerical control exhibited advantages of composite materials and histological anatomy conjunction, which had perfect appearance.CONCLUSION: Currently, there is no material can fully meet the requirement of bone tissue engineering. Thus, promoting adhesion between cells and materials, increasing cellular biological activity, and maintaining biological functions by combing materials or modifying material surface are the research focuses.
4.The study of correlation between balance function and attention of children with attention deficit hyperactivity disorder

Objective To study the relationship between balance function and attention in children with attention deficit hyperactivity disorder(ADHD),and guide the clinical intervention and training of ADHD.Methods The Tetrax balance system and the CPT were conducted among 124 ADHD children (age 6.0-14 years) and the normal controls.Results (1) The balance function was different between ADHD and normal controls.In junior grade of elementary school,the Fourier score of ADHD children was significantly higher than that of the normal control group in F1,F2-F4 and F5-F6 (P<0.05).In senior grade of elementary school,the Fourier score of ADHD children was significantly higher than normal controls in F2-F4 and F5-F6 (P< 0.05).There was no significant difference between ADHD and normal controls in junior high school.(2) There was correlation between balance function and attention in children with ADHD.In junior grade of elementary school,the full scale attention quotient was negatively correlated with F2-4,F5-6,F7-8 (P=0.018,0.009,0.010; rs =-0.146,-0.203,-0.189),and the visual attention quotient were negatively correlated with F1,F2-4,F7-8 (P =0.043,0.039,0.046 ; r s =-0.062,-0.071,-0.112).In senior grade of elementary school,the full scale attention quotient was negatively correlated with F5-6,F7-8 (P=0.015,0.033; rs =-0.217,-0.172),and the visual attention quotient were negatively correlated with F1,F7-8(P=0.037,0.045; rs =-0.051,-0.101).In junior high school,the negative correlation was existed between the visual attention quotient with F7-8 (P=0.038,r s =-0.114).Conclusion ADHD children have deficit balance.The balance function of ADHD children matures gradually.There is internal connection between balance and attention,especially for visual attention.
5.Clone of apoptosis-related genes in prostate cancer cell by means of an improved subtractive hybridization

Objective To set up an apoptosis model of prostate cancer cell line and to clone and study the apoptosis related genes. Methods An apoptosis model of prostate cancer cell line DU-145 has been set up through induction by all transretinoic acid (ATRA).During the process of cell apoptosis the apoptosis-related gene was cloned by means of improved PCR-based subtractive hybridization from the apoptosis prostate cancer cell line DU-145 prostate cancer cells. Results During the process of DU-145 cell apoptosis,c-erb B-2 expression,TNF genes and some unknown apoptosis-related gene were observed.This has been accepted by Genebank,the accession number being AF174394. Conclusions ATRA-induced apoptosis of DU-145 cells is a complex process with multiple genes involved,some of which being unknown yet.
6.Internal pressure in nucleus pulposus and compression degree of adjacent nerve roots following lumbar vertebral herniation

BACKGROUND: Observation and comparison about space-occupying lesions due to herniated nucleus pulposus, as well as nerve root compression in lumbosacral disc herniation and manipulative therapy are easily available; however, there is still lack of related quantitative research regarding the internal pressure in herniated nucleus pulposus.OBJECTIVE: To observe the influence of the internal pressure in herniated nucleus pulposus on nerve root compression in patients with lumbosacral disc herniation.DESIGN: Case control study.SETTING: Bone Setting Therapeutic Center, the Air Force General Hospital of Chinese PLA.PARTICIPANTS: Totally 30 patients with lumbosacral disc herniation received operation in the Orthopedic Department of the Air Force General Hospital between October 2002 and December 2003; meanwhile, 15 patients with lumbosacral disc herniation received manipulative therapy in the Bone Setting Therapeutic Center of the same hospital.ing positive group and negative group according to the results of straight leg raising test. However,the size of herniated nucleus pulposus was not significantly different between positive group and negative group [(4.688±1.991), (4.857±2.033) mm,nipulative therapy, their straight leg raising height was obviously increased compared to that before operation [(54.000±16.388)°, (72.668±15.338)°,P < 0.01], but the size of herniated nucleus pulposus was proved not obviously changed by CT or MRI examination after manipulative therapy (P > 0.05).cleus pulposus on nerve root is related to the iuternal pressure of herniated nucleus pulposus; higher internal pressure in herniated nucleus pulposus would limit straight leg raising height while lower internal pressure has less effect. However, the compression of herniated nucleus pulposus on nerve root has no obvious relationship with the size of herniated nucleus compression by reducing the internal pressure in herniated nucleus pulposus, which may not only depend on changing the space-occupying of herniated nucleus pulposus.
7.Pharmacokinetic study of iguratimod in rats

