1.Research progress of hookworm detection technology

As one of the species of soil-transmitted helminths (STH), hookworm infection is widely prevalent in tropical and subtropical developing countries. Hookworm is harmful to human health mainly including anemia, digestive tract symptoms, female infection can also cause menopause, abortion, infection in infants and young children can lead growth retardation. With the development of economy and implementation of mass drug administration strategies, the infection of hookworm in human body has decreased continuously, but there are still a large number of mild infection cases which clinical symptoms are not obvious, easy to cause missed diagnosis and misdiagnosis. Several zoonotic hookworm species can cause large-scale infections in cats, dogs and other animals, so as to increase the risk of human hookworm infection. Therefore, it is important to find a highly sensitive and rapid detection techniques to identify hookworm species. In this paper, the existing detection techniques of hookworm were reviewed and their application were prospected to provide ideas for hookworm detection at present situation.
2.Myofibrillar myopathy with cytoplasmatic.spheroid bodies: a report of a Chinese family

Objective To report the clinical, myopathological and genetic features in myofibrillar myopathy (MFM) with numerous cytoplasmatic-spheroid bodies. Methods Ten patients in 5 successive generations began to present progressive proximal limbs weakness at 35 to 40 years old. Additionally, 4 cases manifested diarrhea and 6 cases accompanied with cardiorespiratory symptoms. An open biopsy was performed on the proband. In addition to histological, enzymhistochemical staining and ultrastructural examination, immunohistochemical staining with antibody against tau, desmin, ubiquitin, dysferlin, dystrophin-C', dystrophin-N' and dystrophin-R were done. All the exons of the MYOT, CRYAB, DESMIN, LDB3, LMNA, SEPNI gene and the FLNC exon 48 were analysed. Results Cytoplasmatic bodies and spheroid bodies were found in the fibers. The deposited material were positive for tau, desmin, ubiquitin, dysferlin and dystrophin-R, dystrophin-C'. Electron microscope showed granular dense Z-disc material in the inclusions which were surrounded by thin filament. There was no mutation in the above exons of the 7 candidate genes. Conclusions Myofibrillar myopathy involves multiple system impairment. Cytoplasmatic and spheroid bodies contain microtubule and membrane associated protein. The disease might be induced by some unknown genetic abnormities.

4.Survey on application and proportion of different types of dentures in Shanghai

Objective To collect the information about types and clinically-used materials for the dentures, and evaluate the current conditions and characteristics of prosthodontic treatment in Shanghai. Methods Three municipal public hospitals and two private commercial dental labs in Shanghai were served as sample units. The application information on fixed partial dentures, removable partial dentures and complete dentures from January 2005 to December 2007 was collected. The number of dentures in each sample unit, the total proportion of each type of dentures, and the proportion of each type of dentures in public hospitals or private labs were retrospectively analysed. Results From 2005 to 2007, the number of dentures increased by approximately 20% annually. The number of dentures made in private labs increased by 43.58% in 2005 to 2006 and 23.23% in 2006 to 2007.As to the types of dentures, fixed partial dentures constituted the largest proportion (74.49%-76.19% for public hospitals and more than 80% for private labs), while complete dentures accounted for the smallest proportion (5.07% to6.24% for public hospitals and 1.02% to 1.38% for private labs) . Conclusion With an ever-growing awareness of oral health, prosthodontics has been developing rapidly in Shanghai. The rapid increase trend of private dental clinics is noticeable. Fixed partial dentures constitute the largest proportion, while the proportion of complete denture gradually decreases year by year.
5.Meta-analysis of Reinforcing Kidney and Invigorating Spleen Principle in the Treatment of Primary Os-teoporosis

OBJECTIVE:To systematically review the efficacy and safety of reinforcing kidney and invigorating spleen princi-ple in the treatment of primary osteoporosis (POP) in middle and elderly aged patients,and provide evidence-based reference for clinic. METHODS:Retrieved from VIP,Wanfang Database,CJFD,Medline,PubMed,Elsevier,Springer,Web of Science and Ovid,randomized control trials (RCT) about reinforcing kidney and invigorating spleen principle (test group) versus other meth-ods(no reinforcing kidney and invigorating spleen principle,control group)in the treatment of POP were collected. Meta-analysis was performed by using Rev Man 5.3 software provided by Cochrane after data extraction and quality evaluation modified by Jadad scale. RESULTS:Totally 28 RCTs were enrolled,involving 2849 patients. Results of Meta-analysis showed,the total effective rate [OR=3.80,95%CI(2.76,5.23),P<0.001],BGP level [MD=2.23,95%CI(1.80,2.66),P<0.001],BMD level of femoral neck [MD=0.04,95%CI(0.00,0.08),P=0.05] and BMD level of lumbar spine [MD=0.07,95%CI(0.03,0.11),P=0.001] in test group were significantly higher than control group,ALP level [MD=-5.59,95%CI(-10.51,-0.66),P=0.03] and VAS score [MD=-1.38,95%CI(-1.87,-0.89),P<0.001] were significantly lower than control group,with statistical significance. There were no significant differences in U-Ca/Cr [MD=-0.07,95%CI(-0.17,0.02),P=0.12] and serum Ca2+ level [MD=0.16,95%CI (-0.07,0.39),P=0.17] in 2 groups. CONCLUSIONS:Reinforcing kidney and invigorating spleen principle can improve patients' osteogenesis during bone metabolism,reduce the activity of the osteoclastic process and relief patients'pain in the treatment of POP.
6.Analysis of diversity between synoptophore and triple prism strabismometry

? AIM: To investigate the differences between synoptophore and triple prism strabismometry and its possible cause.?METHODS: There were 347 patients with horizontal concomitant strabismus involved, in which 76 patients were esotropia, 37 patients were male while 39 were female, with average age of 13. 27 ± 7. 77 years old. There were 271 patients with exotropia, 131 cases were male while 140 were female, with average age of 15. 43 ± 8. 42 years old. All the patients were examined by synoptophore and prism plus shaded strabismometry in a long distance of 6m. Datas were analyzed by SPSS 17. 0.?RESULTS:In the exotropia patients, the conversions of circular degree(°) and prism degree(△) were:1°=0. 29△ ~1. 78△, which was statistically significant with intermittent strabismus (P=0. 001). While in the esotropia patients, the conversions were:1°=2. 01△ ~2. 15△.?CONCLUSION: The diversity between the two methods is enlarged with the increase of squint angle for exotropia patients. While in esotropia patients, the diversity decreased with the increase of squint angle. Synoptophore equipped with +7. 00D, defects of the triple prism itself and proximal convergence during exam may be the reasons for the diversity.
7.Analysis of iatrogenic urethral injury in 26 cases

Objective To discuss the diagnosis and treatment of iatrogenic urethral injuries and preventive measures.Methods Various clinical parameters in 26 cases of iatrogenic urethral injury between January 2006 to January 2014 were analyzed retro-spectively.Results Twenty-five cases were confirmed in the operation,1 case of confirmed 2 days postoperatively.All patients were follow-up visit for 3-24 months normal renal function,only 1 case of urethral stricture.Conclusion Through effective prevention, early diagnosis and treatment,close follow-up visit can minimize the patient′s occurrence of iatrogenic urethral injury.
8.Development and preliminary evaluation of a fluorescence RPA assay for the rapid detection of Necator americanus

Abstract: Objective To establish a rapid detection assay based on fluorescence recombinase polymerase amplification (RPA) targeting Necator americanus eggs, and to evaluate its efficacy, providing technical support for rapid detection of Necator americanus in fecal samples. Methods The fluorescence RPA primers and probe were designed based on the cox1 gene of Necator americanus and then screened the optimal combination to develop the assay. The genomic DNA of Necator americanus eggs was diluted to 7 concentration gradients including 100 pg/µL, 10 pg/µL, 1 pg/µL, 100 fg/µL, 10 fg/µL, 1 fg/µL, 0.1 fg/µL, to determine the detection limit of the assay. The specificity of the assay was demonstrated by detected genomic DNA from Schistosoma japonicum, Ascaris lumbricoides, Clonorchis sinensis and Fasciola hepatica. A total of 44 fecal samples were collected and DNA extraction was performed, and the modified Kato-Katz method, semi-nest PCR method, and fluorescent RPA method were simultaneously used for detection to evaluate the sensitivity and specificity. Results The established fluorescence RPA assay can specifically amplify a fragment of 194 bp of the Necator americanus cox1 gene within 20 min, with a detection limit of 10 fg/µL. There was no cross-reactivity with Schistosoma japonicum, Ascaris lumbricoides, Clonorchis sinensis, Fasciola hepatica after specificity validation. In 44 fecal samples, 27 positive samples were detected by the fluorescence RPA assay, and 26 positive samples were detected by both the Kato-Katz and the semi-nested PCR. The fluorescence curve of sample number 1 was slightly higher than the negative control in the later stage of the reaction, but did not show a similar trend to the positive control, and was therefore judged to be a suspected negative sample. Compared with the Kato-Katz method and the semi-nest PCR method, The sensitivity of the fluorescent RPA method were 100.00% and the specificity were 94.44%, and the consistency of the detection results was good (Kappa=0.953>0.75). Conclusions The assay based on the fluorescence RPA is an efficient, sensitive and specific technique for detecting Necator americanus and it can be applied for surveillance and early warning of hookworm infection.
9.Relationship between the T staging of the seventh edition and lymph nodes metastasis of lung cancer