ObjectiveTo study the pharmacokinetics of iguratimod in rats. MethodsThe concentration ofiguratirnod in the samples was determined by HPLC method. The pharmacokineties parameters were calculated withDAS softwrare. ResultsThe mainpharmacokineties parameters of normal group(6mg/kg) were as follows:t1/2Ke:3.56h, tpeak: 4.00h, Cmax : 8.87μg/ml, AUC0.24 : 74.76μg· ml-1·h-1. The main pharmacokineties parameters of threemodel groups(3,6,12mg/kg) were as follows: t1/2Ke: 4.54,3.20,3.17h, tpcak:3.83,3.83,4.67h,Cmax:3.84, 8.31,12.69μg/ml, AUC0.24 :40.21,76. 72,117.06μg·ml-1·h-1. Except Cmax and AUC, no significant differenceswere found between the three model groups. And the differences between normal group and model group were notsignificant. ConclusionThe pharmacokinetics of rats ks fit to one-compartment model.
8.Analysis of monitoring results of Keshan disease from 2007 to 2011 in Baoji City of Shaanxi Province

Objective To learn the existing state of Keshan disease and its trend in Baoji City,and to provide a scientific basis for control of the disease.Methods Feng,Qianyang,Linyou and Long Counties,the Keshan disease affected areas were selected to carry out the case study,and Keshan cases were searched from all registered cases of myocardial patients of the past three years at county-level medical institutions and hospitals of townships(towns).Thirteen villages affected by Keshan disease in Qianyang,Fengxiang,Feng,Linyou and Long Counties were selected as monitoring points to carry out a intensified monitoring.All subjects were surveyed by comprehensive clinical physical examination and 12-lead ECG tracings,and they were diagnosed according to the Keshan disease diagnostic criteria.Ten households were selected randomly in each monitoring point(village),and self-produced wheat samples and adult subject's occipital hair samples were collected for detecting selenium levels.Results Seven hundred and forty-six cardiac cases which containing 17 cases of chronic Keshan disease were discovered in 32 medical institutions,and there were no acute and subacute type of Keshan disease patients.Five thousand seven hundred and twenty-three subjects were examined in the 13 monitoring sites,and 65 cases of Keshan disease were diagnosed which including 17 cases of slow and 48 cases of potential Keshan disease,and no acute or subacute case was found.The total detection rate was 1.14%,and the detection rate ranged from 0 to 2.86%.Hair selenium mean was 0.420 mg/kg; wheat selenium mean was 0.079 mg/kg.Conclusions The Keshan disease detection rate of Baoji City is at a low level.The selenium level of local residents has reached the level of non-endemic area.Constant detection of patient with Keshan disease means various control measures should be reinforced.The comprehensive monitoring program,such as monitoring people's in vivo and in vitro selenium level and others,should be established as soon as possible.
9.Meta-analysis on effect of compound Danshen injection in treating neonatal hypoxic-ischemic encephalopathy.

To systematically evaluate the clinical efficacy and safety of compound Danshen injection in treating hypoxic-ischemic encephalopathy (HIE) of newborns. Computer retrievals were made in PubMed, Embase, Cochrane Library, CBM, CNKI, VIP and China info (before May 2014) and relevant literature references, and manual retrievals were made for journals and conference papers, in order to collect randomized or semi-randomized controlled trials concerning compound Danshen injection in the treatment of neonatal HIE. The quality of included references was evaluated according to literatures recommended by Cochrane Handbook. RevMan 5. 3 software was applied in the statistical treatment. Finally, a total of 13 randomized controlled trials were included, covering 1,211 patients (including 639 patients in the compound Danshen injection-treated group and 572 patients in the control group). Meta-analysis results showed that the routine treatment combined with compound Danshen injection can improve the treatment efficiency of neonatal HIE [RR = 1.28; 95% CI (1.21-1.36)], reduce the mortality rate [RR = 0.42; 95% CI (0.23-0.75)] and the incidence of long-term neurological sequelae [RR = 0.48; 95% CI (0.35-0.65)], with statistical differences. No fatal side effect was observed in all of included trials. So far, limited evidences in this study proved that the application of compound Danshen injection in the treatment of neonatal HIE can enhance the clinical efficiency. However, because of the low quality of the included trials, more well-designed and large-scale multi-center randomized controlled trials shall be made in the future.