Objective The new lung cancer TNM staging for T staging the new grading.The aim of this study was to investigate the relationship between the T staging and grading of the Seventh Edition and lymph node metastasis of lung cancer.Methods In 513 cases of non-small cell lung cancer primary tumor size and lymph node metastasis were analyzed,and explore the situation of different size,lymph node metastasis in primary tumors.To analyse the collected data with SPSS software.Results The total lymph node metastatic rates in tumor diameter biggest ≤2 cm(T1a) 、2 cm < ~ ≤3 cm(T1b) 、3 cm < ~ ≤5 cm (T2a) 、5 cm < ~ ≤7 cm(T2b) 、> 7 cm(T3) were 14.47% 、28.89% 、37.59% 、36.37% 、37.89%.The lymph node metastatic rate of T1a was significantly different,compared with T1b 、T2a 、T2b and T3,respectively.There were no differences between every two groups of T1b,T2a,T2b and T3.The N1 metastaic rates of T1a 、T1b 、T2a 、T2b 、T3 were compared by chi-square(P <0.05),The lymph node metastatic rates of T1a and T2a (P =0.001),T1a and T2b (P =0.024).The N2 metastaic rates of T1a 、T1b 、T2a 、T2b 、T3 were compared by chi-square(P <0.05),The lymph node metastatic rate of T 1 a was significantly different,compared with T1b 、T2a and T2b,respectively.The lymph node metastatic rate of T1 b was significantly different,compared with T2a 、T2b and T3,respectively.Conclusion The new T staging of tumor the size of the new classification is associated with lymph node metastasis rate,especially in N2.low T la lymph node metastasis rate.
10.Detection of the immunologic rejection after xeno-islet transplantation:a study by MR imaging enhanced with superparamagnetic iron oxide marking CD4+T cell antibody

Objective To evaluate the feasibility of the diagnosis of the early immunologic rejection after xeno-islet transplantation by MR imaging enhanced with superparamagnetic iron oxide(SPIO)marking CD4+T cell antibody.Methods Two thousand neonatal porcine islets(NPI)were transplanted under the left renal capsule of BALB/C nude mice.When the grafts could be observed bv MRI.107 human PBMC was intraperitoneal injected to nude mouse models to reconstitute the human immunologic system,20 mice were reconstituted.Before and 3,7,14 days after reconstitution of human immunologic system on BALB/C nude mice,MRI imaging Was performed half an hour after intravenous injection of nano-immunomagnetic beads via vena caudalis to observe the grafts'MRI signal.BALB/C nude mice were sacrificed after MRI scanning immediately,the histopathologic examination was assessed on grafts,the results were compared with MRI results.And calculate the sensitivity,specificity,Youden index number and coincidence of the MRI for immunologic rejection.Results Grafts can be observed bv MRI 3 weeks after islet cell transplantation (before immunologic rejection modeling),there is no abnormal MRI signal detected in nude mice'graft region after mierobeads injected.Seven days after building of immunologic rejection model,MRI hypo-signal in graft site is shown in the T2 WI sequence after nano-bioprober injected.Histopathologic assessments were employed on grafts in nude mice immediately(HE and immunohistochemistry staining),the results shown that there are a lot of T lymphocyts infiltrated in graft region.implying the occurrence of immunologic rejection.And the sensitivity,specificity,Youden index number and coincidence is:(72.96±0.24)%,100%,0.73±0.24,(88.46±0.13)%respectively.The correct Kappa between the MRI and the imunohistochemistry staining was 0.76.Conclusion The cellular immunological rejection to xeno-islet grarts can be assessed with nano-bioprobe with anti-CD4+ antibody MR imaging,real time,and noninvasively.
